CD147-spike protein is a novel route for SARS-CoV-2 infection to host cells显示文摘In face of the everlasting battle toward COVID-19 and the rapid evolution of SARS-CoV-2,no specific and effective drugs for treating this disease have been reported until today.Angiotensin-converting enzyme 2(ACE2),a receptor of SARS-CoV-2,mediates the virus infection by binding to spike protein.Although ACE2 is expressed in the lung, kidney, and intestine, its expressing levels are rather low, especially in the lung.Considering the great infectivity of COVID-19,we speculate that SARS-CoV-2 may depend on other routes to facilitate its infection. Here,we first discover an interaction between host cell receptor CD147 and SARS-CoV-2 spike protein. The loss of CD147 or blocking CD147 in Vero E6 and BEAS-2B cell lines by anti-CD147 antibody,Meplazumab, inhibits SARSCoV-2 amplification.Expression of human CD147 allows virus entry into non-susceptible BHK-21 cells, which can be neutralized by CD147 extracellular fragment.Viral loads are detectable in the lungs of human CD147(hCD147)mice infected with SARS-CoV-2,but not in those of virus-infected wild type mice.Interestingly, virions are observed in lymphocytes of lung tissue from a COVID-19 patient.Human T cells with a property of ACE2 natural deficiency can be infected with SARS-CoV-2 pseudovirus in a dosedependent manner, which is specifically inhibited by Meplazumab.Furthermore,CD147 mediates virus entering host cells by endocytosis.Together, our study reveals a novel virus entry route,CD147-spike protein, which provides an important target for developing specific and effective drug against COVID-19.Ke Wang Wei Chen Zheng Zhang Yongqiang Deng Jian-Qi Lian Peng Du Ding Wei Yang Zhang Xiu-Xuan Sun Li Gong Xu Yang Lei He Lei Zhang Zhiwei Yang Jie-Jie Geng Ruo Chen Hai Zhang Bin Wang Yu-Meng Zhu Gang Nan Jian-Li Jiang Ling Li Jiao Wu Peng Lin Wan Huang Liangzhi Xie Zhao-Hui Zheng Kui Zhang Jin-Lin Miao Hong-Yong Cui Min Huang Jun Zhang Ling Fu Xiang-Min Yang Zhongpeng Zhao Shihui Sun Hongjing Gu Zhe Wang Chun-Fu Wang Yacheng Lu Ying-Ying Liu Qing-Yi Wang Huijie Bian Ping Zhu Zhi-Nan Chen 2020Signal Transduction and Targeted Therapy2020,5,1:9
Malnutrition and Quality of Life in Chinese Cancer Patients: a Clinical Study of 23,994 Subjects显示文摘Background Malnutrition is common in patients with cancer, and can negatively impact their quality of life(QoL)and even survival. However, there is currently no large data available on the prevalence of malnutrition in Chinese cancer patients. This study evaluated the prevalence of malnutrition and the QoL of Chinese patients with locoregional, recurrent or metastatic cancer. Methods We conducted a nationwide observational,multi-center,hospital-based cross-sectional study within the Chinese Society of Nutritional Oncology(CSNO)Network. All of the patients were diagnosed with one of the following 18 different types of malignant tumors:lung cancer, gastric cancer, liver cancer, colorectal cancer, breast cancer, esophageal cancer, cervical cancer, endometrial cancer, nasopharyngeal carcinoma, malignant lymphoma, leukemia, pancreatic cancer, ovarian cancer, prostate cancer, bladder cancer, brain cancer, biliary tract malignant tumors or gastrointestinal stromal tumors. These patients were enrolled from 72 hospitals located in different regions of China. The patients’nutritional status was evaluated based on the body mass index(BMI),loss of bodyweight, laboratory measurements and patient generated-subjective global assessment(PG-SGA)scores.The cancer patients’physical status and QoL were assessed by the Karnofsky Performance Status(KPS)questionnaire and the European Organization for Research and Treatment of Cancer(EORTC)QLQ-C30 questionnaire, respectively. Results From December 2013 to April 2016,23,994 patients hospitalized