| 1 | Multi-omics analyses of 398 foxtail millet accessions reveal genomic regions associated with domestication,metabolite traits,and antiinflammatory effects显示文摘Foxtail millet(Setaria italica),which was domesticatedfromthewild speciesgreenfoxtail(Setaria viridis),isa richsource of phytonutrientsfor humans.To evaluate how breeding changed themetabolome offoxtail millet grains,we generated and analyzed the datasets encompassing the genomes,transcriptomes,metabolomes,and anti-inflammatory indices from 398 foxtail millet accessions.We identified hundreds of common variants that influence numerous secondary metabolites.We observed tremendous differences in natural variations of the metabolites and their underlying genetic architectures between distinct sub-groups of foxtail millet.Furthermore,we found that the selection of the gene alleles associated with yellow grains led to altered profiles of metabolites such as carotenoids and endogenous phytohormones.Using CRiSPR-mediated genome editing wevalidated the function of PHYTOENE SYNTHASE1(PSY1)gene in affecting milletgrain colorand quality.Interestingly,our in vitro cell inflammation assays showed that 83 metabolites in millet grains have anti-inflammatory effects.Taken together,ourmulti-omics study illustrates how the breeding history of foxtail millet has shaped its metabolite profile.The datasets we generated in this study also provide important resources for further understanding how millet grain quality is affected by different metabolites,laying the foundations for future millet genetic research and metabolome-assisted improvement. | Xukai Li Jianhua Gao Jingyi Song Kai Guo Siyu Hou Xingchun Wang Qiang He Yanyan Zhang Yakun Zhang Yulu Yang Jiaoyan Tang Hailang Wang Staffan Persson Mingquan Huang Lishuai Xu Linlin Zhong Dongqin Li Yongming Liu Hua Wu Xianmin Diao Peng Chen Xiaowen Wang Yuanhuai Han | 2022 | Molecular Plant2022,15,8: | 8 |
| 2 | Topological and Shape Optimization of Flexure Hinges for Designing Compliant Mechanisms Using the Level Set Method显示文摘A flexure hinge is a major component in designing compliant mechanisms that o ers unique possibilities in a wide range of application fields in which high positioning accuracy is required. Although various flexure hinges with di erent configurations have been successively proposed, they are often designed based on designers' experiences and inspirations. This study presents a systematic method for topological optimization of flexure hinges by using the level set method. Optimization formulations are developed by considering the functional requirements and geometrical constraints of flexure hinges. The functional requirements are first constructed by maximizing the compliance in the desired direction while minimizing the compliances in the other directions. The weighting sum method is used to construct an objective function in which a self-adjust method is used to set the weighting factors. A constraint on the symmetry of the obtained configuration is developed. Several numerical examples are presented to demonstrate the validity of the proposed method. | 3 | Advances in dielectric elastomer actuation technology显示文摘Dielectric elastomer actuators(DEAs) have attracted much interest over the past decades due to the inherent flexibility, large strain, high efficiency, high energy density, and fast response of the material, which are known as one of the most promising candidates for artificial muscle. In this paper, we first introduce the actuation principle and electromechanical modeling approaches of dielectric elastomers(DEs). Then, the performance of different DEs material and existing compliant electrodes that are widely utilized for DEAs are presented. We also highlight the compatibility of DEs, which is suitable for a variety of actuator designs and applications. | 5 | Foxtail millet supplementation improves glucose metabolism and gut microbiota in rats with high-fat diet/streptozotocin-induced diabetes显示文摘Foxtail millet(FM)whole grain has received special attention in recent years.To confirm the hypoglycemic effects of FM,we investigated the effects of FM supplementation on glucose metabolism and gut microbiota in rats with high-fat diet/streptozotocin(HFD/STZ)-induced diabetes.Specifically,we fully