2Analysis of in vivo patterns of caspase 3 gene expression in primary hepatocellular carcinoma and its relationship to p21^(WAF1) expression and hepatic apoptosis显示文摘AIM To detect the expression of caspase 3gene in primary human hepatocellular carcinoma(HCC)and investigate its relationship to p21WAF1gene expression and HCC apoptosis.METHODS In situ hybridization was employedto determine caspase 3 and p21WAF1expression inHCC.In situ end-labeling was used to detecthepatocytic apoptosis in HCC.RESULTS Twenty-one of 39(53.8%)cases ofHCC were found to express caspase 3transcripts,while 45.2% of HCC failed toexpress caspase 3.Non-cancerous adjacent livertissues showed more positive caspase 3(87.5%,7/8)as compared with HCC(P<0.05).The expression of caspase 3 is correlated withHCC differentiation,72.2%(13/18)ofmoderately to highly differentiated HCC showedcaspase 3 transcripts positive,while only 38.1%of poorly differentiated HCC harbored caspase 3transcripts(P<0.05).No relationship wasfound between caspase 3 expression and tumorsize or grade or metastasis,although 52.5%(5/8)of HCC with metastasis were caspase 3positive and a little higher than that with nometastasis(51.6%,P>0.05).Expression of caspase 3 alone did not affect the apoptosisindex(AI)of HCC.The AI was 7.12%o in caspase3-positive tumors(n=21),while in caspase 3-negative cases(n=18)6.59%0(P>0.05).Expression of caspase 3 clearly segregated withp21WAF1positive tumors as compared withp21WAF1-negative cases(16 of 23,69.6% versus5 of 16,31.3%)with statistical significance(P=0.017).In the cases with positive caspase 3and negative p21WAF1,the Al was found slightlyhigher,but with no statistical significance,thanthat with expression of p21WAF1and caspase 3(7.21‰ vs 6.98‰,P>0.05).CONCLUSION Loss of caspase 3 expressionmay contribute to HCC carcinogenesis,althoughthe expression of caspase 3 does not correlatewell with cell apoptosis in HCC.p21WAF1may bemerely one of the inhibitors which can reducecaspase 3 mediated cell apoptosis in HCCs.Bao Hua Sun Jun Zhang Bao JǜWang Xi Ping Zhao You Kun Wang Zhi Qun Yu Dong Liang Yang Lian Jie Hao Department of Clinical Immunology,Tongji Hospital,Tongji Medical University,Wuhan 430030,Hubei Province,China 2000World Journal of Gastroenterology2000,6,3:65
3Design concept of three-dimensional section controllable internal waverider hypersonic inlet显示文摘A new hypersonic inlet named three-dimensional section controllable internal waverider inlet is presented in this paper to achieve the goal of section shape geometric transition and complete capture of the upstream mass. On the basis of the association between hypersonic waverider airframe and streamtraced hypersonic inlet, the waverider concept is extended to yield results for the internal flows, namely internal waverider concept. It is proven theoretically that not osculating cones but osculating axisymmetric theory is appropriate for the design of section controllable internal waverider inlet. And two design methods out of the internal waverider concept are proposed subsequently to construct two inlets with specific section shape request, triangle to ellipse and rectangle to ellipse ones. The calculation results show that the inlets are capable of keeping their shock structures and the main flow characteristics exactly as their derived flowfield. Further, the inlets successfully capture all the upstream mass despite their complicated cross-section transitions. It is believed that the concept proposed ex- plores a new way of designing three-dimensional hypersonic inlets with special demand of section shape transition. 8Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 9Estimates of genetic and phenotypic parameters for weight and length in Paralichthys olivaceus(Temminck et Schlegel)显示文摘The complete diallel cross design was employed to estimate the genetic parameters of the growth of Japanese flounder (Paralichthys olivaceus). A total of 60 full-sib families were cultivated and two growth-related traits, body weight (BW) and body length (BL), were examined at average 2, 3 and 8 months of age respectively, with 1 800 individuals measured in each age group (30 per family). Based on the additive-dominance-maternal-effect genetic analysis model, the restricted maximum likelihood approach was used to estimate various (co)variance components. The results showed that narrow-sense heritability estimates of BW and BL were respectively: 0.29 ± 0.01 and 0.22 ± 0.02 at 2 months of age, 0.32 ± 0.02 and 0.30 ± 0.04 at 3 months of age, 0.48 ± 0.04 and 0.40 ± 0.05 at 8 months of age; broad-sense heritability estimates were respectively: 0.44 ± 0.02 and 0.54 ± 0.04 at 2 months of age, 0.35 ± 0.01 and 0.36 ± 0.03 at 3 months of age, 0.49 ± 0.03 and 0.45 ± 0.04. All heritabilities were statistically significant (P <0.05). Additive genetic correlations between BW and BL at 2, 3 and 8 months of age were consistently positive and highly significant (P <0.01): 0.93 ± 0.02, 0.95 ± 0.03 and 0.92 ± 0.03 respectively. Maternal effect was significant (P <0.05) only at 2 months of age, and was not detected at 3 and 8 months of age. 10Earthquake prediction from China's mobile gravity data显示文摘The relation between plate tectonics and earthquake evolution is analyzed systematically on the basis of 1998e2010 absolute and relative gravity data from the Crustal Movement Observation Network of China. Most earthquakes originated in the plate boundary or within the fault zone. Tectonic deformation was most intense and exhibited discontinuity within the tectonically active fault zone because of the differential movement; the stress accumulation produced an abrupt gravity change, which was further enhanced by the earthquake. The gravity data from China's Mainland since 2000 obviously reflected five major earthquakes(Ms > 7), all of which were better reflected than before 2000. Regional gravity anomalies and a gravity gradient change were observed in the area around the epicenter about 2 or 3 years before the earthquake occurred, suggesting that gravity change may be a seismic precursor. 12Antiviral therapy for hepatitis B virus-related hepatocellular carcinoma after radical hepatectomy显示文摘Objective:To assess the effect of antiviral therapy for hepatitis B virus(HBV)-related hepatocellular carcinoma(HCC)after radical hepatectomy.Methods:A total of 478 HBV-related HCC patients treated by radical hepatectomy were retrospectively collected.Patients in the treatment group(n=141)received postoperative lamivudine treatment(100 mg/d),whereas patients in the control group(n=337)did not.Recurrence-free survival(RFS)rates,overall survival(OS)rates,treatments for recurrent HCC and cause of death were compared between the two groups.Propensity score matching(PSM)analysis was also conducted to reduce confounding bias between the two groups.Results:The 1-,3-,and 5-year RFS rates didn't significantly differ between the two groups(P=0.778);however,the 1-,3-,and 5-year OS rates in the treatment group were significantly higher than those in the control group(P=0.002).Similar results were observed in the matched data.Subgroup analysis showed that antiviral treatment conferred a significant survival benefit for Barcelona Clinical Liver Cancer stage A/B patients.Following HCC recurrence,more people in the treatment group were able to choose curative treatments than those in the control group(P=0.031).For cause of death,fewer people in the treatment group died of liver failure than those in the control group(P=0.041).Conclusion:Postoperative antiviral therapy increases chances of receiving curative treatments for recurrent HCC and prevents death because of liver failure,thereby significantly prolonging OS,especially in early-or intermedian-stage tumors.Yang Ke Liang Ma Xue-Mei You Sheng-Xin Huang Yong-Rong Liang Bang-De Xiang Le-Qun Li 2013Cancer Biology & Medicine2013,10,3:11
