1Thermodynamic Assessment and Experimental Investigation of Fe-Al-C System显示文摘Phase diagram information of the Fe-C,Fe-Al and Al-C systems are reviewed and the Fe-Al-C system is assessed.A FeAlC database is created by combining a set of thermodynamic parameters established by Kumar and SSOL database in Thermo-Calc software package.Ternary phase diagrams are calculated with FeAlC and the newly developed Thermo-Calc databases SSOL2 and SSOL4.The FeAIC database is the best one to describe the Fe-Al-C system.A1 and A3 values on the vertical section containing 1.5 wt pct Al are calculated with the FeAlC database in this work.To validate the thermodynamic calculation,critical temperatures A_(C_1) and A_(C_3) are determined by using dilatometer analysis.There exist some errors between the calculated values and the experimental results.So further optimization of the Fe-Al-C system regarding bcc and fcc phases is necessary.The experimental data in this work could be of some value in further optimization.Weiyan LU Lin LI Li WANG Yanlin HE Shuigen HUANG 2008Journal of Materials Science & Technology2008,24,5:3

6Improved in situ sequencing for high-resolution targeted spatial transcriptomic analysis in tissue sections显示文摘Spatial transcriptomics enables the study of localization-indexed gene expression activity in tissues,providing the transcriptional landscape that in turn indicates the potential regulatory networks of gene expression.In situ sequencing(ISS)is a targeted spatial transcriptomic technique,based on padlock probe and rolling circle amplification combined with next-generation sequencing chemistry,for highly multiplexed in situ gene expression profiling.Here,we present improved in situ sequencing(IISS)that exploits a new probing and barcoding approach,combined with advanced image analysis pipelines for high-resolution targeted spatial gene expression profiling.We develop an improved combinatorial probe anchor ligation chemistry using a 2-base encoding strategy for barcode interrogation.The new encoding strategy results in higher signal intensity as well as improved specificity for in situ sequencing,while maintaining a streamlined analysis pipeline for targeted spatial transcriptomics.We show that IISS can be applied to both fresh frozen tissue and formalin-fixed paraffin-embedded tissue sections for single-cell level spatial gene expression analysis,based on which the developmental trajectory and cell-cell communication networks can also be constructed.Xinbin Tang Jiayu Chen Xinya Zhang Xuzhu Liu Zhaoxiang Xie Kaipeng Wei Jianlong Qiu Weiyan Ma Chen Lin Rongqin Ke 2023Journal of Genetics and Genomics2023,50,9:1

