| 1 | Reduction of the incidence and mortality of rectal cancer bypolypectomy:a prospective cohort study in Haining County显示文摘AIM: To reduce the incidence and morlality of rectal cancerand address the hypothesis that colorectal cancer often arisefrom precursor lesion (s), either adenomas or non-alenomatous polyps, by conducting a population-basedmass screening for colorectal cancer in Haining County,Zhejiang, PRCMETHODS: From 1977 to 1980, physicians screened thepopulation of Haining County using 15 cm rigid endoscopy.Of over 240 000 participants, 4076 of them were diagnoselwith Precursor Iesions, eitler alenomas or non-adenomatous polyps, which were then removedl surgically.All individuals with precursor lesionswere followed up andreexamined by endoscopy every two to five years upto 1998.RESULTS: After the initial screening, 953 metactronousadenomas and 417 non-adenomatous polyps ware detected andremoved from the members of this cohort. Further, 27 cases ofcolorectal cancer were detected and treated. Log-rank tuestsshowed that the survival time among those cancer patients whounderwent mass screening increased significantly comparedtothat of other colorectal cancer patients (P < 0.0001). | 2 | AXL is a candidate receptor for SARS-CoV-2 that promotes infection of pulmonary and bronchial epithelial cells显示文摘The current coronavirus disease 2019(COVID-19)pandemic presents a global public health challenge.The viral pathogen responsible,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),binds to the host receptor ACE2 through its spike(S)glycoprotein,which mediates membrane fusion and viral entry.Although the role of ACE2 as a receptor for SARS-CoV-2 is clear,studies have shown that ACE2 expression is extremely low in various human tissues,especially in the respiratory tract.Thus,other host receptors and/or co-receptors that promote the entry of SARS-CoV-2 into cells of the respiratory system may exist.In this study,we found that the tyrosine-protein kinase receptor UFO(AXL)specifically interacts with the N-terminal domain of SARS-CoV-2 S.Using both a SARS-CoV-2 virus pseudotype and authentic SARS-CoV-2,we found that overexpression of AXL in HEK293T cells promotes SARS-CoV-2 entry as efficiently as overexpression of ACE2,while knocking out AXL significantly reduces SARS-CoV-2 infection in HI 299 pulmonary cells and in human primary lung epithelial cells.Soluble human recombinant AXL blocks SARS-CoV-2 infection in cells expressing high levels of AXL.The AXL expression level is well correlated with SARS-CoV-2 S level in bronchoalveolar lavage fluid cells from COVID-19 patients.Taken together,our findings suggest that AXL is a novel candidate receptor for SARS-CoV-2 which may play an important role in promoting viral infection of the human respiratory system and indicate that it is a potential target for future clinical intervention strategies. | Shuai Wang Zongyang Qiu Yingnan Hou Xiya Deng Wei Xu Tingting Zheng Peihan Wu Shaofang Xie Weixiang Bian Chong Zhang Zewei Sun Kunpeng Liu Chao Shan Aifu Lin Shibo Jiang Youhua Xie Qiang Zhou Lu Lu Jing Huang Xu Li | 2021 | Cell Research2021,31,2: | 39 |
| 3 | The control of syndepositional faulting on the Eogene sedimentary basin fills of the Dongying and Zhanhua sags, Bohai Bay Basin显示文摘The Dongying and Zhanhua sags are the major hydrocarbon exploration and produc- tion subbasins in the Bohai Bay Basin. Integrated analysis of the sedimentary basin fills has shown that the syndepositional faults and their arrangement styles exerted an important influ- ence on the development and distribution of the Eogene depositional systems. The sedimentary filling evolution of the subbasins reflects the general control of the episodic rifting process. The major long-term active normal faults formed a series of paleogeomorphic accident or slopebreak zones that commonly delineated the subsidiary palaeostructural units and the depositional facies tracts and constrained the general distribution of sedimentary facies zones. The central sag boundary fault slopebreak zones usually determined the distribution of the depocenters of ter- restrial clastic depositional systems, particularly the lowstand fans or deltaic depositional sys- tems, and have proven to be the economically important targets for the exploration of subtle sandstone reservoirs. A variety of syndepositional fault arrangement patterns, including the par- allel, en echelon, combo-like, broom-like fault systems and the fault overlap or transfer zones, have been recognized in the subbasins. They generated distinctive geomorphic features and exerted a direct influence on sediment dispersal and sandbody distribution during the Eogene synrift stage. | 5 | Sorafenib in treatment of patients with advanced hepatocellular carcinoma:a systematic review显示文摘BACKGROUND: