1Alteration of P2X3 expression in dorsal root ganglia after sciatic nerve ligation显示文摘BACKGROUND:The expressions of P2X3 receptor in dorsal root ganglia(DRG)after different peripheral nerve injuries are diverse.It indicates the different roles of P2X3 in different models-caused neuropathologic pains.OBJECTIVE:To observe the expressions of P2X3 in corresponding DRG after sciatic nerve ligation in rats.DESIGN:Controlled observation experiment.SETTING:Department of Morphology,Hunan Traditional Chinese Medical College;Department of Human Anatomy and Neurobiology,Xiangya Medical College,Central South University.MATERIALS:Thirty-five healthy adult SD rats of clean grade an d either gender,weighing(200±20)g,were involved.According to the random digits table,the involved rats were randomized into 3 groups:normal group(n =5),sham-operated group(n =5)and experimental group(n =25).The experimental group were subdivided into 3,7,14,21,28 days groups according to different surviving time after operation,5 rats at each time point.Polyclonal rabbit anti-P2X3 antibody(ABCAM company);biotinylated goat anti-rabbit IgG(Zhongshanjingqiao Biotechnical Co.,Ltd.,Beijing);Motic fluorescence microscope(Motic,Germany).METHODS:The experiments were carried out in the Department of Human Anatomy and Neurobiology,Xiangya Medical College,Central South University from June to December 2006.① Rats of experimental group were created into models by ligation of right sciatic nerve according to the method of Seltzer et al.Left sciatic nerve was used as self-control.As for rats in the sham-operated group,ligation of sciatic nerve was omitted,but other procedures were the same as those in the experimental group.Rats of normal group were untouched.② Rats of the normal group and sham-operated group survived for 14 days separately,and those of experimental group survived for corresponding time.After being deeply anesthetized by intraperitoneal injection of over-dose sodium pentobarbital,the rats of experimental group were transcardially perfused.L4-6 corresponding DRG connected to sciatic nerve were taken for preparing transverse sections serially.③P2X3 expression in L4-6 DRG was detected by immunohistochemistry,immunofluorescence and image analysis techniques.MAIN OUTCOME MEASURES:P2X3 expression in L4-6 DRG of rats in each group.RESULTS:Thirty-five SD rats were involved in the final analysis.① P2X3 expression in DRG:In normal DRG of rats,there were abundant P2X3 immuno-positive small-and medium-sized primary sensory neurons,especially the small ones,which mostly received the input from C fibers.There were only a few large neurons expressing P2X3.The immuno-positive products mostly were located in the cytoplasm and processes.The expression of P2X3 had a slight but significant decrease in ipsilateral L4-6 DRG 3 days after sciatic nerve ligation,and a decreasing tendency was observed with the elongation of time.At 28 days,the expression had not returned to base line,and still maintained at a low level.② P2X3 immuno-positive gray scale in DRG:P2X3 immuno-positive gray scale in ipsilateral side L4-6 DRG was 117.74±2.38,129.12± 4.86,133.56±3.79,148.75±6.90 and 150.49±5.15,respectively at 3,7,14,21 and 28 days after sciatic nerve ligation,which was significantly higher than that in the normal group and sham-operated group(105.11±3.52,104.22±5.41,F =78.861,P < 0.05),also significantly higher than that in the contralateral side(105.53±5.85,108.54±3.70,104.07±4.16,106.55±2.02,106.29±5.19,t =3.48-13.95,P < 0.05);There were no significant differences when comparing sham-operated group or contralateral side at each time point with normal group(P > 0.05)CONCLUSION:P2X3 is significantly down regulated in L4-6 DRG after sciatic nerve ligation.It may exert certain effects in neuropathic pain.Guoxing Zhou Lesi Xie Qiben Wang Qingping Yu Xiaofu Liu Qiumei Liu Wei Peng Zhicheng Zeng 2007Neural Regeneration Research2007,2,4:3
2Hybrid Energy Sharing Mechanism for Integrated Energy Systems Based on the Stackelberg Game显示文摘Integrating the sharing economy and the power industry is of positive significance for the development of the energy market.With the energy market transforming from a traditional vertical structure to an interactive and competitive structure,users'roles need to change,along with supply and demand interacting more frequently.Thus,the traditional centralized optimization method for a single energy source can hardly reveal the complex multi-entity behavior of multi-energy coupling.Therefore,this paper establishes a distributed electrical-gas-thermal energy sharing mechanism centered on an energy hub that can converse energy,and build a more applicable integrated energy system body.First,the supply-demand interaction and the energy conversion process is constructed with reference to the operationa丨mode of’a sharing economy and the dual role of prosumers.A Stackelberg model is established with the integrated energy system operator as the leader and prosumers as the followers,to simultaneously optimize the profit of the leader in the upper level and the comfort of the fo!lowers,energy use and utility in the lower level.Furthermore,for protecting the participants’privacy,a distributed algorithm is used to find the optimal solution to equilibrate the model,and the existence and uniqueness of the solution is proved.Finally,a case study validates the effectiveness of the hybrid energy sharing mechanism and provides a reference for the integration of the energy sharing economy with the integrated energy system.Qiao Peng Xiuli Wang Yi Kuang Yifei Wang Hongyang Zhao Zhicheng Wang Jianhu Lyu 2021CSEE Journal of Power and Energy Systems2021,7,5:2

