Spectral tomographic imaging with aplanatic metalens显示文摘Tomography is an informative imaging modality that is usually implemented by mechanical scanning, owing to the limited depth-of-field(DOF)in conventional systems. However, recent imaging systems are working towards more compact and stable architectures;therefore, developing nonmotion tomography is highly desirable. Here,we propose a metalens-based spectral imaging system with an aplanatic GaN metalens(NA=0.78),in which large chromatic dispersion is used to access spectral focus tuning and optical zooming in the visible spectrum. After the function of wavelength-switched tomography was confirmed on cascaded samples, this aplanatic metalens is utilized to image microscopic frog egg cells and shows excellent tomographic images with distinct DOF features of the cell membrane and nucleus. Our approach makes good use of the large diffractive dispersion of the metalens and develops a new imaging technique that advances recent informative optical devices. | Chen Chen Wange Song Jia-Wern Chen Jung-Hsi Wang Yu Han Chen Beibei Xu Mu-Ku Chen Hanmeng Li Bin Fang Ji Chen Hsin Yu Kuo Shuming Wang Din Ping Tsai Shining Zhu Tao Li | 2019 | Light(Science & Applications)2019,8,1: | 12
Cholesterol transport through the peroxisome-ER membrane contacts tethered by PI(4,5)P2 and extended synaptotagmins显示文摘Most mammalian cells take up cholesterol from low-density lipoproteins(LDLs) via receptor-mediated endocytosis. After reaching lysosomes,LDL-derived cholesterol continues to transport to downstream organelles including the ER for specific structural and functional needs.Peroxisomes are recently found to receive cholesterol from lysosomes through lysosomeperoxisome membrane contacts. However, whether and how cholesterol is conveyed from peroxisomes to the ER remain unknown. Here,by combining high-resolution microscopic analyses and in vitro reconstitution of highly purified organelles or artificial liposomes,we demonstrate that peroxisomes form membrane contacts with the ER through the interaction between peroxisomal PI(4,5)P2 and ER-resident extended synaptotagmin-1,2 and 3(E-Syts).Depletion of peroxisomal PI(4,5)P2 or ESyts markedly decreases peroxisome-ER membrane contacts and induces cholesterol accumulation in lysosomes.Furthermore,we show that cholesterol is delivered from 3H-labeled peroxisomes or PI(4,5)P2-containing liposomes to the ER in vitro, and that the presence of peroxisomes augments cholesterol transfer from lysosomes to the ER.Together, our study reveals a new cholesterol transport pathway along the lysosome-peroxisome-ER membrane contacts in the cell. | Jian Xiao Jie Luo Ao Hu Ting Xiao Meixin Li Zekai Kong Luyi Jiang Zimu Zhou Yacheng Liao Chang Xie Beibei Chu Honghua Miao Boliang Li Xiongjie Shi Bao-Liang Song | 2019 | Science China(Life Sciences)2019,62,9: | 8
sLe^(x) expression in invasive micropapillary breast carcinoma is associated with poor prognosis and can be combined with MUC1/EMA as a supplementary diagnostic indicator显示文摘Objective: Mucin 1(MUC1/EMA) and sialyl Lewis X(sLe^(x)) indicate polarity reversal in invasive micropapillary carcinoma(IMPC). The purpose of this study was to evaluate the expression of MUC1/EMA and sLe^(x) and to assess their diagnostic and prognostic value in patients with IMPC.Methods: The expression of sLe^(x) and MUC1/EMA in 100 patients with IMPC and a control group of 89 patients with invasive ductal carcinoma not otherwise specified(IDC-NOS) were analyzed with IHC. Fresh tumor tissues were collected from patients with IMPC or IDC-NOS for primary culture and immunofluorescence analysis. Results: The rate of nodal metastasis was higher in patients with IMPC than those with IDC-NOS, and IMPC cells tended to express more sLe^(x) and MUC1/EMA in the cytomembranes(the stroma-facing surfaces of the micropapillary clusters) than IDC-NOS cells. In IMPC, high