assessed the blood biochemical profiles,pancreatic histopathology,insulin-glucagon immunofluorescence,short-chain fatty acids,and gut microbiota composition of rats with HFD/STZ-induced diabetes before and after FM supplementation.Results showed that both 30% and 48% FM supplementation significantly decreased concentrations of fasting blood glucose,60-min blood glucose,and blood triglycerides(P<0.05);additionally,48% FM supplementation significantly improved blood glucose tolerance and insulin resistance(P<0.05).However,FM supplementation could not effectively repair damage to β-cells over a short period of time.In addition,4 weeks of 48% FM supplementation siginificantly increased the relative abundance of Bifidobacterium and concentration of butyrate,suggesting that the hypoglycemic effects of FM supplementation might be partially mediated by gut microbiota.Collectively,we found a dose-dependent relationship between FM supplementation and improvement of blood glucose metabolism,but did not find a synergistic effect between FM supplementation and metformin(Met)treatment.Our findings provide further support that consuming more whole-grain FM might be beneficial to individuals suffering from type 2 diabetes. | Xin Ren Linxuan Wang Zenglong Chen Min Zhang Dianzhi Hou Yong Xue Xianmin Diao Ruihai Liu Qun Shen | 2022 | Food Science and Human Wellness2022,11,1: | 3 |
| 6 | Tuning charge density of chimeric antigen receptor optimizes tonic signaling and CAR-T cell fitness显示文摘Tonic signaling of chimeric antigen receptor(CAR),i.e.,the spontaneous CAR activation in the absence of tumor antigen stimulation,is considered to be a pivotal event controlling CAR-T efficacy.However,the molecular mechanism underlying the spontaneous CAR signals remains elusive.Here,we unveil that positively charged patches(PCPs)on the surface of the CAR antigenbinding domain mediate CAR clustering and result in CAR tonic signaling.For CARs with high tonic signaling(e.g.,GD2.CAR and CSPG4.CAR),reducing PCPs on CARs or boosting ionic strength in the culture medium during ex vivo CAR-T cell expansion minimizes spontaneous CAR activation and alleviates CAR-T cell exhaustion.In contrast,introducing PCPs into the CAR with weak tonic signaling,such as CD19.CAR,results in improved in vivo persistence and superior antitumor function.These results demonstrate that CAR tonic signaling is induced and maintained by PCP-mediated CAR clustering.Notably,the mutations we generated to alter the PCPs maintain the antigen-binding affinity and specificity of the CAR.Therefore,our findings suggest that the rational tuning of PCPs to optimize tonic signaling and in vivo fitness of CAR-T cells is a promising design strategy for the nextgeneration CAR. | Jian Chen Shizhen Qiu Wentao Li Kun Wang Yu Zhang Han Yang Baichuan Liu Guangfei Li Li Li Min Chen Junjie Lan Jiahua Niu Peijie He Lei Cheng Gaofeng Fan Xin Liu Xianmin Song Chenqi Xu Haitao Wu Haopeng Wang | 2023 | Cell Research2023,33,5: | 2 |
| 7 | Integrated transcriptome analysis of human iPS cells derived from a fragile X syndrome patient during neuronal differentiation显示文摘Fragile X syndrome(FXS) patients carry the expansion of over 200 CGG repeats at the promoter of fragile X mental retardation 1(FMR1), leading to decreased or absent expression of its encoded fragile X mental retardation protein(FMRP). However, the global transcriptional alteration by FMRP deficiency has not been well characterized at single nucleotide resolution, i.e., RNA-seq. Here,we performed in-vitro neuronal differentiation of human induced pluripotent stem(iPS) cells that were derived from fibroblasts of a FXS patient(FXS-iPSC). We then performed RNA-seq and examined the transcriptional misregulation at each intermediate stage during in-vitro differentiation of FXS-iPSC into neurons. After thoroughly analyzing the transcriptomic data and integrating them with those from other platforms, we found up-regulation of many genes encoding TFs for neuronal differentiation(WNT1, BMP4,POU3F4, TFAP2 C, and PAX3), down-regulation of potassium channels(KCNA1, KCNC3, KCNG2, KCNIP4, KCNJ3, KCNK9,and KCNT1) and altered temporal regulation of SHANK1 and NNAT in FXS-iPSC derived neurons, indicating impaired neuronal differentiation and function in FXS patients.
