| 2 | Comparison of clinical effectiveness of acupuncture and a Western drug on allergic rhinitis:study protocol for a randomized controlled trial显示文摘OBJECTIVE: To compare the efficacy of an acupuncture regimen for persistent allergic rhinitis(PER), aimed at improving a patient's mind or Shen in Traditional Chinese Medicine, to that of a second-generation H1-receptor antagonist, cetirizine hydrochloride.METHODS: This multicenter, randomized, controlled clinical trial on PER will be conducted at three institutions in China. The total study period will be 9 weeks. After a 1-week preparatory screening period, 240 eligible participants with PER willbe randomized to receive acupuncture or pharmacotherapy(1∶1) for 4 weeks with a 4-week follow-up. The primary outcome will be changes in7-day average total nasal symptom score. Secondary outcome measures include rhinoconjunctivitis quality of life questionnaire score and total non-nasal symptom score.RESULTS:The presence and seriousness of psychological and emotional impairments should be considered in therapeutic programs for allergic rhinitis.No clinical trial for treating allergic rhinitis via acupuncture regulation of psychological and emotionalactivitieshasbeenreported.CONCLUSION:The findings of the trial will allow us to determine the effects of the mind(Shen)-regulation treatment approach. | 3 | IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. | 4 | The influence of urban park characteristics on bird diversity in Nanjing, China显示文摘Background:Habitat loss,fragmentation and decrease of habitat quality caused by urbanization have led to a dramatic decline in biodiversity worldwide.For highly urbanized areas,parks have become“islands”or habitat fragments for wildlife.As an important indicator group of urban ecosystem health,the response of birds to urbanization has attracted the global attention of ecologists.Understanding the key factors affecting bird diversity in urbanized environment is crucial to the protection of biodiversity in urban ecosystems.Methods:We used the line-transect method to survey birds in 37 urban parks in Nanjing,China.We also measured a number of park characteristics(area,isolation,shape index,environmental noise,distance to city center,and habitat diversity)that are commonly assumed to influence bird diversity.We then used the information-theoretic multi-model inference approach to determine which park characteristics had significant impacts on bird species richness.Results:We found that park area,habitat diversity and the distance to city center were the best positive predictors of bird species richness in Nanjing urban parks.By contrast,park isolation,park shape and environmental noise had little or no influence on bird diversity.Conclusions:Our study highlights the importance of park area,habitat diversity and the distance to city center in determining bird diversity in Nanjing city parks.Therefore,from a conservation viewpoint,we recommend that large parks with complex and diverse habitats far away from the city center should be retained or constructed to increase bird diversity in urban design and planning. | Xueru Yang Xinwei Tan Chuanwu Chen Yanping Wang | 2020 | Avian Research2020,11,4: | 5 |
| 5 | Three-dimensional reconstruction of Picea wilsonii Mast. pollen grains using automated electron microscopy显示文摘Three-dimensional electron microscopy(3 D-EM) has attracted considerable attention because of its ability to provide detailed information with respect to developmental analysis. However, large-scale high-resolution 3 D reconstruction of biological samples remains challenging. Herein, we present a 3 D view of a Picea wilsonii Mast. pollen grain with 100 nm axial and38.57 nm lateral resolution using AutoCUTS-SEM(automatic collector of ultrathin sections-scanning electron microscopy). We established a library of 3,127 100 nm thick serial sections of pollen grains for preservation and observation, demonstrating that the protocol can be used to analyze large-volume samples. After obtaining the SEM images, we reconstructed an entire pollen grain comprising 734 serial sections. The images produced by 3D reconstruction clearly revealed the main components of the P.wilsonii pollen