3Induction of Th1-Type Immune Response by Chitosan Nanoparticles Containing Plasmid DNA Encoding House Dust Mite Allergen Der p2 for Oral Vaccination in Mice显示文摘This study was to prepare the chitosan-pDer p 2 nanoparticles and to investigate the effect of chitosan-DNA nanoparticles on immune response in mice by oral delivery of chitosan-DNA nanoparticles. The nanoparticles were synthesized by complexing chitosan with plasmid DNA. The DNA was fully complexed into chitosan-DNA nanoparticles,suggesting a 100% encapsulation efficiency. Chitosan-DNA complex renders a significant protection of the plasmid. No effect on cell viability was observed in both cell types and average cell viability over 100% was obtained. Oral gene delivery with chitosan-DNA nanoparticles can generate a higher level expression of gene in vivo. Oral chitosan-pDer p 2 nanoparticles in BALB/c mice can induce IFN-γ in serum and prevent subsequent sensitization of Th2 cell-regulated specific IgE responses. 4Therapeutic Effects of DNA Vaccine on Allergen-Induced Allergic Airway Inflammation in Mouse Model显示文摘Vaccination with DNA encoding Dermatophagoides pteronyssinus group 2 (Der p 2) allergen previously showed its effects of immunologic protection on Der p 2 allergen-induced allergic airway inflammation in mice. In present study, we investigated whether DNA vaccine encoding Der p 2 could exert therapeutic role on allergen-induced allergic airway inflammation in mouse model and explored the mechanism of DNA vaccination in asthma specific-allergen immunotherapy. After sensitized and challenged by Der p 2, the BALB/c mice were immunized with DNA vaccine. The degrees of cellular infiltration were scored. IgE levels in serum and IL-4/IL-13 levels in BALF were determined by ELISA. The lung tissues were assessed by histological examinations. Expressions of STAT6 and NF-κB in lung were determined by immunohistochemistry staining. Vaccination of mice with DNA vaccine inhibited the development of airway inflammation and the production of mucin induced by allergen, and reduced the level of Der p 2-specific IgE level. Significant reductions of eosinophil infiltration and levels of IL-4 and IL-13 in BALF were observed after vaccination. Further more, DNA vaccination inhibited STAT6 and NF-κB expression in lung tissue in Der p 2-immunized mice. 5WWOX suppresses KLF5 expression and breast cancer cell growth显示文摘The WW domain-containing oxidoreductase(WWOX) is a tumor suppressor in a variety of cancers, including breast cancer. Reduced WWOX expression is associated with the basal-like subtype and a relatively poor disease-free survival rate among breast cancer patients. Though several WWOX partners have been identified, the functional mechanisms of WWOX's role in cancers have not been fully addressed to date. In the current study, we found WWOX suppresses expression of KLF5—an oncogenic transcription factor—at protein level, and suppresses cancer cell proliferation in both bladder and breast cancer cell lines. Furthermore, we demonstrated that WWOX physically interacts with KLF5 via the former's WW domains and the latter's PY motifs. Interestingly, we found the expression of WWOX negatively correlates with KLF5 expression in a panel of breast cancer cell lines. 8Meanders of the Jialing River in China: Morphology and formation显示文摘As a fundamental category of river system, comparisons in diverse regions and types with quantitative parameters on meander morphology will help to distinguish different characteristics and approach formations and evolutions. It also can provide scientific basis of geomorphology for their protections and exploitation of the meander resources. Based on the images of Google Earth, meanders’ parame- ters are proposed and then measured and calculated. The result suggests that incised meanders in the Jialing River are among the most complex, irregular and sinuous channels. Special river gradient, geological structures, strata and lithology, backwater effect of landform are key factors to shape me- anders of the Jialing River. The evolution of meander in the Jialing River obeys the principle of mini- mum-maximum energy dissipation. Easy acquisitions of remote sensing data make regional and global comparisons possible, and then the differences of formations can be explored. The morphology can probably provide an evidence of antecedent river and demonstrate the channel incisions and crustal uplifts. Meander core in Chinese is named after the isolated hills in the Jialing River. 