1Acupuncture treatment of lung-spleen Qi deficiency in stable chronic obstructive pulmonary disease: a randomized, open-label, controlled trial显示文摘OBJECTIVE:To evaluate the effectiveness and safety of acupuncture that reinforces the spleen to strengthen the lung in patients with stable chronic obstructive pulmonary disease(COPD).METHODS:This was a randomized,open-controlled trial in which the acupuncturist and the participants were not blinded,but the outcome evalu-ators and data analysts were blinded.One-hundred-and-two patients with stable COPD were randomly divided into two groups in a 1∶1 ratio.The acupuncture group received 30-minute sessions of acupuncture therapy at the same acupoints three times weekly for 6 weeks in addition to routine conventional Western Medicine treatment;the control group received routine conventional Western Medicine treatment alone.The primary outcome was the Borg scale score,which was assessed immediately after the 6-minute walk test.The secondary outcomes were the 6-minute walk distance,lung function,and oxygen saturation.Measurements were obtained at baseline and after 6 weeks of treatment.RESULTS:After 6 weeks of treatment,the Borg scale score in the acupuncture group was significantly better than that in the control group(2.02±0.71 versus 5.01±0.34,P<0.05).Furthermore,the post-treatment improvements in the 6-minute walk distance,lung function,and oxygen saturation were significantly greater in the acupuncture group than in the control group,showing that the acupuncture group had better exercise tolerance.CONCLUSION:The findings suggest that acupuncture that aims to reinforce the spleen to strengthen the lung is a safe and effective adjuvant therapy that effectively improves the exercise capacity of patients with stable COPD.Li Yu Xiong Chan Zeng Yan Wei Hua Zhuang Guangtong Zhao Lihong Li Chenyi Li Lin Qin Erqi Chen Xiaoqin Fu Juanjuan 2019Journal of Traditional Chinese Medicine2019,39,6:13
2Potentiating CD8^(+) T cell antitumor activity by inhibiting PCSK9 to promote LDLR-mediated TCR recycling and signaling显示文摘Metabolic regulation has been proven to play a critical role in T cell antitumor immunity.However,cholesterol metabolism as a key component of this regulation remains largely unexplored.Herein,we found that the low-density lipoprotein receptor(LDLR),which has been previously identified as a transporter for cholesterol,plays a pivotal role in regulating CD8+T cell antitumor activity.Besides the involvement of cholesterol uptake which is mediated by LDLR in T cell priming and clonal expansion,we also found a non-canonical function of LDLR in CD8+T cells:LDLR interacts with the T-cell receptor(TCR)complex and regulates TCR recycling and signaling,thus facilitating the effector function of cytotoxic T-lymphocytes(CTLs).Furthermore,we found that the tumor microenvironment(TME)downregulates CD8+T cell LDLR level and TCR signaling via tumor cell-derived proprotein convertase subtilisin/kexin type 9(PCSK9)which binds to LDLR and prevents the recycling of LDLR and TCR to the plasma membrane thus inhibits the effector function of CTLs.Moreover,genetic deletion or pharmacological inhibition of PCSK9 in tumor cells can enhance the antitumor activity of CD8+T cells by alleviating the suppressive effect on CD8+T cells and consequently inhibit tumor progression.While previously established as a hypercholesterolemia target,this study highlights PCSK9/LDLR as a potential target for cancer immunotherapy as well.Juanjuan Yuan Ting Cai Xiaojun Zheng Yangzi Ren Jingwen Qi Xiaofei Lu Huihui Chen Huizhen Lin Zijie Chen Mengnan Liu Shangwen He Qijun Chen Siyang Feng Yingjun Wu Zhenhai Zhang Yanqing Ding Wei Yang 2021Protein & Cell2021,12,4:12

4Organic Carbon in the Surface Sediments from the Intensive Mariculture Zone of Sanggou Bay: Distribution, Seasonal Variations and Sources显示文摘Mariculture has rapidly grown worldwide, which might greatly influence the biogeochemical cycle of organic carbon in coastal seas. In this study, several geochemical parameters, including grain size composition, sedimentary total organic carbon (TOC), total nitrogen (TN), stable carbon (δ13C) and nitrogen (δ15N) isotopic compositions, were analyzed for surface sediments collected from different mariculture zones of Sanggou Bay and in different seasons. We investigated the composition and distribution of or- ganic matter in surface sediments and further evaluated the