1Endovascular therapy for Acute ischemic Stroke Trial (EAST): study protocol for a prospective, multicentre control trial in China显示文摘Background and purpose:5 recent trials have shown the benefit of endovascular treatment for acute ischaemic stroke(AIS)due to large vessel occlusion of the anterior circulation.This study aims to evaluate the safety and efficacy of Solitaire thrombectomy in patients with moderate-to-severe stroke in the Chinese population,which has a high prevalence of intracranial atherosclerosis.Methods and analysis:This multicentre prospective control study will involve 17 stroke centres in China,and plans to recruit 150 patients in the intervention group,and 150 patients in the medical group,in which patients meet enrolment criteria but refuse intervention.Patients with AIS due to large vessel occlusion indicated for treatment with Solitaire stent retriever within 12 hours of symptom onset,and who meet the inclusion and exclusion criteria,will be enrolled in this study.The primary efficacy endpoint is functional independence as defined by a modified Rankin Scale(mRS)score≤2 at 90 days or by functional improvement as defined by mRS,using shift analysis.The procedural efficacy endpoint is arterial recanalisation of the occluded target vessel measured by a modified Thrombolysis in Cerebral Infarction(mTICI)score equal or superior to 2b right following the use of the study device.The primary safety endpoint is symptomatic intracranial haemorrhage(sICH)within 24±3 hours postprocedure.Ethics and dissemination:The protocol was approved by the Ethics Committee at the coordinating centre and by the local Institutional Review Board of each participating centre.Trial registration number:NCT02350283.Zhongrong Miao Xiaochuan Huo Feng Gao Xiaoling Liao Chunjuan Wang Ya Peng Yibin Cao Shengli Chen Meng Zhang Changchun Jiang Xiaoxiang Peng Cunfeng Song Liping Wei Qiyi Zhu Zaiyu Guo Li Liu Hang Lin Hua Yang Wei Wu Hui Liang Anding Xu Kangning Chen Xingquan Zhao Yuesong Pan Hao Li Liping Liu Yilong Wang Yongjun Wang EAST investigators 2016Stroke & Vascular Neurology2016,1,2:8
2Microwave ablation of primary breast cancer inhibits metastatic progression in model mice via activation of natural killer cells显示文摘Surgery is essential for controlling the symptoms and complications of stage IV breast cancer.However,locoregional treatment of primary tumors often results in distant progression,including lung metastasis,the most common type of visceral metastasis.As a minimally invasive thermal therapy,microwave ablation(MWA)has been attempted in the treatment of breast cancer,but the innate immune response after MWA has not yet been reported.Using two murine models of stage IV breast cancer,we found that MWA of primary breast cancer inhibited the progression of lung metastasis and improved survival.NK cells were activated after MWA of the primary tumor and exhibited enhanced cytotoxic functions,and the cytotoxic pathways of NK cells were activated.Depletion experiments showed that NK cells but not CD4+or CD8+T cells played a pivotal role in prolonging survival.Then,we found that compared with surgery or control treatment,MWA of the primary tumor induced completely different NK-cell-related cytokine profiles.Macrophages were activated after MWA of the primary tumor and produced IL-15 that activated NK cells to inhibit the progression of metastasis.In addition,MWA of human breast cancer stimulated an autologous NK-cell response.These results demonstrate that MWA of the primary tumor in metastatic breast cancer inhibits metastatic progression via the macrophage/IL-15/NK-cell axis.MWA of the primary tumor may be a promising treatment strategy for de novo stage IV breast cancer,although further substantiation is essential for clinical testing.Muxin Yu Hong Pan Nan Che Li Li Cong Wang Yue Wang Ge Ma Mengjia Qian Jiawei Liu Mingjie Zheng Hui Xie Lijun Ling Yi Zhao Xiaoxiang Guan Qiang Ding Wenbin Zhou Shui Wang 2021Cellular & Molecular Immunology2021,18,9:5
3Development of an invasive brain machine interface with a monkey model显示文摘Brain-machine interfaces (BMIs) translate neural activities of the brain into specific instructions that can be carried out by external devices. BMIs have the potential to restore or augment motor functions of paralyzed patients suffering from spinal cord damage. The neural activities have been used to predict the 2D or 3D movement trajectory of monkey's arm or hand in many studies. However, there are few studies on decoding the wrist movement from neural activities in center-out paradigm. The present study developed an invasive BMI system with a monkey model using a 10×10-microelectrode array in the primary motor cortex. The monkey was trained to perform a two-dimensional forelimb wrist movement paradigm where neural activities and movement signals were simultaneous recorded. Results showed that neuronal firing rates highly correlated with forelimb wrist movement; > 70% (105/149) neurons exhibited specific firing changes during movement and > 36% (54/149) neurons were used to discriminate directional pairs. The neuronal firing rates were also used to predict the wrist moving directions and continuous trajectories of the forelimb wrist. The four directions could be classified with 96% accuracy using a support vector machine, and the correlation coefficients of trajectory prediction using a general regression neural network were above 0.8 for both horizontal and vertical directions. 