| 1 | Hepatitis B virus X protein enhances hepatocarcinogenesis by depressing the targeting of NUSAP1 mRNA by miR-18b显示文摘Objective: The aim of this study was to investigate the underlying mechanism whereby HBx modulates the targeting of NUSAP1 by miR-18b to enhance hepatocarcinogenesis.Methods: We employed an integrated approach of bioinformatics analysis and molecular experiments in hepatoma cells, HBV transgenic mice, and clinical liver cancer tissues to investigate the role of HBx-regulated miR-18b in the development of liver cancer.Results: In this study, we report that the HBx-mediated tumor suppressor miR-18b modulates hepatocarcinogenesis during the host-HBV interaction. The expression levels of miR-18b were lower in clinical HBV-positive liver cancer tissues and liver tissues of HBV-transgenic mice. Interestingly, HBx inhibited miR-18b expression by inducing the methylation of CpG islands in its promoter. Accordingly, we tested the hypothesis that HBx enhanced hepatocarcinogenesis by increasing the expression of target genes of miR-18b. Moreover, we identified nucleolar spindle-associated protein 1(NUSAP1) as one of the target genes of miR-18b.NUSAP1 was expressed at high levels in liver cancer tissues. Interestingly, HBx up-regulated NUSAP1 by suppressing miR-18b.Functionally, miR-18b significantly inhibited the proliferation of hepatoma cells by depressing NUSAP1 levels in vivo and in vitro.Conclusions: Thus, we conclude that the targeting of NUSAP1 mRNA by the tumor suppressor miR-18b is controlled by HBxmodulated promoter methylation during the host-virus interaction, leading to hepatocarcinogenesis. | 2 | Microvascular pathological features and changes in related injury factors in a rat acute blood stasis model显示文摘OBJECTIVE: To examine the microvascular pathological characteristics and changes in related injury factors in a rat model of acute blood stasis.METHODS: A total of 75 Sprague-Dawley rats were divided randomly and equally into a control group and four experimental groups assessed at different times after the induction of stasis(0, 1, 3 or 6 h after stasis)(n = 15). The acute blood stasis model was established through rat tail-vein injection of high-molecular-weight dextran. After Electrocardiograph(ECG) detection at predetermined times(0,1, 3 and 6 h after induction of stasis), the rats were sacrificed and blood and cardiac samples were harvested for analysis. Hematoxylin-eosin(HE) staining and transmission electron microscopy were used for histopathological detection; an enzyme linked immunosorbent assay(ELISA) was used to detect thromboxane B2(TXB2) and 6-keto-prostaglandin F1α(6-Keto-PGF1α) concentrations; a real-time polymerase chain reaction(PCR) reaction system was used to detect intercellular adhesion molecule1(ICAM-1) and vascular cell adhesion molecule1(VCAM-1) m RNA expression; western blotting was used to detect vascular endothelial cadherin(VE-cadherin) protein expression.RESULTS: The ST segment in the ECG showed gradual elevation after induction of stasis and continued elevation at a high level at 3 and 6 h. The HE staining showed changes in myocardial cell necrosis and tissue dissociation after the induction of stasis, along with inflammatory infiltration. Results of transmission electron microscopy showed immediate changes in blood stasis and lumen occlusion in the microvasculature, along with endothelial cell swelling. After the induction of stasis, TXB2 concentrations gradually increased while 6-Keto-PGF1αconcentrations were immediately significantly reduced. The TXB2/6-Keto-PGF1αratio was maintained at a high level. ICAM-1 m RNA expression showed an unstable elevation while VCAM-1 m RNA expression was significantly reduced after the induction of stasis. Compared with the control group, VE-cadherin protein expression increased at 0 and 3 h after the induction of stasis, while no change occurred at 1 and 6 h.CONCLUSION: The pathological manifestations of acute blood stasis are microvascular blood retention, lumen stenosis and even occlusion. The condition is also called 'blood coagulation and weep' in Traditional Chinese Medicine. | 3 | Corresponding Factors Influencing Crude Oils Assay Using Low-field Nuclear Magnetic Resonance显示文摘In this study, the main factors influencing the measurements by means of the off-line low-field 1H NMR in the lab were discussed base on a robust calibration model established by the PLS algorithm using 255 crude oil samples. The preheating temperature had a great influence on the viscosity of oil samples and the resolution of spectral analysis. The repeatability of spectral measurements was impacted by the metal and wax content of the oil samples. For the case of high wax content oils, the wax species began to crystallize in the course of determination that could affect the repeatability of spectral measurements. These factors have