9Proteomic analysis of heat stress resistance of cucumber leaves when grafted onto Momordica rootstock显示文摘Various biotic and abiotic stresses threaten the cultivation of future agricultural crops.Among these stresses,heat stress is a major abiotic stress that substantially reduces agricultural productivity.Many strategies to enhance heat stress tolerance of crops have been developed,among which is grafting.Here,we show that Momordica-grafted cucumber scions have intrinsically enhanced chlorophyll content,leaf area,and net photosynthetic rate under heat stress compared to plants grafted onto cucumber rootstock.To investigate the mechanisms by which Momordica rootstock enhanced cucumber scions heat stress tolerance,comparative proteomic analysis of cucumber leaves in response to rootstock-grafting and/or heat stress was conducted.Seventy-seven differentially accumulated proteins involved in diverse biological processes were identified by two-dimensional electrophoresis(2-DE)in conjunction with matrix-assisted laser desorption/ionization time-of-flight/time-of-flight mass spectrometry(MALDI-TOF/TOF MS).The following four main categories of proteins were involved:photosynthesis(42.8%),energy and metabolism(18.2%),defense response(14.3%),and protein and nucleic acid biosynthesis(11.7%).Proteomic analysis revealed that scions grafted onto Momordica rootstocks upregulated more proteins involved in photosynthesis compared to scions grafted onto cucumber rootstocks under heat stress and indicated enhanced photosynthetic capacity when seedlings were exposed to heat stress.Furthermore,the expression of photosynthesis-related genes in plants grafted onto Momordica rootstocks significantly increased in response to heat stress.In addition,increased high-temperature tolerance of plants grafted onto Momordica rootstock was associated with the accumulation of ribulose-1,5-bisphosphate carboxylase/oxygenase(Rubisco)and oxygen-evolving enhancer protein 1(OEE1).Taken together,the data indicated that Momordica rootstock might alleviate growth inhibition caused by heat stress by improving photosynthesis,providing valuable insight into enhancing heat stress tolerance in the global warming epoch.Ye Xu Yinghui Yuan Nanshan Du Yu Wang Sheng Shu Jin Sun Shirong Guo 2018Horticulture Research2018,5,1:6
10Scaling behavior of magnitude clusters in aftershock sequence:An example of the Wenchuan Earthquake,China显示文摘The Gutenberg-Richter and Omori Laws, which are generally used to characterize the temporal distribution of aftershock, failed to reflect the statistic properties of climatic outbreak of aftershock energy. Based on a new concept of magnitude clusters describing the fluctuation of aftershock energy release of the Wenchuan Earthquake, we discovered that the pattern of the continuous high-magnitude aftershock follows a power-law rather than a non-Poisson distribution. This suggests that the after-shocks with high magnitudes are statistically clustered. We then divided the aftershock sequences into three sections and demonstrated that though the probability of strong outbreaks decreased with time, there exists a high possibility of the occurrence of isolated high-magnitude aftershocks in the future. Based on self-organized criticality theory, the mechanisms of the power-law pattern of magnitude clusters are discussed. 11FGFR/RACK1 interacts with MDM2, promotes P53 degradation, and inhibits cell senescence in lung squamous cell carcinoma显示文摘Objective:FGFR is considered an important driver gene of lung squamous cell carcinoma(LSCC).Thus,identification of the biological events downstream of FGFR is important for the treatment of this malignancy.Our previous study has shown that the FGFR/RACK1 complex interacts with PKM2 and consequently promotes glycolysis in LSCC cells.However,the biological functions of the FGFR/RACK1 complex remain poorly understood.Methods:Anchorage-independent assays and in vivo tumorigenesis assays were performed to evaluate cancer cell malignancy.Distant seeding assays were performed to evaluate cancer cell metastasis.β-gal staining was used to examine cell senescence,and immunoprecipitation assays were performed to examine the interactions among FGFR,RACK1,and MDM2.Results:FGFR/RACK1 was found to regulate the senescence of LSCC cells.Treatment with PD166866,an inhibitor of FGFR,or knockdown of RACK1 induced senescence in LSCC cells(P<0.01).A molecular mechanistic study showed that FGFR/RACK1/MDM2 form a complex that promotes the degradation of p53 and thus inhibits cell senescence.PD166866 and RG7112,an MDM2/p53 inhibitor,cooperatively inhibited the colony formation and distal seeding of LSCC cells(P<0.01),and upregulated the expression of p53 and p21.Conclusions:Together,our findings revealed the regulatory roles and mechanisms of FGFR/RACK1 in cell senescence.This understanding should be important in the treatment of LSCC.Tao Chen Fei Wang Shupei Wei Yingjie Nie Xiaotao Zheng Yu Deng Xubin Zhu Yuezhen Deng Nanshan Zhong Chengzhi Zhou 2021Cancer Biology & Medicine2021,18,3:3