5Decoding grasp movement from monkey premotor cortex for real-time prosthetic hand control显示文摘Brain machine interfaces (BMIs) have demonstrated lots of successful arm-related reach decoding in past decades, which provide a new hope for restoring the lost motor functions for the disabled. On the other hand, the more sophisticated hand grasp movement, which is more fundamental and crucial for daily life, was less referred. Current state of arts has specified some grasp related brain areas and offline decoding results; however, online decoding grasp movement and real-time neuroprosthetic control have not been systematically investigated. In this study, we obtained neural data from the dorsal premotor cortex (PMd) when monkey reaching and grasping one of four differently shaped objects following visual cues. The four grasp gesture types with an additional resting state were classified asynchronously using a fuzzy k-nearest neighbor model, and an artificial hand was controlled online using a shared control strategy. The results showed that most of the neurons in PMd are tuned by reach and grasp movement, us- ing which we get a high average offline decoding accuracy of 97.1%. In the online demonstration, the instantaneous status of monkey grasping could be extracted successfully to control the artificial hand, with an event-wise accuracy of 85.1%. 8Selection and identification of a novel ssDNA aptamer targeting human skeletal muscle显示文摘Skeletal muscle disorders have posed great threats to health.Selective delivery of drugs and oligonucleotides to skeletal muscle is challenging.Aptamers can improve targeting efficacy.In this study,for the first time,the human skeletal muscle-specific ssDNA aptamers(HSM01,etc.)were selected and identified with Systematic Evolution of Ligands by Exponential Enrichment(SELEX).The HSM01 ssDNA aptamer preferentially interacted with human skeletal muscle cells in vitro.The in vivo study using tree shrews showed that the HSM01 ssDNA aptamer specifically targeted human skeletal muscle cells.Furthermore,the ability of HSM01 ssDNA aptamer to target skeletal muscle cells was not affected by the formation of a disulfide bond with nanoliposomes in vitro or in vivo,suggesting a potential new approach for targeted drug delivery to skeletal muscles via liposomes.Therefore,this newly identified ssDNA aptamer and nanoliposome modification could be used for the treatment of human skeletal muscle diseases.Shuming Sun Han Liu Yan Hu Yanpeng Wang Mingri Zhao Yijun Yuan Yafei Han Yingying Jing Jin Cui Xiaoxiang Ren Xiao Chen Jiacan Su 2023Bioactive Materials2023,,2:3
9Health benefits of Grifola frondosa polysaccharide on intestinal microbiota in type 2 diabetic mice显示文摘Grifola frondosa polysaccharide(GFP)as the natural compounds have been reported to exert diverse bioactivities.The aim of this study was to investigate the regulatory effects of GFP on intestinal microbiota in type 2 diabetic mice.The changes of microbiota in faeces of the diabetic mice upon high fat diet were determined and the V3 region of the 16S rRNA was sequenced by Illumina MiSeq high-throughput sequencing platform.Eighty operational taxonomic unit were identified to be shared by all samples,and the quality and richness of sequencing were assessed via rarefaction and rank abundance curves.The diversity of gut flora was slightly improved in diabetic mice after GFP treatment.The composition and relative abundance of gut microbiota at phylum and genus levels were altered by GFP.The abundance of Robinsoniella,Flavonifractor,Anaerotruncus,and Desulfvibrio were found to concentrate on diabetic mice through LEfSe analysis.Furthermore,Alloprevotella and Burkholderia had strong relevancy with other gut flora.Based on the findings,GFP might be a desired candidate on ameliorating the intestinal unbalance in diabetes.Xiaoxiang Gao Dan Liu Luying Gao Yuezhen Ouyang Yuxi Wen Chao Ai Yuqing Chen Chao Zhao 2022Food Science and Human Wellness2022,11,1:2

16Effect of Phase Structure Evolution on Thermal Expansion and Toughness of (Nd1-xScx)2Zr2O7 (x = 0, 0.025, 0.05, 0.075, 0.1 ) Ceramics显示文摘Nd_2Zr_2O_7 is being explored as an alternate thermal barrier coating(TBC) material for operating above1300 °C, and great effort has been made to enlarge its thermal expansion coefficient(TEC) and improve its toughness. In this study, Sc_2O_3 was doped to Nd_2Zr_2O_7. The phase structure, TECs and toughness of((Nd_(1-x)Scx))_2Zr_2_O7(x= 0, 0.025, 0.05, 0.075, 0.1) compounds were investigated.((Nd_(1-x)Scx))_2Zr_2_O7(x = 0, 0.025,0.05, 0.075) exhibited pyrochlore structure, while((Nd_(0.9)Sc_(0.1)))_2Zr_2O_7 consisted of pyrochlore and fluorite phases. With the increase of the Sc_2O_3 content, the ordering degree of the pyrochlore in the compounds decreased. In((Nd_(1-x)Scx))_2Zr_2O_7 series,((Nd_(0.925)Sc_(0.075)))_2Zr_2O_7 exhibited the largest TEC, while the toughness of the compounds increased with increasing the Sc_2O_3 content. The related mechanisms were discussed based on the crystal structure analysis. 