evidenced why the preheating devices and filter unit are necessary when low field NMR system is used in the online analysis process. | 4 | Effects of Sampling Conditions on Composition and Emission Characteristics of Volatile Organic Compounds in Process Units from Different Refineries显示文摘This study investigates the effects of sampling conditions on volatile organic compound(VOC)compositions including different flow restrictors,SUMMA volumes,sampling heights,and wind speeds.Results show that at the six sampling heights the concentrations of main VOC species were slightly different,while the wind speed had a greater impact on the VOC composition of source profiles.With the increase of wind speed,the weighted percentage of high-carbon aromatic hydrocarbons was higher.Besides,there was an extremely different profile between the normal production and shutdown conditions of the delayed coking unit.To compare the emission characteristics of VOCs in various process units of the S and the C refineries,the samples were collected from the catalytic cracking unit,the continuous catalytic reforming unit,and the delayed coking unit.In the continuous catalytic reforming unit,C3-C5 alkanes and low-carbon aromatic hydrocarbons were the main components collected from the S and the C refineries,accounting for 67.1%and 34.9%,respectively.For the delayed coking unit,the total weighted percentage of high carbon C6-C12 alkanes was significantly higher than other units in the S and the C refineries,accounting for 30.5%and 24.4%,respectively.In the catalytic cracking unit,the low-carbon C2-C5 alkanes were abundant,and the weighted percentage of propylene was higher.The emission characteristics obtained were consistent with the processing technology of production units.The results indicate that the VOC emission characteristics from the same production unit in different refineries have similarities and significant differences which are related to the technological process.The emission characteristics of VOCs could provide the data support for source apportionment work in the production units. | Feng Yunxia Xiao Anshan Li Bo Dong Rui Jia Runzhong Li Mingjun | 2020 | China Petroleum Processing & Petrochemical Technology2020,22,3: | 3 |
| 5 | Changes in nitric oxide,angiotensinⅡ,angiopoietin-like protein 4 mRNA,neuregulin 1 mRNA,and platelet endothelial cell adhesion molecule-1 in rats with acute blood stasis induced by high-molecular-weight dextran显示文摘OBJECTIVE: To investigate the influence of acute blood stasis on nitric oxide(NO), angiotensin Ⅱ(Ang Ⅱ), angiopoietin-like protein 4(ANGPTL4)m RNA, neuregulin 1(NRG-1) m RNA, and platelet endothelial cell adhesion molecule-1(PECAM-1) in rats with stasis induced by high-molecular-weight dextran(HMWD).METHODS: Seventy-five Sprague Dawley rats were divided randomly into five groups(n = 15 in each group): control group, immediate group, 1 h group,3 h group, and 6 h group. A model of acute blood stasis was established via injection of HMWD into the tail vein. After performing electrocardiogram at the predetermined times according to the grouping, we collected blood and cardiac samples for hematoxylin-eosin(HE) staining and histopathological examination via transmission electron microscopy. Enzyme-linked immunosorbent assay was used to detect plasma levels of NO, AngⅡ, and fibrinogen. Real-time polymerase chain reaction was used to detect the expression of ANGPTL4 m RNA and NRG-1 m RNA. Immunohistochemical methods were used to detect PECAM-1 protein expression.RESULTS: The rat model of blood stasis showed blood retention in the capillary lumens. The ST segment showed gradual elevation, and was still elevated at 3 and 6 h after induction of blood stasis.HE staining showed myocardial cell necrosis and dissolution after modeling, along with basement membrane rupture and mitochondrial structural damage. Transmission electron microscopy showed endothelial cell swelling and an increase in absorption vesicles immediately after modeling. Endothelial cell apoptosis was increased at 3 and 6 h after modeling. Cardiac muscle fibers were disordered and intercalated discs were blurred immediately after modeling. There were massive numbers of dissolved cardiac muscle fibers, ruptured basement membranes, and mitochondrial structural damage at 3 and 6 h after modeling. NO plasma concentration was significantly reduced immediately and 1 h after modeling, while it was increased at 3 and 6 h.AngⅡ plasma concentration was decreased immediately after modeling, but increased at 1, 3, and6 h. Fibrinogen plasma concentration was significantly increased at immediate, 1, 3, and 6 h after modeling. PECAM-1 protein expression was obviously increased immediately after modeling, at 1, 6 h was found mild augment. Expression of Ang PTL4 m RNA was increased at immediate, 1, 3, and 6 h after modeling, and was found further