1Effects and safety of Sinomenine in treatment of rheumatoid arthritis contrast to methotrexate: a systematic review and Meta-analysis显示文摘OBJECTIVE: To systematically evaluate the curative clinical efficacy and safety of sinomenine(SIN) in treatment of rheumatoid arthritis(RA) in comparison to methotrexate(MTX).METHODS: We searched the China National Knowledge Infrastructure Database, Chinese Biomedical Literature Database, China Science and Technology Journal Database, Wanfang Database, Pubmed and Cochrane Library electronically up to August 31,2015, without language limitation. Only randomized controlled trials(RCTs) were included. Software Review Manager 5.3 was used for Meta-analysis.RESULTS: A total of 16 eligible studies within 1500RA patients were included. The meta-analysis indicated that on basis of MTX, SIN was more effective in total effective rate(P < 0.000 01). Besides, SIN alone versus MTX also showed advantages in RA therapy(P = 0.04) Taken together, adverse events occurred less frequently in combination of SIN and MTX than MTX alone(P < 0.0001), especially in digestive system(P < 0.000 01),while occurred more in dermato mucosal system with SIN treatment versus MTX(P = 0.02), and were similar for both remedies in nervous system(P = 0.12) and hematological system(P = 0.25).CONCLUTION: Compared to MTX, SIN had better clinical efficacy and relatively fewer adverse events in treatment of RA, especially when it was used together with MTX. 2Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 3The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 4Multigene editing via CRISPR/Cas9 guided by a single-sgRNA seed in Arabidopsis显示文摘We report that a solo single-guide RNA(sg RNA) seed is capable of guiding Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPRàassociated 9(CRISRP/Cas_9) to simultaneously edit multiple genes At RPL_(10)A, At RPL_(10)B and At RPL_(10)C in Arabidopsis. Our results also demonstrate that it is possible to use CRISPR/Cas_9 technology to create At RPL_(10) triple mutants which otherwise cannot be generated by conventional genetic crossing. Compared to other conventional multiplex CRISPR/Cas systems, a single sg RNA seed has the advantage of reducing off-target gene-editing. 5Deficient Rnf43 potentiates hyperactive Kras-mediated pancreatic preneoplasia initiation and malignant transformation显示文摘Background:Largely due to incidental detection,asymptomatic pancreatic cystic le-sions(PCLs)have become prevalent in recent years.Among them,intraductal papillary mucinous neoplasm(IPMN)infrequently advances to pancreatic ductal adenocarcinoma(PDAC).Conservative surveillance versus surgical intervention is a difficult clinical decision for both caregivers and PCL patients.Because RNF43 loss-of-function mutations and KRAS gain-of-function mutations concur in a subset of IPMN and PDAC,their biological significance and therapeutic potential should be elucidated.Methods:Pancreatic Rnf43 knockout and Kras activated mice(Rnf43^(−/−);Kras^(G12D))were generated to evaluate their clinical significance in pancreatic pre-neoplastic initiation and malignant transformation.Results:Loss of Rnf43 potentiated the occurrence and severity of IPMN and PDAC in oncogenic Kras mice.The Wnt/β-catenin signaling pathway was activated in pan-creatic Kras^(G12D)and Rnf43 knockout mice and the PORCN inhibitor LGK974 blocked pancreatic IPMN initiation and progression to PDAC accordingly.Conclusions:Rnf43 is a tumor suppressor in the prevention of pancreatic malignant transformation.This genetically reconstituted autochthonous pancreatic Rnf43^(−/−);Kras^(G12D)preclinical cancer model recapitulates the pathological process from pancreatic cyst to cancer in humans and can be treated with inhibitors of Wnt/β-catenin signaling.Since the presence of RNF43 and KRAS mutations in IPMNs predicts future development of advanced neoplasia from PCLs,patients with these genetic anomalies warrant surveillance,surgery,and/or targeted therapeutics such as Wnt/β-catenin inhibitors.Xian Zhou Zhichao Sun Mengdi Zhang Xiaoyu Qu Shuhui Yang Lianmei Wang Yanling Jing Li Li Weiwei Deng Fangming Liu Jin Di Jie Chen Jian Wu Hongbing Zhang 2022Animal Models and Experimental Medicine2022,5,1:3
6Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 7The Effect of Temperature on Growth and Energy Budget of the Polychaete,Neanthes japonica Izuka显示文摘Growth and energy budget of the polychaete,Neanthes japonica,at various temperatures(17,20,23,26 and 29℃) were investigated in this study. The growth,as indicated by final dry weight and specific growth rate(SGR),increased with increasing temperature,with the maximum level at 26℃,and then decreased significantly at 29℃. A similar trend was observed in feeding rate,food conversion efficiency(FCE) and apparent digestive rate(ADR). However,no significant differences were detected in ADR among all the temperature treatments. In the pattern of energy allocation,faeces energy was only a small component of energy budget and had little influence on the proportion of food energy allocated to growth. The metabolic energy accounted for a large portion of energy intake for each temperature treatment. The nitrogen excretion was appreciable with changing temperature. The two expenditure terms(respiration energy and excretion energy) in energy budget were the major factors influencing the proportion of food energy allocated to growth. These results revealed that temperature affected the growth of N. japonica mainly by influencing feeding rate and FCE. In addition,regression equations describing the relationship between feeding rate,faecal production,SGR,FCE and temperature were obtained. 8Improvement of antibacterial, anti-inflammatory, and osteogenic properties of OGP loaded Co-MOF coating on titanium implants for advanced osseointegration显示文摘The bacterial infection,especially for methicillin-resistant Staphylococcus aureus(MRSA),and the associated severe inflammatory response could extremely limit the crosstalk of RAW264.7 cells and mesenchymal stem cells(MSCs)and lead to the undesirable osseointegration of peri–implants.It is highly demanded to modify the surface of titanium(Ti)implant to improve its anti-bacterial and anti-inflammatory properties and facilitate its disabled osseointegration.Herein,in our study,a multifunctional coating of zeolitic imidazolate frameworks-67 encapsulated osteogenic growth peptide(OGP)(ZO)was fabricated on titanium dioxide nanotubes(TNT)substrates(TNT-ZO)via the electrophoresis deposition(EPD)approach.The TNT-ZO substrates exhibited excellent antibacterial activity indicated by the reactive oxygen species(ROS)generation,outer membrane(OM)and inner membrane(IM)permeabilization change,adenosine triphosphate(ATP)decrease,and intracellular compounds(DNA/RNA)leakage.Importantly,the regulation effects of TNT-ZO coating modified titanium substrates on the RAW264.7-MSCs crosstalk could induce the anti-inflammatory and osteogenic microenvironment via multiple paracrine signaling of Runx2,BMP2,VEGF,and TGF-β1.The promoted effects of coating structure were investigated in vivo,including antibacterial effect,osteogenic differentiation of mesenchymal stem cells,and anti-inflammation of RAW264.7 cells,as well as infected bone regeneration and repair in bone defect transplantation model.The results demonstrated that MRSA was effectively eliminated by the hydrolysis of ZIF-67 nanoparticles on TNT-ZO substrates.Furthermore,the excellent osseointegration of peri–implants was realized simultaneously by modulating the crosstalk of RAW264.7-MSCs.This study could provide a novel approach to designing a multifunctional coating on the Ti implants for infected bone regeneration in orthopedic applications.Bailong Tao Weiwei Yi Xian Qin Junjie Wu Kai Li Ai Guo Jie Hao Lixue Chen 2023Journal of Materials Science & Technology2023,,15:0
9Brpf1 regulates the function of MGE-derived GABAergic interneurons显示文摘Intellectual disability is a broad-spectrum neurodevelopmental disorder characterized by marked impairment of intelligence and adaptive function.Impaired GABA neurotransmission is associated with mental disorders such as epilepsy and mental retardation,and patients with BRPF1 mutations were mentally retarded and often accompanied by epilepsy.Our research is to investigate the role of Brpf1 in the development and function of GABAergic interneurons.We used mouse primary in vitro cultured GABAergic interneurons derived from medial ganglionic eminence(MGE)and utilized AAV-shBrpf1 virus to knockdown Brpf1.The results showed that Brpf1 knockdown led to increased threshold and decreased number of evoked action potentials,and a downward trend of the amplitude and frequency of mIPSC.However,Brpf1 knockdown had lttle effect on the differentiation of GABAergic interneurons as revealed by parvalbumin and somatostatin staining.Moreover,little influence on the morphology was observed as shown by MAP2 and Golgi stain.Finally,it was found that most of the up-regulated genes are involved in protein binding,ubiquitination,and inflammatory response by RNA Seq.Jingli Cao Weiwei Xian Guomin Zhou Linya You 2021解剖学杂志2021,44,S01:0

