| 1 | Bipolar transurethral enucleation and resection of the prostate:Whether it is ready to supersede TURP?显示文摘Objective:According to the EAU Guidelines,transurethral resection of the prostate(TURP)has so far still been considered as the gold standard for surgical treatment for patients with obstructing clinical benign prostate hyperplasia(BPH).However,its relatively high rate of complications and postoperative recurrence necessitates further modification and innovation on the surgery technique.We reported the patient outcomes with our technique.Methods:We retrospectively analyzed 52 patients with obstructing clinical BPH who underwent bipolar transurethral enucleation and resection of the prostate(B-TUERP)between March 2015 and September 2015.Pre-and perioperative parameters were obtained from medical charts.Postoperative follow-ups were administrated at 1,3,6,12 and 24 month(s)after surgery,respectively.Results:All the operations were performed successfully with a mean operative time of 43.1 min and an average tissue removal rate of 74.7%.Qmax was significantly improved immediately after surgery,followed by a continuous improvement throughout the follow-ups.Following a steep decrease in mean prostate specific antigen(PSA)and post void residual(PVR)observed within the first half year after surgery,the serum PSA was then maintained at a constant level of 0.61 ng/mL.Temporary urinary retention was found in four cases(7.7%).Stress urinary incontinence occurred in five patients(9.6%),with the condition resolved in 1e2 weeks without extra treatment.Urethral strictures and bladder neck contractures,as the most commonly observed long-term complications,developed in four patients(7.7%).No recurrence was found during 2 years of follow-ups.An improvement in International Index of Erectile Function(IIEF-5)scores was witnessed in 17 patients preoperatively with normal sexual function during the first 6 months after surgery,and sustained throughout the 24-month period.Conclusions:Enucleation reflects an improvement on surgical technique in many ways with a need for surgical equipment that can be broadly accessible in clinical practice.Currently,bipolar resection is a commonly employed procedure in clinical settings,and its similarity shared with bipolar enucleation technique warrants a quick learning of B-TUERP by urologists.Based on these findings,we believe that the substitution of TURP by TUERP as the gold standard for prostate endoscopic procedure can be expected in the future. | Peng Xu Abai Xu Binshen Chen Shaobo Zheng Yawen Xu Hulin Li Haiyan Shen Chunxiao Liu | 2018 | Asian Journal of Urology2018,5,1: | 19 |
| 2 | Risk analysis for the highly pathogenic avian influenza in China's Mainland using meta-modeling显示文摘A logistic model was employed to correlate the outbreak of highly pathogenic avian influenza (HPAI) with related environmental factors and the migration of birds. Based on MODIS data of the normalized difference vegetation index, environmental factors were considered in generating a probability map with the aid of logistic regression. A Bayesian maximum entropy model was employed to explore the spatial and temporal correlations of HPAI incidence. The results show that proximity to water bodies and national highways was statistically relevant to the occurrence of HPAI. Migratory birds, mainly waterfowl, were important infection sources in HPAI transmission. In addition, the HPAI outbreaks had high spatiotemporal autocorrelation. This epidemic spatial range fluctuated 45 km owing to different distribution patterns of cities and water bodies. Furthermore, two outbreaks were likely to occur with a period of 22 d. | 3 | Selective laser melting of Al–8.5Fe–1.3V–1.7Si alloy: Investigation on the resultant microstructure and hardness显示文摘This article presents the microstructure and hardness variation of an Al–8.5Fe–1.3V–1.7Si(wt%, FVS0812) alloy after selective laser melting(SLM) modification. Three zones were distinguished across the melting pool of the SLM-processed FVS0812 alloy: the laser melted zone(LMZ), the melting pool border, and the heat affected zone(HAZ) in the previously deposited area around the melting pool. Inside the LMZ, either an extremely fine cellular-dendritic structure or a mixture zone