| 1 | Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite. | Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie | 2021 | Cell Research2021,31,7: | 12 |

| 3 | Evaluation of Four Candidate VNTR Loci for Genotyping 225 Chinese Clinical Mycobacterium Tuberculosis Complex Strains显示文摘Objective To evaluate four candidate variable number tandem repeat (VNTR) loci for genotyping Mycobacterium tuberculosis complex strains.Methods Genomic sequences for two M.tuberculosis strains (CCDC5079 and CCDC5180) were generated,and using published sequence data,four candidate VNTR loci were identified.The VNTRs were used to genotype 225 Chinese clinical M.tuberculosis complex strains.The discriminatory power of the VNTRs was evaluated using BioNumerics 5.0 software.Results The Hunter-Gaston Index (HGI) for BJ1,BJ2,BJ3,and BJ4 loci was 0.634,0.917,0.697,and 0.910,respectively.Combining all four loci gave an HGI value of 0.995,thus confirming that the genotyping had good discriminatory power.The HGI values for BJ1,BJ2,BJ3,and BJ4,obtained from Beijing family strain genotyping,were 0.447,0.878,0.315,and 0.850,respectively.Combining all four loci produced an HGI value of 0.988 for genotyping the Beijing family strains.We observed unique patterns for M.bovis and M.africanum strains from the four loci.Conclusion We have shown that the four VNTR loci can be successfully used for genotyping M.tuberculosis complex strains.Notably,these new loci may provide additional information about Chinese M.tuberculosis isolates than that currently afforded by established VNTR loci typing. | JIANG Yi LIU Hai Can ZHENG Hua Jun TANG Biao DOU Xiang Feng ZHAO Xiu Qin ZHU Yong Qiang LU Bing WANG Sheng Yue DONG Hai Yan ZHAO Guo Ping ZHANG Yuan Yuan KAN Biao WAN Kang Lin | 2012 | Biomedical and Environmental Sciences2012,25,1: | 1 |

| 7 | “Dual-Lock-Dual-Key”Controlled Second Near-Infrared Molecular Probe for Specific Discrimination of Orthotopic Colon Cancer and Imaging-Guided Tumor Excision显示文摘Fluorescence imaging in the second infrared window(1000-1700 nm)has emerged as a promising approach to tumor diagnosis.However,the currently available second near-infrared(NIR-II)imaging agents are based on the“always on”modality or single biomarker activation,which are subject to limited imaging contrast,nonspecific response,and even false-positive diagnosis.Here,we developed a H2S/H+dual-stimuli responsive NIR-II fluorescent probe,WH-N3,for precise tumor delimitation and intraoperative fluorescence-guided surgical resection.WH-N3 itself is nonfluorescent,and it can only light up through synergistic activation by H2S and in the tumor acidic environment(TEM).Such a“duallock-dual-key”strategy-based activatable probe exhibited significantly higher tumor-to-normal tissue(T/N)ratios than the“always on”agent(ICG)and single parameter responsive counterpart probes in the imaging of colon tumors,which overexpresses H2S.WH-N3 was also able to visualize the tumor-derived endogenous H2S fluctuation and accurately differentiate tumor types based on H2S content discrepancy.More excitingly,under the guidance of the probe’s highly specific NIR-II fluorescence,a tiny orthotopic colon tumor with diameter down to 0.8 mm was facilely resected.We expect our dual-stimuli responsive strategy will contribute more reliable tools for specific discrimination and imaging-guided excision of tumor. | Kun Dou Chen Fan Wenqi Feng Yao Kong Yunhui Xiang Zijun Wang Zhihong Liu | 2022 | CCS Chemistry2022,4,11: | 0 |
