| 5 | Positive allosteric regulation of PAC1-R up-regulates PAC1-R and its specific ligand PACAP显示文摘PAC1-R is a recognized preferential receptor for the neuropeptide of pituitary adenylate cyclase-activating polypeptide(PACAP),which mediates neuroprotective and nerve regenerative activities of PACAP.In this study,we found that in both PAC1R-CHO cells with high expression of PAC1R-eGFP and retinal ganglion cells(RGC-5)with the natural expression of PAC1-R,oligo-peptide PACAP(28-38)and the positively charged arginine-rich penetrating peptide TAT,as positive allosteric modulators of PAC1-R,significantly trigger the nuclear translocation of PAC1-R.The chromatin immunoprecipitation(ChIP)-PCR results show that the nuclear translocated PAC1-R binds with the promoter regions of PAC1-R and its specific ligand PACAP.The up-regulated promoter activities of PAC1-R and PACAP induced by PACAP(28-38)or TAT are positively correlative with the increase of the expression levels of PAC1-R and PACAP.Moreover,the nuclear translocation of PAC1-R induced by PACAP(28-38)or TAT is significantly inhibited by the mutation of PAC1-R on Cys25 and the palmitoylation inhibitor 2-bromopalmitate.Meanwhile,the increase in both PAC1-R and PACAP levels and the neuroprotective activities of PACAP(28-38)and TAT in MPPinduced cell model of Parkinson’s disease are synchronously inhibited by 2-bromopalmitate,which are positively correlated with the nuclear translocation of PAC1-R induced by PACAP(28-38)or TAT.Bioinformatics analysis and motif enrichment analysis following ChIP-sequencing show that the transcription factors including SP1,Zic2,GATA1,REST and YY1 may be recruited by nuclear PAC1-R and involved in regulating the promoter activities of PAC1-R and PACAP.ChIP-sequencing and related bioinformatics analysis show that the downstream target genes regulated by the nuclear PAC1-R are mostly involved in the process of cellular stress and related to neuroprotection,neuronal genesis and development. | Guangchun Fan Zhengxin Tao Shang Chen Huahua Zhang Rongjie Yu | 2022 | Acta Biochimica et Biophysica Sinica2022,54,5: | 1 |

| 7 | Testing and Modeling of the Dynamic Response Characteristics of Pulsating Heat Pipes during the Start-up Process显示文摘Pulsating heat pipes(PHPs) are two-phase heat transfer heat pipes with high heat transfer capability and simple structure. Heating power is an important factor that affects the start-up response characteristics of PHPs. The operational characteristics during the start-up and stable operating stages were studied through experiments, and the corresponding dynamic response model under a specified heating power was established based on experimental data and flow pattern in the tube. The starting time, starting temperature, and dynamic response characteristic parameters at a certain heating power were calculated by the dynamic response model. The response characteristics of working fluid during the stable operation of PHPs were deduced based on the dynamic response curve of PHPs during the non-operational and stable operation stages. The response characteristics of PHPs for the step effect(given heating power) were quantitatively described by amplification factor K and time constant τ, thereby presenting the basis for the study on heat and mass transfer mechanisms of PHPs from non-operational to steady operation stage. Results showed that the minimum thermal resistance and the minimum time constant of the PHP are approximately 0.28 °C/W and 75, respectively, obtained at a heating power of 160 W. Moreover, these results indicated that the dynamic response of PHPs demonstrates a favourable performance and rapidly reaches another stable working state when their heat transfer performance is stable. | 10 | Heat Leakage Numerical Investigation of a Compound Parabolic ConcentratorPulsating Heat Pipe Solar Collector显示文摘As a new type of equipment for solar medium temperature utilization, the compound parabolic concentrator-pulsating heat pipe solar collector(CPC-PHPSC) uses pulsating heat pipe(PHP) as an endotherm, which can realize efficient energy conversion. The design of proper concentration ratio of compound parabolic concentrator(CPC) ensures that the incident sunlight can be concentrated on the evaporator section surface of PHP without solar tracking system. The objective of the present work is to study the influence of solar radiation intensity, air speed, material thickness(glass and insulation board) and tilt angle on the thermal performance of the new collector, which is difficult to control in the experiment. The heat leakage process and characteristics of the CPC-PHPSC were numerically studied by establishing a 3D numerical model of the collector unit. The results show that the theoretical collector efficiency of CPC-PHPSC reaches 74.5%, which is consistent with the experimental results. During operation, the heat collection performance is the best when the tilt angle is 45° and the solar radiation intensity is 1000 W/m^(2), while the excessive air speed will increase the convective heat loss. | 11 | Variation in Bombyx mori immune response against fungal pathogen Beauveria bassiana with variability in cell wallβ-1,3-glucan显示文摘Entomopathogenic fungi are protected by a cell wall with dynamic structure for adapting to various environmental conditions.β-1,3-Glucan recognition proteins activate the innate immune system of insects by recognizing surface molecules of fungi.However,the associations between pathogenicity and the different components of entomopathogenic fungal cell walls remain unclear.Three Beauveria bassiana strains were selected that have significantly differing virulence against Bombyx mori.The molecular mechanisms underlying the immune response in B.mori were investigated using RNA sequencing,which revealed differences in the immune response to different B.bassiana strains at 12 h post-infection.Immunofluorescence assays revealed thatβ-1,3-glucan content had an opposite trend to that of fungal virulence.β-1,3-Glucan injection upregulated BmβGRP4 expression and significantly reduced the virulence of the high-virulence strain but not that of the medium-virulence or low-virulence strains.BmβGRP4 silencing in B.mori with RNA interference resulted in the opposite virulence pattern,indicating that the virulence of B.bassiana was affected by the cell walls’content ofβ-1,3-glucan,which could be recognized by BmβGRP4.Furthermore,interference with the gene CnA(calcineurin catalytic A subunit)involved inβ-1,3-glucan synthesis eliminated differences in virulence between B.bassiana strains.These results indicate that strains of a single species of pathogenic fungi that have differing cell wall components are recognized differently by the innate immune system of B.mori. | Yulong Wang Zhen Liu Xuebing Yin Shihong Liu Kai Wang Rongjie Wan Haoran Chen Xinyang Li Bo Huang | 2024 | Insect Science2024,31,1: | 0 |

