2Genomic landscape and genetic manipulation of the black soldier fly Hermetia illucens,a natural waste recycler显示文摘The black soldier fly(BSF),Hermetia illucens(Diptera:Stratiomyidae),is renowned for its bioconversion of organic waste into a sustainable source of animal feed.We report a high-quality genome of 1.1 Gb and a consensus set of 16,770 gene models for this beneficial species.Compared to those of other dipteran species,the BSF genome has undergone a substantial expansion in functional modules related to septic adaptation,including immune system factors,olfactory receptors,and cytochrome P450s.We further profiled midgut transcriptomes and associated microbiomes of BSF larvae fed with representative types of organic waste.We find that the pathways related to digestive system and fighting infection are commonly enriched and that Firmicutes bacteria dominate the microbial community in BSF across all diets.To extend its potential practical applications,we further developed an efficient CRISPR/Cas9-based gene editing approach and implemented this to yield flightless and enhanced feeding capacity phenotypes,both of which could expand BSF production capabilities.Our study provides valuable genomic and technical resources for optimizing BSF lines for industrialization.Shuai Zhan Gangqi Fang Minmin Cai Zongqing Kou Jun Xu Yanghui Cao Liang Bai Yixiang Zhang Yongmao Jiang Xingyu Luo Jian Xu Xia Xu Longyu Zheng Ziniu Yu Hong Yang Zhijian Zhang Sibao Wang Jeffery K.Tomberlin Jibin Zhang Yongping Huang 2020Cell Research2020,30,1:9
3High-throughput metagenomic analysis of petroleum-contaminated soil microbiome reveals the versatility in xenobiotic aromatics metabolism显示文摘The soil with petroleum contamination is one of the most studied soil ecosystems due to its rich microorganisms for hydrocarbon degradation and broad applications in bioremediation.However, our understanding of the genomic properties and functional traits of the soil microbiome is limited. In this study, we used high-throughput metagenomic sequencing to comprehensively study the microbial community from petroleum-contaminated soils near Tianjin Dagang oilfield in eastern China. The analysis reveals that the soil metagenome is characterized by high level of community diversity and metabolic versatility. The metageome community is predominated by γ-Proteobacteria and α-Proteobacteria, which are key players for petroleum hydrocarbon degradation. The functional study demonstrates over-represented enzyme groups and pathways involved in degradation of a broad set of xenobiotic aromatic compounds, including toluene, xylene, chlorobenzoate, aminobenzoate, DDT, methylnaphthalene, and bisphenol. A composite metabolic network is proposed for the identified pathways, thus consolidating our identification of the pathways. The overall data demonstrated the great potential of the studied soil microbiome in the xenobiotic aromatics degradation. 4Increased mitochondrial fission drives the reprogramming of fatty acid metabolism in hepatocellular carcinoma cells through suppression of Sirtuin 1显示文摘Background:Mitochondria are dynamic organelles that constantly change their morphology through fission and fusion processes.Recently,abnormally increased mitochondrial fission has been observed in several types of can-cer.However,the functional roles of increased mitochondrial fission in lipid metabolism reprogramming in cancer cells remain unclear.This study aimed to explore the role of increased mitochondrial fission in lipid metabolism in hepa-tocellular carcinoma(HCC)cells.Methods:Lipid metabolism was determined by evaluating the changes in the expressions of core lipid metabolic enzymes and intracellular lipid content.The rate of fatty acid oxidation was evaluated by[PH]-labelled oleic acid.The mito-chondrial morphology in HCC cells was evaluated by fluorescent staining.The expression of protein was determined by real-time PCR,imnmunohistochemistry and Western blotting.Results:Activation of mitochondrial fission significantly promoted de novo fatty acid synthesis in HCC cells through upregulating the expression of lipogenic genes fatty acid synthase(FASN),acetyl-CoA carboxylasel(ACCI),and elonga-tion of very long chain fatty acid protein 6(ELOVL6),while suppressed fatty acid oxidation by downregulating carnitine palmitoyl transferase 1A(CPTIA)and acyl-CoA oxidase 1(ACOX1).Consistently,suppressed mitochondrial fission exhibited the opposite effects.Moreover,in vitro and in vivo studies revealed that mitochondrial fission-induced lipid metabolism reprogramming significantly promoted the proliferation and metastasis of HCC cells.Mechanistically,mito-chondrial fission increased the acetylation level of sterol regulatory element-binding protein 1(SREBPI)and peroxisome proliferator-activated receptor coaC-tivator 1 alpha(PGC-1a)by suppressing nicotinamide adenine dinucleotide(NAD+)/Sirtuin 1(SIRTI)signaling.The elevated SREBP1 then upregulated the expression of FASN,ACC1 and ELOVL6 in HCC cells,while PGC-1c/PPARa sup-pressed the expression of CPTIA and ACOXL Conclusions:Increased mitochondrial fission plays a crucial role in the repro-gramming of lipid metabolism in HCC cells,which provides strong evidence for the use of this process as a drug target in the treatment of this malignancy.Dan Wu Yi Yang Yiran Hou Zifeng Zhao Ning Liang Peng Yuan Tao Yang Jinliang Xing Jibin Li 2022Cancer Communications2022,42,1:4
