| 5 | Celastrus orbiculatus extract induces mitochondrial-mediated apoptosis in human hepatocellular carcinoma cells显示文摘OBJECTIVE:To investigate the apoptotic effects and underlying molecular mechanisms of Celastrus orbiculatus(C.orbiculatus) extract in human hepatocellular carcinoma cells.METHODS:Human hepatocellular carcinoma cells(HCCLM6) were treated with C.orbiculatus extract(COE) at different nontoxic concentrations(10,20,40,80,and 160 μg/mL).The effect of COE on HC-CLM6 viability was examined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assays.Cellular apoptosis following COE treatment was assessed by flow cytometry and western blot analysis.RESULTS:COE significantly inhibited cell viability and induced apoptosis of HCCLM6 cells in a dose-dependent manner.Apoptosis was accompanied by increased Bax expression and decreased Bcl-2 expression.In addition,COE treatment led to the release of cytochrome c,activation of caspase-3,and cleavage of poly(ADP-ribose) polymerase(PARP).Furthermore,activation of extracellular signal-regulated kinase(ERK),p38 kinase,and c-Jun N-terminal kinase(JNK) phosphorylation,and down-regulation of Akt phosphorylation was observed.CONCLUSION:COE induces mitochondrial-mediated,caspase-dependent apoptosis in HCCLM6 cells,which might be attributed to the activation of mitogen-activated protein kinase(MAPK) and inhibition of Akt signaling pathways.These data suggest that COE may be a potential treatment for human hepatocellular carcinoma. | Hua Zhang Yayun Qian Yanqing Liu Guoqing Li Pingfang Cui Yaodong Zhu Hui Ma Xue Ji Shiyu Guo Hisamits Tadashi | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 18 |

In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. | 9 | Gray bootstrap method for estimating frequency-varying random vibration signals with small samples显示文摘During environment testing, the estimation of random vibration signals(RVS) is an important technique for the airborne platform safety and reliability. However, the available methods including extreme value envelope method(EVEM), statistical tolerances method(STM) and improved statistical tolerance method(ISTM) require large samples and typical probability distribution. Moreover, the frequency-varying characteristic of RVS is usually not taken into account. Gray bootstrap method(GBM) is proposed to solve the problem of estimating frequency-varying RVS with small samples. Firstly, the estimated indexes are obtained including the estimated interval, the estimated uncertainty, the estimated value, the estimated error and estimated reliability. In addition, GBM is applied to estimating the single flight testing of certain aircraft. At last, in order to evaluate the estimated performance, GBM is compared with bootstrap method(BM) and gray method(GM) in testing analysis. | 12 | Adsorption of lead on multi-walled carbon nanotubes with different outer diameters and oxygen contents: Kinetics, isotherms and thermodynamics显示文摘The effects of different outer diameters and surface oxygen contents on the adsorption of heavy metals onto six types of multi-walled carbon nanotubes (MWCNTs) were investigated in an aqueous solution and lead was chosen as a model metal ion. The results indicated that the percentage removal and adsorption capacity of lead remarkably increased with decreasing outer diameter due to larger specific surface area (SSA). The SSA-normalized maximum adsorption capacity (qm /SSA) and SSA-normalized adsorption coefficient (Kd /SSA) were strongly positively correlated with surface oxygen content, implying that lead adsorption onto MWCNTs significantly increases with the rise of oxygen content and decreases with decreasing SSA. The calculated thermodynamic parameters indicated that adsorption of lead on MWCNTs was endothermic and spontaneous. When the oxygen content of MWCNTs increased from 2.0% to 5.9%, the standard free energy (△ G0 ) became more negative, which implied that the oxygenated functional groups increased the adsorption affinity of MWCNTs for lead. Through calculation of enthalpy (△ H0 ), G0 and free energy of adsorption (Ea ), lead adsorption onto MWCNTs was recognized as a chemisorption process. | 13 | Qingnaoyizhi decoction suppresses the formation of glial fibrillary acidic protein-positive cells in cultured neural stem cells by inhibiting the Janus kinase 2/signal transducer and activator of transcription 3 signaling pathway显示文摘OBJECTIVE: Inactivation of the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3) signaling axis plays a crucial role in determining the fate of neural stem cells(NSCs).Qingnaoyizhi decoction(QNYZD) has been used for the treatment of vascular dementia and has shown to improve synaptic remodeling. The aim of this study was to evaluate the effect of cerebrospinal fluid(CSF) containing QNYZD(CSF-QNYZD) on the differentiation of cultured NSCs and the involvement of the JAK2/STAT3 pathway.METHODS: The protein