cytomembrane expression of sLe^(x), but not MUC1/EMA, indicated poor prognosis. In addition, among the 100 patients with IMPC, 10 patients had sLe^(x)+/EMA-expression patterns, and 8 patients had sLe^(x)-/EMA+ expression patterns. The primary IMPC cells were suspended, non-adherent tumor cell clusters, whereas the primary IDC cells were adherent tumor cells. Immunofluorescence analysis showed that MUC1/EMA and sLe^(x) were co-expressed on the cytomembranes in IMPC cell clusters and in the cytoplasm in IDC-NOS cells. Conclusions: sLe^(x) can be used as a prognostic indicator and can be combined with MUC1/EMA as a complementary diagnostic indicator to avoid missed IMPC diagnosis. | Yawen Song Hui Sun Kailiang Wu Jianke Lyu Jingyue Zhang Feng Gu Yongjie Ma Beibei Shen Chijuan Wang Xiaojiao Chen Jing Xu Weidong Li Fangfang Liu Li Fu | 2021 | Cancer Biology & Medicine2021,18,2: | 7
The landscape of aging显示文摘Aging is characterized by a progressive deterioration of physiological integrity, leading to impaired functional ability and ultimately increased susceptibility to death. It is a major risk factor for chronic human diseases, including cardiovascular disease, diabetes, neurological degeneration, and cancer. Therefore, the growing emphasis on “healthy aging” raises a series of important questions in life and social sciences. In recent years, there has been unprecedented progress in aging research, particularly the discovery that the rate of aging is at least partly controlled by evolutionarily conserved genetic pathways and biological processes. In an attempt to bring full-fledged understanding to both the aging process and age-associated diseases,we review the descriptive, conceptual, and interventive aspects of the landscape of aging composed of a number of layers at the cellular, tissue, organ, organ system, and organismal levels. | Yusheng Cai Wei Song Jiaming Li Ying Jing Chuqian Liang Liyuan Zhang Xia Zhang Wenhui Zhang Beibei Liu Yongpan An Jingyi Li Baixue Tang Siyu Pei Xueying Wu Yuxuan Liu Cheng-Le Zhuang Yilin Ying Xuefeng Dou Yu Chen Fu-Hui Xiao Dingfeng Li Ruici Yang Ya Zhao Yang Wang Lihui Wang Yujing Li Shuai Ma Si Wang Xiaoyuan Song Jie Ren Liang Zhang Jun Wang Weiqi Zhang Zhengwei Xie Jing Qu Jianwei Wang Yichuan Xiao Ye Tian Gelin Wang Ping Hu Jing Ye Yu Sun Zhiyong Mao Qing-Peng Kong Qiang Liu Weiguo Zou Xiao-Li Tian Zhi-Xiong Xiao Yong Liu Jun-Ping Liu Moshi Song Jing-Dong J.Han Guang-Hui Liu | 2022 | Science China(Life Sciences)2022,65,12: | 4
Hydration, microstructure and autogenous shrinkage behaviors of cement mortars by addition of superabsorbent polymers显示文摘Superabsorbent Polymer(SAP)has emerged as a topic of considerable interest in recent years. The present study systematically and quantitively investigated the effect of SAP on hydration, autogenous shrinkage, mechanical properties, and microstructure of cement mortars.Influences of SAP on hydration heat and autogenous shrinkage were studied by utilizing TAM AIR technology and a non-contact autogenous shrinkage test method.Scanning Electron Microscope(SEM)was employed to assess the microstructure evolution.Although SAP decreased the peak rate of hydration heat and retarded the hydration,it significantly increased the cumulative heat, indicating SAP helps promote the hydration.Hydration promotion caused by SAP mainly occurred in the deceleration period and attenuation period.SAP can significantly mitigate the autogenous shrinkage when the content ranged from 0 to 0.5%.Microstructure characteristics showed that pores and gaps were introduced when SAP was added. The microstructure difference caused by SAP contributed to the inferior mechanical behaviors of cement mortars treated by SAP. | Beibei SUN Hao WU Weimin SONG Zhe LI Jia YU | 2020 | Frontiers of Structural and Civil Engineering2020,14,5: | 3
