| 5 | CmNF-YB8 affects drought resistance in chrysanthemum by altering stomatal status and leaf cuticle thickness显示文摘Drought is a major abiotic stress that limits plant growth and development.Adaptive mechanisms have evolved to mitigate drought stress,including the capacity to adjust water loss rate and to modify the morphology and structure of the epidermis.Here,we show that the expression of CmNF-YB8,encoding a nuclear factor Y(NF-Y)B-type subunit,is lower under drought conditions in chrysanthemum(Chrysanthemum morifolium).Transgenicchrysanthemumlinesinwhich transcript levels of CmNF-YB8 were reduced by RNA interference(CmNF-YB8-RNAi)exhibited enhanced drought resistance relative to control lines,whereas lines overexpressing CmNF-YB8(CmNF-YB8-OX)were less tolerant to drought.Compared to wild type(WT),CmNF-YB8-RNAi plants showed reduced stomatal opening and a thicker epidermal cuticle that correlated with their water loss rate.We also identified genes involved in stomatal adjustment(CBLinteracting protein kinase 6,CmCIPK6)and cuticle biosynthesis(CmSHN3)that are more highly expressed in CmNF-YB8-RNAi lines than in WT,CmCIPK6 being a direct downstream target of CmNF-YB8.Virus-induced gene silencing of CmCIPK6 or CmSHN3 in the CmNFYB8-RNAi background abolished the effects of CmNF-YB8-RNAi on stomatal closure and cuticle deposition,respectively.CmNF-YB8 thus regulates CmCIPK6 and CmSHN3 expression to alter stomatal movement and cuticle thickness in the leaf epidermis,thereby affecting drought resistance. | Tianle Wang Qian Wei Zhiling Wang Wenwen Liu Xin Zhao Chao Ma Junping Gao Yanjie Xu Bo Hong | 2022 | Journal of Integrative Plant Biology2022,64,3: | 1 |
| 6 | Synthesis of AgInS_2 QDs in droplet microreactors:Online fluorescence regulating through temperature control显示文摘For the synthesis of AgInS_2 quantum dots(QDs), a suitable temperature is extremely important for control of the size, shape and fluorescence properties of QDs. Most of synthesis methods for AgInS_2 QDs are based on batch reactors, which bring uneven distribution of temperature, affecting their fluorescence properties and size uniformity. Here we designed a droplet microreactor with a temperature-controllable region, and successfully synthesized water-soluble AgInS_2 QDs. By accurately controlling temperature,we also studied how the reaction temperature affected the fluorescence properties of AgInS_2 QDs. The results showed that with the increasing of reaction temperature, the QDs size increased and the fluorescence peak constantly red-shifted along with enhanced fluorescence intensity. The high correlation between GC12% and GC3% suggested that mutational pressure rather than natural selection was the main factor determining the codon usage and base component in fish rhabdovirus,most of the spots lying below the expected curve( ENC vs GC3%) also suggested that codon usage bias in these 49 genomes was influenced by mutational pressure in codon usage pattern. Through PCA analysis based on RSCU,all ORFs in the 49 fish rhabdoviurs genomes were clustered into two parts,and this result showed that hosts played a role in codon usage pattern of fish rhabdovirus genomes. | 18 | Novel biomarkers identifying hypertrophic cardiomyopathy and its obstructive variant based on targeted amino acid metabolomics显示文摘Background:Hypertrophic cardiomyopathy(HCM)is an underdiagnosed genetic heart disease worldwide.The management and prognosis of obstructive HCM(HOCM)and non-obstructive HCM(HNCM)are quite different,but it also remains challenging to discriminate these two subtypes.HCM is characterized by dysmetabolism,and myocardial amino acid(AA)metabolism is robustly changed.The present study aimed to delineate plasma AA and derivatives profiles,and identify potential biomarkers for HCM.Methods:Plasma samples from 166 participants,including 57 cases of HOCM,52 cases of HNCM,and 57 normal controls(NCs),who first visited the International Cooperation Center for HCM,Xijing Hospital between December 2019 and September 2020,were collected and analyzed by high-performance liquid chromatography-mass spectrometry based on targeted AA metabolomics.Three separate classification algorithms,including random forest,support vector machine,and logistic regression,were applied for the identification of specific AA and derivatives compositions for HCM and the development of screening models to discriminate HCM from NC as well as HOCM from HNCM.Results:The univariate analysis showed that the serine,glycine,proline,citrulline,glutamine,cystine,creatinine,cysteine,choline,and aminoadipic acid levels in the HCM group were significantly different from those in the NC group.Four AAs and derivatives(Panel A;proline,glycine,cysteine,and choline)were screened out by multiple feature selection algorithms for discriminating HCM patients from NCs.The receiver operating characteristic(ROC)analysis in Panel A yielded an area under the ROC curve(AUC)of 0.83(0.75-0.91)in the training set and 0.79(0.65-0.94)in the validation set.Moreover,among 10 AAs and derivatives(arginine,phenylalanine,tyrosine,proline,alanine,asparagine,creatine,tryptophan,ornithine,and choline)with statistical significance between HOCM and HNCM,3 AAs(Panel B;arginine,proline,and ornithine)were selected to differentiate the two subgroups.The AUC values in the training and validation sets for Panel B were 0.83(0.74-0.93)and 0.82(0.66-0.98),respectively.Conclusions:The plasma AA and derivatives profiles were distinct between the HCM and NC groups.Based on the differential profiles,the two established screening models have potential value in assisting HCM screening and identifying whether it is obstructive. | Lanyan Guo Bo Wang Fuyang Zhang Chao Gao Guangyu Hu Mengyao Zhou Rutao Wang Hang Zhao Wenjun Yan Ling Zhang Zhiling Ma Weiping Yang Xiong Guo Chong Huang Zhe Cui Fangfang Sun Dandan Song Liwen Liu Ling Tao | 2022 | Chinese Medical Journal2022,135,16: | 0 |
