1The Chrysanthemum nankingense Genome Provides Insights into the Evolution and Diversification of Chrysanthemum Flowers and Medicinal Traits显示文摘The Asteraceae (Compositae),a large plant family of approximately 24 000-35 000 species,accounts for^10% of all angiosperm species and contributes a lot to plant diversity.The most representative members of the Asteraceae are the economically important chrysanthemums (Chrysanthemum L.)that diversified through reticulate evolution.Biodiversity is typically created by multiple evolutionary mechanisms such as wholegenome duplication 0NGD)or polyploidization and locally repetitive genome expansion.However,the lack of genomic data from chrysanthemum species has prevented an in-depth analysis of the evolutionary mechanisms involved in their diversification.Here,we used Oxford Nanopore long-read technologyto sequence the diploid Chrysanthemum nankingense genome,which represents one of the progenitor genomes of domesticated chrysanthemums.Our analysis revealed that the evolution of the C.nankingense genome was driven by bursts of repetitive element expansion and WGD events including a recentWGD that distinguishes chrysanthemum from sunflower,which diverged from chrysanthemum approximately 38.8 million years ago.Variations of ornamental and medicinal traits in chrysanthemums are linked to the expansion of candidate gene families by duplication events including paralogous gene duplication.Collectively,our study of the assembled reference genome offers new knowledge and resources to dissect the history and pattern of evolution and diversification of chrysanthemum plants,and also to accelerate their breeding and improvement.Chi Song Yifei Liu Aiping Song Gangqiang Dong Hongbo Zhao Wei Sun Shyam Ramakrishnan Ying Wang Shuaibin Wang Tingzhao Li Yan Niu Jiafu Jiang Bin Dong Ye Xia Sumei Chen Zhigang Hu Fadi Chen Shilin Chen 2018Molecular Plant2018,11,12:24

3Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 5Plant HP1 protein ADCP1 links multivalent H3K9 methylation readout to heterochromatin formation显示文摘Heterochromatin Protein 1 (HP1) recognizes histone H3 lysine 9 methylation (H3K9me) through its conserved chromodomain and maintains heterochromatin from fission yeast to mammals. However, in Arabidopsis, Like Heterochromatin Protein 1 (LHP1) recognizes and colocalizes genome-wide with H3K27me3, and is the functional homolog of Polycomb protein. This raises the question whether genuine HP1 homologs exist in plants. Here, we report on the discovery of ADCP1, a plant-specific triple tandem Agenet protein, as a multivalent H3K9me reader in Arabidopsis, and establish that ADCP1 is essential for heterochromatin formation and transposon silencing through modulating H3K9 and DNA methylation levels. Structural studies revealed the molecular basis underlying H3K9me-specific recognition by tandem Agenet of ADCP1. Similar to human HP1α and fly HP1a, ADCP1 mediates heterochromatin phase separation. 6The High Energy X-ray telescope (HE) onboard the Insight-HXMT astronomy satellite显示文摘The Insight-Hard X-ray Modulation Telescope(Insight-HXMT) is a broadband X-ray and γ-ray(1-3000 ke V) astronomy satellite. One of its three main telescopes is the High Energy X-ray telescope(HE). The main detector plane of HE comprises 18 Na I(Tl)/Cs I(Na) phoswich detectors, where Na I(Tl) is used as the primary detector to measure ~ 20-250 ke V photons incident from the field of view(FOV) defined by collimators, and Cs I(Na) is used as the active shielding detector to Na I(Tl) by pulse shape discrimination. Additionally, Cs I(Na) is used as an omnidirectional γ-ray monitor. The HE collimators have a diverse FOV,i.e. 1.1°×5.7°(15 units), 5.7°×5.7°(2 units), and blocked(1 unit). Therefore, the combined FOV of HE is approximately5.7°×5.7°. Each HE detector has a diameter of 190 mm resulting in a total geometrical area of approximately 5100 cm2, and the energy resolution is ~15% at 60 ke V. For each recorded X-ray event by HE, the timing accuracy is less than 10 μs and the deadtime is less than 10 μs. HE is used for observing spectra and temporal variability of X-ray sources in the 20-250 ke V band either by pointing observations for known sources or scanning observations to unveil new sources. Additionally, HE is used for monitoring the γ-ray burst in 0.2-3 Me V band. 7Energy-efficient power allocation for non-regenerative OFDM relay links显示文摘We consider a two-hop non-regenerative (amplify-and-forward) relay link with one source, one destination and one relay, in which orthogonal frequency division multiplexing (OFDM) is used. The power allocation (PA) to maximize the energy efficiency for the whole link is investigated under quality of service (Qos) require- ment and separate power constraints. An approximation method with low complexity is first proposed to convert the PA problem into a quasi-concave problem, which can obtain a tight lower bound in high SNR