5Quantitative proteomic and phosphoproteomic analyses of the hippocampus reveal the involvement of NMDAR1 signaling in repetitive mild traumatic brain injury显示文摘The cumulative damage caused by repetitive mild traumatic brain injury can cause long-term neurodegeneration leading to cognitive impairment.This cognitive impairment is thought to result specifically from damage to the hippocampus.In this study,we detected cognitive impairment in mice 6 weeks after repetitive mild traumatic brain injury using the novel object recognition test and the Morris water maze test.Immunofluorescence staining showed that p-tau expression was increased in the hippocampus after repetitive mild traumatic brain injury.Golgi staining showed a significant decrease in the total density of neuronal dendritic spines in the hippocampus,as well as in the density of mature dendritic spines.To investigate the specific molecular mechanisms underlying cognitive impairment due to hippocampal damage,we performed proteomic and phosphoproteomic analyses of the hippocampus with and without repetitive mild traumatic brain injury.The differentially expressed proteins were mainly enriched in inflammation,immunity,and coagulation,suggesting that non-neuronal cells are involved in the pathological changes that occur in the hippocampus in the chronic stage after repetitive mild traumatic brain injury.In contrast,differentially expressed phosphorylated proteins were mainly enriched in pathways related to neuronal function and structure,which is more consistent with neurodegeneration.We identified N-methyl-D-aspartate receptor 1 as a hub molecule involved in the response to repetitive mild traumatic brain injury,and western blotting showed that,while N-methyl-D-aspartate receptor 1 expression was not altered in the hippocampus after repetitive mild traumatic brain injury,its phosphorylation level was significantly increased,which is consistent with the omics results.Administration of GRP78608,an N-methyl-D-aspartate receptor 1 antagonist,to the hippocampus markedly improved repetitive mild traumatic brain injury-induced cognitive impairment.In conclusion,our findings suggest that N-methyl-D-aspartate receptor 1 signaling in the hippocampus is involved in cognitive impairment in the chronic stage after repetitive mild traumatic brain injury and may be a potential target for intervention and treatment.Zhicheng Tian Zixuan Cao Erwan Yang Juan Li Dan Liao Fei Wang Taozhi Wang Zhuoyuan Zhang Haofuzi Zhang Xiaofan Jiang Xin Li Peng Luo 2023Neural Regeneration Research2023,18,12:1

10Reactive species regulation by interlayered Na^(+)/H^(+)of titanate nanotubes decorated Co(OH)_(2)hollow microsphere for peroxymonosulfate activation and gatifloxacin degradation显示文摘Emerging organic pollutants(EoPs)in water are of great concern due to their high environmental risk,so urgent technologies are needed for effective removal of those pollutants.Herein,a heterogeneous advanced oxidation process(AoP)of peroxymonosulfate(PMS)activation by functional material was developed for degradation of a typical antibiotic,gatifloxacin(GAT).The reactive species including sulfate radical(SO^(4)^(·-))and singlet oxygen(^(1)O_(2))in this AOP were regulated by interlayered ions(Na^(+)/H^(+))of titanate nanotubes that supported on Co(OH)_(2)hollow microsphere.Both the Na-type(NaTi-CoHS)and H-type(HTi-CoHS)materials achieved efficient PMS activation for GAT degradation,and HTi-CoHS even exhibited a relatively high degradation efficiency of 96.6%within 5 min.Co(OH)_(2)was considered the key component for generation of SO_(4)^(·-)after PMS activation,while hydrogen titanate nanotubes(H-TNTs)promoted the transformation of peroxysulfate radical(SO_(5)^(·-))to ^(1)O_(2) by hydrogen bond interaction.Therefore,when the interlayer ion of TNTs transformed from Na^(+) to H^(+),more ^(1)O_(2) was produced for organic pollutant degradation.H-TNTs with lower symmetry preferred to adsorb PMS molecules to achieve interlayer electron transport through hydrogen bonding,rather than electrostatic interaction of Na^(+) for Na-TNTs.In addition,the degradation pathway of GAT mainly proceeded by the cleavage of C-N bond at the 8 N site of the piperazine ring,which was confirmed by condensed Fukui index and mass spectrographic analysis.This work gives new sights into the regulation of reactive species in AoPs by the composition of material and promotes the understanding of pollutant degradation mechanisms in water treatment process.Bin Zhou Long Chen Fan Li Weiliang Sun Zhicheng Pan Yumei Peng Lian Qiu Wen Liu 2023Chinese Chemical Letters2023,34,12:0