Significantly improved hydrogen storage behaviors in MgH_(2) with Nb nanocatalyst显示文摘The study explores the excellent modification effect of Nb nanocatalyst prepared via surfactant assisted ball milling technique(SABM)on the hydrogen storage properties of MgH_(2).Optimal catalyst doping concentration was determined by comparing onset decomposition temperature, released hydrogen capacity, and reaction rate for different MgH_(2)-ywt%Nb(y=0,3,5,7,9)composites.The MgH_(2)-5wt%Nb composite started releasing hydrogen at 186.7℃ and a total of 7.0wt%hydrogen was released in the dehydrogenation process. In addition,5wt%Nb doped MgH_(2) also managed to release 4.2wt%H_(2) within 14 min at 250℃ and had the ability to absorb 4.0wt%hydrogen in 30 min at 100℃.Cycling tests revealed that MgH_(2)-5wt%Nb could retain 6.3wt%H_(2) storage capacity(89.2%)after 20 cycles.Dehydrogenation and hydrogenation activation energy values were decreased from 140.51±4.74 and 70.67±2.07 kJ·mol^(−1) to 90.04±2.83 and 53.46±3.33 kJ·mol^(−1) after doping MgH_(2) with Nb,respectively.Microstructure analysis proved that homogeneously distributed NbH acted as active catalytic unit for improving the hydrogen storage performance of MgH_(2).These results indicate SABM can be considered as an option to develop other nanocatalysts for energy related areas. | Farai Michael Nyahuma Liuting Zhang Mengchen Song Xiong Lu Beibei Xiao Jiaguang Zheng Fuying Wu | 2022 | International Journal of Minerals,Metallurgy and Materials2022,29,9: | 3
Exploring single atom catalysts of transition-metal doped phosphorus carbide monolayer for HER:A first-principles study显示文摘Hydrogen has been identified as one of the most promising sustainable and clean energy. Developing hydrogen evolution reaction(HER) catalyst with high activity is essential for satisfying the future requirements. Considering novel advantages of two-dimensional materials and high catalytic activity of atomic transition metal, in this study, using density functional theory calculation, the HER on single transitionmetal(23 different TM atoms) doped phosphorus carbide monolayer(α-PC) has been investigated. The Volmer–Tafel and Volmer–Heyrovsky reaction mechanisms, and the stability of the most promising HER catalyst are also included. The results show that Ir-αPC with high physical and thermal stability has the most optimal value of Gibbs free adsorption energy for H atom. The relationship of d band center and the HER activity shows a volcano-like curve. The calculation of reaction energy barrier indicates that the Volmer-Heyrovsky step is more favorable than the Volmer-Tafel step. | Dachang Chen Zhiwen Chen Xiaoxing Zhang Zhuole Lu Song Xiao Beibei Xiao Chandra Veer Singh | 2021 | Journal of Energy Chemistry2021,30,1: | 3
Light weight, mechanically strong and biocompatible α-chitin aerogels from different aqueous alkali hydroxide/urea solutions显示文摘Light weight and mechanically strong α-chitin aerogels were fabricated using the sol-gel/self-assembly method from α-chitin in different aqueous alkali hydroxide(KOH, Na OH and Li OH)/urea solutions. All of the α-chitin solutions exhibited temperature-induced rapid gelation behavior. 13 C nuclear magnetic resonance(NMR) spectra revealed that the aqueous alkali hydroxide/urea solutions are non-derivatizing solvents for α-chitin. Fourier transform infrared(FT-IR), X-ray diffraction(XRD) and cross-polarization magic angle spinning(CP/MAS) 13 C NMR confirmed that α-chitin has a stable aggregate structure after undergoing dissolution and regeneration. Subsequently, nanostructured α-chitin aerogels were fabricated by regeneration from the chitin solutions in ethanol and then freeze-drying from t-Bu OH. These α-chitin aerogels exhibited high porosity(87% to 94%), low density(0.09 to 0.19 g/cm^3), high specific surface area(419 to 535 m^2/g) and large pore volume(2.7 to 3.8 cm^3/g). Moreover, the α-chitin aerogels exhibited good mechanical properties under compression and tension models. In vitro studies showed that m BMSCs cultured on chitin hydrogels have good biocompatibility. These nanostructured α-chitin aerogels may be useful for various applications, such as catalyst supports, carbon aerogel precursors and biomedical materials. | Beibei Ding Dan Zhao Jianhui Song Huichang Gao Duoduo Xu Min Xu Xiaodong Cao Lina Zhang Jie Cai | 2016 | Science China Chemistry2016,59,11: | 2

