2Construction of Streptomyces lydicus A01 transformant with the chit33 gene from Trichoderma harzianum CECT2413 and its biocontrol effect on Fusaria显示文摘Streptomyces lydicus A01 resists many plant pathogens (including Fusarium spp.) by producing the antifungal agent natamycin, which binds to the ergosterol of fungal cell membranes and inhibits the growth of pathogens. Trichoderma harzianum CECT2413 is a widely-distributed soil fungus that antagonizes several plant fungal pathogens (including Fusarium spp.) by producing chi-tinase and degrading chitin, a major component of the fungal cell wall. This study attempted to enhance the biocontrol effect of S. lydicus A01 on Fusarium spp. by transforming the chitinase gene of Trichoderma. Chitinase and natamycin could act synergisti-cally on both the cell walls and cell membranes of pathogens. The 33-kD chitinase-encoding gene (chit33) was cloned and conju-gal-transformed from T. harzianum CECT2413 to S. lydicus A01, and then confirmed via polymerase chain reaction (PCR) assays. Subsequent analyses using the 3,5-dinitrosalicylic acid (DNS) method and ultraviolet spectrophotometry showed that compared with its wild type strain (WT), the S. lydicus A01 conjugal transformant (CT) with chit33 gene exhibited substantially higher chi-tinase activity and natamycin production. The resistance of S. lydicus A01-chit33 CT and WT to four Fusaria in crops and vegetables was tested via the cup-plate method. Compared with the WT, the conjugal transformant of S. lydicus A01 with chit33 gene from T. harzianum CECT2413 showed greatly increased biocontrol effect on fusarium disease. 5A Molecular Foaming and Activation Strategy to Porous N-Doped Carbon Foams for Supercapacitors and CO2 Capture显示文摘Hierarchically porous carbon materials are promising for energy storage,separation and catalysis.It is desirable but fairly challenging to simultaneously create ultrahigh surface areas,large pore volumes and high N contents in these materials.Herein,we demonstrate a facile acid-base enabled in situ molecular foaming and activation strategy for the synthesis of hierarchically macro-/meso-/microporous N-doped carbon foams(HPNCFs).The key design for the synthesis is the selection of histidine(His)and potassium bicarbonate(PBC)to allow the formation of 3D foam structures by in situ foaming,the PBC/His acid-base reaction to enable a molecular mixing and subsequent a uniform chemical activation,and the stable imidazole moiety in His to sustain high N contents after carbonization.The formation mechanism of the HPNCFs is studied in detail.The prepared HPNCFs possess 3D macroporous frameworks with thin well-graphitized carbon walls,ultrahigh surface areas(up to 3200 m^2 g^-1),large pore volumes(up to 2.0 cm^3 g^-1),high micropore volumes(up to 0.67 cm^3 g^-1),narrowly distributed micropores and mesopores and high N contents(up to 14.6 wt%)with pyrrolic N as the predominant N site.The HPNCFs are promising for supercapacitors with high specific capacitances(185-240 F g^-1),good rate capability and excellent stability.They are also excellent for CO2 capture with a high adsorption capacity(~4.13 mmol g^-1),a large isosteric heat of adsorption(26.5 kJ mol^-1)and an excellent CO2/N2 selectivity(~24).Mengyuan Zhou Yaqian Lin Huayao Xia Xiangru Wei Yan Yao Xiaoning Wang Zhangxiong Wu 2020Nano-Micro Letters2020,12,5:1

10Prey decline leads to diet shift in the largest population of Indo-Pacific humpback dolphins?显示文摘The Pearl River Delta(PRD)region on the southeast coast of China has long been known as a highly productive fishing ground.Since the late 1980s,fishing pressure in the PRD has been intense,which warrants concerns of potential fishery-related impacts on the food resources and foraging ecology of apex marine predators in this region,such as the Indo-Pacific humpback dolphin(Sousa chinensis).In this study,we examined 54 stomachs with food remains,collected from beached carcasses of humpback dolphins recovered during fifteen years between 2003 and 2017.The 6043 identified prey items represent 62 teleost taxa,primarily small estuarine fish,but also larger reef fish.The dolphins appear to be opportunistic foragers,hunting across the water-column,with preference for shoaling and meaty fishes(e.g.Collichthys lucidus IRI%=38.6%,Johnius belangerii IRI%=23.1%,Mugil cephalus IRI%=14.0%).Our findings suggest a dietary shift