Multiple genetic alterations and behavior of cellular biology in gastric cancer and other gastric mucosal lesions:H.pylori infection, histological types and staging显示文摘AIM To investigate the expression of multiplegenes and the behavior of cellular biology ingastric cancer(GC)and other gastric mucosallesions and their relations to Helicobacter pylori(H.pylori)infection, tumor staging andhistological subtypes.METHODS Three hundred and twenty-sevenspecimens of gastric mucosa obtained viaendoscopy or surgical resection, and ABCimmunohistochemical staining were used todetect the expression of p53,p16,Bcl-2 andCOX-2 proteins.H.pylori was determined byrapid urea test combined with pathologicalstaining or14C urea breath test.Cellular image analysis was performed in 66 patients withintestinal metaplasia(IM)and/or dysplasia(Dys). In 30 of them, both cancer and theparacancerous tissues were obtained at the timeof surgery.Histological pattern, tumor staging, lymph node metastasis, grading ofdifferentiation and other clinical data werestudied in the medical records.RESULTS p16 expression of IM or Dys wassignificantly lower in positive H.pylori chronicatrophic gastritis(CAG)than those withnegative H.pylori(CAG:54.8% vs 88.0%,IM:34.4% vs 69.6%,Dys:23.8% vs 53.6%,allP<0.05),Bcl-2 or COX-2 expression of IM orDys in positive H.pylori cases was significantlyhigher than that without H.pylori(Bcl-2:68.8%vs23.9%,90.5% vs 60.7%;COX-2:50.0% vs10.8%,61.8% vs 17.8%;all P<0.05).Themean number of most parameters of cellularimage analysis in positive H.pylori group wassignificantly higher than that in negative H.pylori group(Ellipser:53±14,40±12μm,Area1:748±572,302±202 μm2,Area2:3050±1661,1681±1990 μm2,all P<0.05;Ellipseb:79±23,58±15 μm,Ratio1:22%±5%,13%±4%,Ratio2:79%±17%,53%±20%,all P<0.01).There was significant correlation between Bcl-2and histologic pattern of gastric carcinoma, andbetween COX-2 and tumor staging or lymph nodemetastasis(Bcl-2:75.0% vs 16.7%;COX-2:76.0% vs 20.0%,79.2% vs 16.7%;allP<0.05).CONCLUSION p1l6, Bcl-2, and COX-2 but not p53 gene may play a role in the early genesis/ progression of gastric carcinoma and are associated with H. pylori infection. p53 gene is relatively late event in gastric tumorigenesis and mainly relates to its progression. There is more cellular-biological behavior of malignant tumor in gastric mucosal lesions with H. pylori infection. Aberrant Bcl-2 protein expression appears to be preferentially associated with the intestinal type cancer. COX-2 seems to be related to tumor staging and lymph node metastasis. | Heng Jun Gao Lian Zhen Yu Jian Feng Bai Yan Shen Peng Gu Sun Han Lin Zhao Kun Miu Xiu Zhen Lü Xiao Yong Zhang Zhi Quan Zhao | 2000 | World Journal of Gastroenterology2000,6,6: | 52
Combined detection tumor markers for diagnosis and prognosis of gallbladder cancer显示文摘AIM:To clarify the value of combined use of markers for the diagnosis of gallbladder cancer and prediction of its prognosis.METHODS:Serum cancer antigens(CA)199,CA242,carcinoembryonic antigen(CEA),and CA125 levels were measured in 78 patients with gallbladder cancer(GBC),78 patients with benign gallbladder diseases, and 78 healthy controls using electrochemiluminescence.CA199,CA242,CEA,and CA125 levels and positive rates were analyzed and evaluated pre-and postoperatively.Receiver operator characteristic curves were used to determine diagnostic sensitivity and specificity of GBC.Survival time analysis, including survival curves, and multivariate survival analysis of a Cox proportional hazards model was performed to evaluate