| 7 | Overexpression of p27^(KIP1)induced cell cycle arrest in G_1 phase and subsequent apoptosis in HCC-9204 cell line显示文摘AIM We have previously reported that inducibleover-expression of Bak may prolong cell cycle inG1 phase and lead to apoptosis in HCC-9204 cells.This study is to investigate whether p27KIP1playsan important role in this process.METHODS In order to elucidate the exactfunction of p27KIP1in this process,a zinc induciblep27KIP1stable transfectant and transient p27KIP1-GFP fusion transfectant were constructed.Theeffects of inducible,p27KIP1on cell growth,cellcycle arrest and apoptosis were examined in themock,control pMD vector,and pMD-KIP1transfected HCC-9204 cells.RESULTS This p27KIP1-GFP transfectant maytransiently express the fusion gene.The cellgrowth was reduced by 35% at 48 h of p27KIP1induction with zinc treatment as determined bytrypan blue exclusion assay.These differencesremained the same after 72 h of p27KIP1expression,p27KIP1caused cell cycle arrest after24 h of induction,with 40% increase in G1population.Prolonged p27KIP1expression in thiscell line induced apoptotic cell death reflected byTUNEL assay.Fourty-eight h and 72 h of p27KIP1expression showed a characteristic DNA ladder onagarose gel electrophoresis. | 9 | Characteristics of atmospheric single particles during haze periods in a typical urban area of Beijing:A case study in October,2014显示文摘To investigate the composition and possible sources of particles, especially during heavy haze pollution, a single particle aerosol mass spectrometer(SPAMS) was deployed to measure the changes of single particle species and sizes during October of 2014, in Beijing. A total of 2,871,431 particles with both positive and negative spectra were collected and characterized in combination with the adaptive resonance theory neural network algorithm(ART-2a). Eight types of particles were classified: dust particles(dust, 8.1%), elemental carbon(EC, 29.0%), organic carbon(OC, 18.0%), EC and OC combined particles(ECOC, 9.5%),Na-K containing particles(Na K, 7.9%), K-containing particles(K, 21.8%), organic nitrogen and potassium containing particles(KCN, 2.3%), and metal-containing particles(metal,3.6%). Three haze pollution events(P1, P2, P3) and one clean period(clean) were analyzed,based on the mass and number concentration of PM_(2.5)and the back trajectory results from the hybrid single particle Lagrangian integrated trajectory model(Hysplit-4 model). Results showed that EC, OC and K were the major components of single particles during the three haze pollution periods, which showed clearly increased ratios compared with those in the clean period. | 12 | Daclatasvir plus asunaprevir in treatment-na?ve patients with hepatitis C virus genotype 1b infection显示文摘AIM To assess daclatasvir plus asunaprevir(d UAL) in treatment-na?ve patients from China's Mainland, Russia and South Korea with hepatitis C virus(HCV) genotype 1 b infection. METHODS Patients were randomly assigned(3:1) to receive 24 wk of treatment with d UAL(daclatasvir 60 mg once daily and asunaprevir 100 mg twice daily) beginning on day 1 of the treatment period(immediate treatment arm) or following 12 wk of matching placebo(placebodeferred treatment arm). The primary endpoint was a comparison of sustained virologic response at posttreatment week 12(SVR12) compared with the historical SVR rate for peg-interferon plus ribavirin(70%) among patients in the immediate treatment arm. The first 12 wk of the study were blinded. Safety was assessed in d UAL-treated patients compared with placebo patients during the first 12 wk(doubleblind phase), and during 24 wk of d UAL in both arms combined.RESULTS In total, 207 patients were randomly assigned to immediate(n = 155) or placebo-deferred(n = 52) treatment. Most patients were Asian(86%), female(59%) and aged < 65 years(90%). Among them, 13% had cirrhosis, 32% had IL28 B non-CC genotypes and 53% had baseline HCV RNA levels of ≥ 6 million IU/m L. Among patients in the immediate treatment arm, SVR12 was achieved by 92%(95% confidence interval: 87.2-96.0), which was significantly higher than the historical comparator rate(70%). SVR12 was largely unaffected by cirrhosis(89%), age ≥ 65 years(92%), male sex(90%), baseline HCV RNA ≥ 6 million(89%) or IL28 B non-CC genotypes(96%), although SVR12 was higher among patients without(96%) than among those with(53%) baseline NS5 A resistanceassociated polymorphisms(at L31 or Y93 H). during the double-blind phase, aminotransferase elevations were more common among placebo recipients than among patients receiving d UAL. during 24 wk of d UAL therapy(combined arms), the most common adverse