contribution of mariculture activities to TOC sources. The TOC and TN contents (mass percentage) in the bay were in the range of 0.14% to 1.45% and 0.03% to 0.20%, respectively. The spatial distribution indicated that sedimentary TOC and TN contents in shellfish monoculture and shellfish-kelp polyculture zones were higher than in other mariculture zones, which might be related to grain size composition and mariculture organisms. Seasonal variations of TOC contents were observed in different mariculture zones. The TOC/TN atomic ratio (C/N),δ13C and δ15N were in the ranges of 5.97 to 10.97, 21.76‰ to 13.14‰ and 2.13‰ to 8.08‰, respectively, implying that sedimentary organic matter in Sanggou Bay was the mixture of marine phytoplankton, terrestrial and maricultural sources. A simple mixing model based on δ13C was applied and the results indicated that the relative contributions of organic carbon sources in Sanggou Bay followed the order kelp (36.6%) marine phytoplankton (28.7%) shellfish bio-deposition (23.8%) terrestrial input (10.9%). 11Association Between Lipid Profiles and Left Ventricular Hypertrophy:New Evidence from a Retrospective Study显示文摘Objective To explore the association between lipid profiles and left ventricular hypertrophy in a Chinese general population.Methods We conducted a retrospective observational study to investigate the relationship between lipid markers[including triglycerides,total cholesterol,low-density lipoprotein cholesterol,high-density lipoprotein(HDL)cholesterol,non-HDL-cholesterol,apolipoprotein A-I,apolipoprotein B,lipoprotein[a],and composite lipid profiles]and left ventricular hypertrophy.A total of 309,400 participants of two populations(one from Beijing and another from nationwide)who underwent physical examinations at different health management centers between 2009 and 2018 in China were included in the cross-sectional study.7,475 participants who had multiple physical examinations and initially did not have left ventricular hypertrophy constituted a longitudinal cohort to analyze the association between lipid markers and the new-onset of left ventricular hypertrophy.Left ventricular hypertrophy was measured by echocardiography and defined as an end-diastolic thickness of the mterventricular septum or left ventricle posterior wall>11 mm.The Logistic regression model was used in the cross-sectional study.Cox model and Cox model with restricted cubic splines were used in the longitudinal cohort.Results In the cross-sectional study for participants in the highest tertile of each lipid marker compared to the respective lowest,triglycerides[odds ratio(OR):1.2S0,95%CI:1.060 to 1.474],HDL-cholesterol(OR:0.780,95%CI:0.662 to 0.918),and lipoprotein(a)(OR:1.311,95%C7:1.115 to 1.541)had an association with left ventricular hypertrophy.In the longitudinal cohort,for participants in the highest tertile of each lipid marker at the baseline compared to the respective lowest,triglycerides[hazard ratio(HR):3.277,95%C/:1.720 to 6.244],HDL-cholesterol(HR:0.516,95%C7:0.283 to 0.940),non-HDL-cholesterol(HR:2.309,95%C/:1.296 to 4.112),apolipoprotein B(HR:2.244,95%CI:1.251 to 4.032)showed an association with new-onset left ventricular hypertrophy.In the Cox model with forward stepwise selection,triglycerides were the only lipid markers entered into the final model.Conclusion Lipids levels,especially triglycerides,are associated with left ventricular hypertrophy.Controlling triglycerides level potentiate to be a strategy in harnessing cardiac remodeling but deserve to be furdier investigated.Xuewei Huang Keqiong Deng Juanjuan Qin Fang Lei Xingyuan Zhang Wenxin Wang Lijin Lin Yuming Zheng Dongai Yao Huiming Lu Feng Liu Lidong Chen Guilan Zhang Yueping Liu Qiongyu Yang Jingjing Cai Zhigang She Hongliang Li 2022Chinese Medical Sciences Journal2022,37,2:1