| 14 | A novel small positive allosteric modulator of neuropeptide receptor PAC1-R exerts neuroprotective effects in MPTP mouse Parkinson's disease model显示文摘As a neuropeptide pituitary adenylate cyclase-activating polypeptide(PACAP)-preferring receptor,PAC1-R mediates effective neuroprotective activity.Based on the finding that the antibiotic doxycycline(DOX)with clinical neuroprotective activity functions as a positive allosteric modulator(PAM)of neuropeptide PACAP receptor 1(PAC1-R),we use virtual and laboratory screening to search for novel small molecule PAMs of PAC1-R.Virtual screening is carried out using a small-molecule library TargetMol.After two-level precision screening with Glide,the top five compounds with the best predicted affinities for PAC1-R are selected and named small positive allosteric modulator 1-5(SPAM1-5).Our results show that only 4-[4-(4-Oxo-3,4-2-yl)butanamido]methyl)benzoic acid(SPAM1)has stronger neuroprotective activity than DOX in the MPP+PD cell model and MPTP PD mouse model.SPAM1 has a higher affinity for PAC1-R than DOX,but has no antibiotic activity.Moreover,both SPAM1 and DOX block the decrease of PAC1-R level in mouse brain tissues induced by MPTP.The successful screening of SPAM1 offers a novel drug for the treatment of neurodegenerative disease targeting the PAC1-R. | Guangchun Fan Shang Chen Zhengxin Tao Huahua Zhang Rongjie Yu | 2022 | Acta Biochimica et Biophysica Sinica2022,54,9: | 0 |

In the present study, we observed the effects of transfecting three amyloid precursor-like protein 2(APLP2) C-terminal fragments(CTFs; C57, C50 and C31) in the p EGFP-N1 vector on S100A9 expression in BV2 cells. Reverse transcription-PCR, western blot assay and immunocytochemistry revealed that S100A9 protein and m RNA expression was greater in BV2 cells after CTF transfection than after mock transfection with an empty vector. Furthermore, transfection of full-length APLP2-751 resulted in low levels of S100A9 protein expression. | 18 | Unprecedent green macroalgae bloom:mechanism and implication to disaster prediction and prevention显示文摘Green macroalgae bloom(GMB),with the dominant species of Ulva prolifera,has regularly occurred since 2007 along the China coast.Although disaster prevention and control achieved favorable results in 2020,the satellite-observed GMB annual maximum coverage(AMC)rebounded sharply in 2021 to an unprecedented level.The reasons for this rebound and the significant interannual variability over past 15 years are still open questions.Here,by using long-term time-series(2007-2022)optical and Synthetic Aperture Radar satellite observations(1000+scenes),meteorological data and water quality statistics,the mechanism analysis was performed by exploring effects from natural factors and human activities.Two key determinants for AMC are successfully identified from numerous potential factors which are the macroalgae distribution in a key area(the Subei Shoal)during a critical period(from April to May 20)and the nutrient availability.Furthermore,by using these two parameters,a novel model for AMC prediction(R^(2)=0.87,p<0.01)is proposed and independently validated,which can reasonably explain the significant interannual variability(2014-2021)and agree well with the latest observation in 2022(percentage difference 12%).Finally,suggestions are proposed for future disaster prevention and alleviation.This work may aid future bloom prediction and management measure optimization. | Mengmeng Cao Xuyan Li Tingwei Cui Xinliang Pan Yan Li Yanlong Chen Ning Wang Yanfang Xiao Xingai Song Yuzhu Xu Runa A Bing Mu Song Qing Rongjie Liu Wenjing Zhao Yuhai Bao Jie Zhang Lan Wei | 2023 | International Journal of Digital Earth2023,16,1: | 0 |
