| 6 | Artesunate synergizes with sorafenib to induce ferroptosis in hepatocellular carcinoma显示文摘Sorafenib is the first-line medication for advanced hepatocellular carcinoma(HCC),but it can only extend limited survival.It is imperative to find a combination strategy to increase sorafenib efficacy.Artesunate is such a preferred candidate,because artesunate is clinically well-tolerated and more importantly both drugs can induce ferroptosis through different mechanisms.In this study we investigated the combined effect of sorafenib and artesunate in inducing ferroptosis of HCC and elucidated the involved molecular mechanisms.We showed that artesunate greatly enhanced the anticancer effects of low dose of sorafenib against Huh7,SNU-449,and SNU-182 HCC cell lines in vitro and against Huh7 cell xenograft model in Balb/c nude mice.The combination index method confirmed that the combined effect of sorafenib and artesunate was synergistic.Compared with the treatment with artesunate or sorafenib alone,combined treatment induced significantly exacerbated lipid peroxidation and ferroptosis,which was blocked by N-acetyl cysteine and ferroptosis inhibitors liproxstatin-1 and deferoxamine mesylate,but not by inhibitors of other types of cell death(z-VAD,necrostatin-1 and belnacasan).In Huh7 cells,we demonstrated that the combined treatment induced oxidative stress and lysosome-mediated ferritinophagy,two essential aspects of ferroptosis.Sorafenib at low dose mainly caused oxidative stress through mitochondrial impairments and SLC7A11-invovled glutathione depletion.Artesunate-induced lysosome activation synergized with sorafenib-mediated pro-oxidative effects by promoting sequential reactions including lysosomal cathepsin B/L activation,ferritin degradation,lipid peroxidation,and consequent ferroptosis.Taken together,artesunate could be repurposed to sensitize sorafenib in HCC treatment.The combined treatment can be easily translated into clinical applications. | Zhong-jie Li Hui-qi Dai Xiao-wei Huang Ji Feng Jing-huan Deng Zi-xuan Wang Xiao-mei Yang Yu-jia Liu Yong Wu Pan-hong Chen Huan Shi Ji-gang Wang Jing Zhou Guo-dong Lu | 2021 | Acta Pharmacologica Sinica2021,42,2: | 31 |
| 7 | Development of single row automatic transplanting device for potted vegetable seedlings显示文摘In China,low degree of automation seriously affects the working efficiency and quality in vegetable transplanting.As one of the most important vegetables in China even in the world,tomato was taken as the research object in this study.An automatic single-row transplanting device was designed,based on the statistical analysis of the physical and mechanical properties of tomato seedlings of a typical variety.Based on the technology of mechatronics,the device integrated the functions of transporting seedling tray,automatic seedling extraction and mechanical planting.The kinematics orthogonality solution combined with the dynamic sequence solution method was used to optimize and analyze the kinematic parameters of the automatic seeding mechanism,and the“sickle”trajectory was obtained.According to the position and movement requirement for taking and dropping seedling,the mechanical conditions and the working parameters of key execution parts were obtained by using analytic drawing method to analyze the mechanical condition of seedling collecting mechanism.The transplanting experiment was conducted at room temperature of 25°C,and the age and moisture content of the seedlings were 40 d and 55%,respectively.The results showed that the highest success rate was 92.59%,and the lowest rate of leakage was 23.13%,when the transplanting frequency was 60 plants/min.The lowest success rate was 77.78%,and the highest rate of leakage was 38.75%,when transplanting frequency was 120 plants/min.When the transplanting frequency is between 60-90 plants/min,the device can meet the requirement of high speed transplanting for potted vegetable seedling. | Xin Jin Daoyi Li Hao Ma Jiangtao Ji Kaixuan Zhao Jing Pang | 2018 | International Journal of Agricultural and Biological Engineering2018,11,3: | 27 |

| 9 | Gegen Qinlian decoction enhances immunity and protects intestinal barrier function in colorectal cancer patients via gut microbiota显示文摘BACKGROUND We previously showed,using the Traditional Chinese Medicine System Pharmacology Database,that Gegen Qinlian decoction(GQD)had a direct antitumor effect,and was combined with programmed cell death protein(PD)-1 inhibitors to treat microsatellite stable(MSS)tumor-bearing mice.However,the effect of GQD on patients with colorectal cancer(CRC)is not clear.AIM To determine the therapeutic mechanism of GQD in improving immune function,reducing inflammation and protecting intestinal barrier function.METHODS Seventy patients with CRC were included in this study:37 in the control group and 33 in the treatment group.The proportions of CD4+T,CD8+T,natural killer(NK),NKT and T regulatory cells were measured by flow cytometry.Levels of the cytokines tumor necrosis