5Precise nanomedicine for intelligent therapy of cancer显示文摘Precise nanomedicine has been extensively explored for efficient cancer imaging and targeted cancer therapy, as evidenced by a few breakthroughs in their preclinical and clinical explorations. Here, we demonstrate the recent advances of intelligent cancer nanomedicine, and discuss the comprehensive understanding of their structure-function relationship for smart and efficient cancer nanomedicine including various imaging and therapeutic applications, as well as nanotoxicity. In particular, a few emerging strategies that have advanced cancer nanomedicine are also highlighted as the emerging focus such as tumor imprisonment, supramolecular chemotherapy, and DNA nanorobot. The challenge and outlook of some scientific and engineering issues are also discussed in future development. 6Non-viral and viral delivery systems for CRISPR-Cas9 technology in the biomedical field显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated protein 9(CRISPR-Cas9) system provides a novel genome editing technology that can precisely target a genomic site to disrupt or repair a specific gene. Some CRISPR-Cas9 systems from different bacteria or artificial variants have been discovered or constructed by biologists, and Cas9 nucleases and single guide RNAs(sgRNA) are the major components of the CRISPR-Cas9 system. These Cas9 systems have been extensively applied for identifying therapeutic targets, identifying gene functions, generating animal models, and developing gene therapies.Moreover, CRISPR-Cas9 systems have been used to partially or completely alleviate disease symptoms by mutating or correcting related genes. However, the efficient transfer of CRISPR-Cas9 system into cells and target organs remains a challenge that affects the robust and precise genome editing activity. The current review focuses on delivery systems for Cas9 mRNA, Cas9 protein, or vectors encoding the Cas9 gene and corresponding sgRNA. Non-viral delivery of Cas9 appears to help Cas9 maintain its on-target effect and reduce off-target effects, and viral vectors for sgRNA and donor template can improve the efficacy of genome editing and homology-directed repair. 8Radiomic features from computed tomography to differentiate invasive pulmonary adenocarcinomas from non-invasive pulmonary adenocarcinomas appearing as part-solid groundglass nodules显示文摘Objective:We aim to investigate radiomic imaging features extracted in computed tomography(CT)images to differentiate invasive pulmonary adenocarcinomas(IPAs)from non-IPAs appearing as part-solid ground-glass nodules(GGNs),and to incorporate significant radiomic features with other clinically-assessed features to develop a diagnostic nomogram model for IPAs.Methods:This retrospective study was performed,with Institutional Review Board approval,on 88 patients with a total of 100 part-solid nodules(56 IPAs and 44 non-IPAs)that were surgically confirmed between February 2014and November 2016 in the First Affiliated Hospital of China Medical University.Quantitative radiomic features were computed automatically on 3D nodule volume segmented from arterial-phase contrast-enhanced CT images.A set of regular risk factors and visually-assessed qualitative CT imaging features were compared with the radiomic features using logistic regression analysis.Three diagnostic models,i.e.,a basis model using the clinical factors and qualitative CT features,a radiomics model using significant radiomic features,and a nomogram model combining all significant features,were built and compared in terms of receiver operating characteristic(ROC)curves.Decision curve analysis was performed for the nomogram model to explore its potential clinical benefit.Results:In addition to three visually-assessed qualitative imaging features,another three quantitative features selected from hundreds of radiomic features were found to be significantly(all P<0.05)associated with IPAs.The diagnostic nomogram model showed a significantly higher performance[area under the ROC curve(AUC)=0.903]in differentiating IPAs from non-IPAs than either the basis model(AUC=0.853,P=0.0009)or the radiomics model(AUC=0.769,P<0.0001).Decision curve analysis indicates a potential benefit of using such a nomogram model in clinical diagnosis.Conclusions:Quantitative radiomic features provide additional information over clinically-assessed qualitative features for differentiating IPAs from non-IPAs appearing as GGNs,and a diagnostic nomogram model including all these significant features may be clinically useful in preoperative strategy planning.Ting Luo Ke Xu Zheng Zhang Lina Zhang Shandong Wu 2019Chinese Journal of Cancer Research2019,31,2:8