for cancer treatment(such as surgery, chemotherapy or radiotherapy)were enrolled in the study. The patients included 12,494(52.9%)males and 11,124(47.1%)females.The mean age was 55.8±13.7 years. The proportions of patients in cancer stagesⅠ,Ⅱ,Ⅲ,Ⅳand uncertain were 11.5%,20.3%,27.5%,30.2%and 10.5%,respectively.Among the 23,994 inpatients, the proportions of patients who were underweight(BMI<18.5 kg/m2),normal(18.5 kg/m2Hong Xia Xu Chun Hua Song Zhen Ming Fu Chang Wang Zeng Qing Guo Yuan Lin Ying Ying Shi Wen Hu Yi Ba Su Yi Li Zeng Ning Li Kun Hua Wang Jing Wu Ying He Jia Jun Yang Cong Hua Xie Fu Xiang Zhou Xin Xia Song Gong Yan Chen Wen Jun Ma Su Xia Luo Zi Hua Chen Ming Hua Cong Hu Ma Chun Ling Zhou Wei Wang Qi Luo Yong Mei Shi Yu Mei Qi Hai Ping Jiang Wen Xian Guan Jun Qiang Chen Jia Xin Chen Yu Fang Lan Zhou Yong Dong Feng Rong Shao Tan Tao Li Jun Wen Ou Qing Chuan Zhao Jian Xiong Wu Min Weng Qing Hua Yao Jiu Wei Cui Wei Li Han Ping Shi the Investigation on Nutrition Status and Clinical Outcome of Common Cancers(INSCOC)Group 2021Journal of Nutritional Oncology2021,6,1:8
Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. The validity of the PG-SGA in Chinese cancer patients warrants further investigation in other cancer types.Zhen Ming Fu Hong Xia Xu Chun Hua Song Wei Li Zeng Qing Guo Yuan Lin Ying Ying Shi Wen Hu Yi Ba Su Yi Li Zeng Ning Li Kun Hua Wang Jing Wu Ying He Jia Jun Yang Cong Hua Xie Xin Xia Song Gong Yan Chen Wen Jun Ma Su Xia Luo Zi Hua Chen Ming Hua Cong Hu Ma Chun Ling Zhou Wei Wang Qi Luo Yong Mei Shi Yu Mei Qi Hai Ping Jiang Wen Xian Guan Jun Qiang Chen Jia Xin Chen Yu Fang Lan Zhou Yong Dong Feng Rong Shao Tan Tao Li Jun Wen Ou Qing Chuan Zhao Jian Xiong Wu Chang Wang Han Ping Shi The Investigation on Nutrition Status and Clinical Outcome of Common Cancers (INSCOC) Group 2016Journal of Nutritional Oncology2016,1,1:7
CD147 antibody specifically and effectively inhibits infection and cytokine storm of SARS-CoV-2 and its variants delta, alpha, beta, and gamma显示文摘SARS-CoV-2 mutations contribute to increased viral transmissibility and immune escape, compromising the effectiveness of existing vaccines and neutralizing antibodies. An in-depth investigation on COVID-19 pathogenesis is urgently needed to develop a strategy against SARS-CoV-2 variants. Here,we identified CD147 as a universal receptor for SARS-CoV-2 and its variants.Meanwhile,Meplazeumab,a humanized anti-CD147 antibody, could block cellular entry of SARS-CoV-2 and its variants-alpha,beta,gamma,and delta, with inhibition rates of 68.7,75.7,52.1,52.1,and 62.3%at 60μg/ml,respectively.Furthermore,humanized CD147 transgenic mice were susceptible to SARS-CoV-2 and its two variants, alpha and beta. When infected, these mice developed exudative alveolar pneumonia, featured by immune responses involving alveoli-infiltrated macrophages, neutrophils, and lymphocytes and activation of IL-17 signaling pathway.Mechanistically,we proposed that severe COVID-19-related cytokine storm is induced by a'spike protein-CD147-CyPA signaling axis':Infection of SARS-CoV-2 through CD147 initiated the JAK-STAT pathway, which further induced expression of cyclophilin A(CyPA);CyPA reciprocally bound to CD147 and triggered MAPK pathway.Consequently, the MAPK pathway regulated the expression of cytokines and chemokines, which promoted the development of cytokine storm.Importantly,Meplazumab could effectively inhibit viral entry and inflammation caused by SARS-CoV-2 and its variants. Therefore, our findings provided a new perspective for severe COVID-19-related pathogenesis.Furthermore, the validated universal receptor for SARS-CoV-2 and its variants can be targeted for COVID-19 treatment.Jiejie Geng Liang Chen Yufeng Yuan Ke Wang Youchun Wang Chuan Qin Guizhen Wu Ruo Chen Zheng Zhang Ding Wei Peng Du Jun Zhang Peng Lin Kui Zhang Yongqiang Deng Ke Xu Jiangning Liu Xiuxuan Sun Ting Guo Xu Yang Jiao Wu Jianli Jiang Ling Li Kun Zhang Zhe Wang Jing Zhang Qingguo Yan Hua Zhu Zhaohui Zheng Jinlin Miao Xianghui Fu Fengfan Yang Xiaochun Chen Hao Tang Yang Zhang Ying Shi Yumeng Zhu Zhuo Pei Fei Huo Xue Liang Yatao Wang Qingyi Wang Wen Xie Yirong Li Mingyan Shi Huijie Bian Ping Zhu Zhi-Nan Chen 2021Signal Transduction and Targeted Therapy2021,6,10:6

