| 1 | Preoperative neutrophil-to-lymphocyte ratio plus platelet-to-lymphocyte ratioin predicting survival for patients with stage Ⅰ-Ⅱgastric cancer显示文摘Background:The preoperative neutrophil-to-lymphocyte ratio(NLR) and the platelet-to-lymphocyte ratio(PLR) are associated with poor prognosis of gastric cancer.We aimed to determine whether the combination of NLR and PLR(NLR-PLR) could better predict survival of patients after curative resection for stage Ⅰ-Ⅱ gastric cancer.Methods:We collected data from the medical records of patients with stage Ⅰ-Ⅱ gastric cancer undergoing curative resection between December 2000 and November 2012 at the Sun Yat-sen Cancer Center.The preoperative NLRPLR was calculated as follows:patients with both elevated NLR(≥2.1) and PLR(≥ 120) were given a score of 2,and patients with only one or neither were given a score of 1 or 0,respectively.Results:Kaplan-Meier analysis and log-rank tests revealed significant differences in overall survival(OS) among patients with NLR-PLR scores of 0,1 and 2(P < 0.001).Multivariate analysis showed that OS was independently associated with the NLR-PLR score[hazard ratio(HR) = 1.51,95%confidence interval(CI) 1.02-2.24,P = 0.039]and TNM stage(HR = 1.36,95%CI 1.01-1.83,P= 0.041).However,other systemic inflammation-based prognostic scores,including the modified Glasgow prognostic score,the prognostic nutritional index,and the combination of platelet count and NLR,were not.In TNM stage-stratified analysis,the prognostic significance of NLR-PLR was maintained in patients with stage Ⅰ(P < 0.001) and stage Ⅱ cancers(P= 0.022).In addition,the area under the receiver operating characteristic curve for the NLR-PLR score was higher than those of other systemic inflammation-based prognostic scores(P = 0.001).Conclusion:The preoperative NLR-PLR score is a useful predictor of postoperative survival in the patients with stage l-ll gastric cancer and may help identify high-risk patients for rational therapy and timely follow-up. | Xiaowei Sun Xuechao Liu Jianjun Liu Shangxiang Chen Dazhi Xu Wei Li Youqing Zhan Yuanfang Li Yingbo Chen Zhiwei Zhou | 2016 | Chinese Journal of Cancer2016,35,7: | 25 |
| 2 | Roles of mechanosensitive channel Piezo 1/2 proteins in skeleton and other tissues显示文摘Mechanotransduction is a fundamental ability that allows living organisms to receive and respond to physical signals from both the external and internal environments.The mechanotransduction process requires a range of special proteins termed mechanotransducers to convert mechanical forces into biochemical signals in cells.The Piezo proteins are mechanically activated nonselective cation channels and the largest plasma membrane ion channels reported thus far.The regulation of two family members,Piezol and Piezo2#has been reported to have essential functions in mechanosensation and transduction in different organs and tissues.Recently,the predominant contributions of the Piezo family were reported to occur in the skeletal system,especially in bone development and mechano-stimulated bone homeostasis.Here we review current studies focused on the tissue-specific functions of Piezol and Piezo2 in various backgrounds with special highlights on their importance in regulating skeletal cell mechanotransduction.In this review,we emphasize the diverse functions of Piezol and Piezo2 and related signaling pathways in osteoblast lineage cells and chondrocytes.We also summarize our current understanding of Piezo channel structures and the key findings about PIEZO gene mutations in human diseases. | Lei Qin Tailin He Sheng Chen Dazhi Yang Weihong Yi Huiling Cao Guozhi Xiao | 2021 | Bone Research2021,9,4: | 14 |
| 3 | Deformation characteristics and formation mechanism of the Yunmengshan metamorphic core complex显示文摘The Yunmengshan metamorphic core complex in the middle part of the Yanshan Fold and Thrust Belt records crust extension processes of the eastern North China Craton during its peak destruction.Development of the metamorphic core complex was controlled by the generally NNE-striking Dashuiyu Shear Zone.The shear zone dips SE and becomes shallower NE-wards,leading to exposures of a ductile shear zone in the southern and middle parts and brittle faults in the northern part.Exposure structures,microstructures,and quartz C-axis fabrics indicate that the ductile shear zone belongs to an extensional shear zone with a top-to-the-SE shear sense.Deformation temperatures of 300–520°C suggest a midcrustal origin for the ductile shear zone.A ductile deformation belt in the footwall of the shear zone is only as wide as 1–3 km,indicating no widespread mid-crustal ductile flow in the region during the deformation.Zircon U–Pb dating of dykes and plutons as well as hornblende and biotite40Ar/39Ar dating demonstrate that the metamorphic core complex originated at 135 Ma and experienced intense shearing of the Dashuiyu Shear Zone,development of the supradetachment basins,and synkinematic intrusion during 135–125 Ma.The metamorphic core complex was subjected to rapid exhumation during 125–114 Ma when the Dashuiyu Shear Zone suffered continuous activity and passive doming.The shear zone and its hanging wall were cut or replaced by a series of brittle faults when they wereuplifted to a brittle regime,showing that exhumation took place in continuous extensional activities.The metamorphic core complex turned into slow exhumation in an extensional regime in the following latest Early Cretaceous.The evolution history suggests that the Yunmengshan metamorphic core complex was developed by the rolling-hinge model,a common formation mechanism for intraplate metamorphic core complexes in the North China Craton,under the continuous NW–SE extension during the Early Cretaceous(135–100 Ma). | Yin Chen Guang Zhu Dazhi Jiang Shaoze Lin | 2014 | Chinese Science Bulletin2014,59,20: | 10 |