region. Furthermore, a more accurate approximation method with higher complexity is raised, which can approach the actual performance both in the low SNR and high SNR regions. We also introduce subcarrier pairing scheme to further enhance the energy efficiency for the link. 8Productive HBV infection of well-differentiated, hNTCP-expressing human hepatoma-derived(Huh7) cells显示文摘Feasible and effective cell models for hepatitis B virus(HBV) infection are required for investigating the complete lifecycle of this virus, including the early steps of viral entry. Resistance to dimethyl sulfoxide/polyethylene glycol(DMSO/PEG), h NTCP expression, and a differentiated state are the limiting factors for successful HBV infection models. In the present study, we used a hepatoma cell line(Hu7^(hDNTCPh)) to overcome these limiting factors so that it exhibits excellent susceptibility to HBV infection. To achieve this goal, different hepatoma cell lines were tested with 2.5% DMSO/4%PEG8000, and one resistant cell line(Huh7 D) was used to construct a stable h NTCP-expressing cell line(Hu7^(hDNTCPh)) using a recombinant lentivirus system. Then, the morphological characteristics and differentiation molecular markers of Hu7^(hDNTCPh) cells with or without DMSO treatment were characterized. Finally, the susceptibility of Hu7^(hDNTCPh) cells to HBV infection was assessed. Our results showed that Huh7 D cells were resistant to 2.5% DMSO/4% PEG8000, whereas the others were not. Hu7^(hDNTCPh) cells were established to express a high level of h NTCP compared to liver extracts, and Hu7^(hDNTCPh) cells rapidly transformed into a non-dividing, well-differentiated polarized phenotype under DMSO treatment. Hu7^(hDNTCPh) cells fully supported the entire lifecycle of HBV infection. 10Donor-Derived CD19-Targeted T Cell Infusion Eliminates B Cell Acute Lymphoblastic Leukemia Minimal Residual Disease with No Response to Donor Lymphocytes after Allogeneic Hematopoietic Stem Cell Transplantation显示文摘Leukemia relapse is still the leading cause of treatment failure after allogeneic hematopoietic stem cell transplantation (allo-HSCT) for B cell acute lymphoblastic leukemia (B-ALL). Relapsed patients with BALL after allo-HSCT have a very short median survival. Minimal residual disease (MRD) is predictive of forthcoming hematological relapse after hematopoietic stem cell transplantation (HSCT);furthermore, eliminating MRD effectively prevents relapse. Donor lymphoblastic infusion (DLI) is the main established approach to treat B-ALL with MRD after allo-HSCT. However, about one-third of patients with MRD are non-responsive to DLI and their prognosis worsens. Although donor-derived cluster of differentiation (CD)19-directed chimeric antigen receptor-modified (CAR) T cells (CART19s) can potentially cure leukemia, the efficiency and safety of infusions with these cells have not yet been investigated in patients with MRD after HSCT. Between September 2014 and February 2018, six patients each received one or more infusions of CART19s from HSCT donors. Five (83.33%) achieved MRD-negative remission, and one case was not responsive to the administration of CAR T cells. Three of the six patients are currently alive without leukemia. No patient developed acute graft-versus-host disease (aGVHD), and no patient died of cytokine release syndrome. 125G: Vision, Scenarios and Enabling Technologies显示文摘This paper presents the authors' vision for 5G wireless systems, which are expected to be standardized around 2020(IMT-2020).In the future, ubiquitous service will be the key requirement from an end-user's prospective, and 5G networks will need to support a vast mesh of human-to-human, human-to-machine, and machine-to-machine connections. Moreover, 5G will need to support these connections in an energy-efficient manner. Various 5G enabling technologies have been extensively discussed. These technologies aim to increase radio link efficiency, expand operating bandwidths, and increase cell density. With these technologies,5G systems can accommodate a massive volume of traffic and a massive number of connections, which is fundamental to providing ubiquitous services. 13Development and prospect of direct‐current circuit breaker in China显示文摘The direct‐current circuit breaker(DCCB)is the most ideal choice for DC fault isolation in DC grids.Despite a late start,China's research and development on the DCCB have made outstanding achievements.This article provides a brief glance of current China's DCCB development status.It begins by sorting out the technical route according to the topology of DCCB.Then it systematically summarises both mechanical and hybrid DCCBs with focussing on the aspects of topology structure and principle,key technology and characteristics,prototype development and application.It is apparent that Chinese scientists and engineers confronted the worldwide problem of large capacity DC breaking,and put forward a comprehensive solution