independent prognostic factors.RESULTS:Serum CA242,CA125,and CA199 levels in the GBC group were significantly higher when compared with those in the benign gallbladder disease and healthy control groups(P<0.01).With a single tumor marker for GBC diagnosis, the sensitivity of CA199 was the highest(71.7%),with the highest specificity being in CA242(98.7%).Diagnostic accuracy was highest with a combination of CA199,CA242,and CA125(69.2%).CA242 could be regarded as a tumor marker of GBC infiltration in the early stage. The sensitivity of CA199 and CA242 increased with progression of GBC and advanced lymph node metastasis(P<0.05).The78 GBC patients were followed up for 6-12 mo(mean:8 mo),during which time serum CA199,CA125,and CA242 levels in the recurrence group were significantly higher than in patients without recurrence(P<0.01).The post-operative serum CA199,CA125,and CA242levels in the non-recurrence group were significantly lower than those in the GBC group(P<0.01).Multivariate survival analysis using a Cox proportional hazards model showed that cancer of the gallbladder neck and CA199 expression level were independent prognostic factors.CONCLUSION:CA242 is a marker of GBC infiltration in the early stage.CA199 and cancer of the gallbladder neck are therapeutic and prognostic markers. | Yun-Feng Wang Fei-Ling Feng Xu-Hong Zhao Zhen-Xiong Ye He-Ping Zeng Zhen Li Xiao-Qing Jiang Zhi-Hai Peng | 2014 | World Journal of Gastroenterology2014,20,14: | 37
Relationship between Cognition Function and Hippocampus Structure after Long-term Microwave Exposure显示文摘Objective To analyze the effects of long-term microwave exposure on hippocampal structure and function in the rat. Methods Experiments were performed on 184 male Wistar rats(three exposure groups and a sham group).Microwaves were applied daily for 6 min over 1 month at average power densities of 2.5,5,and 10 mW/cm2.Learning and memory abilities were assessed by Morris water maze.High performance liquid chromatography was used to detect neurotransmitter concentrations in the hippocampus.Hippocampal structures were observed by histopathological analysis. Results Following long-term microwave exposure there was a significant decrease in learning and memory activity in the 7 d,14 d,and 1 m in all three microwave exposure groups.Neurotransmitter concentrations of four amino acids(glutamate, aspartic acid, glycine, and gamma-aminobutyric acid) in hippocampus were increased in the 2.5 and 5 mW/cm2 groups and decreased in the 10 mW/cm2 group. There was evidence of neuronal degeneration and enlarged perivascular spaces in the hippocampus in the microwave exposure groups.Further, mitochondria became swollen and cristae were disordered. The rough endoplasmic reticulum exhibited sacculated distension and there was a decrease in the quantity of synaptic vesicles. Conclusion These data suggest that the hippocampus can be injured by long-term microwave exposure, which might result in impairment of cognitive function due to neurotransmitter disruption. | ZHAO Li PENG Rui Yun WANG Shui Ming WANG Li Feng GAO Ya Bing DONG Ji LI Xiang SU Zhen Tao | 2012 | Biomedical and Environmental Sciences2012,25,2: | 27

