1Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. 2Restudy of the storage and migration model of the Quaternary groundwater in Beijing Plain area显示文摘Through a wide-spread groundwater sampling mainly at nested monitoring wells in the Beijing Plain area,combined with data collection,the study analyzes water level,hydrochemistry,temperature,tritium,deuterium,18O,14C and other aspects,and indicates that there exits an obvious stratification phenomenon of the groundwater in the alluvial plain area down in the middle part of alluvial-pluvial fans,and the hydraulic connection between layers is very weak.The analysis of the tritium concentration suggests that the average influence depth of the modern precipitation is approximately 120m.Based on the analysis of the 22 paleowater samples through the D18O method and 14C isotopic dating,the deep groundwater circulation in the fine sediments area of Beijing Plain down in the fringe part of alluvial-pluvial fans is relatively slow.On average the paleowater point occurs approximately below the 180m at these points.The main reasons for the formation of stratification and paleowater points are the water-blocking effect of the clay type strata and the compaction effect during the sedimentary process,especially the differential compaction,which could form closed or relatively closed stagnant aquifers.Groundwater in this kind of stagnant aquifers to some degree belongs to non-renewable resource,and should be reconsidered and re-arranged in the water resources management strategy.LIU YuanZhang WU Qiang LIN Pei LIU JiuRong XING LiTing GAO ZhiHui 2012Science China Earth Sciences2012,55,7:7
3Expression and prognosis analyses of Dectin-1 cluster genes in patients with lung adenocarcinoma (LUAD) and the association with immune checkpoint molecules显示文摘Reviews The Dectin-1 cluster comprises seven members:CLEC-12A,CLEC-12B,CLEC-1A,CLEC-7A,CLEC-2,CLEC-9A and OLR1.These members have been demonstrated to be involved in the tumorigenesis,progression,and metastasis of several cancers.However,little is known about their roles in human lung adenocarcinoma(LUAD).The expression patterns of the Dectin-1 cluster were analyzed via the ONCOMINE and GEPIA databases.We evaluated the prognostic value of the Dectin-1 cluster in patients with LUAD using the Kaplan-Meier plotter and GEPIA.Differential expression was validated with the EMBL-EBI database,and protein expression was analyzed with the HPA database.In addition,protein-protein interaction network,GO,and KEGG analyses were conducted.Finally,the correlations between CLEC-12A and immune molecules(immune inhibitors and MHC molecules)were investigated via TISIDB and GEPIA.The expression levels of Dectin-1 cluster genes were downregulated in LUAD tissues compared to those in normal lung tissues.The expression levels of CLEC-12A,CLEC-12B,CLEC-2,and CLEC-9A correlated with tumor stage,and CLEC-12A and CLEC-12B were significantly associated with survival in patients with LUAD.The seven genes mostly participated in immune regulation processes and were involved in autoimmune disorders and hematological malignancies.Finally,correlation analyses revealed CLEC-12A expression was associated with most immune inhibitors and MHCs.CLEC-12A was positively related to PD-1,PD-L1,PD-L2,CTLA4,TIM3,and LAG3.In conclusion,our findings suggest that CLEC-12A and CLEC-12B can be used as prognostic biomarkers in LUAD.CLEC-12A expression was associated with immune checkpoint molecules,and CLEC-12A may be a potential assistant target to improve the efficacy of immune checkpoint inhibitors immunotherapy.LITING YOU FEIFEI NA JUAN ZHOU LIN JIAO YI ZHOU BINWU YING 2021BIOCELL2021,45,3:1