Sorafenib has become the standard first-line treatment for patients with advanced hepatocellular carcinoma (HCC). This study aimed to assess the efficacy and safety of sorafenib in advanced HCC patients and explore its true value for specific subgroups. DATA SOURCES: A computer-based systematic search from January 2005 to June 2011 with 'sorafenib' and 'advanced hepatocellular carcinoma' as search terms was performed for possible clinical trials. Hazard ratios (HR) and their 95% confidence intervals (CI) for overall survival (OS) and time to progression (TTP), rates of partial response (PR), rates of toxicity effects, and details of subgroup analysis were extracted. Meta-analyses were done using the software Review Manager (version 5.0). RESULTS: Six trials with 1164 patients were included. Based on three randomized controlled trials, the pooled HR (sorafenib/ placebo) was 0.66 for OS (95% CI: 0.56-0.78; P<0.00001) and 0.57 for TTP (95% CI: 0.47-0.68; P<0.00001). The pooled odds ratio (OR) for PR was 2.96 (95% CI: 0.96-9.15; P=0.06). For three single-arm trials, the pooled HR was 0.69 for OS (95% CI: 0.56-0.84; P=0.0002) and 0.64 for TTP (95% CI: 0.52-0.78; P<0.00001). The pooled OR for PR in three single-arm trials was 3.56 (95% CI: 1.22-10.39; P=0.02). Subgroup analysis indicated that sorafenib was less effective in patients with extrahepatic spread (with: P=0.13 vs without: P<0.0001), with normal alpha-fetoprotein level (AFP) (P=0.15 vs elevated: P=0.0006), and with elevated level of serum bilirubin (P=0.06 vs normal: P=0.0009). Sorafenib-based therapy significantly increased the risk of grade 3/4 hand-foot skin reaction, diarrhea, fatigue, and rash/desquamation.CONCLUSIONS: Sorafenib-based therapy benefits advanced HCC patients. | 8 | Changes of Antioxidative Enzymes and Lipid Peroxidation in Leaves and Roots of Waterlogging-Tolerant and Waterlogging-Sensitive Maize Genotypes at Seedling Stage显示文摘To better understand the physiological and biochemical mechanisms of waterlogging tolerance, waterlogging effects on lipid peroxidation and the activity of antioxidative enzymes were investigated in leaves and roots of two maize genotypes, HZ32 (waterlogging-tolerant) and K12 (waterlogging-sensitive). Potted maize plants were waterlogged at the second leaf stage under glasshouse conditions. Leaves and roots were harvested 1 d before and 2, 4, 6, 8 and 10 d after the start of waterlogging treatment. Through comparing the activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), glutathione reductase (GR), catalase (CAT) and guaiacol peroxidase (POD) between waterlogging-tolerant and waterloggingsensitive genotype, we deduced that CAT was the most important H2O2 scavenging enzyme in leaves, while APX seemed to play a key role in roots. POD, APX, GR and CAT activities in conjunction with SOD seem to play an essential protective role in the O2-. and H2O2 scavenging process. Lipid peroxidation was enhanced significantly only in K12 (P < 0.001) and there was no difference (P > 0.05) in HZ32 up to 6 d after waterlogging stress. | 13 | Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. | 14 | Long-term outcomes and independent predictors of mortality in patients presenting to emergency departments with acute heart failure in Beijing:a multicenter cohort study with a 5-year follow-up显示文摘Background:Acute heart failure(AHF)is the most common disease in emergency departments(EDs).However,clinical data exploring the outcomes of patients presenting AHF in EDs are limited,especially the long-term outcomes.The purposes of this study were to describe the long-term outcomes of patients with AHF in the EDs and further analyze their prognostic factors.Methods:This prospective,multicenter,cohort study consecutively enrolled 3335 patients with AHF who were admitted to EDs of 14 hospitals from Beijing between January 1,2011 and September 23,2012.Kaplan-Meier and Cox regression analysis were adopted to evaluate 5-year outcomes and associated predictors.Results:The 5-year mortality and cardiovascular death rates were 55.4%and 49.6%,respectively.The median overall survival was 34 months.Independent predictors of 5-year mortality were patient age(hazard ratio[HR]:1.027,95 confidence interval[CI]:1.023-1.030),body mass index(BMI)(HR:0.971,95%CI:0.958-0.983),fatigue(HR:1.127,95%CI:1.009-1.258),ascites(HR:1.190,95%CI:1.057-1.340),hepatic jugular reflux(HR:1.339,95%CI:1.140-1.572),New York Heart Association(NYHA)class III to IV(HR:1.511,95%CI:1.291-1.769),heart rate(HR:1.003,95%CI:1.001-1.005),diastolic blood pressure(DBP)(HR:0.996,95%CI:0.993-0.999),blood urea nitrogen(BUN)(HR:1.014,95%CI:1.008-1.020),B-type natriuretic peptide(BNP)/N-terminal pro-B-type natriuretic peptide(NT-proBNP)level in the third(HR:1.426,95%CI:1.220-1.668)or fourth quartile(HR:1.437,95%CI:1.223-1.690),serum