| 1 | Puerarin reduces apoptosis in rat hippocampal neurons cultured in high glucose medium by modulating the p38 mitogen activated protein kinase and c-Jun N-terminal kinase signaling pathways显示文摘OBJECTIVE: To investigate the neuroprotective effect of puerarin on rat hippocampal neurons cultured in high glucose medium, and to examine the role of the p38 mitogen activated protein kinase(p38 MAPK) and c-Jun N-terminal kinase(JNK) signaling pathways in this effect.METHODS: Primary cultures of hippocampal neurons were prepared from newborn Sprague Dawley rats. Neuron-specific enolase immunocytochemistry was used to identify neurons. The neurons were cultured with normal medium(control group) or with high-glucose medium(high-glucose group),and puerarin(puerarin group), a p38 MAPK inhibitor(SB239063; p38 MAPK inhibitor group) or a JNK inhibitor(SP600125; JNK inhibitor group) were added. After 72 h of treatment, terminal deoxynucleotidyl transferase-mediated d UTP nick end labeling assay was performed to detect apoptosis, and western blotting was used to assess protein levels of p-p38, p38, p-JNK and JNK.RESULTS: In the high-glucose group, the neuronal apoptosis rate and the p-p38/p38 and p-JNK/JNK ratios were higher than in the control group. The p38 MAPK and JNK inhibitors prevented this increase in the apoptosis rate. The apoptosis rates in the puerarin group, the p38 MAPK inhibitor group and the JNK inhibitor group were significantly decreased compared with the high-glucose group.Moreover, protein levels of p-p38 and p-JNK were significantly reduced, and the p-p38/p38 and p-JNK/JNK ratios were decreased in the puerarin group compared with the high-glucose group. | 3 | Methionine deficiency and its hydroxy analogue influence chicken intestinal 3-dimensional organoid development显示文摘Methionine and its hydroxy analogue(MHA)have been shown to benefit mouse intestinal regeneration.The intestinal organoid is a good model that directly reflects the impact of certain nutrients or chemicals on intestinal development.Here,we aimed to establish a chicken intestinal organoid culture method first and then use the model to explore the influence of methionine deficiency and MHA on intestinal organoid development.The results showed that 125-mm cell strainer exhibited the highest efficiency for chicken embryo crypt harvesting.We found that transforming growth factor-b inhibitor(A8301)supplementation promoted enterocyte differentiation at the expense of the proliferation of intestinal stem cells(ISC).The mitogen-activated protein kinase p38 inhibitor(SB202190)promoted intestinal organoid formation and enterocyte differentiation but suppressed the differentiation of enteroendocrine cells,goblet cells and Paneth cells.However,the suppression of enteroendocrine cell and Paneth cell differentiation by SB202190 was alleviated at the presence of A8301.The glycogen synthase kinase 3 inhibitor(CHIR99021),valproic acid(VPA)alone and their combination promoted chicken intestinal organoid formation and enterocyte differentiation at the expense of the expression of Paneth cells and goblet cells.Chicken serum significantly improved organoid formation,especially in the presence of A8301,SB202190,CHIR99021,and VPA,but inhibited the differentiation of Paneth cells and enteroendocrine cells.Chicken serum at a concentration of 0.25%meets the requirement of chicken intestinal organoid development,and the beneficial effect of chicken serum on chicken intestinal organoid culture could not be replaced by fetal bovine serum and insulin-like growth factor-1.Moreover,commercial mouse organoid culture medium supplemented with A8301,SB202190,CHIR99021,VPA,and chicken serum promotes chicken organoid budding.Based on the chicken intestinal organoid model,we found that methionine deficiency mimicked by cycloleucine suppressed organoid formation and organoid size,and this effect was reinforced with increased cycloleucine concentrations.Methionine hydroxy analogue promoted regeneration of ISC but decreased cell differentiation compared with the results obtained with L-methionine.In conclusion,our results provide a potentially excellent guideline for chicken intestinal organoid culture and insights into methionine function in crypt development. | Youli Wang Qihang Hou Yuqin Wu Yanwei Xu Yan Liu Jing Chen Lingling Xu Yuming Guo Shuai Gao Jianmin Yuan | 2022 | Animal Nutrition2022,,1: | 4 |
