1NO and H_2O_2 induced by Verticillium dahliae toxins and its influence on the expression of GST gene in cotton suspension cells显示文摘Nitric oxide (NO) and hydrogen peroxide (H2O2) have been shown to be important signaling molecules that participate in the regulation of several physiological processes. In particular, they have significant role in plant resistance to pathogens by contributing to induction defense genes. Here, whether NO and H2O2 participate in the resistance responses against Verticillium dahliae toxins (VD-toxins) and their effects on the expression of GST gene are studied. The results reveal that NO and H2O2 are produced as part of a complex network of signals that respond to VD-toxins and may converge to function both synergistically and independently by inducing resistant responses. GST gene is potentially involved in the resistance mechanism in the cotton suspension cells. NO induces the expression of GST gene independently of H2O2. 2Inducible Positive Mutant Screening System to Unveil the Signaling Pathway of Late Blight Resistance显示文摘Late blight is the most devastating potato disease and it also causes serious yield loss in tomato.Several disease resistance genes (R genes) to late blight have been cloned from potato in the past decade.However,the resistance mechanisms remain elusive.Tomato and potato belong to the botanical family Solanaceace and share remarkably conserved genome structure.Since tomato is a model system in genetic and plant pathology research,we used tomato to develop a powerful mutant screening system that will greatly facilitate the analysis of the signaling pathway of resistance to Phytophthora infestans.First we proved that the R3a transgenic tomatoes developed specific hypersensitive cell death response (HR) to P.infestans strains carrying the corresponding avirulence gene Avr3a,indicating that the signaling pathway from the R3a-Avr3a recognition to HR is conserved between potato and tomato.Second,we generated transgenic tomatoes carrying both R3a and Avr3a genes,with the latter under the control of a glucocorticiod-inducible promoter.Dexamethasone induced expression of Avr3a and resulted in localized HR.This versatile system can be used to construct a mutant library to screen surviving mutants whose resistance signal transduction was interrupted,providing the basis to identify key genes involved in the resistance to late blight in Solanaceae.Zhiqi Jia Yanhonq Cui Ying Li Xiaoxuan Wang Yonqchen Du Sanwen Huang 2010Journal of Integrative Plant Biology2010,52,5:3

7Integrated microdevice with a windmill-like hole array for the clog-free,efficient,and self-mixing enrichment of circulating tumor cells显示文摘Circulating tumor cells(CTCs)have tremendous potential to indicate disease progression and monitor therapeutic response using minimally invasive approaches.Considering the limitations of affinity strategies based on their cost,effectiveness,and simplicity,size-based enrichment methods that involve low-cost,label-free,and relatively simple protocols have been further promoted.Nevertheless,the key challenges of these methods are clogging issues and cell aggregation,which reduce the recovery rates and purity.Inspired by the natural phenomenon that the airflow around a windmill is disturbed,in this study,a windmill-like hole array on the SU-8 membrane was designed to perturb the fluid such that cells in a fluid would be able to self-mix and that the pressure acting on cells or the membrane would be dispersed to allow a greater velocity.In addition,based on the advantages of fluid coatings,a lipid coating was used to modify the membrane surface to prevent cell aggregation and clogging of the holes.Under the optimal conditions,recovery rates of 93%and 90%were found for A549 and HeLa cells in a clinical simulation test of our platform with a CTC concentration of 20-100 cells per milliliter of blood.The white blood cell(WBC)depletion rate was 98.7%(n=15),and the CTC detection limit was less than 10 cells per milliliter of blood(n=6).Moreover,compared with conventional membrane filtration,the advantages of the proposed device for the rapid(2 mL/min)and efficient enrichment of CTCs without clogging were shown both experimentally and theoretically.Due to its advantages in the efficient,rapid,uniform,and clog-free enrichment of CTCs,our platform offers great potential for metastatic detection and therapy analyses.Hao Li Jinze Li Zhiqi Zhang Zhen Guo Changsong Zhang Zixu Wang Qiuquan Guo Chao Li Chuanyu Li Jia Yao Anran Zheng Jingyi Xu Qingxue Gao Wei Zhang Lianqun Zhou 2022Microsystems & Nanoengineering2022,8,1:1