5Shen Qi Wan attenuates renal interstitial fibrosis through upregulating AQP1显示文摘Renal interstitial fibrosis(RIF)is the crucial pathway in chronic kidney disease(CKD)leading to the end-stage renal failure.However,the underlying mechanism of Shen Qi Wan(SQW)on RIF is not fully understood.In the current study,we investigated the role of Aquaporin 1(AQP1)in SQW on tubular epithelial-to-mesenchymal transition(EMT).A RIF mouse model induced by adenine and a TGF-β1-stimulated HK-2 cell model were etablished to explore the involvement of AQP 1 in the protective effect of SQW on EMT in vitro and in vivo.Subsequently,the molecular mechanism of SQW on EMT was explored in HK-2 cells with AQP1 knockdown.The results indicated that SQW alleviated kidney injury and renal collagen deposition in the kidneys of mice induced by adenine,increased the protein expression of E-cadherin and AQP1 expression,and decreased the expression of vimentin andα-smooth muscle actin(α-SMA).Similarly,treatmement with SQW-containing serum significantly halted EMT process in TGF-β1 stimulated HK-2 cells.The expression of snail and slug was significantly upregulated in HK-2 cells after knockdown of AQP1.AQP1 knockdown also increased the mRNA expression of vimentin andα-SMA,and decreased the expression of E-cadherin.The protein expression of vimentin increased,while the expression of E-cadherin and CK-18 significantly decreased after AQP1 knockdown in HK-2 cells.These results revealed that AQP1 knockdown promoted EMT.Furthermore,AQP1 knockdown abolished the protective effect of SQW-containing serum on EMT in HK-2 cells.In sum,SQW attentuates EMT process in RIF through upregulation of the expression of AQP1.LIN Yiyou WEI Jiale ZHANG Yehui HUANG Junhao WANG Sichen LUO Qihan YU Hongxia JI Liting ZHOU Xiaojie LI Changyu 2023Chinese Journal of Natural Medicines2023,21,5:1

7Effect of Roucongrong(Herba Cistanches Deserticolae) decoction on the substantia nigra through Wnt/β-catenin signaling pathway in rats with Parkinson's disease induced by 6-hydroxydopamine hydrochloride显示文摘OBJECTIVE:To investigate the effect of Roucongrong(Herba Cistanches Deserticolae)decoction on the substantia nigra in rats with Parkinson’s disease(PD)induced by 6-hydroxydopamine hydrochloride(6-OHDA).To further determine whether the Wnt/β-catenin signaling pathway is involved in the action.METHODS:A rat model of PD was established by intracranial injection of 6-OHDA.Subsequently,three concentrations of Roucongrong(Herba Cistanches Deserticolae)decoction were prepared and administered to rats by gavage therapy for 14 d.Behavioral changes were measured in PD rats.In vivo tyrosine hydroxylase(TH)levels in the substantia nigra were examined by immunohistochemistry.Additionally,gene and protein expression levels of members of the Wnt/β-catenin signaling pathway were examined by Western blotting and polymerase chain reaction.Lastly,a Wnt/β-catenin inhibitor was used to investigate the mechanism of action in 1-methyl-4-phenylpyridinium(MPP+)-treated MES23.5 cells in vitro.RESULTS:Roucongrong(Herba Cistanches Deserticolae)decoction improved performance in the stride and gait adjustment tests in PD rats.It also increased TH in the substantia nigra and altered the expression of genes and proteins in the Wnt/β-catenin signaling pathway.Wnt/β-catenin inhibitor reduced the effect of Roucongrong(Herba Cistanches Deserticolae)decoction in MPP+-treated MES23.5 cells.CONCLUSION:Roucongrong(Herba Cistanches Deserticolae)decoction may promote neuronal survival in PD in vivo and in vitro by increasing TH content in the substantia nigra and by activating the Wnt/β-catenin signaling pathway.XU Qian QIN Wei WU Fangzhen LIN Yao HONG Liting CHEN Dan HU Xuefeng CAI Jing 2021Journal of Traditional Chinese Medicine2021,41,5:1
8Shen-Qi-Wan Protects the Renal Peritubular Capillary Injury from Adenine-mediated Damage by Upregulating Aquaporin 1显示文摘Background:Shen-Qi-Wan(SQW),a commonly used prescription against chronic kidney disease(CKD)in Tradi-tional Chinese Medicine(TCM),has a nephroprotective action in adenine-induced kidney injury.However,the mechanism of SQW in renal injury remains unclear.Objective:This study was undertaken to measure the effect of SQW on peritubular capillary injury both in vivo and in vitro assays.Methods:The effect of SQW on the peritubular capillary injury was evaluated according to measuring hypothalamic-pituitary-adrenal(HPA)function,inflammatory cytokines,VEGF(vascular endothelial growth fac-tor),CD34(Cluster of differentiation 34),and AQP1(Aquaporin 1)levels in the kidney,cell migration,and lumen forming capacity.Results:SQW supplementation could ameliorate dysfunction of the HPA and renal