18Expression of recombinant human butyrylcholinesterase in the milk of transgenic mice显示文摘Butyrylcholinesterase(BCHE)is a natural bioscavenger that protects humans against organophosphate toxicity.Due to the limited yield of human BCHE(hBCHE)when purifying from human plasma,it is necessary to find an alternative method to produce this protein.One potential method is to produce transgenic livestock that make modified milk containing high concentration of hBCHE.In this study,we cloned the hBCHEgene into a human lactoferrin(hLF)bacterial artificial chromosome(BAC)construct to make a hLFhBCHE BAC construct.Subsequently,we injected the BAC construct into pronuclei of mouse fertilized embryos and generated transgenic mice.Expression analysis showed that recombinant hBCHE(rhBCHE)was expressed efficiently in the mammary gland of the transgenic mice and the concentration of rhBCHE in the milk of individual mice ranged from 7612 to 15928 mg·L^(–1).Protein function tests showed that rhBCHE has the same enzymatic activity as the native hBCHE.Our results pave the way for making transgenic livestock to produce large quantities of rhBCHE.Dan LU Shengzhe SHANG Shen LIU Ying WU Fangfang WU Tan TAN Qiuyan LI Yunping DAI Xiaoxiang HU Yaofeng ZHAO Ning LI 2014Frontiers of Agricultural Science and Engineering2014,1,3:0
19Depletion of conventional mature B cells and compromised specific antibody response in bovine immunoglobulin μ heavy-chain transgenic mice显示文摘In this study,we introduced the bovine immunoglobulinμheavy-chain gene(the orphaned gene on BTA11)into mouse germline cells.Bovine IgM was highly expressed in selected transgenic lines,and it largely inhibited rearrangements of the endogenous immunoglobulin heavy chain(IgH)genes in these lines.The forced expression of bovine IgM resulted in reduced numbers of pro-and pre-B cells but increased the number of immature B cells in the transgenic mice.Bovine IgM-expressing B cells can migrate from the bone marrow to the spleen,but most of the cells are arrested at the T1 transitional B cell stage,leading to a significantly lower number of T2 transitional and mature B cells in the spleen.Although the serum concentrations of endogenous IgM and IgG in the transgenic mice were significantly decreased,the IgA levels were slightly increased compared to the WT mice.The bovine IgM level in the serum was only one-tenth to one-fifth of that of endogenous mouse IgM,suggesting that most of the serum immunoglobulin were contributed by endogenous IgH gene-expressing B cells.These transgenic mice also exhibited a lower frequency of unique complementarity determining region 3(CDR3)sequences in their VH repertoire and Vκrepertoire but exhibited an increased frequency of unique CDR3 in their Vλrepertoire.Compared to the WT mice,the transgenic mice had a significantly higher percentage of mouse IgMexpressing B cells that expressedλchains.Finally,we showed that the transgenic mice were deficient in a specific antibody response to antigen stimulation.Min ZHANG Xueqian CHENG Dan CHU Jingwen LIANG Yi SUN Li MA Beilei XU Min ZHENG Meili WANG Liming REN Xiaoxiang HU Qingyong MENG Ran ZHANG Ying GUO Yunping DAI Robert AITKEN Ning LI Yaofeng ZHAO 2014Frontiers of Agricultural Science and Engineering2014,1,2:0
20Eff ects of stand condition and root density on fi ne-root dynamics across root functional groups in a subtropical montane forest显示文摘Fine roots play key roles in belowground C cycling in terrestrial ecosystems.Based on their distinct functions,fi ne roots are either absorptive fi ne roots(AFRs)or transport fi ne roots(TFRs).However,the function-based fi ne root dynamics of trees and their responses to forest stand properties remain unclear.Here,we studied the dynamics of AFRs and TFRs and their responses to stand conditions and root density in a subtropical montane mixed forest based on a 2-a root window experiment.Mean(±SE)annual production,mortality,and turnover rate of AFRs were 7.87±0.17 m m^(−2)a^(−1),8.13±0.20 m m^(−2)a^(−1)and 2.96±0.24 a^(−1),respectively,compared with 7.09±0.17 m m^(−2)a^(−1),4.59±0.17 m m^(−2)a^(−1),and 2.01±0.22 a^(−1),respectively,for TFRs.The production and mortality of fi ne roots were signifi cantly higher in high root-density sites than in low-root density sites,whereas the turnover of fi ne roots was faster in the low root-density sites.Furthermore,root density had a larger positive eff ect than other environmental factors on TFR production but had no obvious impact on AFR production.Tree species diversity had an apparent positive eff ect on AFR production and was the crucial driver of AFR production,probably due to a complementary eff ect,but had no evident impact on TFR.Both tree density and tree species diversity were positively correlated with the mortality of AFRs and negatively related to the turnover of TFRs,suggesting that higher root density caused stronger competition for rooting space and that plants tend to reduce maintenance costs by decreasing TFR turnover.These fi ndings illustrated the importance of root functional groups in understanding root dynamics and their responses to changes in environmental conditions.Lin Huang Rudong Zhao Xiaoxiang Zhao Qiuxiang Tian Pengyun Yue Feng Liu 2023Journal of Forestry Research2023,34,3:0