factor(TNF)-α,interferon(IFN)-γ,interleukin(IL)-2,IL-6,IL-10 and serotonin(5-hydroxytryptamine;5-HT)in serum were assessed by enzyme-linked immunosorbent assay(ELISA).The expression of zonula occludens(ZO)-1,occludin,nuclear factor(NF)-κB and TNF-αin tumor and normal tissues was measured by immunohistochemistry.The composition of gut microbiota from patients in the treatment group was assessed using 16S rDNA analysis.RESULTS There were no adverse events in the treatment group.The proportion of CD4+T cells and NKT cells in the post-treatment group was significantly higher than that in the pre-treatment and control groups(P<0.05).The level of TNF-αin the posttreatment group was significantly lower than that in the pre-treatment and control groups(P<0.05).The concentration of 5-HT in the post-treatment group was significantly lower than that in the pre-treatment group(P<0.05).The expression of ZO-1 and occludin in tumor tissues in the treatment group was significantly higher than that in the control group(P<0.05).The expression of ZO-1 in normal tissues of the treatment group was significantly higher than that in the control group(P=0.010).Compared with the control group,expression of NF-κB and TNF-αin tumor tissues of the treatment group was significantly decreased(P<0.05).Compared with the pre-treatment group,GQD decreased the relative abundance of Megamonas and Veillonella.In addition,GQD increased the relative abundance of Bacteroides,Akkermansia and Prevotella.CONCLUSION GQD enhances immunity and protects intestinal barrier function in patients with CRC by regulating the composition of gut microbiota. | Yang Li Zhong-Xin Li Chen-Yang Xie Jing Fan Ji Lv Xin-Jian Xu Jian Lv Wen-Tao Kuai Yi-Tao Jia | 2020 | World Journal of Gastroenterology2020,26,48: | 23 |
| 10 | Effects of Urea and Controlled Release Urea Fertilizers on Methane Emission from Paddy Fields: A Multi-Year Field Study显示文摘A four-year(2008–2011) field study was implemented in a major rice-growing region of China to better understand the effect of urea and controlled release fertilier(CRF, thermoplastic resin-coated urea in this study) on CH4 emission from paddy fields. Over the four years, the average CH4 emission during the rice growing seasons was 76.9, 65.8 and 64.9 kg CH4ha-1in treatments CK(zero N), U(urea) and C(CRF), respectively. Urea and CRF significantly reduced CH4 emission by 14.4% and 15.6%, and increased average rice grain yield by 25.8% and 19.7%(P < 0.05), respectively, compared with treatment CK. Flooding duration would affect CRF's effect on CH4 emission from paddy fields. Under normal aeration conditions, CH4 emission tended to be 3.9%–15.2% lower in treatment C than in treatment U from 2009 to 2011, while it tended to be 4.2% higher under delayed aeration conditions in 2008. The findings suggest that mid-season aeration(MSA) starting on D30(30 days after rice transplanting), just like the local practice, would optimize the CRF's effect on CH4 emission from rice fields in China. | 13 | Asiatic acid inhibits lung cancer cell growth in vitro and in vivo by destroying mitochondria显示文摘Asiatic acid(AA), a pentacyclic triterpene found in Centella asiatica, displays significant antiproliferative effects on cancer cells in vitro although the underlying mechanism of this effect remains unknown. This study investigated the efficacy and mechanism of action of AA against lung cancer both in vivo and in vitro. Using the MTT assay, AA was found to induce apoptosis in a dose- and timedependent manner, an effect enhanced by pretreatment with an autophagy inhibitor. It also elevated expression of microtubule-associated protein 1 light chain 3(LC3) and decreased the expression of p62.Furthermore, exposure to AA resulted in collapse of the mitochondrial membrane potential and generation of reactive oxygen species(ROS), suggesting mitochondria are the target of AA. In the mouse lung cancer xenograft model, oral administration of AA significantly inhibited tumor volume and weight accompanied by significant apoptosis of lung cancer cells. In addition, it led to a significant decrease in the expression of proliferating cell nuclear antigen(PCNA). | 14 | Establishing eutrophication assessment standards for four lake regions, China显示文摘The trophic status assessment of lakes in diferent lake regions may provide important and fundamental information for lake trophic state classification and eutrophication control. In this study, a region-specific lake eutrophication assessment standard was established through a frequency distribution method based on chlorophyll-a concentration. The assessment standards under the oligotrophic state for lakes in the Eastern plain, Yungui Plateau, Northeast Plain and Mountain Mongolia-Xinjiang regions are total phosphorus of 0.068,0.005, 0.011, 0.005 mg/L; total nitrogen of 1.00, 0.16, 0.37, 0.60 mg/L; Secchi depth of 0.60, 8.00, 1.55, 3.00 m; and CODMn of2.24, 1.00, 5.11, 4.00 mg/L, respectively. Moreover, a region-specific comprehensive trophic level