5Dual activated NIR-Ⅱ fluorescence and photoacoustic imaging-guided cancer chemo-radiotherapy using hybrid plasmonic-fluorescent assemblies显示文摘Multimodal imaging in the second near-infrared window(NIR-II)guided cancer therapy is a highly precise and efficient cancer theranostic strategy.However,it is still a challenge to develop activated NIR-II optical imaging and therapy agents.In this study,we develop a pH-responsive hybrid plasmonic-fluorescent vesicle by self-assembly of amphiphilic plasmonic nanogapped gold nanorod(AuNNR)and fluorescent down-conversion nanoparticles(DCNP)(AuNNR-DCNP Ve),showing remarkable and activated NIR-II fluorescence(FL)/NIR-II photoacoustic(PA)imaging performances.The hybrid vesicle also exhibited superior loading capacity of doxorubicin as a superior drug carrier and efficient radiosensitizer for X-ray-induced radiotherapy.Interestingly,the accumulated hybrid AuNNR-DCNP Ve in the tumor resulted in a recovery of NIR-II FL imaging signal and a variation in NIR-II PA imaging signal.Dual activated NIR-II PA and FL imaging of the hybrid vesicle could trace drug release and precisely guided cancer radiotherapy to ultimately reduce the side effects to healthy tissue.Tao Chen Lichao Su Xiaoguang Ge Wenmin Zhang Qingqing Li Xuan Zhang Jiamin Ye Lisen Lin Jibin Song Huanghao Yang 2020Nano Research2020,13,12:4
6Ultrasound-propelled Janus Au NR-mSiO_(2) nanomotor for NIR-II photoacoustic imaging guided sonodynamic-gas therapy of large tumors显示文摘Varieties of contrast agents have been developed for photoacoustic(PA)and ultrasound(US)imaging of cancers in vivo.However,access of traditional contrast agents into the sites of tumors has been principally through passive infiltration without any external force,preventing their deep penetration into the tissues of the tumors,and hindering the use of PA and US for deep tumor imaging.The concept of micro/nanomotors has been the focus of increasing attention as active theranostic agents due to their active movement in particular fluids,thereby conducting assigned tasks.Herein,US-propelled Janus mesoporous SiO_(2)partially coated gold nanorods(Au NR-mSiO_(2))were fabricated for deep tumor NIR-II PA imaging and synergistic sonodynamic-gas therapy.Following US irradiation,2,2-azobis[2-(2-imidazolin-2-yl)propane]dihydrochloride(AIPH)loaded in mSiO_(2)(Au NR-mSiO_(2)/AIPH)generated N_(2)microbubbles with high efficiency to achieve nanomotor drive.Due to the deep penetration of US,the nanomotors exhibited a capability to travel deep within sites of tumors,providing enhanced PA/US imaging inside the tumors.Furthermore,the nanomotor based cancer therapy was demonstrated through synergistic N_(2)gas and sonodynamic therapy.The US-propelled nanomotors demonstrated a novel strategy for the simultaneous PA/US dual imaging deep within tumor tissues and precise therapy of large tumors.Jiamin Ye Qinrui Fu Luntao Liu Lanlan Chen Xuan Zhang Qingqing Li Zhi Li Lichao Su Rong Zhu Jibin Song Huanghao Yang 2021Science China Chemistry2021,64,12:2
7Study and development of high peak power short pulse Nd:YAG laser for peening applications显示文摘Because short pulse Nd:YAG laser of nanosecond pulse-width and high peak power has a unique capability to improve the mechanical properties of metal parts,a study on the development of high peak power short pulse from Nd:YAG laser along with its peening application has been performed.The design scheme of laser and the characteristic of laser beam transmission are presented and discussed.A pulse energy of 25 J with 15 ns pulse-width and a maximum peak power of 1660 k W laser system which use one oscillation and eight amplifiers has been achieved.Laser beam has a max divergence angle of 0.03 mrad,a pulse-to-pulse pulse-width stability of±0.1 ns,and the pulse-to-pulse energy stability factors of less than±2.8%.A low value of divergence means an easier modification of a nearly hat-top laser beam intensity profile and an easier transmission of laser beam.To evaluate the performance of the laser system,several metal materials are processed.Laser peening quality and efficiency are analyzed by using an optical microscope,a transmission electron microscope,and an X-ray diffraction device.The processing results show that the performance of this laser system is excellent.QIAO HongChao ZHAO JiBin YANG Hao 2015Science China(Technological Sciences)2015,58,7:2

19Changes of China’s Status in the Global System and Its Influencing Factors:A Multiple Contact Networks Perspective显示文摘Clarifying China’s position in the global system is an important logical basis for developing national diplomacy.Although much research has been done on China’s development status,most studies have been based on country comparisons or institutional en-vironment.In today’s networked era in which the global economy,trade,personnel,and information are closely connected,studies on China’s global position and its status changes and influencing factors in multiple contact networks are still insufficient.In this study,from the perspective of diverse global contact networks,we constructed economic,cultural,and political influence indices to explore the changes and influencing factors on China’s status in the global system from 2005 to 2018.The results show that during the study period,China’s global influence in the fields of economic ties,cultural exchanges,and political contacts increased significantly,but its influ-ence in the fields of cultural exchanges and political contacts lagged far economic ties.The pattern of China’s economic influence on various economies around the world has shown a transformation from an‘upright pyramid’to an‘inverted pyramid’structure.The proportion of these economies in low-influence zones has decreased from more than 60%in 2005 to less than 20%in 2018.China’s cultural and political influence on various economies around the world has increased significantly;however,for the former,the percentage of high-influence areas is still less than 20%,whereas for the latter the percentage of these economies in medium-and high-influence areas is still less than 50%.Analyses such as a scatter plot matrix show that geographical proximity,economic globalization,close cooperation with developing countries,and a proactive and peaceful foreign policy are important factors in improving China’s status in the diverse global network system.LIU Jian LIU Jibin YANG Qingshan CAI Sikai LIU Jie 2024Chinese Geographical Science2024,34,2:0