| 4 | Distribution of elements extracted from symptom patterns and characteristics of polysomnograph of common symptom patterns of insomnia with Traditional Chinese Medicine显示文摘OBJECTIVE: To analyze the distribution and combined regulation of elements of symptom patterns in the diagnosis of insomnia with Traditional Chinese Medicine(TCM).METHODS: The samples were collected from the patients, diagnosed with insomnia, of Henan Province Hospital of TCM between June 2011 and September 2013. The symptom patterns in insomnia were extracted. Next, symptom differentiation,characteristics of polysomnography(PSG), distribution and combined regulation of these symptom patterns were conducted by tests.RESULTS: In total, 286 eligible patients were recruited. The main locations of the disease symptom elements were the brain and heart, and the main characteristics of the disease symptom elements were phlegm-heat, Yin-deficiency and Qi-stagnation. The elements from two or three symptom patterns were commonly manifested in patients with insomnia, especially from three symptom patterns. We also found that all TCM symptom patterns had an effect on polysomnographic indicators in PSG tests.CONCLUSION: The elements of symptom patterns in insomnia were identified as mainly fire-heat and phlegm-heat. The most common patterns of excess were pathogenic fire derived from stagnation of liver-Qi, and mental disturbance due to phlegm-heat,while the most common patterns of deficiency in both the heart and the spleen. | 5 | Transforming Co3O4 nanosheets into porous N-doped CoxOy nanosheets with oxygen vacancies for the oxygen evolution reaction显示文摘Cobalt oxides have been widely investigated as promising replacements for noble metal-based catalysts for oxygen evolution reaction(OER). Herein, we, for the first time, have obtained porous CoxOy nanosheets with N-doping and oxygen vacancies by etching Co3O4 nanosheets with NH3 plasma. Comparing with the pristine Co3O4 nanosheets(1.79 V), the porous CoxOy nanosheets with N-doping and oxygen vacancies have a much lower potential of 1.51 V versus RHE to reach the current density of 10 mA cm-2. In this work, we report a high-performanee dual-channel phototransistor based on one-dimensional (1D)/two-dimensional (2D) trigonal selenium (t-Se)/ReS2 heterostructures grown by chemical vapor deposition. The injection and separati on efficie ncy of photoge nerated electro n-hole pairs can be greatly improved due to the high-quality in terfacial con tact betwee n t-Se nano belts and ReS2 films. Compared with bare ReS2 film devices, the dual-cha nnel phototra nsistor based on t-Se/ReS2 heterostructure exhibits considerable enhancement with the responsivity (R) and detectivity (D^*) up to 98 A·W^-1 and 6 x 10^10 Jones at 400 nm illumination with an in tensity of 1.7 mW·cm^-2, respectively. Besides, the respo nse time can also be reduced by three times of magnitude to less than 50 ms due to the type-11 band alignment at the in terface. | 9 | A review on COVID-19 transmission,epidemiological features,prevention and vaccination显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has caused hundreds of millions of infections and millions of deaths over past two years.Currently,many countries have still not been able to take the pandemic under control.In this review,we systematically summarized what we have done to mitigate the COVID-19 pandemic,from the perspectives of virus transmission,public health control measures,to the development and vaccination of COVID-19 vaccines.As a virus most likely coming from bats,the SARS-CoV-2 may transmit among people via airborne,faecal-oral,vertical or foodborne routes.Ourmeta-analysis suggested that the R0 of COVID-19 was 2.9(95%CI:2.7–3.1),and the estimates in Africa and Europe could be higher.The median Rt could decrease by 23–96%following the nonpharmacological interventions,including lockdown,isolation,social distance,and face mask,etc.Comprehensive intervention and lockdown were the most effective measures to control the pandemic.According to the pooled R0 in our meta-analysis,there should be at least 93.3%(95%CI:89.9–96.2%)people being vaccinated around the world.Limited amount of vaccines and the inequity issues in vaccine allocation call for more international cooperation to achieve the antiepidemic goals and vaccination fairness. | Yuqin Zhang Gonghua Wu Shirui Chen Xu Ju Wumitijiang Yimaer Wangjian Zhang Shao Lin Yuantao Hao Jing Gu Jinghua Li | 2022 | Medical Review2022,2,1: | 2 |
