3Sintilimab versus docetaxel as second-line treatment in advanced or metastatic squamous non-small-cell lung cancer:an open-label,randomized controlled phase 3 trial(ORIENT-3)显示文摘Background:Treatment options for Chinese patients with locally advanced or metastatic squamous-cell non-small-cell lung cancer(sqNSCLC)after failure of first-line chemotherapy are limited.This study(ORIENT-3)aimed to evaluate the efficacy and safety of sintilimab versus docetaxel as second-line treatment in patients with locally advanced or metastatic sqNSCLC.Methods:ORIENT-3 was an open-label,multicenter,randomized controlled phase 3 trial that recruited patients with stage IIIB/IIIC/IV sqNSCLC after failure with first-line platinum-based chemotherapy.Patients were randomized in a 1:1 ratio to receive either 200 mg of sintilimab or 75 mg/m^(2) of docetaxel intravenously every 3 weeks,stratified by the Eastern Cooperative Oncology Group performance status.The primary endpoint was overall survival(OS)in the full analysis set(FAS).Secondary endpoints included progression-free survival(PFS),objective response rate(ORR),disease control rate(DCR),duration of response(DoR)and safety.Results:Between August 25,2017,and November 7,2018,290 patients were randomized.For FAS,10 patients fromthe docetaxel armwere excluded.Themedian OS was 11.79(n=145;95%confidence interval[CI],10.28-15.57)months with sintilimab versus 8.25(n=135;95%CI,6.47-9.82)months with docetaxel(hazard ratio[HR]:0.74;95%CI,0.56-0.96;P=0.025).Sintilimab treatment significantly prolonged PFS(median 4.30 vs.2.79 months;HR:0.52;95%CI,0.39-0.68;P<0.001)and showed higher ORR(25.50%vs.2.20%,P<0.001)and DCR(65.50%vs.37.80%,P<0.001)than the docetaxel arm.The median DoRwas 12.45(95%CI,4.86-25.33)months in the sintilimab arm and 4.14(95%CI,1.41-7.23)months in the docetaxel arm(P=0.045).Treatment-related adverse events of grade≥3were reported in 26(18.1%)patients in the sintilimab arm and 47(36.2%)patients in the docetaxel arm.Exploratory biomarker analysis showed potential predictive values of expression levels of two transcription factors,including OVOL2(HR:0.35;P<0.001)and CTCF(HR:3.50;P<0.001),for sintilimab treatment.Conclusions:Compared with docetaxel,sintilimab significantly improved the OS,PFS,and ORR of Chinese patients with previously treated locally advanced or metastatic sqNSCLC.Yuankai Shi Lin Wu Xinmin Yu Puyuan Xing Yan Wang Jianying Zhou Airong Wang Jianhua Shi Yi Hu Ziping Wang Guangyu An Yong Fang Sanyuan Sun Caicun Zhou Changli Wang Feng Ye Xingya Li Junye Wang Mengzhao Wang Yunpeng Liu Yanqiu Zhao Ying Yuan Jifeng Feng Zhendong Chen Jindong Shi Tao Sun Gang Wu Yongqian Shu Qisen Guo Yi Zhang Yong Song Shucai Zhang Yuan Chen Wei Li Hongrui Niu Wenwei Hu Lijun Wang Jianan Huang Yang Zhang Ying Cheng Zhengdong Wu Bo Peng Jiya Sun Christoph Mancao Yanqi Wang Luyao Sun 2022Cancer Communications2022,42,12:8
4SUMO E3 Ligases GmSIZ1a and GmSIZ1b regulate vegetative growth in soybean显示文摘SIZ1 is a small ubiquitin-related modifier(SUMO) E3 ligase that mediates post-translational SUMO modification of target proteins and thereby regulates developmental processes and hormonal and environmental stress responses in Arabidopsis. However,the role of SUMO E3 ligases in crop plants is largely unknown. Here, we identified and characterized two Glycine max(soybean) SUMO E3 ligases, GmSIZ1a and GmSIZ1b. Expression of GmSIZ1a and GmSIZ1b was induced in response to salicylic acid(SA), heat, and dehydration treatment, but not in response to cold, abscisic acid(ABA), and Na Cl treatment. Although GmSIZ1a was expressed at higher levels than GmSIZ1b, both genes encoded proteins with SUMO E3 ligase activity in vivo.Heterologous expression of GmSIZ1a or GmSIZ1b rescued the mutant phenotype of Arabidopsis siz1-2, including dwarfism, constitutively activated expression of pathogen-related genes, and ABA-sensitive seed germination.Simultaneous downregulation of GmSIZ1a and GmSIZ1b(GmSIZ1a/b) using RNA interference(RNAi)-mediated gene silencing decreased heat shock-induced SUMO conjugation in soybean. Moreover, GmSIZ1 RNAi plants exhibited reduced plant height and leaf size. However,unlike Arabidopsis siz1-2 mutant plants, flowering time and SA levels were not significantly altered in GmSIZ1 RNAi plants. 