1Isoliquiritigenin inhibits TGF-β1-induced fibrogenesis through activating autophagy via PI3K/AKT/mTOR pathway in MRC-5 cells显示文摘Isoliquiritigenin(ISL),a natural flavonoid derived from the root of liquorice,has been reported to possess anti-inflammatory and antioxidant activities.Previous studies have found that ISL plays a crucial role in anti-fibrosis of adipose tissue and renal tissue;however,its effect on pulmonary fibrogenesis has not been demonstrated.In this study,we aimed to explore the roles and the underlying mechanisms of ISL in TGF-β1-induced fibrogenesis using human lung fibroblast-derived MRC-5 cells.Cell proliferation and migration were determined by MTT and wound healing assay,respectively.The expression levels of alpha-smooth muscle actin(α-SMA),collagen type I alpha 1(COLIA1)and fibronectin(FN),microtubule-associated protein light chain 3(LC3)and related signaling molecules were detected by quantitative real-time PCR,western blot and immunofluorescence assay,correspondingly.EGFP-LC3 transfection was used for autophagy analysis.The results showed that ISL inhibited the TGF-β1-induced proliferation and migration,and down-regulated the expressions ofα-SMA,COLIA1 and FN.ISL treatment led to up-regulation of LC3 in TGF-β1-treated MRC-5 cells,accompanied by significant decrease in the phosphorylation levels of phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT),and mammalian target of rapamycin(mTOR).In addition,the inhibitory effects of ISL on TGF-β1-induced fibrogenic features in MRC-5 cells were enhanced by pretreatment with autophagy activator Rapmycin and PI3K/AKT inhibitor LY294002 and reversed by autophagy inhibitor 3-methyladenine and PI3K/AKT activator IGF-1.Taken together,our results demonstrated that ISL could attenuate the fibrogenesis of TGF-β1-treated MRC-5 cells by activating autophagy via suppressing the PI3K/AKT/mTOR pathway.Therefore,ISL holds a great potential to be developed as a novel therapeutic agent for the treatment of pulmonary fibrosis.Jinjuan He Hao Peng Meifang Wang Ying Liu Xingrong Guo Bin Wang Longjun Dai Xueqin Cheng Zhongji Meng Leyong Yuan Fenglin Cai Yijun Tang 2020Acta Biochimica et Biophysica Sinica2020,52,8:16
2Artificial light-harvesting systems fabricated by supramolecular host–guest interactions显示文摘Artificial light-harvesting systems(LHSs) have drawn increasing research interest in recent times due to the energy crisis worldwide. Concurrently, macrocycle-based host–guest interactions have played an important role in the development of supramolecular chemistry. In recent years, studies towards artificial LHSs driven by macrocycle-based host–guest interactions are gradually being disclosed. In this mini-review, we briefly introduce the burgeoning progress of artificial LHSs driven by host–guest interactions. 3Numerical analysis on the crack propagation and failure characteristics of rocks with double fissures under the uniaxial compression显示文摘The fissures and rock bridges with different dips had different contributions to crack's initiation,propagation,convergence and penetration.In this paper,based on the rock fracture theory,the crack's propagation and evolution process on rock specimen with double fissures under uniaxial compression was simulated.As a result,the crack propagation and evolution law of rocks with different fissure dips(α=0°,15°,30°,45°,60°,75°,90°;β=45°)and different rock bridge dips(β=0°,30°,45°,60°,90°;α=45°)was obtained by numerical tests.Meanwhile,the fissure and rock bridge dips influence on the macro mechanical properties of rock was analyzed.Besides,the paper investigated the influences of different fissure dips and different rock bridge dips on the bridge transfixion.The study is of great significance to reveal the impact of different dips on the mechanical mechanism of multiple-fissures rock under specific conditions,and it also has important theoretical significance for the research on multiple-fissure rock.Leyong Chen Jianjun Liu 2015Petroleum2015,1,4:2

5Supramolecular Prodrug Micelles Constructed by Drug-Drug Conjugate with Water Soluble Pillar[6]arene for Controllable and Rapid Drug Release显示文摘The first attempt of constructing supramolecular prodrug micelles based on the host-guest interaction between water-soluble pillar[6]arene(WP6)and a novel doxorubicin(DOX)-based drug-drug conjugate(G)is reported,in which the drug-drug conjugate is synthesized by conjugating anticancer drug DOX with another drug isoniazide via the acid-cleavable hydrazone bond.The obtained WP6⊃G micelles are stable under physiological conditions,whereas the cumulative release of DOX is approximate to 100%within 30 min at pH 5.5 by simulating the endo-lysosomal environment at 37℃.Therefore,this WP6-based prodrug micelles can achieve efficient DOX ac-cumulation in the acidic tumor cells within a short period of time,which is very important and valuable;meanwhile it can also reduce the unexpected premature burst release under physiological condition.Furthermore,cellular in-ternalization and localization experiments demonstrated that these prodrug micelles enter cancer cells mainly via endocytosis and can lead to significant drug accumulation in SKOV3 cancer cells,implying its promising future for cancer therapy.Yu Cao Xiaochun Zou Shuhan Xiong Yan Li Yingzhong Shen Xiaoyu Hu Leyong Wang 2015Chinese Journal of Chemistry2015,33,3:2

12Cancer cell detection and imaging: MRI-SERS bimodal splat-shaped Fe_3O_4/Au nanocomposites显示文摘The development of multifunctional contrast agents contributes significant character in the diagnosis of cancer. However, still efforts are required to design and improve the biocompatibility of contrast agents for early detection of cancer. Herein, we synthesized splat shape Fe_3O_4/Au nanocomposites for multimode biomedical applications. Transmission electron microscopy(TEM) results showed that the splat-like Fe_3O_4/Au structure was in the range of 130 nm and homogeneously distributed in the aqueous medium. The nanocomposites were incubated with MCF-7 cells to investigate the magnetic resonance imaging(MRI) and surface-enhanced Raman scattering(SERS) activities. The excellent T_2 MRI and enhanced SERS properties were obtained without any cytotoxicity. 20TP53 polymorphisms are involved in inverse colorectal cancer comorbidity in Chinese schizophrenia patients显示文摘The inverse cancer comorbidity in schizophrenia patients may be related to the genetic factors,involving the regulation of apoptosis.The tumour suppressor gene TP53,involved in neural apoptosis,is one of the potential candidate genes associated with schizophrenia which might reduce colorectal cancer risk.We recruited 270 schizophrenia patients and 312 colorectal cancer patients without schizophrenia.To examine the genetic association between schizophrenia and colorectal cancer,we analysed eight SNPs(rs12951053,rs1625895,rs2909430,rs9895829,rs1042522,rs8079544,rs8064946,rs17806770)covering 14.35 kb in the region of TP53.We observed that one of the eight genetic polymorphisms showed statistically significant differences between the colorectal cancer subjects and the schizophrenia subjects(rs12951053,P=0.000 1,OR1.70,95%CI1.30-2.23).In addition,the haplotype of A-G(rs12951053-rs8064946),giving aglobal P=0.001 8,was the most significant.Our data indicate that the polymorphisms of rs12951053 in TP53confer reduced susceptibility to colorectal cancer and suggest a potential protective mechanism against colorectal cancer in the schizophrenia patients of Han Chinese origin.Sun Liming Huang Guoxin Zhao Leyong Chen Xiaofen Chen Wenjiao 2015国际检验医学杂志2015,36,15:0