events(≥ 10%) were elevated alanine aminotransferase and upper respiratory tract infection; emergent grade 3-4 laboratory abnormalities were infrequently observed, and all grade 3-4 aminotransferase abnormalities(alanine aminotransferase, n = 9; aspartate transaminase, n = 6) reversed within 8-11 d. Two patients discontinued d UAL treatment; one due to aminotransferase elevations, nausea, and jaundice and the other due to a fatal adverse event unrelated to treatment. | 14 | Eruption of the Continental Flood Basalts at -259 Ma in the Emeishan Large Igneous Province, SW China: Evidence from Laser Microprobe ^(40)Ar/^(39)Ar Dating显示文摘A suite of continental flood basalts sampled over a vast exposure and stratigraphic thickness in the Emeishan large igneous province (LIP), SW China was investigated for laser microprobe 40Ar/39Ar dating. There are two 40Ar/39Ar age groups for these basalts, corresponding to 259-246 Ma and 177-137 Ma, respectively. A well-defined isochron gives an eruption age of huge quantities of mafic magmas at 258.9±3.4 Ma, which is identical to previous dating and paleontological data. Much younger 40Ar/39Ar ages for some basalts with low-greenschist metamorphic facies probably recorded a late thermo-tectonic event caused by collision between the Yangtze and Qiangtang continental blocks during the Mesozoic, which resulted in the reset of argon isotope system. | 15 | Progress and Prospects of Salt Lake Research in China显示文摘China has unique salt lake resources, and they are distributed in the east of Eurasian salt lake subzone of the Northern Hemisphere Salt Lake Zone, mainly concentrated in the regions with modern mean annual precipitation lower than 500 mm. This paper preliminarily reviews the progress made in salt lake research in China for the past 60 years. In the research of Paleoclimate and paleoenvironment from salt lake sediments, a series of salts have been proposed to be indicators of paleoclimate, and have been well accepted by scholars. The chloride-sulfate depositional regions of the west Qaidam and the east Tarim have been revealed to be the drought center of China since the Quaternary, and more than 6 spreading stages of arid climate(salt forming) have been identified. Five pan-lake periods with highstands have been proved to exist during the late Quaternary on the Tibetan Plateau. In mineral resource prospecting and theories of the forming of salt deposits: the atlas(1:2500000) of hydrochemical zoning of salt lakes on the Tibetan Plateau has been compiled for the first time, revealing the zonal distribution and transition from carbonate type to chloride type from south to north and presenting corresponding mineral assemblages for different type of salt lakes; several large continental salt deposits have been discovered and the theory of continental potash deposition has been developed, including the salt deposition in deep basins surrounded by high mountains, the mineral deposition from multistage evolution through chains of moderate or shallow lakes with multilevels, the origin of potassium rich brines in gravel layers, and the forming of potassium deposits through the inheriting from ancient salt deposits, thus establishing the framework of 'Continental Potash Deposition Theory'; several new types of Mg-borate deposits have been discovered, including the ulexite and pinnoite bed in Da Qaidam Lake, Qinghai, the pinnoite and kurnakovite bed in Chagcam Caka, Tibet, the kurnakovite bed in Lake Nyer, and the corresponding model of borate deposition from the cooling and dissolution of boron rich brines was proposed based on principles of geology, physics and chemistry. The anti-floatation-cold crystallization method developed independently has improved the capacity of KCl production to 3 million tons per year for the Qarham, serving the famous brand of potash fertilizer products. One 1.2 million ton K-sulfate production line, the biggest in the world, has been built in Lop Nor, and K-sulfate of about 1.6 million tons was produced in 2015. Supported by the new technology, i.e. brine preparation in winter-cooling-solarization-isolation-lithium deposition from salt gradient solar pond' the highest lithium production base at Zabuye Lake(4421 m), Tibet, has been established, which is the first lithium production base in China that reaches the year production of 5000 tons of lithium carbonate. The concept of Salt lake agriculture(Salt land agriculture) has been established based on the mass growth of Dunaliella and other bacillus-algae and the occurrence of various halophytes in saltmarsh and salt saline-alkali lands, finding a new way to increase arable lands and