11Comparison of leaf morphological,anatomical,and photosynthetic responses to drought stress among eight apple rootstocks显示文摘The drought resistance of eight commonly used apple rootstocks under natural drought conditions was examined to provide clues for the selection,promotion,and utilization of drought-resistant apple rootstocks.The ultrastructural differences and physiological and biochemical characteristics of the leaves of eight apple rootstocks under drought stress were observed.The index changes were used to rank drought resistance by the membership function method comprehensively.The results showed that the leaf thickness,palisade tissue thickness,sponge tissue thickness,net photosynthetic rate,and chlorophyll content were significantly higher in‘Fupingqiuzi'than those of other rootstocks at various stress conditions.The leaf water content and water use efficiency of'Qingzhen 1'were significantly higher than those of other rootstocks under different stress conditions.The root vigor of‘B.9'was significantly higher than that of other rootstocks.The results of membership function analysis showed that the drought resistance of different rootstocks was in the order:‘Fupingqiuzi'>‘Qingzhen 1'>‘B.9'>‘MM.111'>‘GM256'>‘MM.106'>‘M.26'>‘M.9-Nic29'.‘Fupingqiuzi'had the strongest drought resistance,and‘Qingzhen 1'and‘B.9'were also relatively droughtresistant.These rootstocks can be used as raw materials for drought-resistant apple rootstock breeding and are propagated and utilized in arid areas.Pengpeng Sun Muhammad Mobeen Tahir Xian Lu Zhimin Liu Xiaoyun Zhang Xiya Zuo Yun Shao Xiangyun Xiao Na An Chao Wang Dong Zhang Weiwei Yang 2022Fruit Research2022,2,1:0

13FAST discovery of an extremely radio-faint millisecond pulsar from the Fermi-LAT unassociated source 3FGL J0318.1+0252显示文摘High sensitivity radio searches of unassociated γ-ray sources have proven to be an effective way of finding new pulsars. Using the Five-hundred-meter Aperture Spherical radio Telescope(FAST) during its commissioning phase, we have carried out a number of targeted deep searches of Fermi Large Area Telescope(LAT) γ-ray sources. On February 27, 2018 we discovered an isolated millisecond pulsar(MSP), PSR J0318+0253, coincident with the unassociated γ-ray source 3 FGL J0318.1+0252. PSR J0318+0253 has a spin period of 5.19 ms, a dispersion measure(DM) of 26 pc cm-3 corresponding to a DM distance of about 1.3 kpc, and a period-averaged flux density of(~11±2) μJy at L-band(1.05-1.45 GHz). Among all high energy MSPs, PSR J0318+0253 is the faintest ever detected in radio bands, by a factor of at least ~4 in terms of L-band fluxes. With the aid of the radio ephemeris, an analysis of 9.6 years of Fermi-LAT data revealed that PSR J0318+0253 also displays strong γ-ray pulsations. Follow-up observations carried out by both Arecibo and FAST suggest a likely spectral turn-over around 350 MHz. This is the first result from the collaboration between FAST and the Fermi-LAT teams as well as the first confirmed new MSP discovery by FAST, raising hopes for the detection of many more MSPs. 16Non-desolvation Zn^(2+)storage mechanism enables MoS_(2)anode with enhanced interfacial charge-transfer kinetics for low temperature zinc-ion batteries显示文摘The emerging rocking-chair aqueous zinc-ion battery(AZIB)configuration provides a promising approach for realizing their practical applications by avoiding the critical drawbacks of Zn metal anodes including unsatisfactory Coulombic efficiency and low Zn utilization.Therefore,exploiting appropriate insertion-type anodes with fast charge-transfer kinetics is of great importance,and many modifications focusing on the improvement of electron transport and bulk Zn^(2+)diffusion have been proposed.However,the interfacial Zn^(2+)transfer determined by the desolvation process actually dominates the kinetics of overall battery reactions,which is mainly overlooked.Herein,the interlayer structure of Mo S_(2)is rationally co-intercalated with water and ethylene glycol(EG)molecules(Mo S2@EG),giving rise to a fast non-desolvation Zn^(2+)storage mechanism,which is verified by the extraordinarily smaller activation energy of interfacial Zn^(2+)transfer(4.66 k J mol^(-1))compared with that of pristine Mo S_(2)(56.78 k J mol^(-1)).Furthermore,the results of theoretical calculations,in-situ Raman and ex-situ characterizations also indicate the enhanced structural integrity of Mo S2@EG during cycling due to the enlarged interlayer spacing and charge screening effect induced by interlaminar EG molecules.Consequently,the Mo S_(2)@EG anode enables excellent cycling stability of both high-energy-density Mo_S2@EG||PVO(polyaniline intercalated V_(2)O_(5))and high-voltage Mo S2@EG||Na_(3)V_(2)(PO_(4))_2O_(2)F(NVPF)full batteries with neglectable capacity decay at-20℃.Zeheng Lv Yu Tan Yuanhong Kang Jin Yang Xian Cheng Weiwei Meng Yufei Zhang Cheng Chao Li Jinbao Zhao Yang Yang 2023Science China Chemistry2023,66,5:0