which consists of an innovative topology structure based on coupled negative voltage circuit,breaking throughs on the key technologies such as highly controllable and reliable fault current commutation,millisecond‐level ultra‐fast and efficient electromagnetic repulsion mechanism,high tolerance and high stability power electronic switch,low residual voltage and fast response energy consumption device,etc.The article states that the world's first set of hybrid high‐voltage(HV)DCCB,and the first set of mechanical HV DCCB have been developed.These DCCBs will soon be deployed to the DC grids which have the highest voltage levels therefore require the strongest breaking capacity.These achievements are leading the world in the development and application of DCCB.The article also discusses the overall development trends of DCCB in the areas of new topologies,key techno-logical breakthroughs and application scenarios,etc.These discussions serve as references for DCCB's future technological advancement and its ever‐expanding applications.Weijiang Chen Rong Zeng Junjia He Yi Wu Xiaoguang Wei Taixun Fang Zhanqing Yu Zhao Yuan Yifei Wu Wandi Zhou Bing Yang Lu Qu 2021High Voltage2021,6,1:6
14Cooperative coalition for formation flight scheduling based on incomplete information显示文摘This study analyzes the cooperative coalition problem for formation scheduling based on incomplete information. A multi-agent cooperative coalition framework is developed to optimize the formation scheduling problem in a decentralized manner. The social class differentiation mechanism and role-assuming mechanism are incorporated into the framework, which, in turn, ensures that the multi-agent system(MAS) evolves in the optimal direction. Moreover, a further differentiation pressure can be achieved to help MAS escape from local optima. A Bayesian coalition negotiation algorithm is constructed, within which the Harsanyi transformation is introduced to transform the coalition problem based on incomplete information to the Bayesian-equivalent coalition problem based on imperfect information. The simulation results suggest that the distribution of agents' expectations of other agents' unknown information approximates to the true distribution after a finite set of generations. 15Comparison of lymph node number and prognosis in gastric cancer patients with perigastric lymph nodes retrieved by surgeons and pathologists显示文摘Objective: To compare the numbers of positive and total lymph nodes and prognosis in gastric cancer patients whose perigastric lymph node retrieval was performed by surgeons and pathologists.Methods: We conducted a retrospective analysis of clinical and follow-up data from 1,056 patients who underwent gastric cancer D2 radical lymph node resection between January 2008 and December 2010 in the Gastrointestinal Surgery Department of Yantai Yuhuangding Hospital.The follow-up ended in December 2015.Patients were divided into two groups according to the specialty of physicians who performed the postoperative perigastric lymph node retrieval: the surgeon group(475 cases) and the pathologist group(581 cases).The numbers of positive and total perigastric lymph nodes and the 3- and 5-year survival were compared between gastric cancer patients in the two groups overall and stratified by TNM stage(AJCC 7th Edition).Results: Overall, the numbers of positive and total lymph nodes were significantly higher in the surgeon group than in the pathologist group(6.53±4.07 vs.4.09±3.70, P=0.021; 29.64±11.50 vs.20.71±8.56, P<0.001).Further analysis showed that the total number of lymph nodes in stage Ⅰ patients(19.40±9.62 vs.15.45±8.59, P=0.011) and the numbers of positive and total lymph nodes in stage Ⅱ(1.38±1.08 vs.0.87±1.55, P=0.031; 25.35±10.80 vs.16.75±8.56, P<0.001) and stage Ⅲ patients(8.11±6.91 vs.6.66±5.12, P=0.026; 32.34±12.55 vs.25.45±8.31, P<0.001)were significantly higher in the surgeon group than in the pathologist group.The survival analysis showed that the3- and 5-year survival of stage Ⅱ and Ⅲ patients was significantly higher in the surgeon group than in the pathologist group(82.0% vs.73.1%, 69.5% vs.61.2%, P=0.038; 49.2% vs.38.9%, 36.3% vs.28.0%; P=0.045).Conclusions: Compared with retrieval performed by pathologists, postoperative perigastric lymph node retrieval performed by surgeons was associated with significant increase in the total lymph node number of stage Ⅰ patients, the numbers of positive and total lymph nodes of stage Ⅱ and Ⅲ patients, and the survival of stage Ⅱ and stage Ⅲ gastric cancer patients.Lixin Jiang Zengwu Yao Yifei Zhang Jinchen Hu Dawei Zhao Huiyuan Zhai Xixun Wang Zhenbin Zhang Dong Wang 2016Chinese Journal of Cancer Research2016,28,5:5