12Characteristic Analysis of OC and EC in PM_(2.5) of Typical Haze Weather in Wuhan City显示文摘Long-time large-area haze weather appeared in Wuhan on June 11,2012. It was monitored that PM2. 5hourly concentration obviously rose,and peak value reached 658 μg /m3. OC and PM2. 5presented high correlation,and correlation coefficient was 0. 96. OC daily average concentration occupied 10%-20% in PM2. 5,and difference was big between haze and normal weather. EC occupied 5%,and difference was very small between haze and normal weather. By analyzing change trend of OC/EC,it was found that OC/EC presented increasing trend in late May and was even higher than that during 11-15 June. It was clear that biomass combustion taking straw as the representation has started in late May.But two strong precipitation on May 29 and June 6 inhibited haze weather,and specific meteorological condition caused haze on June 11. Proportion of SOC to OC reached 14%-70%,illustrating that daily difference of secondary photochemical reaction was very big in Wuhan. OC/EC values were respectively 2. 7,3. 5 and 4. 2 in May,June and haze period. SOC daily means were respectively( 5. 33 ±4. 77) and( 32. 5 ±23. 4) μg /m3in May and haze period. 14Hyperechoic demarcation line between a tumor and the muscularis propria layer as a marker for deciding the endoscopic treatment of gastric submucosal tumor显示文摘Minimally invasive endoscopic resection has been rapidly adopted as a new technique for treating patientswith gastric submucosal tumors (SMTs) originating in the muscularis propria (MP) layer. This study was conducted toevaluate the information obtained from endoscopic ultrasonography (EUS) to determine the appropriate endoscopicdissection method for treating SMTs originating in the MP layer. Between February 2014 and May 2016, a total of 50patients with gastric SMTs originating in the MP layer were enrolled in this study. The clinical features of the patientsand their endoscopic, EUS, and histopathologic findings, as well as their postoperative follow-up data, were analyzedin this retrospective study. The mean age of the patients was (55.0±10.2) years, and the male/female ratio was 17:33.Endoscopic submucosal dissection (ESD) was performed on 43 patients and an endoscopic full-thickness resection(EFR) was performed on seven patients. The most frequent location for an SMT was in the upper body region of thestomach (n=16), and the most common pathological diagnosis was a gastrointestinal stromal tumor (GIST) (n=32).The overall rates for complete resection were 95.3% (41/43) and 100.0% (7/7) when the SMTs were treated by ESDand EFR, respectively. The presence of a complete tumor capsule was significantly associated with a complete re-section (P=0.001). Of the cases treated by ESD, nine patients developed perforation, one of whom required laparo-scopic surgery. The remaining patients were closed with clips or purse-string sutures. The presence of an MP2-typetumor (P=0.018) and a wide connection with the MP layer (P=0.044) were significantly associated with perforation. Apreoperative evaluation of the integrity and the location of a tumor capsule and the length of the tumor connection withthe MP layer by EUS can improve the complete resection rate and reduce the occurrence of intraoperative complica-tions. 16Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research.Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao 2021Cancer Biology & Medicine2021,18,1:5
17Optimal waiting period for fresh embryo transfer after hysteroscopic adhesiolysis: a retrospective cohort study显示文摘Background:Very few studies have been conducted regarding the optimal time interval between hysteroscopic adhesiolysis and the embryo transfer(ET).Investigation of this optimal time may be helpful for assisted reproductive technology.Therefore,we investigated effects of the interval between hysteroscopic adhesiolysis and ET upon in vitro fertilization(IVF)cycle outcomes.Methods:Patients were recruited between January 2014 and September 2017 at the Reproductive Hospital Affiliated to Shandong University.Patients who were diagnosed with intra-uterine adhesion(IUA)and underwent hysteroscopic adhesiolysis before fresh IVF-ET or intra-cytoplasmic sperm injection cycles were classified into three groups according to the interval between hysteroscopic adhesiolysis and ET:less than 90 days(Group 1),90 to 180 days(Group 2),and greater than 180 days(Group 3).Baseline characteristics,controlled ovarian stimulation(COS)response,and pregnancy outcomes after ET were compared.Analysis of variance or non-parametric tests were used to test numerical data.The Pearson’s Chi-squared test was used to test categorical data.Results:A total of 312 patients were recruited as follows:112 in Group 1,137 in Group 2,and 63 in Group 3.There were no differences in baseline and COS characteristics among the three groups.The live-birth rate in Group 2(40.1%)was significantly higher than that in Group 1(17.9%;χ^2=14.545,P<0.001).There were no significant differences in the rates of biochemical,ongoing,and clinical pregnancy,and biochemical and clinical pregnancy abortion,as well as stillbirth among the groups.In the mild IUA patients,the live-birth rate was significantly higher in Group 2(42.6%)compared with Group 1(22%;χ^2=8.413,P=0.004).In the moderate IUA patients,Group 2(35.7%)had a higher frequency of live births than Group 1(6.7%;χ^2=8.187,P=0.004).Conclusions:The optimal waiting period for fresh ET after hysteroscopic adhesiolysis was 90 to 180 days in the current study.Ke Deng Xiao-Hua Song Xue-Mei Han Wei-Ya Kong Wei Liu Ting Han Lei Yan 2019Chinese Medical Journal2019,,19:5