| 10 | Sulfonic groups functionalized Zr-metal organic framework for highly catalytic transfer hydrogenation of furfural to furfuryl alcohol显示文摘The highly selective catalytic transfer hydrogenation(CTH)of furfural(FF)to furfuryl alcohol(FOL)is a significant route of biomass valorization.Herein,a series microporous Zr-metal organic framework(ZrMOF)functionalized by sulfonic groups are prepared.Based on the comprehensive structural characterizations by means of X-ray diffraction(XRD),scanning electron microscopy(SEM),transmission electron microscopy(TEM),N2 physisorption,Thermogravimetric(TG)and Fourier transformed infrared spectroscopy(FTIR),we find that sulfonic acid(–SO_(3)H)functional groups are tethered on the UIO-66 without affecting the structure of the framework.Systematic characterizations(NH_(3)-TPD,CO_(2)-TPD,and in-situ FTIR)demonstrate that modifying of sulfonic groups on UIO-66 results in the formation of stronger Lewis acidic-basic and Brnsted acidis sites.The cooperative role of the versatile Lewis acidic-basic and Brnsted acidic sites in 60%mol fraction of sulfonic acid-containing UIO-66(UIO-S_(0.6))retain high surface area and exhibit excellent catalytic performance of 94.7%FOL yield and 16.9 h^(-1).turnover number(TOF)under mild conditions.Kinetic experiments reveal that the activation energy of the CTH of furfural(FF)over UIO-S_(0.6) catalyst is as low as 50.8 k J mol^(-1).Besides,the hydrogen transfer mechanism is investigated through isotope labeling experiments,exhibiting that theβ-H in isopropanol is transferred to the a-C of FF by forming six-membered intermediates on the Lewis acidic-basic and Brnsted acidic sites of the UIO-S_(0.6),which is the rate-determining step in the formation of FOL. | Jingcheng Wu Dong Liang Xiangbo Song Tingsen Liu Tianyi Xu Shuangyin Wang Yuqin Zou | 2022 | Journal of Energy Chemistry2022,31,8: | 2 |

| 19 | Effect and mechanism of CyPA in radiosensitization of lung adenocarcinoma using CRISPR/Cas9 technology显示文摘Objective:To explore the role of cyclophilin A(CyPA)in sensitization of lung adenocarcinoma to radiotherapy using CRISPR/Cas9 technology.Methods:A CyPA knockout human lung adenocarcinoma cell line H1975 was established by CRISPR/Cas9 technology.Groups included a control group(wildtype),CyPA knockout group,traditional Chinese medicine(TCM)extract with Fuzhengzengxiao decoction group,and TCM extract with Fuzhengzengxiao decoction t CyPA knockout group.Each group was exposed to radiation at doses of 0,2,4,6,and 8 Gy.After 24 h,MTT assays were used to determine the survival rate of lung cancer cells and calculate radiosensitivity.The qPCR was used to measure mRNA expression of DDIT3,CDKN1A,and CDC25A associated with DNA damage repair.Results:Without irradiation,Fuzhengzengxiao decoction reduced the survival rate of lung adenocarcinoma cells(P<.0001).After irradiation,TCM extract with Fuzhengzengxiao decoction,CyPA knockout,and TCM extract with Fuzhengzengxiao decoction t CyPA knockout groups had reduced survival rates(P<.0001)and radiosensitivity was increased significantly.Expression of DDIT3,CDKN1A,and CDC25A was upregulated after knockout of CyPA(P<.0001).Expression of DDIT3 and CDC25A was increased after irradiation in wildtype cells treated with TCM extract with Fuzhengzengxiao decoction(DDIT3,P<.0001;CDC25A,P紏.0059).The TCM extract with Fuzhengzengxiao decoction t CyPA knockout group also had increased expression of DDIT3 and CDC25A after irradiation(P<.0001).Conclusion:Fuzhengzengxiao decoction significantly decreases the survival rate of lung cancer cells,and its mechanism may be related to radiosensitization by decreasing expression of CyPA and inducing G1/S cell cycle arrest. | Jinkun Ma Lin Xu Jinchang Huang Qiaoli Zhang Yuqin Qiu Xuewei Qi Lina Wang Yuchan Cao Zeyu Liu Shiyun Liu | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,1: | 1 |
