| 9 | Effects of Subsoiling on Soil Moisture Under No-Tillage for Two Years显示文摘In order to improve the water use efficiency under conservation tillage,the effects of subsoiling on soil moisture under no-tillage were studied. An experiment of 40 cm subsoiling in a field kept under no-tillage for 2 years was operated from 2005 to 2006. Based on the data of the soil moisture and crop yield,the physical basis of subsoiling for water conservation and yield increase was analyzed. The results showed that the soil water storage under subsoiling,from the soil surface to a depth of 100 cm was more than that under no-tillage for the growth season. In the 0-100 cm soil depth,the soil moisture in 50-100 cm depth under subsoiling was more compared with no-tillage,which increased when it's drought and decreased when it's rainy with the increase in soil depth. Compared with no-tillage,subsoiling could reduce the water consumption of oats in the 0-50 cm depth and increase the water consumption in the 50-100 cm depth. Also,subsoiling increased the yield by 18.29% and the water use efficiency by 16.8% in a two-year average. The effects of subsoiling on water conservation and yield increase were affected by precipitation,and a well-proportioned rainfall was better to increase yield and water use efficiency. Meanwhile,subsoiling decreased bulk density,which increased with the available precipitation. | 11 | A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. | 15 | Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases. | Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 29 |

| 17 | Identifying paleoflood deposits archived in Zhongba Site,the Three Gorges reservoir region of the YangtzeRiver,China显示文摘Based on the principle that the present is the key to the past , detailed analyses, such as AMS 14C dating, grain size, component and morphology of heavy minerals, micro-morphology of zircon, Rb/Sr, magnetic susceptibility and total organic carbon (TOC), were conducted to identify paleoflood sediments archived in Zhongba Site. The results indicate that the plaeoflood sediments bear great similarities with modern flood sediments in the following aspects: (1) probability cumulative curves mainly show a pattern of 3―4 segments; (2) grain-size distribution of suspended matter ranges between 3 and 10Ф; (3) the sediments are well-sorted, most of which are suspended matter (>50%); (4) the same species, quantity and morphology of heavy minerals; (5) scanning electronic microscope images show that shapes of zircon are mainly oval and nearly spheral, rounded due to long-distance transport; (6) higher Rb/Sr values (0.55―0.66)than those of sediments from cultural layers (0.03―0.26); (7) magnetic susceptibility values (133.73―433.05 10-6m3/kg) are lower than those of sediments from cultural layers (959.25―2442.44 10?6 m3/kg); (8) TOC (0.14%―0.33%) are lower than those of sediments from cultural lay-ers (1.13%―2.95%). | 20 | Total alkyl dibenzothiophenes content tracing the filling pathway of condensate reservoir in the Fushan Depression,South China Sea显示文摘The condensates are generally characterized by high maturity,low concentration of steranes and ter-panes biomarkers and low content of non-hydrocarbon fraction. In this case commonly used steranes,terpanes and carbazoles parameters cannot be effectively applied in the reservoir-filling tracing. The hydrogen bond formed by sulfur atom in the dibenzothiophenes (DBTs) results in molecule adsorption and fractionation during oil migration in reservoir. Like carbazoles,total DBTs content decreases with the increasing of oil migration distance. Therefore,a new parameter——total DBTs content is proposed to be used to trace the oil migration orientation and filling pathway. In present study,total DBTs con-tents of condensates and light oils are obtained by adding internal standard——eight deuterium atoms substituted DBT during Gas Chromatography-Mass Spectrometry analysis of aromatic fraction. Except for a few samples with much lower content of non-hydrocarbon fraction,the total DBTs content shows a fine positive correlation with that of carbazoles. Large errors can be caused in the process of pyrrolic nitrogen compounds separation. The application of this new parameter in the Fushan Depression of Beibu Gulf Basin,South China Sea indicates that this parameter is a reliable one to trace filling pathway in condensate reservoirs. Combined with other DBTs-related parameters,such as 4-/1-methydibenzo-thiophene and 2,4-/1,4-dimethyldibenzothiophene,oil migration orientation and filling pathway of the Fushan Depression was determined. The accumulations of Huachang oil field in the Fushan Depres-sion are mainly migrated and charged from northeast to southeast along the Huachang uplift. It can be predicated that the light oil and condensates in the Huachang oil field should be sourced from the source kitchen at the Bailian Sag.
