1Naringenin reduces lung metastasis in a breast cancer resection model显示文摘Metastasis is the main cause of death in cancer patients. To improve the outcomes of patients undergoing a surgery, new adjuvant therapies that can effectively inhibit metastases have to be developed. Studies have shown that flavonoid naringenin, a natural product that is mainly present in grapes and citrus, may contribute to cancer prevention. It has many advantages compared to traditional chemotherapeutic drugs, such as low toxicity. To determine whether naringenin can also inhibit metastases, a breast cancer resection model that mimics clinical situations was established. We found that orally administered naringenin significantly decreased the number of metastatic tumor cells in the lung and extended the life span of tumor resected mice. Flow cytometry analysis revealed that T cells displayed enhanced antitumor activity in naringenin treated mice, with an increased proportion of IFN-γ and IL-2 expressing T cells. In vitro studies further demonstrated that relief of immunosuppression caused by regulatory T cells might be the fundamental mechanism of metastasis inhibition by naringenin. These results indicate that orally administered naringenin can inhibit the outgrowth of metastases after surgery via regulating host immunity. 2Chrysin promotes osteogenic differentiation via ERK/MAPK activation显示文摘T he effect of the anti-infl ammatory fl avonoid chrysin on osteogenesis was determined in preosteoblast MC3T3-E1 cells.Results demonstrated that chrysin could induce osteogenic differentiation in the absence of other osteo-genic agents.Chrysin treatment promoted the expres-sion of transcription factors(Runx2 and Osx)and bone formation marker genes(Col1A1,OCN,and OPN)as well as enhanced the formation of mineralized nodules.During osteogenic differentiation,chrysin preferentially activated ERK1/2,but not JNK nor the p38 MAPKs.Further experi-ments with inhibitors revealed the co-treatment of U0126,PD98059,or ICI182780(a general ER antagonist)with chrysin effectively abrogated the chrysin-induced osteo-genesis and ERK1/2 activation.Thus,the effect of chrysin on osteogenesis is ERK1/2-dependent and involves ER.Therefore,chrysin has the signifi cant potential to enhance osteogenesis for osteoporosis prevention and treatment.Wenfeng Zeng Yan Yan Fayun Zhang Chunling Zhang Wei Liang 2013Protein & Cell2013,4,7:8
3Asymptotic regulation of cascade systems with unknown control directions and nonlinear parameterization显示文摘A robust partial-state feedback asymptotic regulating control scheme is developed for a class of cascade systems with both nonlinear uncertainties and unknown control directions. A parameter separation technique is introduced to separate the time-varying uncertainty and the unmeasurable state from nonlinear functions. Then, the Nussbaum-type gain method together with the idea of changing supply functions is adopted in the design of a smooth partial-state regulator that can ensure all the signals of the closed-loop system are globally uniformly bounded. Especially, the system state asymptotically converges to zero. 4Heat-fluid-solid coupling model for gas-bearing coal seam and numerical modeling on gas drainage promotion by heat injection显示文摘Improving the absorbed gas to active desorption and seepage and delaying gas drainage attenuation are considered as key methods for increasing drainage efficiency and gas output.According to the solid mechanics theory,the nonlinear Darcy seepage theory and thermodynamics,the heat-fluid-solid coupling model for gassy coal has been improved.The numerical model was founded from the improved multi-field coupling model by COMSOL Multiphysics and gas drainage by borehole down the coal seam enhanced by heat injection was modelled.The results show that the heatfluid-solid model with adsorption effects for gassy coal was well simulated by the improved multi-field model.The mechanism of coal seam gas desorption seepage under the combined action of temperature,stress and adsorption can be well described.Gas desorption and seepage can be enhanced by heat injection into coal seams.The gas drainage rate was directly proportional to the temperature of injected heat in the scope of 30-150 ℃ and increasing in the whole modelleddrainage process (0-1000 d).The increased level was maximum in the initial drainage time and decreasing gradually along with drainage time.The increasing ratio of drainage rate was