1Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. 2Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 3The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 4Superabsorbent Fibers for Comfortable Disposable Medical Protective Clothing显示文摘Disposable medical protective clothing for 2019-nCoV mainly consists of stacked layers with nanopore films,polymer coated nonwoven fabrics and melt-blown nonwoven fabrics against anti-microbial and anti-liquid penetration.However,such structures lack moisture permeability and breathability leading to an uncomfortable,stuffy wearing experience.Here,we propose a novel medical protective clothing material with a superabsorbent layer to enhance moisture absorption.Poly(acrylic acid-co-acrylamide)/polyvinyl alcohol superabsorbent fibers(PAAAM/PVA fibers)were prepared via wet spinning.And the superabsorbent composite layer was stacked from PAAAM/PVA fibers,bamboo pulp fibers(BPF)and ethylene-propyl-ene side by side fibers(ESF).The novel disposable medical protective composite fabric was obtained through gluing the superabsorbent layer to the inner surface of strong antistatic polypropylene nonwoven fabric.The resultant composite fabric possesses excellent absorption and retention capacity for sweat,up to 12.3 g/g and 63.8%,and a maximum hygroscopic rate of 1.04 g/h,higher than that of the conventional material(only 0.53 g/h).The moisture permeability of the novel material reached 12,638.5 g/(m^(2) d),which was 307.6%of the conventional material.The novel material can effectively reduce the humidity inside the protective clothing and significantly improve the comfort of medical staff.Lin Yang Hong Liu Shuai Ding Jiawei Wu Yan Zhang Zhongzhen Wang Lili Wei Mingwei Tian Guangming Tao 2020Advanced Fiber Materials2020,2,3:11
5The chromosome-level reference genome assembly for Panax notoginseng and insights into ginsenoside biosynthesis显示文摘Panax notoginseng,a perennial herb of the genus Panax in the family Araliaceae,has played an important role in clinical treatment in China for thousands of years because of its extensive pharmacological effects.Here,we report a high-quality reference genome of P.notoginseng,with a genome size up to 2.66 Gb and a contig N50 of 1.12 Mb,produced with third-generation PacBio sequencing technology.This is the first chromosome-level genome assembly for the genus Panax.Through genome evolution analysis,we explored phylogenetic and whole-genome duplication events and examined their impact on saponin biosynthesis.We performed a detailed transcriptional analysis of P.notoginseng and explored genelevel mechanisms that regulate the formation of characteristic tubercles.Next,we studied the biosynthesis and regulation of saponins at temporal and spatial levels.We combined multi-omics data to identify genes that encode key enzymes in the P.notoginseng terpenoid biosynthetic pathway.Finally,we identified five glycosyltransferase genes whose products catalyzed the formation of different ginsenosides in P.notoginseng.The genetic information obtained in this study provides a resource for further exploration of the growth characteristics,cultivation,breeding,and saponin biosynthesis of P.notoginseng.Zhouqian Jiang Lichan Tu Weifei Yang Yifeng Zhang Tianyuan Hu Baowei Ma Yun Lu Xiuming Cui Jie Gao Xiaoyi Wu Yuru Tong Jiawei Zhou Yadi Song Yuan Liu Nan Liu Luqi Huang Wei Gao 2021Plant Communications2021,2,1:11

8Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8

10Modeling and simulation of urea-water-solution droplet evaporation and thermolysis processes for SCR systems显示文摘A reliable mathematical model of urea-water-solution(UWS) droplet evaporation and thermolysis is developed.The well known Abramzon–Sirignano evaporation model is corrected by introducing an adjustment coefficient considering the different evaporation behaviors of UWS droplet at different ambient temperatures. A semidetailed kinetic scheme of urea thermolysis is developed based on Ebrahimian's work. Sequentially, the evaporation characteristics, decomposition efficiency of a single UWS droplet and deposit formation are simulated. As a result, the relation of evaporation time, relative velocity, exhaust temperature and droplet initial diameter is presented. Synchronously, it indicates that temperature is the decisive factor for urea thermolysis. Different temperatures result in different deposit components, and deposit yield is significantly influenced by temperature and decomposition time. 11Mg-Zn-Y-Nd coated with citric acid and dopamine by layer-by-layer self-assembly to improve surface biocompatibility显示文摘Magnesium alloy has been generally accepted as an important biodegradable material on cardiovascular stent development for a long time. However, its limited biocompatibility, especially delayed endothelialization process restricts its further application. In this contribution, we modified the Mg-Zn-Y-Nd alloy surface with citric acid and dopamine via a layer-by-layer self-assembly assay, aiming at improving the biocompatibility of the magnesium alloy. The citric acid/dopamine(CA/PDA) layer exhibited a remarkable suppression of platelet activation/aggregation and thrombosis under 15 dyn/cm2 blood flowing. Inhibition on vascular smooth muscle cells growth and macrophages attachment/activation were also observed on this layer. In particular, the CA/PDA layer presented a promoted property for the vascular endothelial cells growth and spreading compared with the bare magnesium alloy, suggesting the pro-endotelialized function. 12Crystal Structure of the MH2 domain of Drosophila Mad显示文摘The decapentaplegic(Dpp),a member of the TGF-β superfamily,plays a pivotal role in the control of proliferation,global patterning and induction of specific cell fates during Drosophila development.Mother against Dpp(Mad) is the founding member of the conserved Smad protein family which spe-cifically transduces the intracellular TGF-β signaling cascade.Here we report the 2.80 structure of the MH2 domain of Mad(Mad-MH2) that was readily superposed to the mammal Smad-MH2 structures.This unphosphorylated Mad-MH2 forms a symmetric homotrimer in crystals,consistent with the result of the size-exclusion chromatography that Mad-MH2 exhibited a propensity for concentration-dependent oligomerization prior to phosphorylation.Structural analysis revealed that the formation of homotrimeric Mad-MH2 is mainly mediated by contacts involving the extreme C-terminal SSVS motif,and is strengthened by phosphorylation of the last two Ser residues which was confirmed by the gel filtration analysis of the pseudophosphorylated Mad-MH2(DVD).Intriguingly,the homotrimer within an asymmetric unit only possesses two ordered C-terminal tails,reminiscent of the arrangement of the R-Smad/Smad4 complexes,indicating that the subunit with a flexible SSXS motif would be readily replaced by Co-Smad to form a functional heterotrimer.WANG Chong,CHEN Lei,WANG Le & WU JiaWei MOE Key Laboratory of Bioinformatics,Department of Biological Sciences and Biotechnology,Tsinghua University,Beijing 100084,China 2009Science China(Life Sciences)2009,52,6:5