18Progress and challenges in blocked impurity band infrared detectors for space-based astronomy显示文摘Astronomical detection at infrared wavelengths is crucial in astrophysics due to the critical information in this wavelength range.Blocked impurity band(BIB) infrared detectors are desirable for space-based astronomical observation due to their broad response range, low dark currents, high quantum efficiencies, and excellent radiation resistance. In this review, typical BIB device structures and device physics development are first introduced. Subsequently, we discuss progress in Si-based BIB detectors with different doping types and emphasize their applications in space-based infrared detection. Additionally, we discuss recent efforts on pixel performance optimization, response extension, and higher operating temperature devices. 19Generation of tooth-like structures from integration-free human urine induced pluripotent stem cells显示文摘Background:Tooth is vital not only for a good smile,but also good health.Yet,we lose tooth regularly due to accidents or diseases.An ideal solution to this problem is to regenerate tooth with patients’own cells.Here we describe the generation of tooth-like structures from integration-free human urine induced pluripotent stem cells(ifhU-iPSCs).Results:We first differentiated ifhU-iPSCs to epithelial sheets,which were then recombined with E14.5 mouse dental mesenchymes.Tooth-like structures were recovered from these recombinants in 3 weeks with success rate up to 30%for 8 different iPSC lines,comparable to H1 hESC.We further detected that ifhU-iPSC derived epithelial sheets differentiated into enamel-secreting ameloblasts in the tooth-like structures,possessing physical properties such as elastic modulus and hardness found in the regular human tooth.Conclusion:Our results demonstrate that ifhU-iPSCs can be used to regenerate patient specific dental tissues or even tooth for further drug screening or regenerative therapies.Jinglei Cai Yanmei Zhang Pengfei Liu Shubin Chen Xuan Wu Yuhua Sun Ang Li Ke Huang Rongping Luo Lihui Wang Ying Liu Ting Zhou Shicheng Wei Guangjin Pan Duanqing Pei 2013Cell Regeneration2013,2,1:4
20Soil properties and herbaceous characteristics in an age sequence of Haloxylon ammodendron plantations in an oasis-desert ecotone of northwestern China显示文摘Haloxylon ammodendron,a typical desert shrub with C4 pathway of photosynthesis,possessing a strong ability to adapt to an extreme drought environment,has a rapid growth rate in sandy lands and is widely used in sand-fixing shelter-forest systems in oasis-desert ecotones.To assess the effects of H.ammodendron plantation on the soil,we measured soil properties and herbaceous characteristics along a nearly 40-year chronosequence after H.ammodendron was planted in shifting sand dunes in an oasis-desert ecotone.Results showed that silt and clay fractions increased significantly in the topsoil.The accumulation rates of soil organic carbon(SOC),total nitrogen(TN) and total phosphorus(TP) were faster in the early stages(0–9 years) and slower in the late stages(9–39 years).The soil p H and electrical conductivity(EC) were higher than those in the non-vegetation dunes.Moreover,the soil properties in the topsoil(0–5 cm) showed larger variation scope than those in the deeper soil layers(5–20 cm).The significant relationships of the soil silt+clay content with the chemical properties mainly appeared in the topsoil.The wind erosion susceptibility of the soil,evaluated by erodible fraction(EF),decreased significantly with increasing H.ammodendron plantation age.Additionally,the annual pioneer herb,Agriophyllum squarrosum,was gradually substituted by the annual salt-tolerant herb,Bassia dasyphylla,with increasing plantation age.These results showed beneficial effects of H.ammodendron plantation on improving soil conditions.However,the dynamics of the herbaceous species also reminded us that the longterm effects of H.ammodendron plantation,especially on changes in vegetation composition,still need further evaluation.ZHANG Ke SU Yongzhong WANG Ting LIU Tingna 2016Journal of Arid Land2016,8,6:4