13Engineering of Reversible Luminescent Probes for Real-Time Intravital Imaging of Liver Injury and Repair显示文摘As the major organ for drug metabolism and detoxification,the liver is prone to damage and severely impaired functionality.The treatment of liver diseases is based on a clear understanding of the process underlying liver injury and repair.However,intravital real-time imaging of liver injury and repair is still limited due to the lack of in vivo reversible visualization methods.To this end,we proposed a rational design strategy for the development of a reversible upconversion luminescence nanoprobe that allows real-time and in vivo imaging of liver injury and repair processes.As a proof of concept,we first developed a small molecule probe NB3 which can reversibly respond to related analytes of early liver injury[peroxynitrite(ONOO−)]and liver repair[glutathione(GSH)].The small molecule probe was then integrated with a core–shell upconversion nanoparticle to form a sophisticated nanoprobe.Compared with traditional small molecule probes,this nanoprobe exhibited a higher selectivity to ONOO−,longer retention time in liver,and wider dynamic response range to GSH after oxidation by ONOO−.The novel nanoprobe facilitated the successful monitoring and discrimination among the different degrees of liver injury and repair in a mouse model.Xiao Liu Linhui He Xiangyang Gong Yue Yang Dan Cheng Juanjuan Peng Lu Wang Xiao-Bing Zhang Lin Yuan 2022CCS Chemistry2022,4,1:1

15Biophysical characterization and ligand-binding properties of the elongation factor Tu from Mycobacterium tuberculosis显示文摘Mycobacterium tuberculosis(Mtb)is the key devastating bacterial pathogen responsible for tuberculosis.Increasing emergence of multi-drug-resistant,extensively drug-resistant,and rifampicin/isoniazid-resistant strains of Mtb makes the discovery of validated drug targets an urgent priority.As a vital translational component of the protein biosynthesis system,elongation factor Tu(EF-Tu)is an important molecular switch responsible for selection and binding of the cognate aminoacyl-tRNA to the acceptor site on the ribosome.In addition,EF-Tu from Mtb(MtbEF-Tu)is involved in the initial step of trans-translation which is an effective system for rescuing the stalled ribosomes from non-stop translation complexes under stress conditions.Given its crucial role in protein biosynthesis,EF-Tu is identified as an excellent molecular target for drug design.Here,we reported the recombinant expression,purification,biophysical characterization,and structural modeling of the MtbEF-Tu protein.Our results demonstrated that prokaryotic expression plasmids of pET28a-MtbEF-Tu could be expressed efficiently in Escherichia coli.We successfully purified the 6× His-tagged proteins with a yield of 16.8 mg from 1 l of Luria Bertani medium.Dynamic light scattering experiments showed that MtbEF-Tu existed in a monomeric form,and circular dichroism experiments indicated that MtbEF-Tu was well structured.Moreover,isothermal titration calorimetry experiments displayed that the purified MtbEF-Tu protein possessed intermediate binding affinities for guanosine-5′-triphosphate(GTP)and GDP.The GTP/GDP-binding sites were predicted by flexible molecular docking approach which reveals that GTP/GDP binds to MtbEF-Tu mainly through hydrogen bonds.Our work lays the essential basis for further structural and functional studies of MtbEF-Tu as well as MtbEF-Tu-related novel drug developments.Juanjuan Yang Jing Hong Ling Luo Ke Liu Chun Meng Zhi-liang Ji Donghai Lin 2019Acta Biochimica et Biophysica Sinica2019,51,2:1

18Electrocatalytic oxidation of GMP on an ITO electrode modified by the photodeposition of Pd nanoparticles onto a monolayer TiO_2 nanosheets/[Ru(phen)_2(dC18bpy)]^(2+) hybrid film显示文摘An indium tin oxide(ITO)electrode coated with monolayer TiO2/[Ru(phen)2(dC18bpy)] 2+ (phen=1,10-phenanthroline, dC18bpy=4,4′-dioctadecyl-2,2′-bipyridyl)hybrid film(denoted as ITO/TiO2-Ru)has been prepared using the modified Langmuir-Blodgett(LB)method,and the electrocatalytic oxidation of mononucleotide of guanosine 5′-monophosphate(GMP)on an ITO/TiO2-Ru electrode after Pd-photodeposition(denoted as ITO/TiO2-Ru/Pd)has been studied.Atomic force microscopy reveals that the single-layered hybrid film of TiO2 nanosheets/[Ru(phen)2(dC18bpy)] 2+is closely packed at a surface pressure of 25 mN m 1and has a thickness of(3.20±0.5)nm.X-ray photoelectron spectra show the formation of Pd nanoparticles on the surface of hybrid film with radii of 20–200 nm by the reduction of[Pd(NH3)4] 2+ under light irradiation.When it is applied to oxidize GMP,a larger catalytic oxidative current is achieved on the ITO/TiO2-Ru/Pd electrode at the external potential above 700 mV(vs.Ag|AgCl|KCl)in comparison with the naked ITO electrode and ITO/TiO2-Ru electrode.Such a result indicates that the Pd nanoparticles are able to hamper the combination of electron hole pairs and reduce the counterwork of insulating long alkyl chains of amphiphilic Ru(II)complexes,and thus develops the electron transfer efficiency and produces the enhanced redox current.CHEN Xi' An WANG Shun LIN JuanJuan LIU AiLi HUANG ShaoMing 2011Science China Chemistry2011,54,3:0