grain, i.e., two sacci and pollen corpus, tube cell, generative cell, and two degenerated prothallial cells, and their internal organization. In addition, we performed a quantitative analysis of the different pollen grain cells, including sacci, and found that there were 202 connections within a saccus SEM image. | 9 | Nestedness and underlying processes of bird assemblages in Nanjing urban parks显示文摘Nestedness is an important pattern frequently reported for species assemblages on islands or fragmented systems.However,to date,there are few studies that comprehensively investigated faunal nestedness and underlying processes in urbanized landscapes.In this study,we examined the nestedness of bird assemblages and its underlying causal mechanisms in 37 urban parks in Nanjing,China.We used the line-transect method to survey birds from April 2019 to January 2020.We used the Weighted Nestedness metric based on Overlap and Decreasing Fill(WNODF)to estimate the nestedness of bird assemblages.We applied spearman partial correlation test to examine the relationships between nestedness ranks of sites and park characteristics(area,isolation,anthropogenic noise,number of habitat types,and building index),as well as between nestedness ranks of species and their ecological traits(body size,geographic range size,clutch size,minimum area requirement,dispersal ratio,and habitat specificity).We found that bird assemblages in urban parks were significantly nested.Park area,habitat diversity,building index,habitat specificity,and minimum area requirement of birds were significantly correlated with nestedness.Therefore,the nestedness of bird assemblages was caused by selective extinction,habitat nestedness,and urbanization.However,the nestedness of bird assemblages did not result from passive sampling,selective colonization,or human disturbance.Overall,to maximize the number of species preserved in our system,conservation priority should be given to parks with large area,rich habitat diversity,and less building index.From a species perspective,we should focus on species with large area requirement and high habitat specificity for their effective conservation. | Xinwei Tan Xueru Yang Chuanwu Chen Yanping Wang | 2021 | Current Zoology2021,67,4: | 2 |

[Method]Oxford cup double-plate method was used for preliminary screening of broad-spectrum bacteriocin-producing Lactobacillus strain; after eliminating the interference of organic acid and hydrogen peroxide,antibacterial substances containing protein properties produced by the screened strain were detected; finally,the obtained strain was identified. [Result]After elimination of interference factors such as organic acid and hydrogen peroxide,the fermentation supernatant of strain CW3 could still inhibit the growth of indicator strains; the antibacterial activity of fermentation broth was significantly reduced after treatment with trypsase and pepsase,which indicated that the produced antibacterial substances were bacteriocins. Results of physiological and biochemical identification showed that strain CW3 belonged to Lactobacillus plantarum. | 11 | Expression of multidrug resistance 1 gene and C3435T genetic polymorphism in peripheral blood of patients with intractable epilepsy显示文摘BACKGROUND:Increased expression of multidrug resistance 1 (MDR1) mRNA in peripheral blood of pa-tients with intractable epilepsy is not due to epilepsy drugs,but epilepsy behavior. Monitoring MDR1 ex-pression in peripheral blood is a target for MDR1 gene evaluation. OBJECTIVE: To investigate the influence of antiepileptic drugs and seizures on MDR expression in intrac-table epilepsy,and to analyze the genetic polymorphisms of C3435T in the MDR1 gene. DESIGN,TIME AND SETTING: Factorial designs and comparative observations at the experimental cen-ter of the Affiliated Hospital of Qingdao Medical College,Qingdao University between October 2003 and October 2004. PARTICIPANTS: A total of 120 subjects were recruited from the epilepsy clinical department of the Affili-ated Hospital of Qingdao Medical College. Four groups (n=30) were classified according to statistical fac-torial design: intractable epilepsy,treatment response,no treatment,and normal control