in recent years,from primarily demersal(as previously reported)to greater intake of neritic and pelagic fish.Dolphin foraging group size has decreased in recent years,which corresponds with declining size and numbers of prey items retrieved from dolphin stomachs.We suggest that these are indicators of declining food resources.Faced with a shortage of preferred prey,humpback dolphins may have broadened their dietary spectrum to maintain their daily energy intake,while their foraging group size decreased in response to the altered tradeoff between the costs and benefits of group foraging.Wenzhi LIN Leszek KARCZMARSKI Ruilian ZHOU Yaqian MO Lang GUO Sam King Fung YIU Xi NING Tak-Cheung WAI Yuping WU 2021Integrative Zoology2021,16,4:0

12Driving force of biomolecular liquid–liquid phase separation probed by nuclear magnetic resonance spectroscopy显示文摘The assembly of biomolecular condensates is driven by liquid–liquid phase separation. To understand the structure and functions of these condensates, it is essential to characterize the underlying driving forces, e.g., protein–protein and protein–RNA interactions. As both structured and low-complexity domains are involved in the phase separation process, NMR is probably the only technique that can be used to depict the binding topology and interaction modes for the structured and nonstructured domains simultaneously. Atomic-resolution analysis for the intramolecular and intermolecular interactions between any pair of components sheds light on the mechanism for phase separation and biomolecular condensate assembly and disassembly. 13Ice storm damage to oak forests in subtropical China显示文摘Ice storms,as important sources of frequent and injurious disturbances,drive forest dynamics in the Northern Hemisphere.However,stand-level differential vulnerability to ice storms and the associated factors that predispose forest stands remain unclear.This is particularly concerning in the subtropics where the frequency of ice storms is predicted to increase with global warming.Here we assessed how the impact on three forest stands(early and late secondary-growth forests,and old-growth forests)differed after an extreme ice storm during 20–21 March 2022,and identified the abiotic and biotic factors that determine the damage intensity in the Shennongjia World Natural Heritage Site,a biodiversity conservation hotspot in central China.We found a stand-specific‘middomain effect’where the late secondary-growth forest sustained the most severe damage,the early secondarygrowth forest sustained the least,and the old-growth forest suffered an intermediate amount.‘Crown broken’was the most severe damage type across all three forest stands,although the proportion of‘branch broken’was also high in the old-growth forest.Topography played a significant role in determining the vulnerability of the early secondary-growth forest to severe ice storms whereas the forest structure and composition were important factors in explaining the damage rates in the old-growth forest,although they differed among the damage categories.In contrast,topography,forest structure and composition generally explain the intensity of damage in the late secondary-growth forests.Our results highlight that,in subtropical forests,the intensity of damage caused by severe ice storms and related determining factors are stand-level dependent.We also suggest exploring potential management strategies(e.g.,slow-growing hardwood species that can resist storms should be the main species for reforestation in early secondary-growth forests)to mitigate the risk of future severe ice storms,as well as other wind-related climatic extremes.Jinyu Guo Jifa Cui Nan Wu Yaqian Zhang Jie Wang Hanyu Xiang Baoshuang Hu Youbing Zhou 2023Forest Ecosystems2023,10,3:0