develop related green industry in salt rich environments. | 16 | Effect of small interfering RNA targeting survivin gene on biological behaviour of bladder cancer显示文摘Background Bladder cancer is the most common type of urinary system tumours. It is frequently associated with genetic mutations that deregulate the cell cycle and render these tumours resistant to apoptosis. Survivin, a newly discovered member inhibitor of apoptosis protein (IAP) family in several human cancers, by inducing cell proliferation and inhibiting apoptosis is frequently activated in bladder cancer. We studied the influence of small interfering RNA (siRNA) targeting survivin on the biological behaviour of bladder cancer cells. Methods A double strand survivin target sequence specific siRNA was designed and synthesized. After transfection of bladder cancer cell line T24 by siRNA/liposome complex with increasing concentrations (50-200 nmol/L), the transfectant cells were intratumourally injected at different doses (5 μg or 50 μg). The effects were measured in vitro and in vivo. Results The selected siRNA efficiently down-regulated survivin mRNA expression in a dose and time dependent manner. The maximal effect was achieved at the concentration of 100 nmol/L, at which survivin expression level was down-regulated by 75.91%. The inhibition rate of cell growth was 55.29% (P<0.01) and the markedly increased apoptotic rate was 45.70% (P<0.01). In vivo intratumoural injection of 50 μg siRNA-survivin could notably prevent the growth of bladder cancer (P<0.01) in xenografted animals. Conclusion The application of siRNA-survivin could markedly inhibit survivin expression in bladder cancer cell line by inducing apoptosis and inhibiting the growth of the tumour. | 17 | Electrical stimulation of acupoint combinations against deep venous thrombosis in elderly bedridden patients after major surgery显示文摘OBJECTIVE:To compare the effects of electrical stimulation of different acupoint combinations among postoperative bedridden elderly patients on hemorheology and deep venous blood flow velocity and investigate the role of electrical stimulation against deep vein thrombosis(DVT).METHODS:From November 2010 to October 2011,a total of 160 elderly bedridden patients after major surgery were divided into the conventional care group,invigorating and promoting Qi group,blood-activating and damp-eliminating group,and acupoint-combination stimulation group.Whole blood viscosity,plasma viscosity,D-dimer levels,lower limb skin temperature,lower limb circumference,and flow velocities of the external iliac vein,femoral vein,popliteal vein,and deep calf veins in all patients were documented and compared among the four groups.RESULTS:Whole blood viscosity,plasma viscosity,D-dimer levels,and lower limb circumference were significantly reduced in the blood-activating and damp-eliminating group compared with the conventional care group(P<0.05) and were almost equal to those in the acupoint-combination stimulation group(P>0.05).Lower limb venous flow velocities were accelerated in the invigorating and promoting Qi group compared with the other groups,excluding the acupoint-combination stimulation group(P<0.05).CONCLUSION:Hemorheological indices in postoperative bedridden elderly patients were improved after combined electrical stimulation at Yinlingquan(SP 9) and Sanyinjiao(SP 6).Combined electrical stimulation at Zusanli(ST 36) and Taichong(LR 3),on the other hand,accelerated lower limb venous flow. | Lili Hou Cuiping Chen Lei Xu PeihaoYin Wen Peng | 2013 | Journal of Traditional Chinese Medicine2013,33,2: | 13 |
| 18 | Effects of water salinity and N application rate on water- and N-use efficiency of cotton under drip irrigation显示文摘In arid and semi-arid regions, freshwater scarcity and high water salinity are serious and chronic problems for crop production and sustainable agriculture development. We conducted a field experiment to evaluate the effect of irrigation water salinity and nitrogen(N) application rate on soil salinity and cotton yield under drip irrigation during the 2011 and 2012 growing seasons. The experimental design was a 3×4 factorial with three irrigation water salinity levels(0.35, 4.61 and 8.04 dS/m) and four N application rates(0, 240, 360 and 480 kg N/hm2). Results showed that soil water content increased as the salinity of the irrigation water increased, but decreased as the N application rate increased. Soil salinity increased as the salinity of the irrigation water increased. Specifically, soil salinity measured in 1:5 soil:water extracts was 218% higher in the 4.61 dS/m treatment and 347% higher in the 8.04 dS/m treatment