CD4+T cells memorize obesity and promote weight regain显示文摘Body weight regain often causes failure of obesity therapies while the underlying mechanism remains largely unknown. In this study,we report that immune cells, especially CD4+T cells, mediate the‘memory’of previous obese status. In a weight gain-loss-regain model,we found that C57BL/6J mice with an obesity history showed a much faster rate of body weight regain. This obesity memory could last for at least 2 months after previously obese mice were kept at the same body weight as non-obese mice.Surprisingly, such obesity memory was abrogated by dexamethasone treatment, whereas immunodeficient Rag1−/−and H2A−/−mice failed to establish such memory.Rag1−/−mice repossessed the obesity memory when immune cells or CD4+T cells isolated from previously obese mice were transferred.Furthermore, depletion of CD4+T cells led to obesity memory ablation.Taken together,we conclude that CD4+T cells mediate obesity memory and promote weight regain. | Jianghuan Zou Beibei Lai Mingzhu Zheng Qin Chen Shujun Jiang Anying Song Zan Huang Peiliang Shi Xin Tu Di Wang Linrong Lu Zhaoyu Lin Xiang Gao | 2018 | Cellular & Molecular Immunology2018,15,6: | 2
Motivating Ru-bri site of RuO_(2)by boron doping toward high performance acidic and neutral oxygen evolution显示文摘The electrocatalysis of oxygen evolution reaction(OER)plays a key role in clean energy storage and transfer.Nonetheless, the sluggish kinetics and poor durability under acidic and neutral conditions severely hinder practical applications such as electrolyzer compatible with the powerful proton exchange membrane and biohybrid fuel production. Here,we report a borondoped ruthenium dioxide electrocatalyst(B-RuO_(2))fabricated by a facile boric acid assisted strategy which demonstrates excellent acidic and neutral OER performances.Density functional theory calculations and advanced characterizations reveal that the boron species form an anomalous B–O covalent bonding with the oxygen atoms of RuO_(2)and expose the fully coordinately bridge ruthenium site(Ru-bri site),which seems like a switch that turns on the inactive Ru-bri site into OER-active,resulting in more exposed active sites, modified electronic structure, and optimized binding energy of intermediates. Thus, the B-RuO_(2)exhibits an ultralow overpotential of 200 mV at 10 mA/cm^(2)and maintains excellent stability compared to commercial RuO_(2)in 0.5 M sulfuric acid. Moreover, the superior performance is as well displayed in neutral electrolyte, surpassing most previously reported catalysts. | Chongjing Liu Beibei Sheng Quan Zhou Dengfeng Cao Honghe Ding Shuangming Chen Pengjun Zhang Yujian Xia Xiaojun Wu Li Song | 2022 | Nano Research2022,15,8: | 1

Orchestrating antigen delivery and presentation efficiency in lymph node by nanoparticle shape for immune response显示文摘Activating humoral and cellular immunity in lymph nodes(LNs)of nanoparticle-based vaccines is critical to controlling tumors. However, how the physical properties of nanovaccine carriers orchestrate antigen capture, lymphatic delivery, antigen presentation and immune response in LNs is largely unclear. Here,we manufactured gold nanoparticles(AuNPs)with the same size but different shapes(cages, rods, and stars),and loaded tumor antigen as nanovaccines to explore their disparate characters on above four areas. Results revealed that star-shaped AuNPs captured and retained more repetitive antigen epitopes. On lymphatic delivery, both