of the a-Al matrix and nanoscale Al_(12)(Fe,V)_3Si particles appeared. With a decreased laser beam scanning speed, the cellular-dendritic structure zone within the LMZ shrank significantly while the mixture zone expanded. The a-Al and Al_(12)(Fe,V)_3Si mixture zone was also observed in the HAZ, but another phase, submicron h-Al_(13)Fe_4 particles with rectangular or hexagonal shapes,formed along the melting pool border. | 6 | Structure-function relationship of the mammarenavirus envelope glycoprotein显示文摘Mammarenaviruses, including lethal pathogens such as Lassa virus and Junín virus, can cause severe hemorrhagic fever in humans. Entry is a key step for virus infection, which starts with binding of the envelope glycoprotein(GP) to receptors on target cells and subsequent fusion of the virus with target cell membranes. The GP precursor is synthesized as a polypeptide, and maturation occurs by two cleavage events, yielding a tripartite GP complex(GPC) formed by a stable signal peptide(SSP), GP1 and GP2. The unique retained SSP interacts with GP2 and plays essential roles in virion maturation and infectivity. GP1 is responsible for binding to the cell receptor, and GP2 is a class I fusion protein. The native structure of the tripartite GPC is unknown.GPC is critical for the receptor binding, membrane fusion and neutralization antibody recognition.Elucidating the molecular mechanisms underlining the structure–function relationship of the three subunits is the key for understanding their function and can facilitate novel avenues for combating virus infections. This review summarizes the basic aspects and recent research of the structure–function relationship of the three subunits. We discuss the structural basis of the receptor-binding domain in GP1, the interaction between SSP and GP2 and its role in virion maturation and membrane fusion, as well as the mechanism by which glycosylation stabilizes the GPC structure and facilitates immune evasion. | 8 | CILP-2 is a novel secreted protein and associated with insulin resistance显示文摘Genetic association studies have implicated that cartilage intermediate layer protein 2 (CILP-2) confers the risk susceptibility for type 2 diabetes (T2DM). However, it is still unknown whether CILP-2 is involved in the regulation of glucose homeostasis and insulin resistance (IR). In the current study, we initially observed that CILP-2 as a secreted protein was detected in both conditioned medium and lysates of cells transfected with an overexpressed vector. We then found that circulating CILP-2 levels had a progressive increase from normal to impaired glucose tolerance (a pre-diabetic status) and then to diabetes, which was correlated positively with waist-to-hip ratio, triglyceride, fasting blood glucose, 2-h blood glucose after glucose overload, HbA1c, fasting insulin, 2-h plasma insulin after glucose overload, and homeostasis model assessment of insulin resistance but negatively with HDL-C. CILP-2 expression was increased in the liver and muscle but decreased in adipose tissues of obese mice or T2DM patients. Furthermore, we demonstrated that CILP-2 circulating levels were affected by OGTT and Exenatide. CILP-2 overexpression resulted in impaired glucose tolerance and hepatic IR in vivo and increased PEPCK expression whereas suppressed phosphorylation of insulin receptor and Akt kinase in vitro. | 9 | A high-quality sponge gourd(Luffa cylindrica)genome显示文摘Sponge gourd(Luffa cylindrica)is an important cultivated vegetable and medicinal plant in the family Cucurbitaceae.In this study,a draft genome sequence of the sponge gourd inbred line P93075 was analyzed.Using Illumina,PacBio,and 10×Genomics sequencing techniques as well as new assembly techniques such as FALCON and chromatin interaction mapping(Hi-C),a chromosome-scale genome of approximately 656.19 Mb,with an N50 scaffold length of 48.76 Mb,was generated.From this assembly,25,508 protein-coding gene loci were identified,and 63.81%of the whole-genome consisted of transposable elements,which are major contributors to the expansion of the sponge gourd genome.According to a phylogenetic analysis of conserved genes,the sponge gourd lineage diverged from the bitter gourd lineage approximately 41.6 million years ago.Additionally,many