maximum when the temperature raised from 30 to 60 ℃.Although the drainage rate would increase along with increasing temperature,when exceeding 60 ℃,the increasing ratio of drainage rate with rising temperature would decrease.Gas drainage promotion was more effective in coal seams with lower permeability than with higher permeability.The coal seam temperature in a 5 m distance surrounding the heat injection borehole would rise to around 60 ℃ in 3 months.That was much less than the time of gas drainage in the coal mines in sites with low permeability coal seams.Therefore,it is valuable and feasible to inject heat into coal seams to promote gas drainage,and this has strong feasibility for coal seams with low permeability which are widespread in China.Ruifu Yuan Chunling Chen Xiao Wei Xiaojun Li 2019International Journal of Coal Science & Technology2019,6,4:4
5Genome and transcriptome of Papaver somniferum Chinese landrace CHM indicates that massive genome expansion contributes to high benzylisoquinoline alkaloid biosynthesis显示文摘Opium poppy(Papaver somniferum)is a source of morphine,codeine,and semisynthetic derivatives,including oxycodone and naltrexone.Here,we report the de novo assembly and genomic analysis of P.somniferum traditional landrace‘Chinese Herbal Medicine’.Variations between the 2.62 Gb CHM genome and that of the previously sequenced high noscapine 1(HN1)variety were also explored.Among 79,668 protein-coding genes,we functionally annotated 88.9%,compared to 68.8%reported in the HN1 genome.Gene family and 4DTv comparative analyses with three other Papaveraceae species revealed that opium poppy underwent two whole-genome duplication(WGD)events.The first of these,in ancestral Ranunculales,expanded gene families related to characteristic secondary metabolite production and disease resistance.The more recent species-specific WGD mediated by transposable elements resulted in massive genome expansion.Genes carrying structural variations and large-effect variants associated with agronomically different phenotypes between CHM and HN1 that were identified through our transcriptomic comparison of multiple organs and developmental stages can enable the development of new varieties.These genomic and transcriptomic analyses will provide a valuable resource that informs future basic and agricultural studies of the opium poppy.Li Pei Baishi Wang Jian Ye Xiaodi Hu Lihong Fu Kui Li Zhiyu Ni Zhenlong Wang Yujie Wei Luye Shi Ying Zhang Xue Bai Mengwan Jiang Shuhui Wang Chunling Ma Shujin Li Kaihui Liu Wanshui Li Bin Cong 2021Horticulture Research2021,8,1:4
6On the boundedness of running-in attractors based on recurrence plot and recurrence qualification analysis显示文摘A feature parameter was proposed to quantitatively explore the boundedness of running-in attractors;its variation throughout the friction process was also investigated. The enclosing radius R was built with recurrence plots(RPs) and recurrence qualification analysis(RQA) by using the time delay embedding and phase space reconstruction. Additionally, the typology of RPs and the recurrence rate(RR) were investigated to verify the applicability of R in characterizing the friction process. Results showed that R is larger at the beginning, but exhibits a downward trend in the running-in friction process;R becomes smooth and trends to small steady values during the steady-state friction period, and finally shows an upward trend until failure occurs. The evolution of R, which corresponded with the typology of RPs and RR during friction process, can be used to quantitatively analyze the variation of the running-in attractors and friction state identifacation. 7Expression of Pseudorabies Virus gE Core Epitopes in Escherichia coli Strain BL21 and Utilization of Indirect PRV gE-ELISA显示文摘Pseudorabies virus glycoprotein E( PRV gE) has been recognized as a suitable diagnostic antigen for pseudorabies. In order to produce gE antigen in large quantities and at low cost,a gene fragment encoding PRV gE core epitopes was expressed in E. coli BL21 expression system. SDS-PAGE and Western Blotting revealed that the expression product in culture supernatant of E. coli BL21 was a recombinant protein,approximately 51. 8 Kd. At 5 h post-induction,protein concentration assay showed that the expression product amounted to 1. 65 mg /ml,accounting for 24. 