Comparison of Undernutrition Prevalence of Children under 5 Years in China between 2002 and 2013显示文摘Objective To describe the undernutrition status of children under 5-year in China, and study the trend between 2002 and 2013. Methods The study was based on two national surveys. Undernutrition was determined against WHO's 2006 growth standards. The prevalence in 2013 and 2002 was weighted by China sixth National Population Census(2010). The relationship between undernutrition and gender/age groups/different areas use weighted logistic regression. Results The results indicated the overall prevalence of stunting, underweight, and wasting of Chinese children under 5-year was 8.1%, 2.4%, and 1.9% in 2013, respectively. The prevalence of stunting was higher for children aged 12-47 month, while underweight was higher for children aged 48-59 month. The prevalence of undernutrition was higher in rural areas than in urban areas, especially in poor rural areas. There was a decline of stunting, underweight, and wasting between 2002 and 2013 among the children, with greater reduction in rural areas than in urban areas. Conclusion The prevalence of undernutrition of children under 5-year remains high in rural areas especially in poor rural areas in China. It is urgent to take action to control undernutrition in the vulnerable areas and subgroups. | YU Dong Mei ZHAO Li Yun YANG Zhen Yu CHANG Su Ying YU Wen Tao FANG Hong Yun WANG Xun YU Dan GUO Qi Ya XU Xiao Li FANG Yue Hui ZHAO Wen Hua YANG Xiao Guang DING Gang Qiang LIANG Xiao Feng | 2016 | Biomedical and Environmental Sciences2016,29,3: | 18
COVID-19 Outbreak Caused by Contaminated Packaging of Imported Cold-Chain Products—Liaoning Province,China,July 2020显示文摘Summary What is known about this topic?Few major outbreaks of coronavirus disease 2019(COVID-19)have occurred in China after major nonpharmaceutical interventions and vaccines have been deployed and implemented. However, sporadic outbreaks that had high possibility to be linked to cold chain products were reported in several cities of China..What is added by this report? In July 2020,a COVID-19 outbreak occurred in Dalian,China. The investigations of this outbreak strongly suggested that the infection source was from COVID-19 virus-contaminated packaging of frozen seafood during inbound unloading personnel contact. What are the implications for public health practice?Virus contaminated paper surfaces could maintain infectivity for at least 17–24 days at-25℃.Exposure to COVID-19 virus-contaminated surfaces is a potential route for introducing the virus to a susceptible population.Countries with no domestic transmission of COVID-19 should consider introducing prevention strategies for both inbound travellers and imported goods.Several measures to prevent the introduction of the virus via cold-chain goods can be implemented. | Huilai Ma1 Jianqun Zhang Ji Wang Ying Qin Cao Chen Yang Song Liang Wang Jun Meng Lingling Mao Fengqin Li Ning Li Jian Cai Yong Zhang Dayan Wang Yunting Xia Hong Wang Shaofeng Jiang Xiang Zhao Peihua Niu Wenjie Tan Tao Ma Yecheng Yao Naiying Mao Zhen Zhu Tianjiao Ji Qian Yang Baoying Huang Li Zhao Jianxing Yu Li Bai Shuangli Zhu Dongyan Wang Yan Zhang Yingwei Sun Mingchun Luan Yanhai Wang Haibo Sun Shihong Yang Zhijian Bo Xiang Ren Zhongjie Li George Fu Gao Wei Yao Wenqing Yao Zijian Feng Wenbo Xu | 2021 | China CDC weekly2021,3,21: | 16

How to identify dislocations in molecular dynamics simulations?