| 1 | Fine mapping of locus S-b for F_1 pollen sterility in rice (Oryza sativa L.)显示文摘Hybrid sterility is the main barrier in util-izing the heterosis of subspecies in rice. A knowledge of the underlying molecular mechanism of the hybrid sterility will be useful for overcoming the barrier. In this research, the F1 pollen sterility locus, S-b, was mapped between SSR markers PSM8 and PSM202. To fine map the locus, one F2 mapping population of 3910 plants was developed using the near-isogenic lines of the locus. Ninety-seven recombinants be-tween two markers were selected. Moreover, a series of markers, including two SSR markers, two InDel markers and four CAPS markers, were developed on the region. Linkage analysis showed that marker W4 was co-segregated with locus S-b, while makers A8 and A14 were located on the two sides of the locus with a distance of 0.026 and 0.038 cM, respectively. The markers were then integrated with the se-quences of the clones of the region. Results showed that all the polymorphic markers were anchored on the three end-to-end jointed clones AC093089, AC079021 and AC134931. According to the physical information of the markers, locus S-b was finally de-limited to a region of 27 kb between A8 and A14. Seven ORFs were identified on the region based on the annotation results of RiceGAAS system. | 3 | Pathways related to PMA-differentiated THP1 human monocytic leukemia cells revealed by RNA-Seq显示文摘Previous analyses have reported that the human monocytic cell line THP1 can be differentiated into cells with macrophage-like characteristics by phorbol 12-myristate 13-acetate(PMA). However, little is known about the mechanism responsible for regulating this differentiation process. Here, we performed high-throughput RNA-Seq analysis to investigate the genes differently expressed in THP1 cells treated with and without PMA and examined those that may be responsible for the PMA-induced differentiation of monocytes into macrophages. We found 3,000 genes to be differentially expressed after PMA treatment. Gene ontology analysis revealed that genes related to cellular processes and regulation of biological processes were significantly enriched. KEGG analysis also demonstrated that the differentially expressed genes(DEGs) were significantly enriched in the PI3K/AKT signaling pathway and phagosome pathway. Importantly, we reveal an important role of the PI3K/AKT pathway in PMA-induced THP1 cell differentiation. The identified DEGs and pathways may facilitate further study of the detailed molecular mechanisms of THP1 differentiation. In this work, we construct a robust functional interface layer on the modified LiB electrode which considerably improves the electrochemical stability of lithium metal electrode in solid state batteries. It is found that the functional interface layer consisting of polydioxolane, polyiodide ion and Li TFSI effectively restrains the growth of lithium dendrites through the redox shuttle reaction of I-/I3-and maintains a good contact between lithium anode and solid electrolyte during cycles. | 18 | The m6A writers regulated by the IL-6/STAT3 inflammatory pathway facilitate cancer cell stemness in cholangiocarcinoma显示文摘Objective:Investigation of the regulatory mechanisms of cell stemness in cholangiocarcinoma(CCA)is essential for developing effective therapies to improve patient outcomes.The purpose of this study was to investigate the function and regulatory mechanism of m6A modifications in CCA cell stemness.Methods:Interleukin 6(IL-6)treatment was used to induce an inflammatory response,and loss-of-function studies were conducted using mammosphere culture assays.Chromatin immunoprecipitation,polysome profiling,and methylated RNA immunoprecipitation analyses were used to identify signaling pathways.The in vitro findings were verified in a mice model.Results:We first identified that m6A writers were highly expressed in CCAs and further showed that STAT3 directly bound to the gene loci of m6A writers,showing that IL-6/STAT3 signaling regulated expressions of m6A writers.Downregulating m6A writers prevented cell proliferation and migration in vitro and suppressed CCA tumorigenesis in vivo.Notably,the knockdown of m6A writers inhibited CCA cell stemness that was triggered by IL-6 treatment.Mechanistically,IGF2BP2 was bound to CTNNB1 transcripts,significantly enhancing their stability and translation,and conferring stem-like properties.Finally,we confirmed that the combination of m6A writers,IGF2BP2,and CTNNB1 distinguished CCA tissues from normal tissues.Conclusions:Overall,this study showed that the IL-6-triggered inflammatory response facilitated the expressions of m6A writers and cell stemness in an m6A-IGF2BP2-dependent manner.Furthermore,the study showed that m6A modification was a targetable mediator of the response to inflammation factor exposure,was a potential diagnostic biomarker for CCA,and was critical to the progression of CCA. | Hua Ye Tianqi Chen Zhancheng Zeng Bo He Qianqian Yang Qi Pan Yueqin Chen Wentao Wang | 2022 | Cancer Biology & Medicine2022,19,3: | 0 |