17% of total proteins in the culture supernatant. An indirect PRV gE-ELISA was established by using the recombinant expression product as a coating antigen. Cross-reactivity assay showed that this antigen was PRV specific. In addition,the assay was consistently reproducible. 8Q-Learning-Based Target Selection for Bearings-Only Autonomous Navigation显示文摘This paper presents a Q-learning-based target selection algorithm for spacecraft autonomous navigation using bearing observations of known visible targets.For the considered navigation system,the position and velocity of the spacecraft are estimated using an extended Kalman filter(EKF)with the measurements of inter-satellite line-of-sight(LOS)vectors obtained via an onboard star camera.This paper focuses on the selection of the appropriate target at each observation period for the star camera adaptively,such that the performance of the EKF is enhanced.To derive an effective algorithm,a Q-function is designed to select a proper observation region,while a U-function is introduced to rank the targets in the selected region.Both the Q-function and the U-function are constructed based on the sequence of innovations obtained from the EKF.The efficiency of the Q-learning-based target selection algorithm is illustrated via numerical simulations,which show that the presented algorithm outperforms the traditional target selection strategy based on a Cramer-Rao bound(CRB)in the case that the prior knowledge about the target location is inaccurate.XIONG Kai WEI Chunling 2021Journal of Systems Science & Complexity2021,34,4:2
9Structural vaccinology: structure-based design of influenza A virus hemagglutinin subtype-specific subunit vaccines显示文摘There is a great need for new vaccine development against influenza A viruses due to the drawbacks of traditional vaccines that are mainly prepared using embryonated eggs.The main component of the current split influenza A virus vaccine is viral hemagglutinin(HA)which induces a strong antibody-mediated immune response.To develop a modern vaccine against influenza A viruses,the current research has been focused on the universal vaccines targeting viral M2,NP and HA proteins.Crystallographic studies have shown that HA forms a trimer embedded on the viral envelope surface,and each monomer consists of a globular head(HA1)and a“rod-like”stalk region(HA2),the latter being more conserved among different HA subtypes and being the primary target for universal vaccines.In this study,we rationally designed the HA head based on the crystal structure of the 2009-pandemic influenza A(H1N1)virus HA as a model,tested its immunogenicity in mice,solved its crystal structure and further examined its immunological characteristics.The results show that the HA globular head can be easily prepared by in vitro refolding in an E.coli expression system,which maintains its intact structure and allows for the stimulation of a strong immune response.Together with recent reports on some similar HA globular head preparations we conclude that structure-based rational design of the HA globular head can be used for subtype-specific vaccines against influenza viruses.Chunling Xuan Yi Shi Jianxun Qi Wei Zhang Haixia Xiao George F.Gao 2011Protein & Cell2011,2,12:2

11Matrix attachment regions included in a bicistronic vector enhances and stabilizes follistatin gene expressions in both transgenic cells and transgenic mice显示文摘In the present study, follistatin(FST) gene expression vectors with either a bicistronic gene transfer cassette alone, or a bicistron gene cassette carrying a matrix attachment region(MAR) were constructed and transfected to bovine fetal fibroblasts. Evaluations of both the integration and expression of exogenous FST indicated that the p MAR-CAG-FST-IRES-Ac GFP1-poly A-MAR(pMAR-FST) vector had higher capacity to form monoclonal transgenic cells than the vector without MAR,though transient transfection and integration efficiency were similar with either construct. Remarkably, protein expression in transgenic cells with the p MAR-FST vector was significantly higher than that from the bicistronic vector. Exogenous FST was expressed in all of the p MARFST transgenic mice at F_0, F_1 and F_2. Total muscle growth in F_0 mice was significantly greater than in wild-type mice,with larger muscles in fore and hind limbs of transgenic mice. p MAR-FST transgenic mice were also found with more evenly distributed muscle bundles and thinner spaces between sarcolemma, which suggests a correlation between transgene expression-associated muscle development and the trend of muscle growth.