16Clusterin in Alzheimer's disease: a player in the biological behavior of amyloid-beta显示文摘Alzheimer's disease (AD) remains a major killer, and although its pathogenesis varies, one dominant feature is an increase in the expression, formation, and sedimentation of senile plaques of amyloid-beta (Aβ) peptides in the brain. The chaperone protein clusterin has, since its first discovery at the end of the 20th century, been labeled as a cytoprotector. However, epigenetic studies showing that clusterin is associated with the severity and risk of AD, especially in the hippocampus, triggered studies to clarify its role in the pathogenesis of AD. It is true that clusterin can inhibit the aggregation of Aβ and therefore prevent further formation of senile plaques in the AD brain, yet it induces the formation of soluble forms of Aβ which are toxic to neurons. Another problematic finding is that clusterin is involved in a pathway through which Aβ has neurodegenerative effects intracellularly. 17Modeling of oil near-infrared spectroscopy based on similarity and transfer learning algorithm显示文摘Near-infrared spectroscopy mainly reflects the frequency-doubled and total-frequency absorption information of hydrogen-containing groups (0 -H, C-H, N-H, S-H) in organic molecules for near-infrared lights with different wavelengths, so it is applicable to testing of most raw materials and products in the field of petrochemicals. However, the modeling process needs to collect a large number of laboratory analysis data. There are many oil sources in China, and oil properties change frequently. Modeling of each raw material is not only unfeasible but also will affect its engineering application efficiency. In order to achieve rapid modeling of near-infrared spectroscopy and based on historical data of different crude oils under different detection conditions, this paper discusses about the feasibility of the application of transfer learning algorithm and makes it possible that transfer learning can assist in rapid modeling using certain historical data under similar distributions under a small quantity of new data. In consideration of the requirement of transfer learning for certain similarity of different datasets, a transfer learning method based on local similarity feature selection is proposed. The simulation verification of spectral data of 13 crude oils measured by three different probe detection methods is performed. 19Complementation of Wild-Type and Drug-Resistant Hepatitis B Virus Genomes to Maintain Viral Replication and Rescue Virion Production under Nucleos(t)ide Analogs显示文摘As the open reading frames of hepatitis B virus(HBV)genomes are overlapping,resistance mutations(MTs)in HBV polymerase may result in stop codon MTs in hepatitis B surface proteins,which are usually detected as a mixed population with wild-type(WT)HBV.The question was raised how the coexistence of nucleos(t)ide analogs(NAs)resistance MTs and WT sequences affects HBV replication.In the present study,HBV genomes with frequently detected reverse transcriptase(RT)/surface truncation MTs,rtA181T/sW172^*,rtV191I/sW182^*and rtM204I/sW196^*,were phenotypically characterized alone or together with their WT counterparts in different ratios by transient transfection in the absence or presence of Nas.In the absence of Nas,RT/surface truncation MTs impaired the expression and secretion of HBV surface proteins,and had a dose-dependent negative effect on WT HBV virion secretion.However,in the presence of Nas,coexistence of MTs with WT maintained viral replication,and the presence of WT was able to rescue the production of MT HBV virions.Our findings reveal that complementation of WT and MT HBV genomes is highly effective under drug treatment.Chunchen Wu Baolin Li Xiaoyong Zhang Kaitao Zhao Yingshan Chen Yifei Yuan Yan Liu Rongjuan Chen Dongping Xu Xinwen Chen Mengji Lu 2019Virologica Sinica2019,34,4:4
20Sustained‑Release Nanocapsules Enable Long‑Lasting Stabilization of Li Anode for Practical Li‑Metal Batteries显示文摘A robust solid-electrolyte interphase(SEI)enabled by electrolyte additive is a promising approach to stabilize Li anode and improve Li cycling efficiency.However,the self-sacrificial nature of SEI forming additives limits their capability to stabilize Li anode for long-term cycling.Herein,we demonstrate nanocapsules made from metal–organic frameworks for sustained release of LiNO3 as surface passivation additive in commercial carbonate-based electrolyte.The nanocapsules can offer over 10 times more LiNO3 than the solubility of LiNO3.Continuous supply of LiNO3 by nanocapsules forms a nitride-rich SEI layer on Li anode and persistently remedies SEI during prolonged cycling.As a result,lifespan of thin Li anode in 50μm,which experiences drastic volume change and repeated SEI formation during cycling,has been notably improved.By pairing with an industry-level thick LiCoO2 cathode,practical Li-metal full cell demonstrates a remarkable capacity retention of 90%after 240 cycles,in contrast to fast capacity drop after 60 cycles in LiNO3 saturated electrolyte.Qianqian Liu Yifei Xu Jianghao Wang Bo Zhao Zijian Li Hao Bin Wu 2020Nano-Micro Letters2020,12,12:4