显示文摘Dislocations are of great importance in revealing the underlying mechanisms of deformed solid crystals. With the development of computational facilities and technologies, the observations of dislocations at atomic level through numerical simulations are permitted.Molecular dynamics(MD)simulation suggests itself as a powerful tool for understanding and visualizing the creation of dislocations as well as the evolution of crystal defects. However, the numerical results from the large-scale MD simulations are not very illuminating by themselves and there exist various techniques for analyzing dislocations and the deformed crystal structures. Thus,it is a big challenge for the beginners in this community to choose a proper method to start their investigations. In this review,we summarized and discussed up to twelve existing structure characterization methods in MD simulations of deformed crystal solids.A comprehensive comparison was made between the advantages and disadvantages of these typical techniques. We also examined some of the recent advances in the dynamics of dislocations related to the hydraulic fracturing. It was found that the dislocation emission has a significant effect on the propagation and bifurcation of the crack tip in the hydraulic fracturing. | LI Duo WANG Feng Chao YANG Zhen Yu ZHAO Ya Pu | 2014 | Science China(Physics,Mechanics & Astronomy)2014,57,12: | 11
Association Between Diurnal Temperature Range and Respiratory Tract Infections显示文摘Objective This study aimed to assess the association between emergency-room visits for respiratory tract infection (RTI) with diurnal temperature range (DTR), a weather parameter closely associated with urbanization and global climate change. Methods We conducted a semiparametric time-series analysis to estimate the percentage increase in emergency-room visits for RTI associated with changes in DTR after adjustment for daily weather conditions (temperature and relative humidity) and outdoor air pollution. Results DTR was significantly associated with daily emergency-room visits for RTI. An increase of 1 °C in the current-day (L0) and in the 2-day moving average (L01) DTR corresponded to a 0.94% [95% confidence interval (CI), 0.34%-1.55%] and 2.08% (95% CI, 1.24%-2.93%) increase in emergency-room visits for RTI, respectively. Conclusion DTR was associated with increased risk of RTI. | 14 | Malnutrition and Quality of Life in Chinese Cancer Patients: a Clinical Study of 23,994 Subjects显示文摘Background Malnutrition is common in patients with cancer, and can negatively impact their quality of life(QoL)and even survival. However, there is currently no large data available on the prevalence of malnutrition in Chinese cancer patients. This study evaluated the prevalence of malnutrition and the QoL of Chinese patients with locoregional, recurrent or metastatic cancer. Methods We conducted a nationwide observational,multi-center,hospital-based cross-sectional study within the Chinese Society of Nutritional Oncology(CSNO)Network. All of the patients were diagnosed with one of the following 18 different types of malignant tumors:lung cancer, gastric cancer, liver cancer, colorectal cancer, breast cancer, esophageal cancer, cervical cancer, endometrial cancer, nasopharyngeal carcinoma, malignant lymphoma, leukemia, pancreatic cancer, ovarian cancer, prostate cancer, bladder cancer, brain cancer, biliary tract malignant tumors or gastrointestinal stromal tumors. These patients were enrolled from 72 hospitals located in different regions of China. The patients’nutritional status was evaluated based on the body mass index(BMI),loss of bodyweight, laboratory measurements and patient generated-subjective global assessment(PG-SGA)scores.The cancer patients’physical status and QoL were assessed by the Karnofsky Performance Status(KPS)questionnaire and the European Organization for Research and Treatment of Cancer(EORTC)QLQ-C30 questionnaire, respectively. Results From December 2013 to April 2016,23,994 patients hospitalized for cancer treatment(such as