| 4 | Prognostic value of programmed death.1, programmed death-ligand 1, programmed death-ligand 2 expression, and CD8(+) T cell density in primary tumors and metastatic lymph nodes from patients with stage T1.4N+M0 gastric adenocarcinoma显示文摘Background: Anti-programmed death-1/programmed death-ligand 1(PD-1/PD-L1) immunotherapy has been proved to be effective on gastric cancer in ongoing clinical trials. However, the value of PD-L1 in predicting responses of patients with gastric cancer to anti-PD-1/PD-L1 immunotherapy is controversial. Some studies suggested that intra-and inter-tumoral heterogeneity of PD-L1 expression might explain the controversy.This study aimed to analyze the expression of PD-L1, PD-L2, and PD-1 as well as CD8(+) T-cell density in primary tumors and lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma to explore the heterogeneity of PD-1 signaling pathway molecules.Methods: In primary tumors and metastatic as well as non-metastatic lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma, we detected PD-L1 and PD-L2 expression with immunohistochemistry. CD8(+)T-cell density in primary tumors and PD-1 expression on CD8(+)T cells were detected with immunofluorescence. Univariate analysis was used to determine the prognostic values of them. Cox proportional hazard regression model was used to identify independent risk factors that affect patients' overall survival and disease-free survival.Results: Among 119 eligible patients who had undergone surgical resection, the positive rate of PD-L1 was higher in metastatic lymph nodes than in primary tumors(45.4% vs. 38.7%, P = 0.005); the positive rate of PD-1 on CD8(+)T cells was significantly higher in primary tumors and metastatic lymph nodes than in tumor-free lymph nodes(both P < 0.001). The intensity of PD-1 expression on CD8(+) T cells in primary tumors and in metastatic lymph nodes were stronger than that in tumor-free lymph nodes from the same patient. Beside, the positive rate of PD-L2 did not show any differences between primary tumors and metastatic lymph nodes. | 5 | Circular RNA circStag1 promotes bone regeneration by interacting with HuR显示文摘Postmenopausal osteoporosis is a common bone metabolic disorder characterized by deterioration of the bone microarchitecture,leading to an increased risk of fractures.Recently,circular RNAs(circ RNAs)have been demonstrated to play pivotal roles in regulating bone metabolism.However,the underlying functions of circ RNAs in bone metabolism in postmenopausal osteoporosis remain obscure.Here,we report that circ Stag1 is a critical osteoporosis-related circ RNA that shows significantly downregulated expression in osteoporotic bone marrow mesenchymal stem cells(BMSCs)and clinical bone tissue samples from patients with osteoporosis.Overexpression of circ Stag1 significantly promoted the osteogenic capability of BMSCs.Mechanistically,we found that circ Stag1 interacts with human antigen R(Hu R),an RNA-binding protein,and promotes the translocation of Hu R into the cytoplasm.A high cytoplasmic level of Hu R led to the activation of the Wnt signaling pathway by stabilizing and enhancing low-density lipoprotein receptor-related protein 5/6(Lrp5/6)andβ-catenin expression,thereby stimulating the osteogenic differentiation of BMSCs.Furthermore,overexpression of circ Stag1 in vivo by circ Stag1-loaded adeno-associated virus(circ Stag1-AAV)promoted new bone formation,thereby preventing bone loss in ovariectomized rats.Collectively,we show that circ Stag1 plays a pivotal role in promoting the regeneration of bone tissue via Hu R/Wnt signaling,which may provide new strategies to prevent bone metabolic disorders such as postmenopausal osteoporosis. | Gaoyang Chen Canling Long Shang Wang Zhenmin Wang Xin Chen Wanze Tang Xiaoqin He Zhiteng Bao Baoyu Tan Jin Zhao Yongheng Xie Zhizhong Li Dazhi Yang Guozhi Xiao Songlin Peng | 2022 | Bone Research2022,10,3: | 7 |

| 7 | Dispersion–tribological property relationship in mineral oils containing 2D layered α-zirconium phosphate nanoplatelets显示文摘Inorganic nanomaterials exhibit superior friction-reduction and anti-wear properties in oils. In this study, 2D layered α-zirconium phosphate (α-ZrP) nanosheets intercalated with different amines have been synthesized to study their dispersion stabilities in lubricating oil and tribological applications. The intercalated amines should be sufficiently long and lipophilic to provide stabilization to α-ZrP nanosheets in mineral oil. The results of tribological tests illustrate that with the addition of well-dispersed nanosheets, the coefficient of friction (COF) and pin volume loss reduce by ~47% and 75%, respectively. The excellent dispersion stability enables the nanosheets to flow into the contact area at the beginning, and thereby protect the rubbing surface. A reduction in the van der Waals forces between the adjacent layers induced by the intercalated amines transforms the friction between adjacent layers from pin disk to sliding, leading to a decrease in the COF under hydrodynamic lubrication.
