1Long-term follow-up of the effects of fecal microbiota transplantation in combination with soluble dietary fiber as a therapeutic regimen in slow transit constipation显示文摘As some studies have reported that strategies targeting the gut microbiota such as fecal microbiota transplantation(FMT) with or without other microecological therapy might have efficacy in treating slow transit constipation(STC), we conducted a singlecenter, open-label trial to study the long-term effect of FMT combined with soluble dietary fiber(pectin) on STC. Thirty-one adult patients with STC were enrolled into the trial. Patients received 6-day FMT procedures repeatedly for the first 3 months and soluble dietary fiber(pectin) daily during the follow-up. The rate of clinical remission and improvement, stool consistency, the Wexner constipation scale, and assessment of constipation-related symptoms were evaluated at week 4 and 1 year later. The clinical remission and improvement rates at week 4 were 69.0%(20/29) and 75.9%(22/29), respectively. At the end of the study,48.3%(14/29) of patients continued to have at least three complete spontaneous bowel movements per week and 58.6%(17/29)of patients showed clinical improvements. Stool consistency, the Wexner constipation scale, and constipation symptoms improved both at short-term and long-term follow-up. 3DDIT4 promotes gastric cancer proliferation and tumorigenesis through the p53 and MAPK pathways显示文摘Background:Gastric cancer(GC)is one of the most common malignancies worldwide,particularly in China.DNA damage-inducible transcript 4(DDIT4)is a mammalian target of rapamycin inhibitor and is induced by various cellular stresses;however,its critical role in GC remains poorly understood.The present study aimed to investigate the poten-tial relationship and the underlying mechanism between DDIT4 and GC development.Methods:We used western blotting,real-time polymerase chain reaction,and immunohistochemical or immunoflu-orescence to determine DDIT4 expression in GC cells and tissues.High-content screening,cell counting kit-8 assays,colony formation,and in vivo tumorigenesis assays were performed to evaluate cell proliferation.Flow cytometry was used to investigate cell apoptosis and cell cycle distribution.Results:DDIT4 was upregulated in GC cells and tissue.Furthermore,downregulating DDIT4 in GC cells inhibited proliferation both in vitro and in vivo and increased 5-fluorouracil-induced apoptosis and cell cycle arrest.In contrast,ectopic expression of DDIT4 in normal gastric epithelial cells promoted proliferation and attenuated chemosensitivity.Further analysis indicated that the mitogen-activated protein kinase and p53 signaling pathways were involved in the suppression of proliferation,and increased chemosensitivity upon DDIT4 downregulation.Conclusion:DDIT4 promotes GC proliferation and tumorigenesis,providing new insights into the role of DDIT4 in the tumorigenesis of human GC.Feng Du Lina Sun Yi Chu Tingyu Li Chao Lei Xin Wang Mingzuo Jiang Yali Min Yuanyuan Lu Xiaodi Zhao Yongzhan Nie Daiming Fan 2018Cancer Communications2018,38,1:10

6Coastal plain evolution in southern Hainan Island,China显示文摘The coast of southern Hainan Island is characterized by wide sandy embayments, which consist of ( i) drowned valleys bounded by steep bedrock hills and only locally receiving sediments, and embayments of various dimensions covered either by (ii) alluvial-deltaic deposits or by (iii) sands of coastal beach ridges/barriers and associated elongated lagoons. During the late Tertiary-Pleistocene the area has experienced isostatic and eustatic movements associated with neotectonics and climatic changes. Such history isrecorded in terraces at various altitudes (SO, 40, 20 m asl) and sequences of coastal sand ridges/baymouth bars. The Holocene variations in sea level and climate are recorded in the dated coastal ridges, coral reef and beachrock. Conditions suitable for reef development started about 8000 a BP. 8Association of systemic inflammation and body mass index with survival in patients with resectable gastric or gastroesophageal junction adenocarcinomas显示文摘Objective:The systemic inflammation index and body mass index(BMI)are easily accessible markers that can predict mortality.However,the prognostic value of the combined use of these two markers remains unclear.The goal of this study was therefore to evaluate the association of these markers with outcomes based on a large cohort of patients with gastric cancer.Methods:A total of 2,542 consecutive patients undergoing radical surgery for gastric or gastroesophageal junction adenocarcinoma between 2009 and 2014 were included.Systemic inflammation was quantified by the preoperative neutrophil-to-lymphocyte ratio(NLR).High systemic inflammation was defined as NLR≥3,and underweight was defined as BMI<18.5 kg/m2.Results:Among 2,542 patients,NLR≥3 and underweight were common[627(25%)and 349(14%),respectively].In the entire cohort,NLR≥3 or underweight independently predicted overall survival(OS)[hazard ratio(HR):1.236,95%confidence interval(95%CI):1.069–1.430;and HR:1.600,95%CI:1.350–1.897,respectively]and recurrence-free survival(RFS)(HR:1.230,95%CI:1.054–1.434;and HR:1.658,95%CI:1.389–1.979,respectively).Patients with both NLR≥3 and underweight(vs.neither)had much worse OS(HR:2.445,95%CI:1.853–3.225)and RFS(HR:2.405,95%CI:1.802–3.209).Furthermore,we observed similar results in subgroup analyses according to pathological stage,age,and postoperative chemotherapy.Conclusions:Our results showed that preoperative elevated NLR and decreased BMI had a significant negative effect on survival.Underweight combined with severe inflammation could enhance prognostication.Taking active therapeutic measures to reduce inflammation and increase nutrition may help improve outcomes.Xianchun Gao Yanan Pan Weili Han Caie Hu Chenchen Wang Ling Chen Yong Guo Yupeng Shi Yan Pan Huahong Xie Liping Yao Jianjun Yang Jianyong Zheng Xiaohua Li Xiaonan Liu Liu Hong Jipeng Li Mengbin Li Gang Ji Zengshan Li Jielai Xia Qingchuan Zhao Daiming Fan Kaichun Wu Yongzhan Nie 2021Cancer Biology & Medicine2021,18,1:2

