| 6 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 10 | Bn.YCO affects chloroplast development in Brassica napus L.显示文摘Mature chloroplasts,as the main sites of photosynthesis,are essential for seedling growth in higher plants.Loss of function of genes involved in chloroplast development changes plant phenotype.We obtained a YELLOW COTYLEDON (YCO) mutant in rapeseed (Brassica napus L.) using CRISPR-Cas9.Bn.YCO,a gene of unknown function,has two homologous copies (Bna A01.YCO and Bna C01.YCO) in B.napus.Homozygous mutation of these two homologs resulted in yellow cotyledons and chlorotic true leaves.Transmission electron microscopy revealed that the formation of thylakoid membranes was inhibited in yellow cotyledons.Sequence similarity search revealed that YCO was conserved in different species,and a subcellular location assay verified that Bn.YCO was located in the chloroplast.Bn.YCO was expressed in multiple tissues,most highly in cotyledons.Knockout of Bn.YCO blocked the transcription of plastid genes,especially those of photosystem genes transcribed by plastid-encoded polymerase.Transcriptome sequencing showed that the majority of genes involved in ribosome assembly and photosynthesis were down-regulated in Bn.yco mutants.These results suggested that loss of function of Bn.YCO affected plastid gene transcription,which influenced chloroplast biogenesis in rapeseed seedlings. | Tingting Liu Baolong Tao Hanfei Wu Jing Wen Bin Yi Chaozhi Ma Jinxing Tu Tingdong Fu Lixia Zhu Jinxiong Shen | 2021 | The Crop Journal2021,9,5: | 1 |

| 14 | Comparative phosphoproteomic analysis of compatible and incompatible pollination in Brassica napus L.显示文摘Self-incompatibility(SI)promotes outbreeding and prevents self-fertilization to promote genetic diversity in angiosperms.Several studies have been carried to investigate SI signaling in plants;however,protein phosphorylation and dephosphorylation in the fine-tuning of the SI response remain insufficiently understood.Here,we performed a phosphoproteomic analysis to identify the phosphoproteins in the stigma of self-compatible‘Westar’and self-incompatible‘W-3’Brassica napus lines.A total of 4109 phosphopeptides representing 1978 unique protein groups were identified.Moreover,405 and 248 phosphoproteins were significantly changed in response to SI and self-compatibility,respectively.Casein kinaseⅡ(CKⅡ)phosphorylation motifs were enriched in self-incompatible response and identified 127 times in 827 dominant SI phosphorylation residues.Functional annotation of the identified phosphoproteins revealed the major roles of these phosphoproteins in plant-pathogen interactions,cell wall modification,mRNA surveillance,RNA degradation,and plant hormone signal transduction.In particular,levels of homolog proteins ABF3,BKI1,BZR2/BSE1,and EIN2 were significantly increased in pistils pollinated with incompatible pollens.Abscisic acid and ethephon treatment partially inhibited seed set,while brassinolide promoted pollen germination and tube growth in SI response.Collectively,our results provided an overview of protein phosphorylation during compatible/incompatible pollination,which may be a potential component of B.napus SI responses. | Zhiqiang Duan Shengwei Dou Zhiquan Liu Bing Li Bin Yi Jinxiong Shen Jinxing Tu Tingdong Fu Cheng Dai Chaozhi Ma | 2020 | Acta Biochimica et Biophysica Sinica2020,52,4: | 0 |