expression levels of glial fibrillary acidic protein(GFAP), tubulin, drosophila mothers against decapentaplegic protein(SMAD-1), STAT3, and phosphorylated-STAT3 were detected by western immunoblot analysis in the groups: control, CSF, JAK/STAT inhibitor(AG490),CSF-QNYZD, and CSF-XDZ(CSF-Xidezhen). The differentiation of NSCs was determined by immunofluorescence staining. The proliferation of NSCs was measured using the Cell Counting Kit-8 proliferation assay.RESULTS: Compared with the control group,CSF-QNYZD and AG490 significantly increased the number and expression of tubulin-positive cells, reduced the number and expression of GFAP-positive cells, and down-regulated the expression of p-STAT3. However, CSF-QNYZD also decreased the expression of SMAD-1 and STAT3.CONCLUSION: Enhanced neuronal differentiation may be associated with the down-regulation of glial differentiation instead of promoting proliferationin treated NSCs. Furthermore, QNYZD may play a direct role in suppressing the formation of GFAP-positive cells and enhancing neuronal differentiation by inhibiting JAK2/STAT3 activation. | 17 | Integrating a weighted-average method into the random walk framework to generate individual friend recommendations显示文摘Friend recommendation is a fundamental service in both social networks and practical applications,and is influenced by user behaviors such as interactions, interests, and activities. In this study, we first conduct in-depth investigations on factors that affect recommendation results. Next, we design Friend++, a hybrid multi-individual recommendation model that integrates a weighted average method(WAM) into the random walk(RW) framework by seamlessly employing social ties, behavior context, and personal information. In Friend++, the first plus signifies recommending a new friend through network features, while the second plus stands for using node features. To verify our method, we conduct experiments on three social datasets crawled from the Sina microblog system(Weibo). Experimental results show that the proposed method significantly outperforms six baseline methods in terms of recall, precision, F1-measure, and MAP. As a final step, we describe a case study that demonstrates the scalability and universality of our method. | 18 | Porphyromonas gingivalis infection promotes mitochondrial dysfunction through Drp1-dependent mitochondrial fission in endothelial cells显示文摘Porphyromonas gingivalis(P.gingivalis),a key pathogen in periodontitis,has been shown to accelerate the progression of atherosclerosis(AS).However,the definite mechanisms remain elusive.Emerging evidence supports an association between mitochondrial dysfunction and AS.In our study,the impact of P.gingivalis on mitochondrial dysfunction and the potential mechanism were investigated.The mitochondrial morphology of EA.hy926 cells infected with P.gingivalis was assessed by transmission electron microscopy,mitochondrial staining,and quantitative analysis of the mitochondrial network.Fluorescence staining and flow cytometry analysis were performed to determine mitochondrial reactive oxygen species(mtROS)and mitochondrial membrane potential(MMP)levels.Cellular ATP production was examined by a luminescence assay kit.The expression of key fusion and fission proteins was evaluated by western blot and immunofluorescence.Mdivi-1,a specific Drp1 inhibitor,was used to elucidate the role of Drp1 in mitochondrial dysfunction.Our findings showed that P.gingivalis infection induced mitochondrial fragmentation,increased the mtROS levels,and decreased the MMP and ATP concentration in vascular endothelial cells.We observed upregulation of Drp1(Ser616)phosphorylation and translocation of Drp1 to mitochondria.Mdivi-1 blocked the mitochondrial fragmentation and dysfunction induced by P.gingivalis.Collectively,these results revealed that P.gingivalis infection promoted mitochondrial fragmentation and dysfunction,which was dependent on Drp1.Mitochondrial dysfunction may represent the mechanism by which P.gingivalis exacerbates atherosclerotic lesions. | Tong Xu Qin Dong Yuxiao Luo Yanqing Liu Liang Gao Yaping Pan Dongmei Zhang | 2021 | International Journal of Oral Science2021,13,3: | 7 |

But this SEM may be inappropriate in some cases where random errors or latent variables are highly nonnormal. The authors develop a new SEM, called as quantile SEM(QSEM), by allowing for a quantile regression-type structural equation and without distribution assumption of random errors and latent variables. A Bayesian empirical likelihood(BEL) method is developed to simultaneously estimate parameters and latent variables based on the estimating equation method. A hybrid algorithm combining the Gibbs sampler and Metropolis-Hastings algorithm is presented to sample observations required for statistical inference. Latent variables are imputed by the estimated density function and the linear interpolation method.