11Silencing of ER-resident oxidoreductase PDIA3 inhibits malignant biological behaviors of multidrug-resistant gastric cancer显示文摘Glycosylation is a common posttranslational modification of proteins,which plays a role in the malignant transformation,growth,progression,chemoresistance,and immune response of tumors.Disulfide isomerase family A3(PDIA3)specifically acts on newly synthesized glycoproteins to promote the correct folding of sugar chains.Studies have shown that PDIA3 participates in multidrug-resistant gastric cancer(MDR-GC).In this study,we performed western blot analysis and immunohistochemistry to identify PDIA3 expression.Cell proliferation was assessed by CCK-8 assay.Transwell assays were used to detect the migration and invasion abilities of cells.Immunoprecipitation coupled to mass spectrometry(IP-MS)analysis was employed to identify PDIA3-interacting proteins and the associated pathways in MDR-GC cells.Glycoprotein interactions and translocation were detected by immunofluorescence assay.The results showed that PDIA3 knockdown significantly inhibited the proliferation,invasion,and migration abilities of MDR-GC cells.Kyoto Encyclopedia of Genes and Genomes analysis of the IP-MS results showed that PDIA3 was closely associated with focal adhesion pathways in MDR-GC cells.Additionally,important components of focal adhesion pathways,including fibronectin-1(FN1)and integrinα5(ITGA5),were identified as pivotal PDIA3-binding glycoproteins.Knockdown of PDIA3 altered the cellular locations of FN1 and ITGA5,leading to abnormal accumulation.In conclusion,our results suggest that knockdown of PDIA3 inhibited the malignant behaviors of MDR-GC cells and influenced the translocation of FN1 and ITGA5.Danyang Song Meng Guo Kaichu Wu Jianyu Hao Yongzhan Nie Daiming Fan 2021Acta Biochimica et Biophysica Sinica2021,53,9:2

19Effect of black and white sesame on lowering blood lipids of rats with hyperlipidemia induced by high-fat diet显示文摘Sesamum indicum has various biological effects,and this study aimed to investigate its hypolipidemic effect.A hyperlipidemia rat model was established by feeding rats with high-fat diets in this study,then the hyperlipidemia rats were fed with different doses of black and white sesame seeds and black and white sesame kernels.The serum and liver biochemical indicators of the rats were determined,and liver pathology analysis was conducted to evaluate the hypolipidemic effect of sesame.The results showed that the blood lipid of SD(Sprague-Dawley)rats fed with high-fat diet for three weeks was higher than 200 mg/d L,indicating that the hyperlipidemia rat model was successfully established.At the same dose,the final body weights of the rats of white sesame seed and kernel groups were higher than those of the black sesame seed and kernel groups;and the weights of the black and white sesame seed groups were lower than those of the black and white sesame kernel groups.The liver indexes of the normal control group and the experimental groups were significantly lower than the model control group(P<0.05),indicating that black and white sesame seed and kernel could reduce the lipid accumulation on the liver.The malondialdehyde(MDA)values of the liver in the normal control group and the experimental groups were significantly lower than that in the model control group(P<0.05).The total antioxidant capacity(T-AOC),high density lipoprotein cholesterol(HDL-C),glutathione peroxidase(GSH-Px)and total superoxide dismutase(T-SOD)in the liver of each experimental group were significantly higher than those in the model control group(P<0.05).Based on the above results as well as pathological observation and analysis,we found that the black and white sesame seeds and kernels could improve fatty liver lesions and lipid metabolism disorder and could reduce lipid accumulation on the liver,thereby protecting the liver from damage and reducing the risk of fatty liver.Moreover,black and white sesame seeds,black and white sesame kernels can reduce the levels of total cholesterol(TC)and triglyceride(TG)in serum,and high dose of black sesame seeds(30%black sesame seeds)and low dose of black sesame kernels(10%black sesame kernels)were more beneficial for the recovery of hyperlipidemia and fatty liver.The above results indicated that black and white sesame has hypolipidemic effect.Chenxi Li Yahui Li Yuxiang Ma Dongying Wang Yongzhan Zheng Xuede Wang 2020Grain & Oil Science and Technology2020,3,2:1