function loss induced by adenine.SQW also significantly inhibited the inflammatory cytokines including MCP-1(monocyte chemotactic protein-1)and VCAM-1(vascular cell adhesion molecule-1)level.Alternatively,SQW administration showed an ameliorating effect from the toxicity and alleviated the injury of capillaries around renal tubules instigated by adenine through increasing AQP1 mRNA and protein level.SQW medicated the serum enhanced the migration and lumen formation ability of HMEC-1 cells,and significantly increased AQP1 protein level.Moreover,AQP1 knockdown efficiently inhibited the migration and lumen formation ability in HMEC-1 cells,and weakened the effect of SQW medicated serum.Conclusion:These results suggested that SQW attenuated peritubular capillary injury in adenine-induced CKD model rats through boosting angiogenesis in endothelial cell,and AQP1 may be a potential target of SQW for treating the renal injury.Yuting Bao Yehui Zhang Yuanxiao Yang Xueming Chen Luning Lin Yunbo Fu Liting Ji Changyu Li 2022Clinical Complementary Medicine and Pharmacology2022,2,1:1

10Establishment of a prognostic scoring model for regional recurrent nasopharyngeal carcinoma after neck dissection显示文摘Objective:The main aim of this study was to establish a scoring model to predict risk of progression and survival in patients with regionally recurrent nasopharyngeal carcinoma(NPC).Methods:Three hundred and forty-eight patients subjected to neck dissection from 2003 to 2017 were included for study.Clinicopathologic information for each patient was analyzed.Independent prognostic factors were selected using the Cox proportional hazards model and incorporated into the scoring model.Concordance index(C-index)and calibration curves were used to verify discrimination and calibration,respectively and the results validated using bootstrap resampling.Results:Microscopic positive lymph node>2[hazard ratio(HR),2.19;95%confidence interval(CI),1.30–3.68;P=0.003],extranodal extension(HR,2.75;95%CI,1.69–4.47;P<0.001),and lower neck involvement(HR,1.78;95%CI,1.04–3.04;P=0.034)were identified from multivariate analysis as independent factors for overall survival(OS).A qualitative 4-point scale was generated to stratify patients into 4 risk groups for predicting OS and progression-free survival(PFS).The novel scoring model demonstrated enhanced discrimination(C-index=0.69;95%CI,0.62–0.76)relative to the original recurrent tumor-node-metastasis(rTNM)staging system(C-index=0.56;95%CI,0.50–0.62),and was internally validated with a bootstrap-adjusted C-index of 0.70.The calibration curve showed good agreement between predicted probabilities and actual observations.Conclusions:The scoring system established in this study based on a large regionally recurrent NPC cohort fills a gap regarding assessment of risk and prediction of survival outcomes after neck dissection in this population and could be further applied to identify high-risk patients who may benefit from more aggressive intervention.Xiaoyun Li Chao Lin Jinjie Yan Qiuyan Chen Xuesong Sun Sailan Liu Shanshan Guo Liting Liu Haojun Xie Qingnan Tang Yujing Liang Ling Guo Hao Li Xuekui Liu Xiang Guo Linquan Tang Haiqiang Mai 2020Cancer Biology & Medicine2020,17,1:0

12Carbon Monoxide Promotes the Catalytic Hydrogenation on Metal Cluster Catalysts显示文摘Size effect plays a crucial role in catalytic hydrogenation.The highly dispersed ultrasmall clusters with a limited number of metal atoms are one candidate of the next generation catalysts that bridge the single-atom metal catalysts and metal nanoparticles.However,for the unfavorable electronic property and their interaction with the substrates,they usually exhibit sluggish activity.Taking advantage of the small size,their catalytic property would be mediated by surface binding species.The combination of metal cluster coordination chemistry brings new opportunity.CO poisoning is notorious for Pt group metal catalysts as the strong adsorption of CO would block the active centers.In this work,we will demonstrate that CO could serve as a promoter for the catalytic hydrogenation when ultrasmall Pd clusters are employed.By means of DFT calculations,we show that