index was developed to provide an understandable assessment method for the public. The results indicated that the frequency distribution analysis based on chlorophyll-a combined with trophic level index provided a useful metric for the assessment of the lake trophic status. In addition, the diference of eutrophication assessment standards in diferent lake regions was analyzed, which suggested that the sensitivities of algae to nutrients and the assessment standard of trophic status possessed significant regional diferences for the four lake ecoregions. | 17 | Dynamic performance of angle-steel concrete columns under low cyclic loading-II:parametric study显示文摘Tests of nine angle-steel concrete column (ASCC) specimens under low cyclic loading are described in a companion paper (Zheng and Ji, 2008). In this paper, the skeleton curves from the numerical simulation are presented, and show good agreement with the test results. Furthermore, parametric studies are conducted to explore the influence of factors such as the axial compression ratio, shear steel plate ratio, steel ratio, prismatic concrete compression strength, yield strength of angle steel and shear span ratio, etc., on the monotonic load-displacement curves of the ASCCs. Based on a statistical analysis of the calculated results, hysteretic models for load-displacement and moment-curvature are proposed, which agree well with the test results. | 18 | Microwave Exposure Impairs Synaptic Plasticity in the Rat Hippocampus and PC12 Cells through Over-activation of the NMDA Receptor Signaling Pathway显示文摘Objective The aim of this study is to investigate whether microwave exposure would affect the N-methyl-D-aspartate receptor(NMDAR) signaling pathway to establish whether this plays a role in synaptic plasticity impairment. Methods 48 male Wistar rats were exposed to 30 m W/cm2 microwave for 10 min every other day for three times. Hippocampal structure was observed through H&E staining and transmission electron microscope. PC12 cells were exposed to 30 m W/cm2 microwave for 5 min and the synapse morphology was visualized with scanning electron microscope and atomic force microscope. The release of amino acid neurotransmitters and calcium influx were detected. The expressions of several key NMDAR signaling molecules were evaluated. Results Microwave exposure caused injury in rat hippocampal structure and PC12 cells, especially the structure and quantity of synapses. The ratio of glutamic acid and gamma-aminobutyric acid neurotransmitters was increased and the intracellular calcium level was elevated in PC12 cells. A significant change in NMDAR subunits(NR1, NR2 A, and NR2B) and related signaling molecules(Ca2+/calmodulin-dependent kinase II gamma and phosphorylated c AMP-response element binding protein) were examined. Conclusion 30 m W/cm2 microwave exposure resulted in alterations of synaptic structure, amino acid neurotransmitter release and calcium influx. | 20 | Adipose-derived mesenchymal stem cells protect against CMS-induced depression-like behaviors in mice via regulating the Nrf2/HO-1 and TLR4/NF-κB signaling pathways显示文摘Increasing studies show that inflammatory processes may be involved in depressive disorders.Nuclear factor erythroid-2 related factor 2(Nrf2)modulates tissue microglial M1 phenotypic changes to the M2 phenotype,which is implicated in protection against inflammatory diseases.We have reported that the adipose-derived mesenchymal stem cells(ADSCs)display anti-inflammatory activity.In this study we explored whether the mechanism of anti-inflammatory activity of ADSCs was related to Nrf2.ADSCs were isolated from mouse fat pads and intravenously administered to chronic mild stress(CMS)-exposed C57BL/6 mice at the dose of 1×106 once a week for 3 weeks.We showed that ADSC administration significantly remedied CMS-induced depressive-like behaviors in sucrose preference test,tail suspension test,and forced swim test accompanied by suppressing microglial activation and the expression of inflammatory factors including MCP-1,TNF-α,IL-1β,and IL-6.Furthermore,ADSC administration promoted both the expression of BDNF and TrkB,and promoted Nrf2/HO-1 signaling but suppressed TLR4/NF-κB signaling in brain tissue.In order to elucidate the role of Nrf2/HO-1 signaling in ADSC-caused neuroprotection,Nrf2-modified ADSCs were cocultured with BV2 microglial cells,then exposed to lipopolysaccharide(LPS).Downregulation of Nrf2 in ADSCs decreased the protective effects of ADSCs against LPS-induced microglial activation and M1 polarization.Nrf2 overexpression in ADSCs markedly suppressed LPS-induced TLR4 and NF-κB expression in microglial cells.These results suggest a possible antidepressive mechanism correlated with microglial polarization for anti-inflammatory agents,which may provide a new microglia-targeted strategy for depression therapy. | Xiao Huang Guo-qiang Fei Wen-juan Liu Jing Ding Yuan Wang Hao Wang Jian-lin Ji Xin Wang | 2020 | Acta Pharmacologica Sinica2020,41,5: | 15 |
