| 1 | Immunity-and-matrix-regulatory cells derived from human embryonic stem cells safely and effectively treat mouse lung injury and fibrosis显示文摘Lung injury and fibrosis represent the most significant outcomes of severe and acute lung disorders,including COVID-19.However,there are still no effective drugs to treat lung injury and fibrosis.In this study,we report the generation of clinical-grade human embryonic stem cells(hESCs)-derived immunity-and matrix-regulatory cells(IMRCs)produced under good manufacturing practice requirements,that can treat lung injury and fibrosis in vivo.We generate IMRCs by sequentially differentiating hESCs with serum-free reagents.IMRCs possess a unique gene expression profile distinct from that of umbilical cord mesenchymal stem cells(UCMSCs),such as higher expression levels of proliferative,immunomodulatory and anti-fibrotic genes.Moreover,intravenous delivery of IMRCs inhibits both pulmonary inflammation and fibrosis in mouse models of lung injury,and significantly improves the survival rate of the recipient mice in a dose-dependent manner,likely through paracrine regulatory mechanisms.IMRCs are superior to both primary UCMSCs and the FDA-approved drug pirfenidone,with an excellent efficacy and safety profile in mice and monkeys.In light of public health crises involving pneumonia,acute lung injury and acute respiratory distress syndrome,our findings suggest that IMRCs are ready for clinical trials on lung disorders. | Jun Wu Dingyun Song Zhongwen Li Baojie Guo Yani Xiao Wenjing Liu Lingmin Liang Chunjing Feng Tingting Gao Yanxia Chen Ying Li Zai Wang Jianyan Wen Shengnan Yang Peipei Liu Lei Wang Yukai Wang Liang Peng Glyn Nigel Stacey Zheng Hu Guihai Feng Wei Li Yan Huo Ronghua Jin Ng Shyh-Chang Qi Zhou Liu Wang Baoyang Hu Huaping Dai Jie Hao | 2020 | Cell Research2020,30,9: | 7 |

| 3 | The effects of asymmetric potential vorticity forcing on the instability of South Asia High and Indian summer monsoon onset显示文摘Based on the theory of potential vorticity(PV),the unstable development of the South Asia High(SAH)due to diabatic heating and its impacts on the Indian Summer Monsoon(ISM)onset are studied via a case diagnosis of 1998.The Indian Summer Monsoon onset in 1998 is related to the rapidly strengthening and northward moving of a tropical cyclone originally located in the south of Arabian Sea.It is demonstrated that the rapid enhancement of the cyclone is a consequence of a baroclinic development characterized by the phase-lock of high PV systems in the upper and lower troposphere.Both the intensification of the SAH and the development of the zonal asymmetric PV forcing are forced by the rapidly increasing latent heat released from the heavy rainfall in East Asia and South East Asia after the onsets of the Bay of Bengal(BOB)monsoon and the South China Sea(SCS)monsoon.High PV moves southwards along the intensified northerlies on the eastern side of the SAH and travels westwards on its south side,which can reach its northwest.Such a series of high PV eddies are transported to the west of the SAH continuously,which is the main source of PV anomalies in the upper troposphere over the Arabian Sea from late spring to early summer.A cyclonic curvature on the southwest of the SAH associated with increasing divergence,which forms a strong upper tropospheric pumping,is generated by the anomalous positive PV over the Arabian Sea on 355 K.The cyclone in the lower troposphere moves northwards from low latitudes of the Arabian Sea,and the upper-layer high PV extends downwards and southwards.Baroclinic development thus occurs and the tropical low-pressure system develops into an explosive vortex of the ISM,which leads to the onset of the ISM.In addition,evolution of subtropical anticyclone over the Arabian Peninsula is another important factor contributing to the onset of the ISM.Before the onset,the surface sensible heating on the Arabian Peninsula is very strong.Consequently the subtropical anticyclone which dominated the Arabian Sea in spring retreats westwards to the Arabian Peninsula and intensifies rapidly.The zonal asymmetric PV forcing develops gradually with high PV eddies moving southwards along northerlies on the eastern side of the anticyclone,and a high PV trough is formed in the middle troposphere over the Arabian Sea,which is favorable to the explosive barotropic development of the tropical cyclone into the vortex.Results from this study demonstrate that the ISM onset,which is different from the BOB and the SCS monsoon onset,is a special dynamical as well as thermodynamic process occurring under the condition of fully coupling of the upper,middle,and lower tropospheric circulations. | ZHANG YaNi WU GuoXiong LIU YiMin GUAN Yue | 2014 | Science China Earth Sciences2014,57,2: | 5 |