augment at 3,and 6 h. Expression of NRG-1 m RNA was increased at immediate, 1, 3, and 6 h after modeling, and the strongest expression was at 1 h.CONCLUSION: The pathological manifestation of acute blood stasis is characterized by microvascular blood retention. | 7 | Synthesis and electrochemical properties of Zn_3V_3O_8 as novel anode material显示文摘Zn_3V_3O_8 two-dimensional micro sheets are successfully synthesized by combination of solvothermal method and heat treatment. The X-ray diffraction(XRD) and field emission scanning electron microscopy(SEM) are used to characterize the structure and morphology of the samples, and the battery testing system is used to investigate its electrochemical performance. The results show that the Zn_3V_3O_8 is successfully obtained using mix solvent(water:ethylene glycol =1:1), and the sample takes on morphology of two-dimensional sheets. The initial discharge capacity for Zn_3V_3O_8 two-dimensional micro sheets is 752.0 mAh/g at 100 mA/g in the voltage range of 0.01-3 V. After heat treatment in a tube furnace at 550 ℃, the sample had the initial discharge capacity as 1152.0 mAh/g and maintained it at 901.4 mAh/g after 100 cycles. | 10 | A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. | 11 | Correlation analysis of the North Equatorial Current bifurcation and the Indonesian Throughflow显示文摘Based on monthly mean Simple Ocean Data Assimilation(SODA) products from 1958 to 2007,this study analyzes the seasonal and interannual variability of the North Equatorial Current(NEC) bifurcation latitude and the Indonesian Throughflow(ITF) volume transport. Further,Empirical Mode Decomposition(EMD) method and lag-correlation analysis are employed to reveal the relationships between the NEC bifurcation location,NEC and ITF volume transport and ENSO events. The analysis results of the seasonal variability show that the annual mean location of NEC bifurcation in upper layer occurs at 14.33°N and ITF volume transport has a maximum value in summer,a minimum value in winter and an annual mean transport of 7.75×106 m3/s. The interannual variability analysis indicates that the variability of NEC bifurcation location can be treated as a precursor of El Ni?o. The correlation coefficient between the two reaches the maximum of 0.53 with a time lag of 2 months. The ITF volume transport is positively related with El Ni?o events with a maximum coefficient of 0.60 by 3 months. Moreover,the contrast test of a variety of control measures was conducted. [Result] A. piceacola needed one year or more to complete a generation; there were significant differences in insect plant rate and extremely significant differences in population density among different plots. Trunk injection and foliar spraying conducted before the forth larval stage had good protection effects on needles of Picea mongolica; trunk injection with 14% imidacloprid · dichlorvos at egg stage received the best control effect,the loss rate of needles was the lowest of 8. 43%,so the method could be massively popularized in the future. | 20 | In vivo and in situ real-time fluorescence imaging of peripheral nerves in the NIR-II window显示文摘The peripheral nervous system(PNS)is essential for performing and maintaining various motor and sensory functions.Abnormalities can lead to a series of peripheral neurological conditions,such as paraesthesia,pain,or spasms,which are debilitating and lowering the quality of life.Thecurrent guidelines for diagnosis rely predominantly on clinical symptoms resulting from PNS dysfunction,which occur already at an advancedstage.There are currently no effective methods that visually reflect the extent of peripheral neuropathy.In our study,we present a novel in vivoand in situ real-time imaging of peripheral nerves based on the second near-infrared window(NIR-II)fluoresce nee.In NIR-II system,lead sulfidequa ntum dots(PbS Qds)with NIR-II fluoresce nee specifically bound to motor neuro rvspecific protein agrin,acting as image con trast.In micemodel,peripheral nerves were visible as soon as after 2 h post injection.We provide evidenee for the efficacy of this approach,which allows todirectly dem on strate peripheral nerves,their structure,and pote ntial damagesites and degree.Furthermore,our products were of goodbiocompatibility,while the n eural fluoresce nee signal was solid,bright and stable for 4 h in vivo.Thus,overall,our results suggest that NIR-II isan effective new method for direct imaging of peripheral nerves in vivo,opening new horizons on early,improved and more precise,targeteddiag no sis.A resulti ng more rapid installatio n of perso nalized therapy facilitates a better prognosis of clinical peripheral neuropathy. | Zhujun Feng Yimeng Yang Jian Zhang Kan Wang Yunxia Li Heng Xu Zhen Wang Ewelina Biskup Shixian Dong Xing Yang Yuefeng Hao Jun Chen Yan Wo | 2019 | Nano Research2019,12,12: | 1 |