Autophagy Attenuates MnCl2-induced Apoptosis in Human Bronchial Epithelial Cells显示文摘Objective To investigate the role of autophagy in MnC l2-induced apoptosis in human bronchial epithelial 16 HBE cells. Methods Cell proliferation was measured by MTT assay.Mitochondrial membrane potential(MMP) and apoptosis were measured by flow cytometry.Autophagic vacuoles were detected by fluorescence microscopy.Cellular levels of apoptosis and autophagy-related proteins were measured by western blotting. Results 16 HBE cell proliferation was inhibited by Mn Cl2 in a dose-and time-dependent manner.Mn Cl2-induced 16 HBE cell growth inhibition was related to MMP depolarization prior to the induction of apoptosis. Our data revealed that Mn Cl2-induced apoptosis in 16 HBE cells was mediated by decreased expression of Bcl-2 and increased levels of cleaved caspase-3.It was observed that when we exposed 16 HBE cells to MnCl_2 in a dose-dependent manner, the formation of autophagic vacuoles and the levels of LC-3B-II were elevated.RNA interference of LC3 B in these Mn Cl2-exposed cells demonstrated that MMP loss and apoptosis were enhanced.Additionally, the pan-caspase inhibitor Z-VAD-FMK increased the cellular levels of Bcl-2 and decreased apoptosis, but did not affect the cellular levels of LC3 B in Mn Cl2-treated 16 HBE cells. Conclusion Mn Cl2 dose-and time-dependently inhibits 16 HBE cell proliferation and induces MMP loss and apoptosis.Autophagy acts in a protective role against Mn Cl2-induced apoptosis in 16 HBE cells.YUAN Zhun YING Xian Ping ZHONG Wei Jian TIAN Shi Min WANG Yu JIA Yong Rui CHEN Wen FU Juan Ling ZHAO Peng ZHOU Zong Can 2016Biomedical and Environmental Sciences2016,29,7:5
Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Gastric Cancer Patients显示文摘Purpose Malnutrition is severe among gastric cancer patients in China. While the Patient-Generated Subjective Global Assessment (PG-SGA) is a widely used nutritional assessment tool, the validation for Chinese patients with gastric cancer has not been performed yet. The aim of this research is to validate the Chinese version of the PGSGA in Chinese gastric cancer patients by conducting analyses of nutrition screening and assessment data. Methods Two thousand consecutive adult patients with gastric cancer were included in this observational, cross-sectional study in several tertiary hospitals in China. A survey was made among those patients, which included several questionnaires and some anthropometric parameters. We calculated the parameters to validate the use of PG-SGA categorizing the nutritional status of gastric cancer patients. Then we performed statistical analysis to evaluate the internal consistency, external consistency, and construct validity. Results This study verified the ability of the PG-SGA to differentiate gastric cancer patients into different nutritional groups. The confirmatory factor analysis verified the original four-factor structure of the PG-SGA. The internal and external consistency was measured by a concordance analysis which showed an intraclass correlation coefficients (ICC) of 0.987. The correlations of the Box 4 score with the Karnofsky performance status score demonstrated alternative form validity with good external consistency (Pearson, r = 0.643, P < 0.001). All three anthropometric dimensions of the PG-SGA were supported by principal component analysis. Spearman correlations (P < 