8Vascular Endothelial Growth Factor‑Recruiting Nanofiber Bandages Promote Multifunctional Skin Regeneration via Improved Angiogenesis and Immunomodulation显示文摘Tissue injury leads to gradients of chemoattractants,which drive multiple processes for tissue repair,including the inflam-matory response as well as endogenous cell recruitment.However,a limited time window for the gradients of chemoattract-ants as well as their poor stability at the injury site may not translate into healthy tissue repair.Consequently,intelligent multifunctional scaffolds with the capability to stabilize injury-induced cytokines and chemokines hold great promise for tissue repair.Vascular endothelial growth factor(VEGF)plays a significant role in wound healing by promoting angiogen-esis.The overarching objective of this research was to develop intelligent multifunctional scaffolds with the capability to endogenously recruit VEGF and promote wound healing via angiogenic and immunomodulatory dual functions.Prominin-1-derived peptide(PR1P)was encapsulated into electrospun poly(L-lactide-coglycolide)/gelatin(P/G)-based bandages.The sustained release of PR1P recruited VEGF in situ,thereby stabilizing the protein concentration peak in vivo and affording a reparative microenvironment with an adequate angiogenic ability at the wound site.Meanwhile,PR1P-recruited VEGF-induced macrophage reprogramming towards M2-like phenotypes further conferred immunomodulatory functions to the bandages.These dual functions of proangiogenesis and immunomodulation formed a cascade amplification,which regulated matrix metalloproteinases(MMP-9)as well as inflammatory factors(nuclear factor(NF)-κb,tumor necrosis factor(TNF)-α)in the wound microenvironment via the VEGF/macrophages/microenvironment axis.Consequently,the bandages realized multifunctional regeneration in splinted excisional wounds in rats,with or without diabetes,affording a higher skin append-age neogenesis,sensory function,and collagen remodeling.Conclusively,our approach encompassing in situ recruitment of VEGF at the injury site with the capability to promote immunomodulation-mediated tissue repair affords a promising avenue for scarless wound regeneration,which may also have implications for other tissue engineering disciplines.Yi Chen Zhengchao Yuan Weiyan Sun Muhammad Shafiq Jun Zhu Jiafei Chen Hai Tang Ling Hu Weikang Lin Yanxi Zeng Long Wang Lei Zhang Yunlang She Hui Zheng Guofang Zhao Dong Xie Xiumei Mo Chang Chen 2023Advanced Fiber Materials2023,5,1:1
9Zinc-finger protein 382 antagonises CDC25A and ZEB1 signaling pathway in breast cancer显示文摘Our previous studies found that Zinc-finger protein 382(ZNF382)played as a tumor suppressor gene in esophageal and gastric cancers,and a positive correlation between the high expression of ZNF382 and better outcome in breast cancer patients.However,the biological roles and mechanisms of ZNF382 in breast cancer remains unclear.We detected ZNF382 expression by reverse-transcription PCR(RT-PCR)and real-time quantitative PCR(qRT-PCR)in breast cancer cells and tissues,and explored the impacts and mechanisms of ectopic ZNF382 expression in breast cancer cells in vitro and in vivo,respectively.Our results revealed that ZNF382 was significantly down-regulated in breast cancer tissues compared with adjacent non-cancer tissues.Restoration of ZNF382 expression in silenced breast cancer cells not only inhibited tumor cell colony formation,viability,migration and invasion,and epithelial-mesenchymal-transition(EMT),but also induced apoptosis and G0/G1 arrest.In conclusion,ZNF382 could induce G0/G1 cell cycle arrest through inhibiting CDC25A signaling,and,inhibit cell migration,invasion and EMT by antagonizing ZEB1 signaling in breast cancer cells.Shuman Li Xiaoqian He Yan Wang Weihong Chen Ran Sun Shaorong Tian Sanxiu He Chunyun Pu Chen Li Dishu Zhou Yu Jiang Qian Tao Lili Li Lin Ye Yue Wu Weiyan Peng Tingxiu Xiang 2023Genes & Diseases2023,10,2:1
10Direct visualization of percolating metal-insulator transition in V_(2)O_(3) using scanning microwave impedance microscopy显示文摘Using the extensively studied V_(2)O_(3) as a prototype system, we investigate the role of percolation in metal-insulator transition(MIT). We apply scanning microwave impedance microscopy to directly determine the metallic phase fraction p and relate it to the macroscopic conductance G, which shows a sudden jump when p reaches the percolation threshold. Interestingly, the conductance G exhibits a hysteretic behavior against p, suggesting two different percolating processes upon cooling and warming. Based on our image analysis and model simulation, we ascribe such hysteretic behavior to different domain nucleation and growth processes between cooling and warming, which is likely caused by the decoupled structural and electronic transitions in V_(2)O_(3) during MIT. 12Norditerpenoids biosynthesized by variediene synthase-associated P450 machinery along with modifications by the host cell Aspergillus oryzae显示文摘The chemical diversity of terpenoids is typically established by terpene synthase-catalyzed cyclization and diversified by post-tailoring modifications.Fungal bifunctional terpene synthase(BFTS)associated P450 enzymes have shown significant catalytic potentials through the development of various new terpenoids with different biological activities.This study discovered the BFTS and its related gene cluster from the plant endophytic fungus Didymosphaeria variabile 17020.Heterologous expression of the BFTS in Saccharomyces cerevisiae resulted in the characterization of a major product diterpene variediene(1),along with two new minor products neovariediene and neoflexibilene.Further heterologous expression of the BFTS and one cytochrome P450 enzyme VndE(CYP6138B1)in Aspergillus oryzae NSAR1 led to the identification of seven norditerpenoids(19 carbons)with a structurally unique 5/5 bicyclic ring system.Interestingly,in vivo experiments suggested that the cyclized terpene variediene(1)was modified by VndE along with the endogenous enzymes from the host cell A.oryzae through serial chemical conversions,followed by multi-site hydroxylation via A.oryzae endogenous enzymes.Our work revealed that the two-enzymes biosynthetic system and host cell machinery could produce structurally unique terpenoids.Lan Jiang Kangjie Lv Guoliang Zhu Zhi Lin Xue Zhang Cuiping Xing Huanting Yang Weiyan Zhang Zhixin Wang Chengwei Liu Xudong Qu Tom Hsiang Lixin Zhang Xueting Liu 2022Synthetic and Systems Biotechnology2022,7,4:0

14On‑Ramp Merging for Highway Autonomous Driving:An Application of a New Safety Indicator in Deep Reinforcement Learning显示文摘This paper proposes an improved decision-making method based on deep reinforcement learning to address on-ramp merging challenges in highway autonomous driving.A novel safety indicator,time difference to merging(TDTM),is introduced,which is used in conjunction with the classic time to collision(TTC)indicator to evaluate driving safety and assist the merging vehicle in finding a suitable gap in traffic,thereby enhancing driving safety.The training of an autonomous driving agent is performed using the Deep Deterministic Policy Gradient(DDPG)algorithm.An action-masking mechanism is deployed to prevent unsafe actions during the policy exploration phase.The proposed DDPG+TDTM+TTC solution is tested in on-ramp merging scenarios with different driving speeds in SUMO and achieves a success rate of 99.96%without significantly impacting traffic efficiency on the main road.The results demonstrate that DDPG+TDTM+TTC achieved a higher on-ramp merging success rate of 99.96%compared to DDPG+TTC and DDPG.Guofa Li Weiyan Zhou Siyan Lin Shen Li Xingda Qu 2023Automotive Innovation2023,6,3:0
