1Cage slip characteristics of a cylindrical roller bearing with a trilobe-raceway显示文摘Based on dynamic analysis of rolling bearings, the nonlinear dynamic differential equations of a cylindrical roller bearing with a trilobe-raceway were established and solved by the GSTIFF(gear stiff) integer algorithm with a variable step. The influences of structural parameters and the tolerance of the trilobe-raceway, working conditions of the bearing, and the outer ring installation method on cage slip characteristics were investigated. The results show that:(i) The cage slip ratio and bearing rating life of a cylindrical roller bearing with a trilobe-raceway would reduce when the low-radius(radius of the outer raceway contour at the lowest point) and D-value(difference value between the high and low points of the outer raceway contour) decrease, and the former(low-radius) contributes more significantly.(ii) The cage slip ratio of a cylindrical roller bearing with a trilobe-raceway rises with the increase of the bearing speed, and decreases with the increase of the radial force; the variation range increases with the increase of the low-radius.(iii) When the installation angle of the outer ring increases in a period, the cage slip ratio remains unchanged while the bearing rating life rises up a little. 3LncDACH1 promotes mitochondrial oxidative stress of cardiomyocytes by interacting with sirtuin3 and aggravates diabetic cardiomyopathy显示文摘Diabetic cardiomyopathy(DCM)is a common complication in diabetic patients.The molecular mechanisms of DCM remain to be fully elucidated.The intronic long noncoding RNA of DACH1(lnc DACH1)has been demonstrated to be closely associated with heart failure and cardiac regeneration.In this study,we investigated the role of lnc DACH1 in DCM and the underlying molecular mechanisms.The expression of lnc DACH1 was increased in DCM hearts and in high glucose-treated cardiomyocytes.Knockout of lnc DACH1 reduced mitochondrial oxidative stress,cell apoptosis,cardiac fibrosis and hypertrophy,and improved cardiac function in DCM mice.Overexpression of lnc DACH1 exacerbated mitochondria-derived reactive oxygen species(ROS)level and apoptosis,decreased activity of manganese superoxide dismutase(Mn-SOD);while silencing of lnc DACH1 attenuated ROS production,mitochondrial dysfunction,cell apoptosis,and increased the activity of Mn-SOD in cardiomyocytes treated with high glucose.Lnc DACH1 directly bound to sirtuin3(SIRT3)and facilitated its degradation by ubiquitination,therefore promoting mitochondrial oxidative injury and cell apoptosis in mouse hearts.In addition,SIRT3 silencing abrogated the protective effects of lnc DACH1 deficiency in cardiomyocytes.In summary,lnc DACH1 aggravates DCM by promoting mitochondrial oxidative stress and cell apoptosis via increasing ubiquitination-mediated SIRT3 degradation in mouse hearts.Inhibition of lnc DACH1 represents a novel therapeutic strategy for the intervention of diabetic cardiomyopathy.Qi Zhang Danyang Li Xue Dong Xiaowen Zhang Junwu Liu Lili Peng Bo Meng Qi Hua Xinyu Pei Lu Zhao Xiaoxi Hu Yang Zhang Zhenwei Pan Yanjie Lu Baofeng Yang 2022Science China(Life Sciences)2022,65,6:4

5Safety of endovascular therapy for symptomatic intracranial artery stenosis:a national prospective registry显示文摘Introduction The safety outcomes of endovascular therapy for intracranial artery stenosis in a real-world stetting are largely unknown.The Clinical Registration Trial of Intracranial Stenting for Patients with Symptomatic Intracranial Artery Stenosis(CRTICAS)was a prospective,multicentre,real-world registry designed to assess these outcomes and the impact of centre experience.Methods 1140 severe,symptomatic intracranial arterial stenosis(ICAS)patients treated with endovascular therapy were included from 26 centres,further divided into three groups according to the annual centre volume of intracranial angioplasty and stent placement procedures over 2 years:(1)high volume for≥25 cases/year;(2)moderate volume for 10–25 cases/year and(3)low volume for<10 cases/year.Results The rate of 30-day stroke,transient ischaemic attack or death was 9.7%(111),with 5.4%,21.1%and 9.7%in high-volume,moderate-volume and low-volume centres,respectively(p<0.05).Multivariable logistic regression confirmed high-volume centres had a significantly lower primary endpoint compared with moderate-volume centres(OR=0.187,95%CI:0.056 to 0.627;p≤0.0001),while moderate-volume and low-volume centres showed no significant difference(p=0.8456).Conclusion Compared with the preceding randomised controlled trials,this real-world,prospective,multicentre registry shows a lower complication rate of endovascular treatment for symptomatic ICAS.Non-uniform utilisation in endovascular technology,institutional experience and patient selection in different volumes of centres may have an impact on overall safety of this treatment.Yabing Wang Tao Wang Adam Andrew Dmytriw Kun Yang Liqun Jiao Huaizhang Shi Jie Lu Tianxiao Li Yujie Huang Zhenwei Zhao Wei Wu Jieqing Wan Qinjian Sun Bo Hong Yongli Li Liyong Zhang Jianfeng Chu Qiong Cheng Yiling Cai Pengfei Wang Qi Luo Hua Yang Baijing Dong Yang Zhang Jun Zhao Zuoquan Chen Wei Li Xiaoxin Bai Weiwen He Xueli Cai Maimai Ti Osama O Zaidat 2022Stroke & Vascular Neurology2022,7,2:3