14Experimental free-space quantum secure direct communication and its security analysis显示文摘We report an experimental implementation of free-space quantum secure direct communication based on single photons.The quantum communication scheme uses phase encoding,and the asymmetric Mach–Zehnder interferometer is optimized so as to automatically compensate phase drift of the photons during their transitions over the free-space medium.At a 16 MHz pulse repetition frequency,an information transmission rate of 500 bps over a 10 m free space with a mean quantum bit error rate of 0.49%±0.27%is achieved.The security is analyzed under the scenario that Eve performs the collective attack for single-photon state and the photon number splitting attack for multi-photon state in the depolarizing channel.Our results show that quantum secure direct communication is feasible in free space.DONG PAN ZAISHENG LIN JIAWEI WU HAORAN ZHANG ZHEN SUN DONG RUAN LIUGUO YIN AND GUI LU LONG 2020Photonics Research2020,8,9:5
15Design and Implementation of an Adaptive Feedback Queue Algorithm over Open Flow Networks显示文摘The concurrent presence of different types of traffic in multimedia applications might aggravate a burden on the underlying data network, which is bound to affect the transmission quality of the specified traffic. Recently, several proposals for fulfilling the quality of service(QoS) guarantees have been presented. However, they can only support coarse-grained QoS with no guarantee of throughput, jitter, delay or loss rate for different applications. To address these more challenging problems, an adaptive scheduling algorithm for Parallel data Processing with Multiple Feedback(PPMF) queues based on software defined networks(SDN) is proposed in this paper, which can guarantee the quality of service of high priority traffic in multimedia applications. PPMF combines the queue bandwidth feedback mechanism to realise the automatic adjustment of the queue bandwidth according to the priority of the packet and network conditions, which can effectively solve the problem of network congestion that has been experienced by some queues for a long time. Experimental results show PPMF significantly outperforms other existing scheduling approaches in achieving 35--80% improvement on average time delay by adjusting the bandwidth adaptively, thus ensuring the transmission quality of the specified traffic and avoiding effectively network congestion.Jiawei Wu Xiuquan Qiao Junliang Chen 2018China Communications2018,15,7:5

17City-specific vehicle emission control strategies to achieve stringent emission reduction targets in China's Yangtze River Delta region显示文摘The Yangtze River Delta(YRD) region is one of the most prosperous and densely populated regions in China and is facing tremendous pressure to mitigate vehicle emissions and improve air quality.Our assessment has revealed that mitigating vehicle emissions of NOx would be more difficult than reducing the emissions of other major vehicular pollutants(e.g.,CO,HC and PM_(2.5)) in the YRD region.Even in Shanghai,where the emission control implemented are more stringent than in Jiangsu and Zhejiang,we observed little to no reduction in NOx emissions from 2000 to 2010.Emission-reduction targets for HC,NOx and PM_(2.5) are determined using a response surface modeling tool for better air quality.We design city-specific emission control strategies for three vehicle-populated cities in the YRD region:Shanghai and Nanjing and Wuxi in Jiangsu.Our results indicate that even if stringent emission control consisting of the Euro 6/VI standards,the limitation of vehicle population and usage,and the scrappage of older vehicles is applied,Nanjing and Wuxi will not be able to meet the NOx emissions target by 2020.Therefore,additional control measures are proposed for Nanjing and Wuxi to further mitigate NOx emissions from heavy-duty diesel vehicles.Shaojun Zhang Ye Wu Bin Zhao Xiaomeng Wu Jiawei Shu Jiming Hao 2017Journal of Environmental Sciences2017,29,1:4

19Rational design of hierarchically porous Fe-N-doped carbon as efficient electrocatalyst for oxygen reduction reaction and Zn-air batteries显示文摘The rational design and construction of hierarchically porous nanostructure for oxygen reduction reaction (ORR) electrocatalysts is crucial to facilitate the exposure of accessible active sites and promote the mass/electron transfer under the gas-solid-liquid triple-phase condition. Herein, an ingenious method through the pyrolysis of creative polyvinylimidazole coordination with Zn/Fe salt precursors is developed to fabricate hierarchically porous Fe-N-doped carbon framework as efficient ORR electrocatalyst. The volatilization of Zn species combined with the nanoscale Kirkendall effect of Fe dopants during the pyrolysis build the hierarchical micro-, meso-, and macroporous nanostructure with a high specific surface area (1,586 m^(2)·g^(−1)), which provide sufficient exposed active sites and multiscale mass/charge transport channels. The optimized electrocatalyst exhibits superior ORR activity and robust stability in both alkaline and acidic electrolytes.