Pd_(n)(n=2-147)clusters display sluggish activity for hydrogenation due to the too strong binding of hydrogen atom and reaction intermediates thereon,whereas introducing CO would reduce the binding energies of vicinal sites,thus enhancing the hydrogenation reaction.Experimentally,supported Pd_(2)CO catalysts are fabricated by depositing preestablished[Pd_(2)(μ-CO)_(2)Cl_(4)]2-clusters on oxides and demonstrated as an outstanding catalyst for the hydrogenation of styrene.The promoting effect of CO is further verified experimentally by removing and reintroducing a proper amount of CO on the Pd cluster catalysts.Ruixuan Qin Pei Wang Pengxin Liu Shiguang Mo Yue Gong Liting Ren Chaofa Xu Kunlong Liu Lin Gu Gang Fu Nanfeng Zheng 2020Research2020,,1:0

14Large Eddy Simulation of the Particle Coagulation in High Concentration Particle-Laden Planar Jet Flow显示文摘Particle coagulation by Brownian motion is an important but difficult research topic.When particle volume concentration is larger than 0.1%,the classic SMOLUCHOWSKI equation is not applicative anymore.The high concentration coagulation,with HEINE's correction,source terms for the Taylor-series expansion method of moments(TEMOM) are firstly driven in this paper.Ultra-fine particle(d0?100 mm) with initial volume fraction f?1% coagulation in a planar jet turbulence flow is simulated via the large eddy simulation(LES).The instantaneous and time-averaged particle distributions and the high concentration enhancement are given out.The particle number concentration distribution results show that the coagulation is more intense comparing to dilute case in previous research,especially near the nozzle exit.After jet flow is fully developed,the effect is much more obvious at the region between vortexes.The time-averaged γ(the high concentration enhance factor) distributes sharply and symmetrically about the jet centerline at the upstream,but becomes broad and flat at downstream where the cross-stream averaged γ fluctuates drastically.As a new attempt,this paper shows Brownian coagulation with high concentration also can be calculated via TEMOM appropriately,and the coagulation at the region between vortexes is about 1.38 times intensive of the dilute result calculated by the classic Smoluchowski theory.LIN Peifeng WU Dichong YU Mingzhou WANG Chao ZHANG Lite ZHU Zefei 2011Chinese Journal of Mechanical Engineering2011,24,6:0

17Comparison of the anxiety level between the medical staff and the public during the early phase of the coronavirus disease 2019 pandemic显示文摘Background:A sudden outbreak of the coronavirus disease 2019(COVID-19)started in December 2019 in Wuhan,China.Up-to-date,there have been limited studies examining the anxiety status of Chinese individuals in the early phase of the pandemic period(January 30,2020–February 15,2020).This survey aimed to compare the level of anxiety of the medical staff with that of the public and to provide a theoretical basis for developing an effective psychological intervention.Method:Questionnaires were sent on the Internet(http://gffzz603dbffde9e44a22h0qu6cnbkn6qf6wb0.ffgz.tsg.suse.edu.cn)during this period.The anxiety levels of Chinese people were investigated using the Self-Rating Anxiety Scale(SAS),and the demographic data were collected simultaneously.Results:A total of 1110 participants were enrolled in this study,with an effective response rate of 100%.A total of 482 respondents were medical staff(43.4%),while 628 were members of the general public(56.6%).The medical staff itself had a higher SAS score than the general public(48.36±13.40 vs.45.74±11.79,P<0.01),while the medical staff in Wuhan were more anxious than the public in Wuhan with a higher SAS score(54.17±14.08 vs.48.53±11.92,P<0.01).Conclusion:The COVID-19 pandemic has had a significant impact on the anxiety levels of the medical staff and the public,with the medical personnel showing a higher anxiety level than the public,especially female medical staff in Wuhan.Therefore,urgent intervention programs to reduce anxiety should be implemented.Guanguan Luo Yumin Liu Dongai Yao Shaozhou Ni Bangsheng Wu Liting Lin Yunming Wang Xiaoqing Jin 2022Emergency and Critical Care Medicine2022,2,3:0