13Protective Effect of Resveratrol on the Brain in a Rat Model of Epilepsy显示文摘Accumulating evidence has suggested resveratrol as a promising drug candidate for the treatment of epilepsy. To validate this, we tested the protective effect of resveratrol on a kainic acid(KA)-induced epilepsy model in rats and investigated the underlying mechanism. We found that acute resveratrol application partially inhibited evoked epileptiform discharges in the hippocampal CA1 region. During acute, silent and chronic phases of epilepsy,the expression of hippocampal kainate glutamate receptor(GluK2) and the GABAAreceptor alpha1 subunit(GABAAR-alpha1) was up-regulated and down-regulated,respectively. Resveratrol reversed these effects and induced an antiepileptic effect. Furthermore, in the chronic phase, resveratrol treatment inhibited the KA-induced increased glutamate/GABA ratio in the hippocampus. 16Synthesis and Spectrophotometric Studies of 2-(2-Imidazolylazo)-5-Dicthylamino Phenol显示文摘In this paper, 2-(2-imidazolylazo)-5-diethylami phenol (IZAPN) was prepared, theprotonation behaviour and the condition of spectrophotometric determination of cobalt with thisreagent are reported. The results show that the new colour reagent reacts with cobalt to from astable purple red complex (l:3). However, the complex formed could be changed into anotherspecies with stable absorptivity by addition of mineral acid (1+1). 17The Chinese Hα Solar Explorer(CHASE) mission: An overview显示文摘The Chinese Hα Solar Explorer(CHASE), dubbed “Xihe”—Goddess of the Sun, was launched on October 14, 2021 as the first solar space mission of China National Space Administration(CNSA). The CHASE mission is designed to test a newly developed satellite platform and to acquire the spectroscopic observations in the Hα waveband. The Hα Imaging Spectrograph(HIS)is the scientific payload of the CHASE satellite. It consists of two observational modes: raster scanning mode and continuum imaging mode. The raster scanning mode obtains full-Sun or region-of-interest spectral images from 6559.7 to 6565.9 ? and from 6567.8 to 6570.6 ? with 0.024 ? pixel spectral resolution and 1 min temporal resolution. The continuum imaging mode obtains photospheric images in continuum around 6689 ? with the full width at half maximum of 13.4 ?. The CHASE mission will advance our understanding of the dynamics of solar activity in the photosphere and chromosphere. 18Macrophage-NLRP3 Inflammasome Activation Exacerbates Cardiac Dysfunction after Ischemic Stroke in a Mouse Model of Diabetes显示文摘Ischemic stroke is one of the leading causes of death worldwide.In the post-stroke stage,cardiac dysfunction is common and is known as the brain-heart interaction.Diabetes mellitus worsens the post-stroke outcome.Stroke-induced systemic inflammation is the major causative factor for the sequential complications,but the mechanism underlying the brain-heart interaction in diabetes has not been clarified.The NLRP3(NLR pyrin domain-containing 3) inflammasome,an important component of the inflammation after stroke,is mainly activated in M1-polarized macrophages.In this study,we found that the cardiac dysfunction induced by ischemic stroke is more severe in a mouse model of type 2 diabetes.Meanwhile,M1-polarized macrophage infiltration and NLRP3 inflammasome activation increased in the cardiac ventricle after diabetic stroke.Importantly,the NLRP3 inflammasome inhibitor CY-09 restored cardiac function,indicating that the Ml-polarized macrophage-NLRP3 inflammasome activation is a pathway underlying the brain-heart interaction after diabetic stroke.Hong-Bin Lin Guan-Shan Wei Feng-Xian Li Wen-Jing Guo Pu Hong Ya-Qian Weng Qian-Qian Zhang Shi-Yuan Xu Wen-Bin Liang Zhi-Jian You Hong-Fei Zhang 2020Neuroscience Bulletin2020,36,9:7

20Efficacy of hepatic resection vs transarterial chemoembolization for solitary huge hepatocellular carcinoma显示文摘AIM: To compare the efficacy of hepatic resection(HR) and transarterial chemoembolization(TACE) for patients with solitary huge(≥ 10 cm) hepatocellular carcinoma(HCC).METHODS: Records were retrospectively analyzed of 247 patients with solitary huge HCC, comprising 180 treated by HR and 67 by TACE. Long-term overall survival(OS) was compared between the two groups using the Kaplan-Meier method, and independent predictors of survival were identified by multivariate analysis. These analyses were performed using all patients in both groups and/or 61 pairs of propensity score-matched patients from the two groups. RESULTS: OS at 5 years was significantly higher in the HR group than the TACE group, across all patients(P = 0.002) and across propensity score-matchedpairs(36.4% vs 18.2%, P = 0.039). The two groups showed similar postoperative mortality and morbidity.