groups. METHODS: One-step semi-quantitative reverse-transcription polymerase chain reaction technology was used to test expressions of the MDR1 gene in 120 subjects. C3435T polymorphisms in intractable epilepsy group and normal control groups were analyzed by polymerase chain reaction-restriction fragment length polymorphism. MAIN OUTCOME MEASURES: Expression of MDR1 mRNA in the four groups,and C3435T genetic polymorphisms in intractable epilepsy and normal control groups. RESULTS: MDR1 gene expression was increased in the intractable epilepsy group,due to the factor sei-zures,but not the antiepileptic drugs. However,the interaction between the two factors was not statistically significant. Of the 30 subjects in the intractable epilepsy group,the following genotypes were exhibited: 3 (10%) C/C genotype,9 (30%) C/T genotype,and 18 (60%) T/T genotype at the site of C3435T,while 4 (13%),10 (33%),and 16 (53%) subjects were determined to express these genotypes in the normal control group,respectively. C and T allele frequency were 25% and 75% in the intractable epilepsy group,and 30% and 70% in the normal control group,respectively. However,there was no statistical difference between the groups. CONCLUSION: Results demonstrated that seizures,not antiepileptic drugs,induced MDR1 gene expres-sion in intractable epilepsy. Since its practical equivalent phase centers are usually distributed sparsely and nonuniformly, traditional 3D SAR algorithms suffer from low resolution and high sidelobes in cross-track dimension. To deal with this problem, this paper introduces a method based on back-projection and convex optimization to achieve 3D high accuracy imaging reconstruction. Compared with traditional SAR algorithms, the proposed method sufficiently utilizes the sparsity of the 3D SAR imaging scene and can achieve lower sidelobes and higher resolution in cross-track dimension. | 15 | 槲皮素对人肺癌细胞株A549化疗药物增敏作用的实验研究(英文)显示文摘Objective: The present study was designed to investigate whether quercetin exerts increasing chemosensitivity on human lung adenocarcinoma cells when quercetin combined with cisplatin (DDP) and vincristine (VCR) in vitro respectively and its possible antitumor mechanism. To provide experimental proof for clinical combination application. Methods: Using intermittent administration of high dose VCR, human lung adenocarcinoma sensitive cell line (A549/S) was induced to VCR-resistant human lung adenocarcinoma cell line (A549/VCR). MTT assay was adapted for examing the 50% inhibition (IC50) value of DDP and VCR on A549/S and A549/VCR when quercetin combined with DDP and VCR respectively. Results: IC50 of DDP on A549/S and A549/VCR was 10.18 and 12.35 mg/L, and the IC50 of VCR on the two cell lines was 1.21 and 12.77 mg/L, respectively. The resistance fold of A549/VCR on VCR and DDP was 10.55 and 1.21, respectively. When quercetin at concentration of 50, 100 and 200 μmol/L in combination with DDP and VCR respectively, the IC50 of DDP and VCR on A549/S and A549/VCR were obvious decreased (P < 0.05 - P < 0.01). A polarlike or pseudopolar format grid is introduced to sample data close to the resolution, which presents the design of a separable kernel for efficient FFT implementation.The proposed imaging algorithm formulates the reflectivity image of the target scene as an interpolation-free double image series expansion with two usual approximation-induced phase error terms being taken into account,whereby more generalized application scenarios with high frequency, large bandwidth or larger aperture length for imaging a target scene located within either the far-field or the near-field of the radar aperture are processable with high accuracy. In addition, convergence acceleration methods in computational mathematics are introduced to accelerate the convergence of the image series expansion, so as to make the algorithm more efficient. | 18 | Effects of drip irrigation and cropping on soil salinity,ionic composition and waxy corn production in a severely saline and calcareous and gypsiferous soil显示文摘Drip technologies have been suggested as practical for irrigation under conditions of high salinity and for reclamation of saline soils.Drip irrigation triggered by soil