genes that respond to biotic and abiotic stresses were found to be lineage specific or expanded in the sponge gourd genome,as demonstrated by the presence of 462 NBS-LRR genes,a much greater number than are found in the genomes of other cucurbit species;these results are consistent with the high stress resistance of sponge gourd.Collectively,our study provides insights into genome evolution and serves as a valuable reference for the genetic improvement of sponge gourd. | Haibin Wu Gangjun Zhao Hao Gong Junxing Li Caixia Luo Xiaoli He Shaobo Luo Xiaoming Zheng Xiaoxi Liu Jinju Guo Junqiu Chen Jianning Luo | 2020 | Horticulture Research2020,7,1: | 2 |

| 11 | Differences in the primary culture, purification and biological characteristics between endothelial cells and smooth muscle cells from rat aorta显示文摘Objective: To investigate the differences of primary culture, purification and biological characteristics between endothelial cells and smooth muscle cells from rat aorta. Methods: Endothelial cells were obtained using the vascular ring adherence, collagenase digestion method and an improved vascular ring adherence method, while smooth muscle cells were separated from tissue sections of rat aorta. Clones of endothelial cells were selected by limiting dilution assay. Both cell types were identified using specific cell immunofluorescent markers, and phase contrast microscopy was used to observe the morphological disparity between endothelial cells and smooth muscle cells at the single cell and colony level. Cell proliferation was determined by the cell counting kit-8. Differences between endothelial cells and smooth muscle cells were evaluated in trypsin digestion time, attachment time and recovery after cryopreservation. Results: Endothelial cells were obtained by all three methods. The improved vascular ring method provided the most reproducible results. Cells were in good condition, and of high purity. Smooth muscle cells were cultured successfully by the tissue fragment culture method. Clonal expansion of single endothelial cells was attained. The two cell types expressed their respective specific markers, and the rate of proliferation of smooth muscle cells exceeded that of endothelial cells. Endothelial cells were more sensitive to trypsin digestion than smooth muscle cells. In addition, they had a shorter adherence time and better recovery following cryopreservation than smooth muscle cells. Conclusion: The improved vascular ring method was optimal for yielding endothelial cells. Limiting dilution is a novel and valid method for purifying primary endothelial cells from rat aorta. Primary rat endothelial cell and vascular smooth muscle cell cultures exhibited different morphological characteristics, proliferation rate, adherence time, susceptibility to trypsin digestion and recovery after cryopreservation. | 15 | 肝细胞肝癌经肝动脉化疗栓塞后破裂:2例报告及系统回顾(英文)显示文摘Objective: Rupture of hepatocellular carcinoma (HCC) following transarterial embolization/chemoembolization (TAE/TACE) is a rare but life-threatening complication. The aim of the study was to explore the incidence, risk factors, clinical characteristics, treatment, and outcomes of this complication. Methods: We described two cases and reviewed all cases of ruptured HCC after TAE/TACE reported in the literature. Results: Our search yielded 32 cases of ruptured HCC after TAE/TACE. The overall incidences were 0.45% per patient and 0.21% per session. The mean age of the patients was 57.4 years (range 28-90 years, n=26, No. of cases with available information). Males accounted for 81% of cases (21/26). The 50% of the cases had histories of primary hypertension, diabetes or peripheral artery disease (6/12). Mean diameter of the tumors was 11.4 cm (range 3-20 cm, n=27). The 100% of cases had superficial or exophytic tumors (23/23). Portal vein thrombosis was presented in 61.5% of patients (8/13). The median interval between TAE/TACE and rupture was 2 days (range 0 hour-30 days, n=31). Management choices included emergency TAE, surgery, and conservative treatment. The overall median survival time was 7 days (n=19). Conclusion: Rupture of HCC following TAE/TACE is relatively rare but potentially life-threatening. The management is difficult and prognosis is poor.