sodium(HR:0.980,95%CI:0.972-0.988),serum albumin(HR:0.981,95%CI:0.971-0.992),ischemic heart diseases(HR:1.195,95%CI:1.073-1.331),primary cardiomyopathy(HR:1.382,95%CI:1.183-1.614),diabetes(HR:1.118,95%CI:1.010-1.237),stroke(HR:1.252,95%CI:1.121-1.397),and the use of diuretics(HR:0.714,95%CI:0.626-0.814),(3-blockers(HR:0.673,95%CI:0.588-0.769),angiotensin-converting enzyme inhibitors(ACEIs)(HR:0.714,95%CI:0.604-0.845),angiotensin-II receptor blockers(ARBs)(HR:0.790,95%CI:0.646-0.965),spironolactone(HR:0.814,95%CI:0.663-0.999),calcium antagonists(HR:0.624,95%CI:0.531-0.733),nitrates(HR:0.715,95%CI:0.631-0.811),and digoxin(HR:0.579,95%CI:0.465-0.721).Conclusions:The results of our study demonstrate poor 5-year outcomes of patients presenting to EDs with AHF.Age,BMI,fatigue,ascites,hepatic jugular reflux,NYHA class III to IV,heart rate,DBP,BUN,BNP/NT-proBNP level in the third or fourth quartile,serum sodium,serum albumin,ischemic heart diseases,primary cardiomyopathy,diabetes,stroke,and the use of diuretics,P-blockers,ACEIs,ARBs,spironolactone,calcium antagonists,nitrates,and digoxin were independently associated with 5-year all-cause mortality. | Yang Li Xiao-Lu Sun Hong Qiu Jian Qin Chun-Sheng Li Xue-Zhong Yu Guo-Xing Wang Yan Fu Ya-An Zheng Bin Zhao Dong-Ming Yu Si-Jia Wang Guo-Gan Wang | 2021 | Chinese Medical Journal2021,,15: | 18 |

| 16 | Screening Methods for Waterlogging Tolerance at Maize (Zea mays L.) Seedling Stage显示文摘Waterlogging strongly affects agronomic performance of maize (Zea mays L.). In order to investigate the suitable selection criteria of waterflooding tolerant genotypes, and identify the most susceptible stage and the best continuous treatment time to waterlogging, 20 common maize inbred lines were subjected to successive artificial waterflooding at seedling stage, and waterlogging tolerance coefficient (WTC) was used to screen waterflooding tolerant genotypes. In addition, peroxidase (POD) activities and malondialdehyde (MDA) contents were measured for 6 of 20 lines. The results showed that the second leaf stage (V2) was the most susceptible stage, and 6 d after waterflooding was the best continuous treatment time. Dry weight (DW) of both shoots and roots of all lines were significantly reduced at 6 d time-point of waterlogging, compared to control. POD activities and MDA contents were negatively and significantly correlated, and the correlation coefficient was -0.9686 (P < 0.0001). According to the results, WTC of shoot DW can be used for practical screening as a suitable index, which is significantly different from control and waterlogged plants happened 6 d earlier. Furthermore, leaf chlorosis, MDA content and POD activities could also be used as reference index for material screening. | 19 | Prevalence of HIV Indeterminate Western Blot Tests and Follow-up of HIV Antibody Sero-Conversion in Southeastern China显示文摘HIV-indeterminate Western blotting(WB)results are typically obtained in WB confirmatory assays,and the number of indeterminate samples may increase with the detection of HIV infections,which will present considerable challenges for the management of HIV/AIDS.Nucleic acid detection has been used as a laboratory test for screening suspected or indeterminate samples.However,the effectiveness of these assays for the differential diagnosis of HIV-indeterminate WB samples remained undetermined.In this study,210 subjects with HIV-indeterminate WB results were detected from 6360 positive HIV screening samples between 2015 and 2016 in southeastern China,in which HIV-indeterminate WB results accounted for 3.30%.The highest proportion of indeterminate results was observed in pregnant and lying-in women receiving physical examinations(16.67%),followed by that in voluntary blood donors(8.82%).The most common WB band patterns were p24,gpl60 and p24,and gpl60.The follow-up study revealed that the highest negative and positive conversion rates of HIV antibodies were in samples with a single p24 band(80.28%),and with gpl60 and p24 bands(86.21%),respectively.Among the Env,Gag,and Pol antibodies,samples with a Gag band showed the highest negative conversion rate(81.25%),whereas the highest positive conversion rate was observed in samples with an Env band(56.76%).In addition,quantitative and qualitative HIV nucleic acid testing exhibited the highest sensitivity(96.3%)and specificity(97.85%),respectively.Our results indicate a lower proportion of HIV indeterminate WB results in southeastern China compared to previous reports,and the follow-up re-examination of patients with HIV indeterminate results should be performed.Nucleic acid testing facilitates the identification of HIV infections. | Shouli Wu Min Gao Jian Zheng Pingping Yan Wei Wang Xiaoli Lu Yuefeng Qiu Yansheng Yan | 2019 | Virologica Sinica2019,34,4: | 15 |