19Impact of Cold Spells on Ischemic Stroke Severity and Clinical Outcomes in Tianjin, China显示文摘There are several studies on the impact of extreme weather on the incidence and mortality of stroke, but the research on the effect of extreme weather events on severity and outcome of ischemic stroke is scarce. This study aimed to evaluate the impact of cold spells on the severity and clinical outcomes of patients with initial acute ischemic stroke in a hospital-based study. We enrolled 553 patients with initial ischemic stroke during the cold seasons between 2016 and 2019. Patients were separated into the mild stroke group and moderate-to-severe stroke group according to the National Institutes of Health Stroke Scale(NIHSS) of the U.S., and good outcome group and poor outcome group according to modified Rankin Scale(mRS) scores. There are nine different definitions of cold spells according to intensities and duration time. After adjustment for climate factors, air quality index, and common risk factors, it is found that cold spells were associated with moderate-to-severe neurological deficits and clinical outcomes in overall ischemic stroke patients. Furthermore, the delay effect for stroke severity started at the beginning of the cold wave(Lag 0) and lasted up to 14 days(Lags 0–14). In addition, when a cold spell was set as a daily mean temperature < 3rd or 5th percentile and with a duration ≥ 2 or 3 consecutive days, cold spells had a significant impact on clinical outcomes, and there was a definite delay effect of at least 7 days(Lags 0–7) and it lasted up to 14days(Lags 0–14). 20MAPK4 facilitates angiogenesis by inhibiting the ERK pathway in non-small cell lung cancer显示文摘Background:Angiogenesis plays an important role in the occurrence and development of non-small cell lung cancer(NSCLC).The atypical mitogen-activated protein kinase 4(MAPK4)has been shown to be involved in the pathogenesis of various diseases.However,the potential role of MAPK4 in the tumor angiogenesis of NSCLC remains unclear.Methods:Adult male C57BL/6 wild-type mice were randomly divided into the control group and p-siMAPK4 intervention group,respectively.The cell proliferation was analyzed with flow cytometry and immunofluorescence staining.The vascular density in tumor mass was analyzed by immuno-fluorescence staining.The expressions of MAPK4 and related signaling molecules were detected by western blot analysis and immunofluorescence staining,and so on.Results:We found that the expression of MAPK4,which was dominantly expressed in local endothelial cells(ECs),was correlated with tumor angiogenesis of NSCLC.Furthermore,MAPK4 silencing inhibited the proliferation and migration abilities of human umbilical vein ECs(HUVECs).QKHZC-2020-4Y156,QKH-JC-2018-1428,QKH-RC-2019-5612;Collaborative Innovation Center of Chinese Ministry of Education,Grant/Award Number:2020-39;Program for Science and Technology Joint Fund Project in Zunyi Science and Technology Bureau and Zunyi Medical University,Grant/Award Numbers:ZSKH-RPT-2020-6,ZSKH-HZ-2021-193;Graduate Research Fund of Zunyi Medical University,Grant/Award Number:2023-ZYK-171 Global gene analysis showed that MAPK4 silencing altered the expression of multiple genes related to cell cycle and angiogenesis pathways,and that MAPK4 silencing increased transduction of the extracellular regulated protein kinases 1/2(ERK1/2)pathway but not Akt and c-Jun n-terminal kinase pathways.Further analysis showed that MAPK4 silencing inhibited the proliferation and migration abilities of HUVECs cultured in tumor cell supernatant,which was accompanied with increased transduction of the ERK1/2 pathway.Clinical data analysis suggested that the higher expression of MAPK4 and CD34 were associated with poor prognosis of patients with NSCLC.Targeted silencing of MAPK4 in ECs using small interfering RNA driven by the CD34 promoter effectively inhibited tumor angiogenesis and growth of NSCLC in vivo.Conclusion:Our results reveal that MAPK4 plays an important role in the angiogenesis and development of NSCLC.MAPK4 may thus represent a new target for NSCLC.Jing Chen Jing Yang Yufang Liu Xu Zhao Juanjuan Zhao Lin Tang Mengmeng Guo Ya Zhou Chao Chen Dongmei Li Zhenke Wen Guiyou Liang Lin Xu 2024Cancer Innovation2024,3,3:0