0.001) were significant in the majority of tests. Conclusion The Chinese version of the PG-SGA is a valid tool to access the patients’ nutritional status for Chinese gastric patients.Zhen Ming Fu Hong Xia Xu Chun Hua Song Wei Li Zeng Qing Guo Yuan Lin Ying Ying Shi Wen Hu Yi Ba Su Yi Li Zeng Ning Li Kun Hua Wang Jing Wu Ying He Jia Jun Yang Cong Hua Xie Xin Xia Song Gong Yan Chen Wen Jun Ma Su Xia Luo Zi Hua Chen Ming Hua Cong Hu Ma Chun Ling Zhou Wei Wang Qi Luo Yong Mei Shi Yu Mei Qi Hai Ping Jiang Wen Xian Guan Jun Qiang Chen Jia Xin Chen Yu Fang Lan Zhou Yong Dong Feng Rong Shao Tan Tao Li Jun Wen Ou Qing Chuan Zhao Jian Xiong Wu Chang Wang Han Ping Shi The Investigation on Nutrition Status and Clinical Outcome of Common Cancers (INSCOC) Group 2018Journal of Nutritional Oncology2018,3,4:5
Post-marketing safety surveillance and reevaluation of Motherwort injection:A clinical study of 10094 cases显示文摘OBJECTIVE:To investigate the safety profiles of Motherwort injection(MI).METHODS:A multi-center,prospective and drugderived hospital intensive monitoring method was conducted to assess the safety of MI in real world applications. This study was based on a very large population after the injection was approved and marketed in China. All patients using the injection in participating hospitals were monitored to determine the incidence, pattern, severity and outcome of associated adverse events.RESULTS:The post-marketing surveillance was performed in 10 094 female patients from April to December,2015.The incidence of adverse drug reactions(ADRs) was 0.79‰(8/10 094).Among the 8 patients, the reported adverse events mainly included systemic abnormalities, such as fever, chills and eyelid edema;skin and appendages disorders, such as pruritus and rash;gastrointestinal disorders, such as nausea, abdominal distension and pain;heart rate and rhythm disorders, such as palpitation and increased heart rate. All of these ADRs were mild in severity.CONCLUSION:In this study the ADRs incidence rate of MI is very low, which supports that it is generally safe for use in obstetric and gynecological diseases. However, the total number of 8 ADRs recorded over a relatively short time span seems limited, and the low number of reports could not represent an absolute guarantee of safety.Cao Shan Zhang Wenhao Zhao Ziwei Heng Mingli Bu Huaien Wang Hongwu Liu Xinghui Wang Zhong Cai Yan Ma Yuyan Cui Shihong Deng Jihong Ding Guifeng Ding Yajuan Dong Linhong Duan Zhentao Fan Ling Fan Yang Fu Fen He Jing Ji Shuying Jin Lin Li Hong Li Hongying Liao Tao Lu Wei Luo Xiucui Lü Zhihui Ma Fengchun Ma Dafeng Shi Tianyun Sun Juying Sun Xiaotong Teng Hong Wang Jinhua Wang Ruihua Wang Ying Wang Zhengling Xi Jie Xu Minjuan Xu Zhihong Yan Qian Yang Cuirong Yang Yimei Yin Jie Yu Jinhua Yuan Wenjun Zhang Guanli Zhang Meihua Zhao Renfeng Zhong Yonghong Zhou Jian 2018Journal of Traditional Chinese Medicine2018,38,4:4
Studies on The Adsorption Capacity for Bilirubin of The Adsorbent Chitosan-β-Cyclodextrin显示文摘The adsorbent crosslinked chitosan-β-cyclodextrin (β-CD) was prepared by the reaction of glutaraldehyde with chitosan and β-cyclodextrin. This type of adsorbent has high adsorption capacity for unconjugated bilirubin. The adsorption capacity was related to the β-CD content of the adsorbent; phosphate buffer concentration; temperature; pH value; ionic strength and the adsorbent beads. The results indicated that the chitosan-β-CD was a good adsorbent for unconjugated bilirubin with high capacity.Wei Liang XU, Chun Ling FU, Jun LI Department of Chemistry, Zhejiang University, Hua jia chi section, Hangzhou 310029 2001Chinese Chemical Letters2001,12,4:3