| 5 | Novel soybean peptide iglycin ameliorates insulin resistance of high-fat diet fed C57BL/6J mice and differentiated 3T3L1 adipocytes with improvement of insulin signaling and mitochondrial function显示文摘Soy consumption has been associated with potential health benefits in reducing chronic diseases.These physiological functions have been attributed to soy proteins or more commonly to bioactive peptides.Thus,more studies are required to identify these bioactive peptides,and elucidate their biological mechanisms of action.In the present study,a novel peptide iglycin was purifi ed from soybean seeds with a molecular mass of 3.88 k Da.Thereafter,iglycin reduced fasting blood glucose and restored insulin sensitivity of C57 BL/6 J mice on a high-fat diet with increased phosphorylation of insulin receptor substrate 1(IRS1)and AKT in adipose tissue.Furthermore,it improved glucose uptake,induced translocation of intracellular GLUT4 to plasma membrane and activation of insulin signaling in adipocytes under insulin-resistant condition.In addition,it decreased reactive oxygen species production,lipid peroxidation and inhibited adipocyte apoptosis with improved mitochondrial function as evidenced by up-regulation of succinate dehydrogenase activity,mitochondrial membrane potential and intracellular ATP store.These data suggested that iglycin ameliorated insulin resistance via activation of insulin signaling,which was associated with inhibition of oxidative stress,adipocyte apoptosis,and improvement of mitochondrial function. | Yinghuan Wu Ran Zhao Minxia Li Huiyun Li Zhengwang Chen Yanying Zhao | 2022 | Food Science and Human Wellness2022,11,6: | 3 |
| 6 | Associated risk factors in the early stage of diabetic retinopathy显示文摘Background:To investigate the retinal capillary density(RCD)of the macula using optical coherence tomography angiography(OCT-A)in type 2 diabetic patients and to further determine the association with risk factors.Methods:A total of 212 eyes from 212 subjects were recruited;subjects included diabetics with no retinopathy(NDR,n=90 eyes),diabetics with mild retinopathy DR(MDR,n=36 eyes),and healthy participants(Control,n=86 eyes).All participants underwent OCT-A scanning.RCD was quantified by superficial and deep retinal capillary layers(SRCL and DRCL)from OCT-A images.Results:RCD in SRCL and DRCL was lower in NDR(P<0.001)as well as in MDR(P<0.001)when compared with control eyes.Diabetic patients were subdivided according to individual risk factors,complications related to diabetes,and hyperglycemia.Diabetic patients showed lower RCD in both the SRCL and DRCL when compared with healthy controls.Diabetics with age>55y,HbA1c>7%had significantly reduced DRCL(P<0.05)when compared with the other group of diabetics(age<55y,HbA1c<7%).Diabetics with a blood urea nitrogen(BUN)>8.2 mmol/L had significantly reduced SRCL and DRCL when compared to the other group of diabetics.Conclusions:Risk factors including older age,higher level of HbA1c,LDL-C and BUN,were associated with lower RCDs found in type 2 diabetic patients with and without mild DR by OCT-A.The impairment of retinal capillary by OCT-A may play a key role in the early monitoring of management in diabetes. | Fan Tan Qi Chen Xiran Zhuang Chaoming Wu Yanying Qian Yuanyuan Wang Jianhua Wang Fan Lu Meixiao Shen Yingzi Li | 2019 | Eye and Vision2019,6,1: | 2 |

| 18 | Body-centered-cubic structure and weak anharmonic phonon scattering in tungsten显示文摘It was recently found that the anharmonic phonon–phonon scattering in tungsten is extremely weak at high frequencies,leading to a predominance of electron-phonon scattering and consequently anomalous phonon transport behaviors.In this work,we calculate the phonon linewidths of W along high-symmetry directions from first principles.We find that the weak phonon–phonon scattering can be traced back to two factors.The first is the triple degeneracy of the phonon branches at the P and H points,a universal property of elemental body-centered-cubic(bcc)structures.The second is a relatively isotropic character of the phonon dispersions.When both are met,phonon–phonon scattering rates must vanish at the P and H points.The weak phonon–phonon scattering feature is also applicable to Mo and Cr.However,in other elemental bcc substances like Na,the isotropy condition is violated due to the unusually soft character of the lower transverse acoustic phonon branch along the Γ-N direction,opening emission channels and leading to much stronger phonon–phonon scattering.We also look into the distributions of electron meanfree paths(MFPs)at room temperature in tungsten,which can help engineer the resistivity of nanostructured W for applications such as interconnects. | Yani Chen Jinlong Ma Shihao Wen Wu Li | 2019 | npj Computational Materials2019,,1: | 0 |
| 19 | Influences of Echinacea Polysaccharide on Secretion of IL-8mRNA by LPS-injured IEC-6 Cells显示文摘[Objective] The paper aimed to study the effects of Echinacea polysaccharide on secretion of IL-8m RNA by LPS-injured IEC-6 cells,in order to figure out the mechanism of EPS on injured cells. [Method]Total RNA was extracted with TRIzon reagent. IL-8 m RNA was amplified by RT-PCR and detected by agarose gel electrophoresis. Furthermore,electrophoresis and image was analyzed. [Result] The 50 μg/m L EPS could partially inhibit IL-8 m RNA level produced by LPS-stimulated IEC-6; with the increasing concentration of EPS(200 and 500 μg/m L),the inhibitory effect against IL-8 m RNA gradually enhanced. IEC-6 was pretreated by 50,100,200 and 500 μg/m L EPS for 24 h,then stimulated by 10 μg/m L LPS for 1 and 4 h,respectively. RT-PCR analysis showed that the expression of IL-8 m RNA induced by LPS was effectively inhibited by EPS; the inhibitory effect of EPS on expression of IL-8m RNA after stimulated by LPS for 4 h was more intense and the expression concentration was lower; the inhibitory rate at 4 h was higher than that at 1 h.