6Corynoxine B targets at HMGB1/2 to enhance autophagy forα-synuclein clearance in fly and rodent models of Parkinson's disease显示文摘Parkinson's disease(PD)is the most common neurodegenerative movement disease.It is featured by abnormal alphα-synuclein(α-syn)aggregation in dopaminergic neurons in the substantia nigra.Macroautophagy(autophagy)is an evolutionarily conserved cellular process for degradation of cellular contents,including protein aggregates,to maintain cellular homeostasis.Corynoxine B(Cory B),a natural alkaloid isolated from Uncaria rhynchophylla(Miq.)Jacks.,has been reported to promote the clearance ofα-syn in cell models by inducing autophagy.However,the molecular mechanism by which Cory B induces autophagy is not known,and theα-syn-lowering activity of Cory B has not been verified in animal models.Here,we report that Cory B enhanced the activity of Beclin 1/VPS34 complex and increased autophagy by promoting the interaction between Beclin 1 and HMGB1/2.Depletion of HMGB1/2 impaired Cory B-induced autophagy.We showed for the first time that,similar to HMGB1,HMGB2 is also required for autophagy and depletion of HMGB2 decreased autophagy levels and phosphatidylinositol 3-kinaseⅢactivity both under basal and stimulated conditions.By applying cellular thermal shift assay,surface plasmon resonance,and molecular docking,we confirmed that Cory B directly binds to HMGB1/2 near the C106 site.Furthermore,in vivo studies with a wild-typeα-syn transgenic drosophila model of PD and an A53Tα-syn transgenic mouse model of PD,Cory B enhanced autophagy,promotedα-syn clearance and improved behavioral abnormalities.Taken together,the results of this study reveal that Cory B enhances phosphatidylinositol 3-kinaseⅢactivity/autophagy by binding to HMGB1/2 and that this enhancement is neuroprotective against PD.Qi Zhu Juxian Song Jia-Yue Chen Zhenwei Yuan Liangfeng Liu Li-Ming Xie Qiwen Liao Richard D.Ye Xiu Chen Yepiao Yan Jieqiong Tan Chris Soon Heng Tan Min Li Jia-Hong Lu 2023Acta Pharmaceutica Sinica B2023,13,6:2

12Metagenomic Analysis Reveals A Possible Association Between Respiratory Infection and Periodontitis显示文摘Periodontitis is an inflammatory disease that is characterized by progressive destruction of the periodontium and causes tooth loss in adults.Periodontitis is known to be associated with dysbiosis of the oral microflora,which is often linked to various diseases.However,the complexity of plaque microbial communities of periodontitis,antibiotic resistance,and enhanced virulence make this disease difficult to treat.In this study,using metagenomic shotgun sequencing,we investigated the etiology,antibiotic resistance genes(ARGs),and virulence genes(VirGs)of periodontitis.We revealed a significant shift in the composition of oral microbiota as well as several functional pathways that were represented significantly more abundantly in periodontitis patients than in controls.In addition,we observed several positively selected ARGs and VirGs with the Ka/Ks ratio>1 by analyzing our data and a previous periodontitis dataset,indicating that ARGs and VirGs in oral microbiota may be subjected to positive selection.Moreover,5 of 12 positively selected ARGs and VirGs in periodontitis patients were found in the genomes of respiratory tract pathogens.Of note,91.8%of the background VirGs with at least one non-synonymous single-nucleotide polymorphism for natural selection were also from respiratory tract pathogens.These observations suggest a potential association between periodontitis and respiratory infection at the gene level.Our study enriches the knowledge of pathogens and functional pathways as well as the positive selection of antibiotic resistance and pathogen virulence in periodontitis patients,and provides evidence at the gene level for an association between periodontitis and respiratory infection.Zhenwei Liu Tao Zhang Keke Wu Zhongshan Li Xiaomin Chen Shan Jiang Lifeng Du Saisai Lu Chongxiang Lin Jinyu Wu Xiaobing Wang 2022Genomics, Proteomics & Bioinformatics2022,20,2:0

16Effectiveness of Traditional Chinese Medicine on coronavirus disease 2019 in 92 patients:a retrospective study显示文摘OBJECTIVE:To investigate the antipyretic effect of early treatment with Traditional Chinese Medicine(TCM) on coronavirus disease 2019(COVID-19) patients.METHODS:We retrospectively analyzed 369 patients from January 26th,2020 to April 15th,2020,who had been diagnosed with COVID-19.Among 92 eligible cases,45 cases were identified as treatment group Ⅰ(n = 45) and 47 cases were identified as treatment group Ⅱ.Patients in the treatment group Ⅰ were treated with TCM herbal decoction within 5 d after admission.Patients in the treatment group Ⅱ were treated with TCM herbal decoction after the 6th admission day.The onset time of antipyretic effect,the antipyretic time,the time of negative oropharyngeal swab nucleic acid conversion,and the changes of cell count in blood routine test were compared.RESULTS:The treatment group