Effects of molybdenum on nutrition, quality, and flavour compounds of strawberry (Fragaria×ananassa Duch. Cv. Akihime) fruit显示文摘Molybdenum(Mo) is an essential trace element in plant nutrition and physiology. It affects photosynthesis and photosynthate accumulation, therefore also affecting fruit quality and nutritional content. This study assessed the effects of different sodium molybdate(Na_2MoO_4) concentrations on strawberry. Five different Mo concentrations were applied in this experiment, including 0, 67.5, 135, 168.75, 202.5 g ha^(-1), respectively. The mineral concentration, including nitrogen(N), Mo, iron(Fe), copper(Cu), and selenium(Se) was assessed in strawberry fruit, as well as chlorophyll content, nutrition quality, taste and aroma. Results showed that net photosynthetic rate(P_n) and chlorophyll content for the strawberry plants increased with an increase in Mo concentration; and the contents of N, Mo, Fe, Cu, total soluble solids(TSS), titratable acidity(TA), sweetness, some sugars, organic acids, and some volatile compounds in the fruit all increased, as well. However, the Mo concentration did not significantly affect the concentrations of Se, sucrose, lactic acid, acetic acid, and some aroma compounds. Fruit sprayed with 135 g ha^(-1) Mo exhibited the highest TSS and sweetness values, as well as the highest N and Fe concentrations among all the treatments. P_n value and chlorophyll content, fructose, glucose, sorbitol and total sugar contents in fruit supplied with 135 g ha^(-1) Mo were also higher than that in other treatments. Fruit sprayed with a Mo concentration of 67.5 g ha^(-1) exhibited significantly higher ascorbic acid(AsA) values than that of control. Ninety-seven volatile compounds were identified in fruit extracted by head-space solid-phase microextraction(HS-SPME) coupled with gas chromatography-mass spectrometry(GC-MS). Fruits sprayed with 135 g ha^(-1) Mo had the highest concentrations of six characteristic aroma compounds, including methyl butanoate, γ-decalactone, ethyl butanoate, methyl hexanoate, γ-dodecalactone, and ethyl caproate.LIU Li XIAO Wei JI Mei-ling YANG Chao LI Ling GAO Dong-sheng FU Xi-ling 2017Journal of Integrative Agriculture2017,16,7:3
Exceptional co-catalyst free photocatalytic activities of B and Fe co-doped SrTiO3 for CO2 conversion and H2 evolution显示文摘CO2 should be converted into chemical-fuels,and to reduce H2O to H2 over SrTiO3 (STO) owing to its negative conduction band position vs.NHE.Herein a novel B and Fe co-doped SrTiO3 (B,F-STO) photocatalyst was successfully fabricated via a single-step sol-hydrothermal process.Various experiments confirmed that B and Fe are effectively doped into the STO matrix.Boron substituted oxygen anions, while Fe substituted Ti cations.UV-visible diffuse reflectance spectra (UV-vis DRS) and valence-band X-ray photoelectron spectroscopy (XPS) spectra confirmed that the band gap of STO significantly reduced from 3.4 to 1.9 eV upon co-doping with B and Fe.Hence, the B,F-STO photocatalyst exhibits more absorption (λ ≤ 650 nm) compared to pure STO (λ ≤ 360 nm).Further, from photoluminescence spectra, fluorescence spectra, and photoelectrochemical measurements, charge separation in STO is considerably enhanced by co-doping B and Fe.This resulted in the improved UV-vis light catalytic activities for CO2 conversion to CH4 and CO and H2O splitting to evolve H2.The amounts of CH4 and CO produced over B,F-STO are ~ 17.2 and 21 μmol, respectively, about 5-fold enhanced compared to that of STO (~ 3.4 μmol CH4 and 5.2 μmol CO),and the calculated quantum efficiency at λ =420 run is ~ 2.16%.Similarly,the amount of H2 produced over B,F-STO is ~ 61 μmol, about 6.7-fold enhanced compared to that over STO (9 μmol),and the calculated quantum efficiency at λ =420 nm is ~ 2.12%.This work provides feasible routes to fabricate highly efficient SrTiO3-based nanophotocatalysts for solar-fuel production.Muhammad Humayun Lei Xu Ling Zhou Zhiping Zheng Qiuyun Fu Wei Luo 2018Nano Research2018,11,12:3