10Changes in the Photosynthetic Pigment Contents and Transcription Levels of Phycoerythrin-Related Genes in Three Gracilariopsis lemaneiformis Strains Under Different Light Intensities显示文摘Three Gracilariopsis lemaneiformis strains,including wild type and high-temperature-resistant cultivars 981 and 2007,were studied with the changes in their photosynthetic pigment contents and related gene transcription levels under different light intensities(10,60,100,and 200μmolm^(−2)s^(−1)).The three G.lemaneiformis strains had the following photosynthetic pigments with high-to-low contents:phycoerythrin(PE),phycocyanin(PC),allophycocyanin(APC),and chlorophyll a(Chl a).Among the three strains,cultivar 981 had the highest PE content,followed by cultivar 2007.The PC and APC contents were similar among the three strains,but they were higher in cultivars 981 and 2007 than in the wild type.The Chl a contents in the three G.lemaneiformis strains were equal.A low light intensity(10μmolm^(−2)s^(−1))promoted photosynthetic pigment accumulation in G.lemaneiformis and improved the relative PE gene transcription(peA and peB)in a short period(≤6 d).A high light intensity decreased the PE content.PebA and PebB,which catalyzed phycoerythrobilin synthesis,showed no compensatory upregulation at a low light intensity among the strains except for the wild type.At a high light intensity,transcription levels of pebA and pebB in the three strains were upregulated.This study provided an experimental basis for elucidating the photosynthesis of G.lemaneiformis.As key genes of algal growth,photo-synthesis-related genes served as useful gene markers for screening elite varieties with good traits in breeding.Cultivar 2007 was superior to cultivar 981 in terms of maintaining high pigment levels in a wide range of light intensities,which is the most suitable for aquaculture.CAO Xuexue WANG Haitao ZANG Xiaonan LIU Zhu XU Di JIN Yuming ZHANG Feng WANG Zhendong 2021Journal of Ocean University of China2021,20,3:1

17Quantitative morphometric analysis of a deep-water channel in the Taranaki Basin, New Zealand显示文摘The morphological changes of deep-water channels have an important influence on the distributions of channel sand reservoirs,so it is important to explore the morphological change process of deep-water channel for the exploration and development of deep-water oil and gas.Based on a typical sinuous Quaternary channel(Channel I)in the Taranaki Basin,New Zealand,a variety of seismic interpretation techniques were applied to quantitatively characterize the morphological characteristics of the Channel I,and the relationships between the quantitative parameters and the morphological changes of the Channel I,as well as the controlling factors affecting those morphological changes,were discussed.The results are as follows:(1)in the quantitative analysis,six parameters were selected:the channel depth,width,sinuosity,and aspect ratio(width/depth),the channel swing amplitude(λ)and the channel bend frequency(ω);(2)according to the quantitative morphological parameters of the channel(mainly including three parameters such as channel sinuosity,ωandλ),the Channel I was divided into three types:the low-sinuous channel(LSC),the high-sinuous channel(HSC),the moderate-sinuous channel(MSC).U-shaped channel cross-sections developed in the LSC,V-shaped channel cross-sections developed in the HSC,including inclined-V and symmetric-V cross-sections,and dish-shaped channel cross-sections developed in the MSC;(3)the morphological characteristics of the LSC and MSC were related to their widths and depths,while the morphology of the HSC was greatly affected by the channel width,a change in depth did not affect the HSC morphology;(4)the morphological changes of the Channel I were controlled mainly by the slope gradient,the restricted capacity of the channel and the differential in fluid properties.Wei Wu Guangxu Wang Changsong Lin Weiqing Liu Quan Li Zhendong Feng Shuyuan Ning 2023Acta Oceanologica Sinica2023,42,5:0

19Automatic segmentation of gas plumes from multibeam water column images using a U-shape network显示文摘Cold seeps are widely developed on the seabed of continental margins and can form gas plumes due to the upward migration of methane-rich fluids.The detection and automatic segmentation of gas plumes are of great significance in locating and studying the cold seep system that is usually accompanied by hydrate layers in the subsurface.A multibeam echo-sounder system(MBES)can record the complete backscatter intensity of the water column,and it is one of the most effective means for detecting cold seeps.However,the gas plumes recorded in multibeam water column images(WCI)are usually blurred due to the interference of the complicated water environment and the sidelobes of the MBES,making it difficult to obtain the effective segmentation.Therefore,based on the existing UNet semantic segmentation network,this paper proposes an AP-UNet network combining the convolutional block attention module and the pyramid pooling module for the automatic segmentation and extraction of gas plumes.Comparative experiments are conducted among three traditional segmentation methods and two deep learning methods.The results show that the AP-UNet segmentation model can effectively suppress complicated water column noise interference.The segmentation precision,the Dice coefficient,and the recall rate of this model are 92.09%,92.00%,and 92.49%,respectively,which are 1.17%,2.10%,and 2.07%higher than the results of the UNet.Fanlin YANG Feng WANG Zhendong LUAN Xianhai BU Sai MEI Jianxing ZHANG Hongxia LIU 2023Journal of Oceanology and Limnology2023,41,5:0