15Gambogic acid-encapsulated polymeric micelles improved therapeutic effects on pancreatic cancer显示文摘Gambogic acid(GA) is a natural product with potent anticancer activity in vitro. However, poor water solubility and systematic toxicity limit the further clinical application of GA. Micellization of hydrophobic molecule could effectively ameliorate aqueous dispersity of GA and induce better blood retention and tumor accumulation, hence lead to improved stability and therapeutic effect of GA. In this study, monomethyl poly(ethylene glycol)-poly(ε-caprolactone)-poly(trimethylene carbonate)(MPEG-P(CL-ran-TMC)) was used to encapsulate GA by a single-step solid dispersion method to form a GA encapsulated MPEG-P(CLran-TMC) micelles(GA micelles). GA micelles were characterized with a small particle size(44 ±1 nm),high drug loading content(26.28% ± 0.12%) and high-efficiency encapsulation(87.59% ± 0.41 %). Compared with free GA, GA micelles showed better dispersion in water, prolonged release behavior in vitro, and enhanced tumor cellular uptake. GA micelles could also effectively induce apoptosis in AsPC-1 cells.Compared with free GA, GA micelles exhibited superior antitumor efficacy and better apoptosis induced effect in a subcutaneous xenograft mouse model of AsPC-1 cells. 16Association between soft drink consumption types and risk of lung cancer and all-cancer: A prospective study of PLCO data显示文摘Diet/sugar-free soft drinks are considered to be healthier than regular soft drinks.However,few studies have examined the relationship between the types of soft drinks(regular and diet/sugar-free)and lung cancer(LC)/all-cancer(AC)risk.In this study,we comprehensively assessed the influence of the type of soft drink consumption on LC/AC risk based on the Prostate,Lung,Colorectal,and Ovarian(PLCO)Cancer Screening Trial.Multivariable Cox proportional hazards and competing risks Fine-Gray regression models adjusted for relevant confounders were used to estimate hazard ratios(HRs)and subdistribution HRs for different types of soft drink consumption.In the PLCO population,female subgroup,and the ever/current smoker subgroup,consumption of both regular and diet soft drinks was associated with a significantly reduced risk of LC compared with no soft drinks at all.For the non-lung cancer(NLC)risk,consumption of only diet soft drinks had a significant positive association for the total population and female subgroup.Based on our findings,it was suggested that partial replacement of regular soft drinks with diet soft drinks might be beneficial to LC prevention,especially for females and ever/current smokers.Additionally,completely replacing regular soft drinks with diet soft drinks might be detrimental to NLC prevention,especially for females.Dongfang You Hongyang Xu Xin Chen Jiawei Zhou Yaqian Wu Yingdan Tang Zhongtian Wang Yang Zhao Fang Shao 2022The Journal of Biomedical Research2022,36,6:0
17PA1426 regulates Pseudomonas aeruginosa quorum sensing and virulence:an in vitro study显示文摘Objective:Pseudomonas aeruginosa(P.aeruginosa)contains a hierarchy of quorum sensing(QS)network,consisting of the las,rhl and pqs systems,which play a key role in coordinating the expression of virulence factors.PA2146 was found to be associated with P.aeruginosa pathogenicity in macrophage and host Immune response.The aim of this study was to investigate the effects of PA2146 on the virulence of P.aeruginosa and explore its mechanism.Methods:PA2146 gene knockout strain and complement strain of P.aeruginosa PAO1 were constructed.The biomass of biofilm was detected by crystal violet staining;the virulence factors were measured,including pyocyanin,rhamnolipid,LasA elastase,LasB elastase and hemolytic activity;RNA-seq and label-free relative quantitative proteomics analyses were carried out to test the influence of PA2146 on transcriptomics and proteomics.This study was approved by the Institutional Review Board of the Third Xiangya Hospital,Central South University,China(approval No.2019-S021).Results:PA2146-deficient strains showed reduced biofilm formation and increased pyocyanin,rhamnolipid,LasA elastase,LasB elastase,and hemolytic activity,as well as increased motility,compared with the wild-type strain.RNA-seq and label-free relative quantitative proteomics analyses revealed that PA2146 repressed the transcription of several genes that are integral to the pqs system and to pyocyanin biosynthesis,and increased the expression of MexEF-OprN efflux pump components at the gene and protein level.Conclusion:PA2146 gene was found to inhibit the pqs system.PA2146 may affect quorum sensing by directly inhibiting the pqs system or by enhancing the expression of MexEF-OprN efflux pump components,thereby promoting efflux of 2-heptyl-4(1H)-quinolone,a Pseudomonas quinolone signal precursor,and thus affecting P.aeruginosa virulence.Fang Tan Pengfei She Linying Zhou Shijia Li Xianghai Zeng Lanlan Xu Yaqian Liu Zubair Hussain Yong Wu 2021Journal of Bio-X Research2021,4,1:0