Ⅰ showed shorter average antipyretic duration(4 vs 7 d;P < 0.05),and shorter average time for polymerase chain reaction(PCR) nucleic acid test results to turn negative(7 vs 11 d;P < 0.05) than the treatment group II.For patients(n = 54) with body temperature > 38 ℃,patients in the treatment group I had shorter median onset time of antipyretic effect than those in the treatment group Ⅱ(3 vs 4 d;P < 0.05).The absolute lymphocyte(LYMPH) count and absolute eosinophil(EOS) count on the 3rd day after admission and the neutrophil/lymphocyte ratio on the 6th day after admission of patients in the treatment group I were notably different from those in the treatment group Ⅱ at the same time point(P < 0.05).Based on Spearman’s rank correlation analysis,the change of body temperature on the 3rd day after admission was positively correlated with the increase of EOS count and the increase of EOS count and LYMPH counts on the 6th day after admission(P < 0.01).CONCLUSIONS:Early TCM intervention within 5 d after hospital admission shortened the onset time of antipyretic effect and fever duration of COVID-19 patients,reduced the time required for PCR test results to turn negative.Moreover,early TCM intervention also improved the results of inflammatory markers for COVID-19 patients.LYMPH and EOS counts can be used as indicators of TCM antipyretic effect.XU Guihua CHEN Feifei ZHANG Wei WU Yingen CHEN Xiaorong SHI Kehua WANG Zhenwei SHI Miaoyan ZHANG Xing LU Yunfei YUAN Weian LYU Hua CHEN Xuan 2023Journal of Traditional Chinese Medicine2023,43,3:0
17Seasonal diet and microbiome shifts in wild rhesus macaques are better correlated at the level of nutrient components than food items显示文摘Food supply is one of the major drivers of animal behavior,and the gut microbiome is an important mediator be-tween food supply and its effects on physiology.However,predicting the outcome of diet change on microbiome and consequences for the animal has proven extremely challenging.We propose this reflects processes occurring at different scales.Inadequate accounting for the multi-level complexity of nutrition(nutrients,foods,diets)obscures the diet influence on microbiome and subsequently animal.Here,we present a detailed year-round,multi-level analysis of diet and microbiome changes in a wild population of a temperate primate,the rhesus macaque(Macaca mulatta).Total daily food and nutrient intake of 6 male and 6 female macaques was monitored in each of the 4 sea-sons(total 120 days observations).For each individual,we found significant variation in the microbiome between all 4 seasons.This response was more strongly correlated with changes in macronutrient intake than with food items and much of the response could be explained at the level of 6 ecological guilds—sets of taxa sharing similar responses to nutrient intake.We conclude that study of diet,microbiome,and animal performance in ecology will more effectively identify patterns if diet is recorded at the level of nutrient intake.Although microbiome response to diet does show variation in species-level taxa in response to food items,there is greater commonality in response at the level of guilds.A goal for microbiome researchers should be to identify genes encoding microbial attributes that can define such guilds.Zhenwei CUI Andrew JHOLMES Wenjuan ZHANG Dalong HU Qi SHAO Zhenlong WANG Jiqi LU David RAUBENHEIMER 2022Integrative Zoology2022,17,6:0
18Single-atomic Zn-(C/N/O)lithiophilic sites induced stable lithium plating/stripping in anode-free lithium metal battery显示文摘For anode-free lithium metal battery,lithiophilic surface modification on the current collector can effectively reduce the lithium nucleation barrier,so as to regulate the electrodeposition of lithium.Here,atomically dispersed Zn-(C/N/O)lithiophilic sites in the amorphous carbon medium were introduced onto Cu by an in-situ induced ion coordination chemistry strategy to get the modified Zn@NC@RGO@Cu current collector.X-ray absorption spectroscopy(XAS)combined with scanning transmission electron microscopy in high angle annular dark field(STEM-HAADF)analysis proved the single atomic state of the zinc sites surrounded by C,N,and O with a coordination number of~3.According to the electrochemical tests and first principle calculations,the ultra-uniformly dispersed Zn-(C/N/O)sites at the atomic level can effectively improve the lithium affinity,reduce the energy barrier for lithium nucleation,homogenize the lithium nucleation,and enhance an inorganic lithium compounds rich solid electrolyte interphase layer.As a result,the nucleation overpotential of lithium on the modified current collector was reduced to 7.7 mV,which was 5.4 times lower than that on bare Cu.Uniform lithium nucleation and deposition enabled stable Li plating/stripping and elevated Coulombic efficiency of 98.95%in Li||Cu cell after>850 cycles.Capacity retention of 89.7%was successfully achieved in the anode-free lithium metal battery after 100 cycles.Shifei Huang Sirong Lu Yao Lv Nanrui Li Zhenwei Wu Geng Zhong Xiaolong Ren Yufeng Wang Bo Sun Yuxiong Huang Feiyu Kang Yidan Cao 2023Nano Research2023,16,8:0