10Responses of nitrification performance,triclosan resistome and diversity of microbes to continuous triclosan stress in activated sludge system显示文摘Triclosan(TCS)is commonly found in wastewater treatment plants,which often affects biological treatment processes.The responses of nitrification,antibiotic resistome and microbial community under different TCS concentrations in activated sludge system were evaluated in this study.The experiment was conducted in a sequencing batch reactor(SBR)for 240 days.Quantitative PCR results demonstrated that the abundance of ammonium oxidizing bacteria could be temporarily inhibited by 1 mg/L TCS and then gradually recovered.And the abundances of nitrite oxidizing bacteria(NOB)under 2.5 and 4 mg/L TCS were three orders of magnitude lower than that of seed sludge,which accounted for partial nitrification.When the addition of TCS was stopped,the abundance of NOB increased.The mass balance experiments of TCS demonstrated that the primary removal pathway of TCS changed from adsorption to biodegradation as TCS was continuously added into the SBR system.Moreover,TCS increased the abundance of mexB,indicating the efflux pump might be the main TCS-resistance mechanism.As a response to TCS,bacteria could secrete more protein(PN)than polysaccharide.Three-dimensional excitation-emission matrix revealed that tryptophan PN-like substances might be the main component in PN to resist TCS.High-throughput sequencing found that the relative abundances of Paracoccus,Pseudoxanthomonas and Thauera increased,which could secrete extracellular polymeric substances(EPS).And Sphingopyxis might be the main TCS-degrading bacteria.Overall,TCS could cause partial nitrification and increase the relative abundances of EPS-secreting bacteria and TCS-degrading bacteria.Da Zhang Jingfeng Gao Lifang Zhang Wenzhi Zhang Jingxin Jia Huihui Dai Zhiqi Wang 2020Journal of Environmental Sciences2020,32,6:1
11Electrically programmable phase-change photonic memory for optical neural networks with nanoseconds in situ training capability显示文摘Optical neural networks (ONNs), enabling low latency and high parallel data processing withoutelectromagnetic interference, have become a viable player for fast and energy-efficient processing andcalculation to meet the increasing demand for hash rate. Photonic memories employing nonvolatile phase-change materials could achieve zero static power consumption, low thermal cross talk, large-scale, andhigh-energy-efficient photonic neural networks. Nevertheless, the switching speed and dynamic energyconsumption of phase-change material-based photonic memories make them inapplicable for in situ training.Here, by integrating a patch of phase change thin film with a PIN-diode-embedded microring resonator,a bifunctional photonic memory enabling both 5-bit storage and nanoseconds volatile modulation wasdemonstrated. For the first time, a concept is presented for electrically programmable phase-changematerial-driven photonic memory integrated with nanosecond modulation to allow fast in situ training and zerostatic power consumption data processing in ONNs. ONNs with an optical convolution kernel constructedby our photonic memory theoretically achieved an accuracy of predictions higher than 95% when testedby the MNIST handwritten digit database. 12Climatic Change Characteristics of Thunderstorm in Zhangjiakou City显示文摘In order to understand the climatic change trend and characteristics of thunderstorm in Zhangjiakou City,and provide climate background basis for the thunder prevention and lightning disaster reduction in the region,observation data of the thunderstorm at Zhangjiakou ground meteorological station from 1960 to 2013 were statistically analyzed by linear fitting,climate trend rate,M-K test,guaranteed rate method and so on. The results showed that the average annual thunderstorm days reached 37. 7 d in Zhangjiakou City from 1960 to 2013,and was in the moderate thunderstorm range. In nearly 54 years,thunderstorm day was reducing at the rate of about 1. 