surgery, chemotherapy or radiotherapy)were enrolled in the study. The patients included 12,494(52.9%)males and 11,124(47.1%)females.The mean age was 55.8±13.7 years. The proportions of patients in cancer stagesⅠ,Ⅱ,Ⅲ,Ⅳand uncertain were 11.5%,20.3%,27.5%,30.2%and 10.5%,respectively.Among the 23,994 inpatients, the proportions of patients who were underweight(BMI<18.5 kg/m2),normal(18.5 kg/m2 | Hong Xia Xu Chun Hua Song Zhen Ming Fu Chang Wang Zeng Qing Guo Yuan Lin Ying Ying Shi Wen Hu Yi Ba Su Yi Li Zeng Ning Li Kun Hua Wang Jing Wu Ying He Jia Jun Yang Cong Hua Xie Fu Xiang Zhou Xin Xia Song Gong Yan Chen Wen Jun Ma Su Xia Luo Zi Hua Chen Ming Hua Cong Hu Ma Chun Ling Zhou Wei Wang Qi Luo Yong Mei Shi Yu Mei Qi Hai Ping Jiang Wen Xian Guan Jun Qiang Chen Jia Xin Chen Yu Fang Lan Zhou Yong Dong Feng Rong Shao Tan Tao Li Jun Wen Ou Qing Chuan Zhao Jian Xiong Wu Min Weng Qing Hua Yao Jiu Wei Cui Wei Li Han Ping Shi the Investigation on Nutrition Status and Clinical Outcome of Common Cancers(INSCOC)Group | 2021 | Journal of Nutritional Oncology2021,6,1: | 8
Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear. Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks. Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. Women with comorbidities were more likely to have severe disease, while men with comorbidities were more likely to have unfavorable treatment outcomes. | MA Yan ZHU Dong Shan CHEN Ren Bo SHI Nan Nan LIU Si Hong FAN Yi Pin WU Gui Hui YANG Pu Ye BAI Jiang Feng CHEN Hong CHEN Li Ying FENG Qiao GUO Tuan Mao HOU Yong HU Gui Fen HU Xiao Mei HU Yun Hong HUANG Jin HUANG Qiu Hua HUANG Shao Zhen JI Liang JIN Hai Hao LEI Xiao LI Chun Yan LI Min Qing LI Qun Tang LI Xian Yong LIU Hong De LIU Jin Ping LIU Zhang MA Yu Ting MAO Ya MO Liu Fen NA Hui WANG Jing Wei SONG Fang Li SUN Sheng WANG Dong Ting WANG Ming Xuan WANG Xiao Yan WANG Yin Zhen WANG Yu Dong WU Wei WU Lan Ping XIAO Yan Hua XIE Hai Jun XU Hong Ming XU Shou Fang XUE Rui Xia YANG Chun YANG Kai Jun YUAN Sheng Li ZHANG Gong Qi ZHANG Jin Bo ZHANG Lin Song ZHAO Shu Sen ZHAO Wan Ying ZHENG Kai ZHOU Ying Chun ZHU Jun Teng ZHU Tian Qing ZHANG Hua Min WANG Yan Ping WANG Yong Yan 无 | 2020 | Biomedical and Environmental Sciences2020,33,12: | 8
Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. The validity of the PG-SGA in Chinese cancer patients warrants further investigation in other cancer types. | Zhen Ming Fu Hong Xia Xu Chun Hua Song Wei Li Zeng Qing Guo Yuan Lin Ying Ying Shi Wen Hu Yi Ba Su Yi Li Zeng Ning Li Kun Hua Wang Jing Wu Ying He Jia Jun Yang Cong Hua Xie Xin Xia Song Gong Yan Chen Wen Jun Ma Su Xia Luo Zi Hua Chen Ming Hua Cong Hu Ma Chun Ling Zhou Wei Wang Qi Luo Yong Mei Shi Yu Mei Qi Hai Ping Jiang Wen Xian Guan Jun Qiang Chen Jia Xin Chen Yu Fang Lan Zhou Yong Dong Feng Rong Shao Tan Tao Li Jun Wen Ou Qing Chuan Zhao Jian Xiong Wu Chang Wang Han Ping Shi The Investigation on Nutrition Status and Clinical Outcome of Common Cancers (INSCOC) Group | 2016 | Journal of Nutritional Oncology2016,1,1: | 7

The result showed that: The grey relational grade (GRG) of the number of flowers/plant (NF), the number of branches/plant (NB), the width of floral bud (WB), and the weight/flower (WF) to the FY were larger (> 0.5); FY improved with the increase of NF and NB. Moreover, the indirect selection of either trait could not achieve improvement of FY. It is necessary to improve FY by multi-trait selection. The integration selection index (ISI) equation of FY was established with the characters NF, NB, WB, and WF: I = 0.3187x1 - 318.6x2 + 670.1x4 + 6.3x8, index heritability = 0.8014, selective response of the integration breeding value = 245.8811. This will provide a theoretic basis for the genetic breeding of rugosa rose. | LI Yan-yan FENG Zhen ZHAO Lan-yong MO Zhen-hua ZHANG Bao | 2007 | Agricultural Sciences in China2007,6,12: | 6 |
Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. Conclusion The Chinese version of the PG-SGA is a valid tool to access the patients’ nutritional status for Chinese gastric patients. | Zhen Ming Fu Hong Xia Xu Chun Hua Song Wei Li Zeng Qing Guo Yuan Lin Ying Ying Shi Wen Hu Yi Ba Su Yi Li Zeng Ning Li Kun Hua Wang Jing Wu Ying He Jia Jun Yang Cong Hua Xie Xin Xia Song Gong Yan Chen Wen Jun Ma Su Xia Luo Zi Hua Chen Ming Hua Cong Hu Ma Chun Ling Zhou Wei Wang Qi Luo Yong Mei Shi Yu Mei Qi Hai Ping Jiang Wen Xian Guan Jun Qiang Chen Jia Xin Chen Yu Fang Lan Zhou Yong Dong Feng Rong Shao Tan Tao Li Jun Wen Ou Qing Chuan Zhao Jian Xiong Wu Chang Wang Han Ping Shi The Investigation on Nutrition Status and Clinical Outcome of Common Cancers (INSCOC) Group | 2018 | Journal of Nutritional Oncology2018,3,4: | 5
Mesenchymal stem cell treatment improves outcome of COVID-19 patients via multiple immunomodulatory mechanisms显示文摘The infusion of coronavirus disease 2019(COVID-19)patients with mesenchymal stem cells(MSCs)potentially improves clinical symptoms, but the underlying mechanism remains unclear. We conducted a randomized,single-blind,placebo-controlled(29 patients/group)phase II clinical trial to validate previous findings and explore the potential mechanisms.Patients treated with umbilical cord-derived MSCs exhibited a shorter hospital stay(P=0.0198)and less time required for symptoms remission(P=0.0194)than those who received placebo.Based on chest images, both severe and critical patients treated with MSCs showed improvement by day 7(P=0.0099)and day 21(P=0.0084).MSC-treated patients had fewer adverse events.MSC infusion reduced the levels of C-reactive protein, proinflammatory cytokines, and neutrophil extracellular traps(NETs)and promoted the maintenance of SARS-CoV-2-specific antibodies. To explore how MSCs modulate the immune system,we employed single-cell RNA sequencing analysis on peripheral blood. Our analysis identified a novel subpopulation of VNN2+hematopoietic stem/progenitorlike(HSPC-like)cells expressing CSF3R and PTPRE that were mobilized following MSC infusion.Genes encoding chemotaxis factors—CX3CR1 and L-selectin—were upregulated in various immune cells.MSC treatment also regulated B cell subsets and increased the expression of costimulatory CD28 in T cells in vivo and in vitro. In addition,an in vivo mouse study confirmed that MSCs suppressed NET release and reduced venous thrombosis by upregulating kindlin-3 signaling.Together, our results underscore the role of MSCs in improving COVID-19 patient outcomes via maintenance of immune homeostasis. | Rongjia Zhu Tingdong Yan Yingmei Feng Yan Liu Hongcui Cao Gongxin Peng Yanlei Yang Zhen Xu Jingqi Liu Wei Hou Xiaoyue Wang Zhe Li Luchan Deng Shihua Wang Jing Li Qin Han Hongling Li Guangliang Shan Yinghao Cao Xingyan An Jianshe Yan Zhonghui Zhang Huafei Li Xuebin Qu Jiaqi Zhu Shumin Zhou Jiao Wang Fengchun Zhang Jinming Gao Ronghua Jin Dayong Xu Yan-Qing Ma Tao Huang Shuang Peng Zhi Zheng Ilia Stambler Eric Gilson Lee Wei Lim Alexey Moskalev Antonio Cano Sasanka Chakrabarti Brun Ulfhake Huanxing Su Haoying Xu Sihuan Xu Feng Wei Holly MBrown-Borg Kyung-Jin Min Georgina Ellison-Hughes Calogero Caruso Kunlin Jin Robert Chunhua Zhao | 2021 | Cell Research2021,31,12: | 5