than in the 0.35 dS/m treatment. Nitrogen fertilizer application had relatively little effect on soil salinity, increasing salinity by only 3%–9% compared with the unfertilized treatment. Cotton biomass, cotton yield and evapotranspiration(ET) decreased significantly in both years as the salinity of irrigation water increased, and increased as the N application rate increased regardless of irrigation water salinity; however, the positive effects of N application were reduced when the salinity of the irrigation water was 8.04 dS/m. Water use efficiency(WUE) was significantly higher by 11% in the 0.35 dS/m treatment than in the 8.04 dS/m treatment. There was no significant difference in WUE between the 0.35 dS/m treatment and the 4.61 dS/m treatment. The WUE was also significantly affected by the N application rate. The WUE was highest in the 480 kg N/hm2 treatment, being 31% higher than that in the 0 kg N/hm2 treatment and 12% higher than that in the 240 kg N/hm2 treatment. There was no significant difference between the 360 and 480 kg N/hm2 treatments. The N use efficiency(NUE) was significantly lower in the 8.04 dS/m treatment than in either the 4.61 dS/m or the 0.35 dS/m treatment. There was no significant difference in NUE between the latter two treatments. These results suggest that irrigation water with salinity <4.61 dS/m does not have an obvious negative effect on cotton production, WUE or NUE under the experimental conditions. | 19 | Combined hepatectomy and radiofrequency ablation versus TACE in improving survival of patients with unresectable BCLC stage B HCC显示文摘BACKGROUND: Combined hepatectomy and radiofrequency ablation(RFA) provides an additional treatment for patients with Barcelona Clinic Liver Cancer(BCLC) stage B hepatocellular carcinoma(HCC) who are conventionally deemed unresectable. This study aimed to analyze the outcome of this combination therapy by comparing it with transarterial chemoembolization(TACE). METHODS: We retrospectively reviewed 51 patients with unresectable BCLC stage B HCC who had received the combination therapy. We compared the survival of these patients with that of 102 patients in the TACE group(control). Prognostic factors associated with worse survival in the combination group were analyzed.RESULTS: No differences in tumor status and liver function were observed between the TACE group and combination group. The median survival time for the combination group and TACE group was 38(6-54) and 17(3-48) months, respectively(P<0.001). The combination group required longer hospitalization than the TACE group [8(5-14) days vs 4(2-9) days,P<0.001]. More than two ablations decreased the survival rate in the combination group. CONCLUSIONS: Combined hepatectomy and RFA yielded a better long-term outcome than TACE in patients with unresectable BCLC stage B HCC. | 20 | Recent progress on the recycling technology of Li-ion batteries显示文摘Lithium-ion batteries(LIBs)have been widely applied in portable electronic devices and electric vehicles.With the booming of the respective markets,a huge quantity of spent LIBs that typically use either LiFePO_(4) or Li N_(x)Co_(y)Mn_(z)O_(2) cathode materials will be produced in the very near future,imposing significant pressure for the development of suitable disposal/recycling technologies,in terms of both environmental protection and resource reclaiming.In this review,we firstly do a comprehensive summary of the-state-of-art technologies to recycle Li N_(x)Co_(y)Mn_(z)O_(2) and LiFePO_(4)-based LIBs,in the aspects of pretreatment,hydrometallurgical recycling,and direct regeneration of the cathode materials.This closed-loop strategy for cycling cathode materials has been regarded as an ideal approach considering its economic benefit and environmental friendliness.Afterward,as for the exhausted anode materials,we focus on the utilization of exhausted anode materials to obtain other functional materials,such as graphene.Finally,the existing challenges in recycling the LiFePO_(4) and Li N_(x)Co_(y)Mn_(z)O_(2) cathodes and graphite anodes for industrial-scale application are discussed in detail;and the possible strategies for these issues are proposed.We expect this review can provide a roadmap towards better technologies for recycling LIBs,shed light on the future development of novel battery recycling technologies to promote the environmental benignity and economic viability of the battery industry and pave way for the large-scale application of LIBs in industrial fields in the near future. | Yuqing Wang Ning An Lei Wen Lei Wang Xiaotong Jiang Feng Hou Yuxin Yin Ji Liang | 2021 | Journal of Energy Chemistry2021,30,4: | 10 |