21 d /10 a in the city. There was thunderstorm in other three seasons except winter,and thunderstorm frequently occurred in summer. Summer thunderstorm day was reducing at the rate of about 1. 29 d /10 a,which caused the reduction of the annual one. In summer,thunderstorm day of July reduced most significantly. There were two key points of abrupt reduction,which occurred in 1971 and 1998,on the change of thunderstorm days in Zhangjiakou in recent 54 years. The first thunderstorm date had an advancing trend in recent years,while the final date had not an obvious change. The possibility of thunderstorm was very small from late October to next March in Zhangjiakou City. The first thunderstorm date occurred mostly before middle May,and the period from middle and late May to early and middle September was the high incidence of thunderstorm. 14Efficient Simultaneous Detection of Metabolites Based on Electroenzymatic Assembly Strategy显示文摘Objective and Impact Statement:We describe an electroenzymatic mediator(EM)sensor based on an electroenzymatic assembly peak separation strategy,which can efficiently realize the simultaneous detection of 3 typical cardiovascular disease(CVD)metabolites in 5μl of plasma under one test.This work has substantial implications toward improving the efficiency of chronic CVD assessment.Introduction:Monitoring CVD of metabolites is strongly associated with disease risk.Independent and time-consuming detection in hospitals is unfavorable for chronic CVD management.Methods:The EM was flexibly designed by the cross-linking of electron mediators and enzymes,and 3 EM layers with different characteristics were assembled on one electrode.Electrons were transferred under tunable potential;3 metabolites were quantitatively detected by 3 peak currents that correlated with metabolite concentrations.Results:In this study,the EM sensor showed high sensitivity for the simultaneous detection of 3 metabolites with a lower limit of 0.01 mM.The linear correlation between the sensor and clinical was greater than 0.980 for 242 patients,and the consistency of risk assessment was 94.6%.Conclusion:Metabolites could be expanded by the EM,and the sensor could be a promising candidate as a home healthcare tool for CVD risk assessment.Anran Zheng Chao Li Shengkai Xu Zhen Guo Chuanyu Li Changsong Zhang Jia Yao Zhiqi Zhang Jinze Li Lutao Du Shasha Zhao Chuanxin Wang Wei Zhang Lianqun Zhou 2023Biomedical Engineering Frontiers2023,4,1:0
15Dual-bionic regenerative microenvironment for peripheral nerve repair显示文摘Autologous nerve grafting serves is considered the gold standard treatment for peripheral nerve defects;however,limited availability and donor area destruction restrict its widespread clinical application.Although the performance of allogeneic decellularized nerve implants has been explored,challenges such as insufficient human donors have been a major drawback to its clinical use.Tissue-engineered neural regeneration materials have been developed over the years,and researchers have explored strategies to mimic the peripheral neural microenvironment during the design of nerve catheter grafts,namely the extracellular matrix(ECM),which includes mechanical,physical,and biochemical signals that support nerve regeneration.In this study,polycaprolactone/silk fibroin(PCL/SF)-aligned electrospun material was modified with ECM derived from human umbilical cord mesenchymal stem cells(hUMSCs),and a dual-bionic nerve regeneration material was successfully fabricated.The results indicated that the developed biomimetic material had excellent biological properties,providing sufficient anchorage for Schwann cells and subsequent axon regeneration and angiogenesis processes.Moreover,the dual-bionic material exerted a similar effect to that of autologous nerve transplantation in bridging peripheral nerve defects in rats.In conclusion,this study provides a new concept for designing neural regeneration materials,and the prepared dual-bionic repair materials have excellent auxiliary regenerative ability and further preclinical testing is warranted to evaluate its clinical application potential.Yanjun Guan Zhiqi Ren Boyao Yang Wenjing Xu Wenjun Wu Xiangling Li Tieyuan Zhang Dongdong Li Shengfeng Chen Jun Bai Xiangyu Song Zhibo Jia Xing Xiong Songlin He Chaochao Li Fanqi Meng Tong Wu Jian Zhang Xiuzhi Liu Haoye Meng Jiang Peng Yu Wang 2023Bioactive Materials2023,,8:0