rods and star-shaped nanovaccines mainly drain into the LN follicles region while cage-shaped showed stronger paracortex retention.A surprising finding is that the star-shaped nanovaccines elicited potent humoral immunity, which is mediated by CD4^(+)T helper cell and follicle B cell cooperation significantly preventing tumor growth in the prophylactic study.Interestingly,cage-shaped nanovaccines preferentially presented peptide-MHC I complexes to evoke robust CD8^(+)T cell immunity and showed the strongest therapeutic efficacy when combined with the PD-1 checkpoint inhibitor in established tumor study. These results highlight the importance of nanoparticle shape on antigen delivery and presentation for immune response in LNs,and our findings support the notion that different design strategies are required for prophylactic and therapeutic vaccines. | Hongjuan Zhao Yatong Li Beibei Zhao Cuixia Zheng Mengya Niu Qingling Song Xinxin Liu Qianhua Feng Zhenzhong Zhang Lei Wang | 2023 | Acta Pharmaceutica Sinica B2023,13,9: | 1
Probing self-optimization of carbon support in oxygen evolution reaction显示文摘Despite acknowledgment of structural reconstruction of materials following oxygen evolution reaction (OER) reaction, the role of support during the reconstruction process has been ignored. Given this, we directly in situ transform the residual iron present in raw single-walled carbon nanotubes (SWCNT) into Fe_(2)O_(3) and thus build Fe_(2)O_(3)-CNT as the model system. Intriguingly, an anomalous self-optimization occurred on SWCNT and the derived components show satisfactory electrochemical performance. Soft X-ray absorption spectroscopy (sXAS) analysis and theory calculation correspondingly indicate that self-optimization yields stronger interaction between SWCNT and Fe_(2)O_(3) nanoparticles, where the electrons migrate from Fe_(2)O_(3) to optimized SWCNT. Such polarization will generate a positive charge center and thus boost the OER activity. This finding directly observes the self-optimization of support effect, providing a new perspective for OER and related electrochemical reactions. | Wenjie Xu Dengfeng Cao Oyawale Adetunji Moses Beibei Sheng Chuanqiang Wu Hongwei Shou Xiaojun Wu Shuangming Chen Li Song | 2021 | Nano Research2021,14,12: | 1
Combined zero valent iron and hydrogen peroxide conditioning significantly enhances the dewaterability of anaerobic digestate显示文摘The importance of enhancing sludge dewaterability is increasing due to the considerable impact of excess sludge volume on disposal costs and on overall sludge management. This study presents an innovative approach to enhance dewaterability of anaerobic digestate(AD) harvested from a wastewater treatment plant. The combination of zero valent iron(ZVI, 0–4.0 g/g total solids(TS)) and hydrogen peroxide(HP, 0–90 mg/g TS) under pH 3.0 significantly enhanced the AD dewaterability. The largest enhancement of AD dewaterability was achieved at 18 mg HP/g TS and 2.0 g ZVI/g TS, with the capillary suction time reduced by up to 90%. Economic analysis suggested that the proposed HP and ZVI treatment has more economic benefits in comparison with the classical Fenton reaction process. The destruction of extracellular polymeric substances and cells as well as the decrease of particle size were supposed to contribute to the enhanced AD dewaterability by HP + ZVI conditioning. | Qilin Wang Jing Sun Kang Song Xu Zhou Wei Wei Dongbo Wang Guo-Jun Xie Yanyan Gong Beibei Zhou | 2018 | Journal of Environmental Sciences2018,30,5: | 1