water potential thresholds was applied to both reclaim a severely saline calcareous gypsiferous soil and irrigate a waxy corn crop(Zea mays L.sinesis Kulesh).However,there is a lack of knowledge on the sustainability of reclamation of saline soils with drip irrigation and the changes in soil salinity and salt ion composition during the amelioration process.Therefore,effects on soil salinity,its ionic composition,and on crop growth and yields were evaluated in an experiment conducted in the Yinchuan Plain,northwest China.Treatments included fields in their first to fourth years of the drip irrigation reclamation-cropping scheme and adjacent native,non-cropped or irrigated salinesodic land as control.Yield of waxy corn increased and days of growth to maturity decreased as a function of time and reclamation management.The improvement in crop performance could be largely credited to the reduction of soil salinity and changes in salt composition under the drip-irrigated reclamation protocols.The drip irrigation regime created a region of low salinity proximal to the emitters conducive to germination and plant growth.Deleterious ions for crop growth such as Na+and Cl^(-)were reduced while Ca2+and Mg2+concentration increased,especially in the upper 40 cm of soil.After only a single season of drip-irrigated waxy corn production,both Cl-SO_(4)^(2-)ratios and sodium adsorption ratio(SAR)decreased dramatically.The results suggested that drip irrigation is an effective technology for reclamation of severely saline-affected soils,such as those widely distributed over the Ningxia Plain in China and that this or similar reclamation strategy could be appropriate for reclamation of other hard to manage calcareous and gypsiferous soils. | Junli Tan Yaohu Kang Yanping Jiao Shuqin Wan Xina Wang Juncang Tian Ran Erel Alon Ben-Gal | 2023 | International Journal of Agricultural and Biological Engineering2023,16,5: | 0 |
| 19 | An RNA-RNA crosstalk network involving HMGB1 and RICTOR facilitates hepatocellular carcinoma tumorigenesis by promoting glutamine metabolism and impedes immunotherapy by PD-L1+ exosomes activity显示文摘Hepatocellular carcinoma(HCC)is the global leading cause of cancer-related deaths due to the deficiency of targets for precision therapy.A new modality of epigenetic regulation has emerged involving RNA-RNA crosstalk networks where two or more competing endogenous RNAs(ceRNAs)bind to the same microRNAs.However,the contribution of such mechanisms in HCC has not been well studied.Herein,potential HMGB1-driven RNA-RNA crosstalk networks were evaluated at different HCC stages,identifying the mT0RC2 component RICTOR as a potential HMGB1 ceRNA in HBV^(+)early stage HCC.Indeed,elevated HMGB1 mRNA was found to promote the expressio n of RICTOR mRNA through competitively bin ding with the miR-200 family,especially miR-429.Functio nal assays emplo ying overexpressi on or in terference strategies dem on strated that the HMGB1 and RICTOR 3zuntra nslated regions(UTR)epigenetically promoted the malignant proliferation,self-renewal,and tumorigenesis in HCC cells.Intriguingly,in terference agai nst HMGB1 and RICTOR in HCC cells promoted a stron ger an ti-PD-L1 immuno therapy resp on se,which appeared to associate with the production of PD-L1^(+)exosomes.Mechanistically,the HMGB1-driven RNA-RNA crosstalk network facilitated HCC cell glutamine metabolism via dual mechanisms,activating a positive feedback loop involving mT0RC2-AKT-C-MYC to upregulate glutamine synthetase(GS)expression,and inducing mTORCI signaling to derepress SIRT4 on glutamate dehydrogenase(GDH).Meanwhile,this crosstalk network could impede the efficacy of immunotherapy through mTORCI-P70S6K dependent PD-L1 production and PD-L1^(+)exosomes activity.In conclusion,our study highlights the non-coding regulatory role of HMGB1 with implicatio ns for RNA-based therapeutic targeting together with a predictio n of an ti-PD-L1 immuno therapy in HCC. | Yanping Wei Xuewu Tang Yibin Ren Yun Yang Fengliang Song Jingbo Fu Shuowu Liu Miao Yu Jing Chen Suyang Wang Kecheng Zhang Yexiong Tan Zhipeng Han Lixjn Wei Baohua Zhang Zhangjun Cheng Liang Li Hongyang Wang | 2022 | Signal Transduction and Targeted Therapy2022,7,1: | 0 |