15Analysis of Ecosystem Degradation Factors in Yuanmou Arid-Hot Valleys Based on Interpretative Structural Model显示文摘For ecological restoration and reconstruction of the degraded area, it is an important premise to correctly understand the degradation factors of the ecosystem in the arid-hot valleys. The factors including vegetation deg-radation, land degradation, arid climate, policy failure, forest fire, rapid population growth, excessive deforestation, overgrazing, steep slope reclama-tion, economic poverty, engineering construction, lithology, slope, low cul-tural level, geological hazards, biological disaster, soil properties etc, were selected to study the Yuanmou arid-hot valleys. Based on the interpretative structural model (ISM), it has found out that the degradation factors of the Yuanmou arid-hot valleys were not at the same level but in a multilevel hier-archical system with internal relations, which pointed out that the degradation mode of the arid-hot valleys was 'straight (appearance)-penetrating-back-ground' . 16SARS-CoV-2 spike protein receptor-binding domain perturbates intracellular calcium homeostasis and impairs pulmonary vascular endothelial cells显示文摘Exposure to the spike protein or receptor-binding domain(S-RBD)of SARS-CoV-2 significantly influences endothelial cells and induces pulmonary vascular endotheliopathy.In this study,angiotensin-converting enzyme 2 humanized inbred(hACE2 Tg)mice and cultured pulmonary vascular endothelial cells were used to investigate how spike protein/S-RBD impacts pulmonary vascular endothelium.Results show that S-RBD leads to acute-to-prolonged induction of the intracellular free calcium concentration([Ca^(2+)]i)via acute activation of TRPV4,and prolonged upregulation of mechanosensitive channel Piezo1 and store-operated calcium channel(SOCC)key component Orai1 in cultured human pulmonary arterial endothelial cells(PAECs).In mechanism,S-RBD interacts with ACE2 to induce formation of clusters involving Orai1,Piezo1 and TRPC1,facilitate the channel activation of Piezo1 and SOCC,and lead to elevated apoptosis.These effects are blocked by Kobophenol A,which inhibits the binding between S-RBD and ACE2,or intracellular calcium chelator,BAPTA-AM.Blockade of Piezo1 and SOCC by GsMTx4 effectively protects the S-RBDinduced pulmonary microvascular endothelial damage in hACE2 Tg mice via normalizing the elevated[Ca^(2+)]i.Comparing to prototypic strain,Omicron variants(BA.5.2 and XBB)of S-RBD induces significantly less severe cell apoptosis.Transcriptomic analysis indicates that prototypic S-RBD confers more severe acute impacts than Delta or Lambda S-RBD.In summary,this study provides compelling evidence that S-RBD could induce persistent pulmonary vascular endothelial damage by binding to ACE2 and triggering[Ca^(2+)]i through upregulation of Piezo1 and Orai1.Targeted inhibition of ACE2-Piezo1/SOCC-[Ca^(2+)]i axis proves a powerful strategy to treat S-RBD-induced pulmonary vascular diseases.Kai Yang Shiyun Liu Han Yan Wenju Lu Xiaoqian Shan Haixia Chen Changlei Bao Huazhuo Feng Jing Liao Shuxin Liang Lei Xu Haiyang Tang Jason X-J.Yuan Nanshan Zhong Jian Wang 2023Signal Transduction and Targeted Therapy2023,8,8:2
17Impeding the combination of astrocytic ASCT2 and NLRP3 by talniflumate alleviates neuroinflammation in experimental models of Parkinson's disease显示文摘Alanine-serine-cysteine transporter 2(ASCT2)is reported to participate in the progression of tumors and metabolic diseases.It is also considered to play a crucial role in the glutamate-glutamine shuttle of neuroglial network.However,it remains unclear the involvement of ASCT2 in neurological diseases such as Parkinson’s disease(PD).In this study,we demonstrated that high expression of ASCT2 in the plasma samples of PD patients and the midbrain of MPTP mouse models is positively correlated with dyskinesia.We further illustrated that ASCT2 expressed in astrocytes rather than neurons significantly upregulated in response to either MPP+or LPS/ATP challenge.Genetic ablation of astrocytic ASCT2 alleviated the neuroinflammation and rescued dopaminergic(DA)neuron damage in PD models in vitro and in vivo.Notably,the binding of ASCT2 to NLRP3 aggravates astrocytic inflammasometriggered neuroinflammation.Then a panel of 2513 FDA-approved drugs were performed via virtual molecular screening based on the target ASCT2 and we succeed in getting the drug talniflumate.It is validated talniflumate impedes astrocytic inflammation and prevents degeneration of DA neurons in PD models.Collectively,these findings reveal the role of astrocytic ASCT2 in the pathogenesis of PD,broaden the therapeutic strategy and provide a promising candidate drug for PD treatment.Yang Liu Ting Liu Yuanzhang Zhou Wenjie Li Min Wang Nanshan Song Wenbin Zhang Jingwei Jiang Shengtao Yuan Jianhua Ding Gang Hu Ming Lu 2023Acta Pharmaceutica Sinica B2023,13,2:2