Electrical Stimulation Enabled via Electrospun Piezoelectric Polymeric Nanofibers for Tissue Regeneration显示文摘Electrical stimulation has demonstrated great effectiveness in the modulation of cell fate in vitro and regeneration therapy in vivo.Conventionally, the employment of electrical signal comes with the electrodes, battery, and connectors in an invasive fashion. This tedious procedure and possible infection hinder the translation of electrical stimulation technologies in regenerative therapy.Given electromechanical coupling and flexibility, piezoelectric polymers can overcome these limitations as they can serve as a self-powered stimulator via scavenging mechanical force from the organism and external stimuli wirelessly.Wireless electrical cue mediated by electrospun piezoelectric polymeric nanofibers constitutes a promising paradigm allowing the generation of localized electrical stimulation both in a noninvasive manner and at cell level.Recently, numerous studies based on electrospun piezoelectric nanofibers have been carried out in electrically regenerative therapy. In this review, brief introduction of piezoelectric polymer and electrospinning technology is elucidated first.Afterward,we highlight the activating strategies(e.g.,cell traction, physiological activity, and ultrasound)of piezoelectric stimulation and the interaction of piezoelectric cue with nonelectrically/electrically excitable cells in regeneration medicine. Then, quantitative comparison of the electrical stimulation effects using various activating strategies on specific cell behavior and various cell types is outlined.Followingly, this review explores the present challenges in electrospun nanofiber-based piezoelectric stimulation for regeneration therapy and summarizes the methodologies which may be contributed to future efforts in this field for the reality of this technology in the clinical scene. In the end,a summary of this review and future perspectives toward electrospun nanofiber-based piezoelectric stimulation in tissue regenerationare elucidated. | Guangbo Xia Beibei Song Jian Fang | 2022 | Research2022,,4: | 1

Intercellular transfer of activated STING triggered by RAB22A-mediated non-canonical autophagy promotes antitumor immunity显示文摘STING,an endoplasmic reticulum(ER)transmembrane protein, mediates innate immune activation upon cGAMP stimulation and is degraded through autophagy. Here,we report that activated STING could be transferred between cells to promote antitumor immunity,a process triggered by RAB22A-mediated non-canonical autophagy.Mechanistically,RAB22A engages PI4K2A to generate PI4P that recruits the Atg12–Atg5–Atg16L1 complex, inducing the formation of ER-derived RAB22A-mediated non-canonical autophagosome,in which STING activated by agonists or chemoradiotherapy is packaged. This RAB22A-induced autophagosome fuses with RAB22A-positive early endosome, generating a new organelle that we name Rafeesome(RAB22A-mediated non-canonical autophagosome fused with early endosome).Meanwhile,RAB22A inactivates RAB7 to suppress the fusion of Rafeesome with lysosome, thereby enabling the secretion of the inner vesicle of the autophagosome bearing activated STING as a new type of extracellular vesicle that we define as R-EV(RAB22A-induced extracellular vesicle).Activated STING-containing R-EVs induce IFNβrelease from recipient cells to the tumor microenvironment, promoting antitumor immunity.Consistently,RAB22A enhances the antitumor effect of the STING agonist diABZI in mice, and a high RAB22A level predicts good survival in nasopharyngeal cancer patients treated with chemoradiotherapy. Our findings reveal that Rafeesome regulates the intercellular transfer of activated STING to trigger and spread antitumor immunity, and that the inner vesicle of non-canonical autophagosome originated from ER is secreted as R-EV,providing a new perspective for understanding the intercellular communication of organelle membrane proteins. | Ying Gao Xueping Zheng Boyang Chang Yujie Lin Xiaodan Huang Wen Wang Shirong Ding Weixiang Zhan Shang Wang Beibei Xiao Lanqing Huo Youhui Yu Yilin Chen Run Gong Yuanzhong Wu Ruhua Zhang Li Zhong Xin Wang Qiuyan Chen Song Gao Zhengfan Jiang Denghui Wei Tiebang Kang | 2022 | Cell Research2022,32,12: | 1
