| 1 | Cell–cell contact with proinflammatory macrophages enhances the immunotherapeutic effect of mesenchymal stem cells in two abortion models显示文摘Mesenchymal stem cells(MSCs),which are pluripotent cells with immunomodulatory properties,have been considered good candidates for the therapy of several immune disorders,such as inflammatory bowel diseases,concanavalin A-induced liver injury,and graft-versus-host disease.The embryo is a natural allograft to the maternal immune system.A successful pregnancy depends on the timely extinction of the inflammatory response induced by embryo implantation,followed by the switch to a tolerant immune microenvironment in both the uterus and the system.Excessive infiltration of immune cells and serious inflammatory responses are triggers for embryo rejection,which results in miscarriage.Here,we demonstrated that adoptive transfer of MSCs could prevent fetal loss in a lipopolysaccharide(LPS)-induced abortion model and immune response-mediated spontaneous abortion model.The immunosuppressive MSCs alleviated excessive inflammation by inhibiting CD4+T cell proliferation and promoting the decidual macrophage switch to M2 in a tumor necrosis factor-stimulated gene-6(TSG-6)-dependent manner.Cell-tocell contact with proinflammatory macrophages increased the TSG-6 production by the MSCs,thereby enhancing the suppressive regulation of T cells and macrophages.Moreover,proinflammatory macrophages in contact with the MSCs upregulated the expression of CD200 on the stem cells and facilitated the reprogramming of macrophages towards an anti-inflammatory skew through the interaction of CD200 with CD200R on proinflammatory macrophages.Therefore,the results demonstrate that a TSG-6-mediated paracrine effect,reinforced by cell-to-cell contact between MSCs and proinflammatory macrophages,is involved in the mechanism of MSC-mediated abortion relief through the induction of immune tolerance.Our study also indicates the potential application of MSCs in clinical recurrent miscarriages. | Yanhong Li Di Zhang Ling Xu Lin Dong Ji Zheng Yikong Lin Jiefang Huang Yanyun Zhang Yu Tao Xingxing Zang Dajin Li Meirong Du | 2019 | Cellular & Molecular Immunology2019,16,12: | 23 |
| 2 | BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. | 5 | Effects of maize root exudates and organic acids on the desorption of phenanthrene from soils显示文摘The eects of maize root exudates and low-molecular-weight-organic anions (LMWOAs) on the desorption of phenanthrene from eight artificially contaminated soils were evaluated. A significant negative correlation was observed between the amounts of phenanthrene desorbed and the soil organic carbon (SOC) contents (P < 0.01), and the inuences of soil pH and clay content on phenanthrene desorption were insignificant (P > 0.1). Neither maize root exudates nor oxalate and citrate anions inuenced desorption of phenanthrene with the addition of NaN3. A faster phenanthrene desorption occurred without the addition of NaN3 in the presence of maize root exudates than oxalate or citrate due to the enhanced degradation by root exudates. Without the addition of NaN3, oxalate or citrate at dierent concentrations could inhibit phenanthrene desorption to dierent extents and the inhibiting eect by citrate was more significant than by oxalate. | 6 | Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. The influence of preparation technology on the structure and phase composition of the coating was investigated by scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS) and X-ray diffraction (XRD) analyses, and then their relationship was discussed. The results indicate that the Si/Mo ratio of the slurry and sintering processing were two main factors that significantly affected the structure and phase composition of the multi-coating. Appropriate sintering process and relatively high Si/Mo ratio were essential for preparing the multi-coating with dense structure and favorable phase composition. After being sintered at 1723 K for 2 h and with the Si/Mo ratio of the slurry being 4.5 (weight ratio), a dense structure accompanied by favorable phase composition of the coating can be obtained. When heat treated at 2373 K for 1 h, this coating became more compact and continuous. | 18 | High-efficiency extraction synthesis for high-purity copper nanowires and their applications in flexible transparent electrodes显示文摘Copper nanowires(Cu NWs)are considered an excellent alternative to indium tin oxide(ITO)in flexible transparency electrodes(FTEs).However,the mixed particles and surface oxidation of Cu NWs degrade the transmittance and conductivity of the electrodes.Therefore,highly purified Cu NWs without oxidation are vital for high-performance FTEs.Herein,a facile and effective purification process is introduced to purify Cu NWs in a water and n-hexane system,which takes advantage of the differences in hydrophilicity between Cu NWs and Cu NPs caused by their different adsorption affinities to octadecylamine(ODA).At the same sheet resistance,the transmittance of the purified Cu NW-based FTEs improved approximately 2%compared to that of non-purified Cu NW-based FTEs.Immersion of the electrode in glacial acetic acid removed the surface organics and oxides.After only 40 s of treatment,the sheet resistance dramatically decreased from 10^5 Ohm/sq to 31 Ohm/sq with a transmittance of 85%.In addition,the Cu NW-based FTE conductors showed excellent flexibility(remaining stable after 1000 bending cycles).The Cu NW-based FTEs were further applied to fabricate a flexible transparent heater.At a voltage of 10 V,the temperature of the heater reached 73℃,demonstrating the potential applications of this material in various fields. | He Zhang Shang Wang Yanhong Tian Jiayue Wen Chunjin Hang Zhen Zheng Yilong Huang Su Ding Chenxi Wang | 2020 | Nano Materials Science2020,2,2: | 4 |

| 20 | Effect of thermal exposure on the microstructure and creep properties of a fourth-generation Ni-based single crystal superalloy显示文摘The effect of thermal exposure on microstructure and creep properties of a fourth-generation nickelbased single crystal superalloy was investigated.The thermal exposure of samples after the full heat treatment was carried out at 1000℃,1100℃ and 1140℃ for 100 h and 200 h.The γ’ coarsening,γ’ rafting and γ channel widening were observed in samples after thermal exposure.When the thermal exposure time was constant,the morphology of γ’ phase in the alloy evolved significantly with increasing aging temperature.The interracial dislocation networks in aged samples after creep ruptured gradually became irregular and sparse with the increase of exposure temperature.When the higher exposure temperature was used,enla rgement of the defect pores was observed in samples,the microcracks were more likely to initiate and propagate at the corner of these pores.After aging at 1000℃ for 100 h,the creep life at 1140℃/137 MPa was slightly longer than that of heat-treated sample,which could be attributed to the slightly coarsened γ’ phase,homogenization of refractor elements.In contrast,the creep life of sample exposed at 1140℃ for 100 h was greatly decreased.The decrease of creep life was dominated by the rafting of γ’phase,the irregular interfacial dislocation networks as well as the enlargement of homogenization pores. | Yeshun Huang Xinguang Wang Chuanyong Cui Zihao Tan Jinguo Li Yanhong Yang Jinlai Liu Yizhou Zhou Xiaofeng Sun | 2021 | Journal of Materials Science & Technology2021,,10: | 4 |