13Robust sequential design for piecewise-stationary multi-armed bandit problem in the presence of outliers显示文摘The multi-armed bandit(MAB)problem studies the sequential decision making in the presence of uncertainty and partial feedback on rewards.Its name comes from imagining a gambler at a row of slot machines who needs to decide the best strategy on the number of times as well as the orders to play each machine.It is a classic reinforcement learning problem which is fundamental to many online learning problems.In many practical applications of the MAB,the reward distributions may change at unknown time steps and the outliers(extreme rewards)often exist.Current sequential design strategies may struggle in such cases,as they tend to infer additional change points to fit the outliers.In this paper,we propose a robust change-detection upper confidence bound(RCD-UCB)algorithm which can distinguish the real change points from the outliers in piecewise-stationary MAB settings.We show that the proposed RCD-UCB algorithm can achieve a nearly optimal regret bound on the order of O(√SKT log T),where T is the number of time steps,K is the number of arms and S is the number of stationary segments.We demonstrate its superior performance compared to some state-of-the-art algorithms in both simulation experiments and real data analysis.(See http://gffzz188fe103f8f1460as60pncqvkf5vc6uxc.ffgz.tsg.suse.edu.cn/woaishufenke/MAB_STRF.git for the codes used in this paper.)Yaping Wang Zhicheng Peng Riquan Zhang Qian Xiao 2021Statistical Theory and Related Fields2021,5,2:0
14A Monitoring Method for Transmission Tower Foots Displacement Based on Wind-Induced Vibration Response显示文摘The displacement of transmission tower feet can seriously affect the safe operation of the tower,and the accuracy of structural health monitoring methods is limited at the present stage.The application of deep learning method provides new ideas for structural health monitoring of towers,but the current amount of tower vibration fault data is restricted to provide adequate training data for Deep Learning(DL).In this paper,we propose a DT-DL based tower foot displacement monitoring method,which firstly simulates the wind-induced vibration response data of the tower under each fault condition by finite element method.Then the vibration signal visualization and Data Transfer(DT)are used to add tower fault data samples to solve the problem of insufficient actual data quantity.Subsequently,the dynamic response test is carried out under different tower fault states,and the tower fault monitoring is carried out by the DL method.Finally,the proposed method is compared with the traditional online monitoring method,and it is found that this method can significantly improve the rate of convergence and recognition accuracy in the recognition process.The results show that the method can effectively identify the tower foot displacement state,which can greatly reduce the accidents that occurred due to the tower foot displacement.Zhicheng Liu Long Zhao Guanru Wen Peng Yuan Qiu Jin 2023Structural Durability & Health Monitoring2023,17,6:0

16Single-cell RNA-seq Revealed that Altered Tumor Infiltrating Lymphocytes in Cirrhotic Liver Indicate Development of Hepatocellular Carcinoma显示文摘Background and Aims:Cirrhosis is the precursor lesion for most hepatocellular carcinoma(HCC)cases.However,no biomarker effectively predicted HCC initiation before diagnosis by imaging.We aimed to investigate the hallmarks of immune microenvironments in healthy,cirrhotic livers and HCC tumor tissues and to identify immune biomarkers of cirrhosis-HCC transition.Methods:Expression matrices of singlecell RNA sequencing studies were downloaded and integrated with Seurat package vignettes.Clustering was performed to analyze the immune cell compositions of different sample types.Results:The cirrhotic liver and HCC tumors had distinct immune microenvironments,but the immune landscape of cirrhotic livers was not markedly modified compared with healthy livers.Two subsets of B cells and three subsets of T cells were identified in the samples.Among the T cells,naïve T cells were more prominent in the cirrhotic and healthy liver samples than in the HCC samples.In contrast,the neutrophil count was lower in cirrhotic livers.Two macrophage clusters were identified,one that actively interacted with T cells and B cells and was enriched in cirrhotic blood compared with HCC blood samples.Conclusions:Decreased naïve T cell infiltration and increased neutrophil infiltration in the liver may indicate the development of HCC in cirrhotic patients.Alterations in blood-resident immune cells may also be a sign of HCC development in cirrhotic patients.The dynamics of the immune cell subsets may serve as novel biomarkers to predict the transition from cirrhosis to HCC.Jingxian Duan Peng Zhu Yong Zhang Tianhao Mu Yingqiang Li Rui Xiong Su Chen Yingmei Li Zhicheng Li Shifu Chen Lei Zhang 2023Journal of Clinical and Translational Hepatology2023,11,4:0
17Numerical study on the ceiling gas temperature in a subway train with different fire locations显示文摘Ceiling gas temperature rise is an important evaluation indicator determining the level of risk in a subway tunnel fire.However,very little literature has been found that has addressed the emergency when a fired subway train with lateral multiple openings stops in the interval tunnel.Hence,a battery of full-scale numerical simulations were employed to address the impact of train fire location on the gas temperature beneath the train ceiling.Numerical results showed that the ceiling gas temperature rise is affected by the pressure difference on both sides of fire source and the backflow from the end wall,which depends on the heat release rate and the fire location.The ceiling gas temperature rise decays exponentially in the process of longitudinal spread,and it can be predicted by a dimensionless model with a sum of two exponential equations.Finally,based on a critical fire location(L'cr=0.667),two exponential equations were developed to quantitatively express the influences of the fire size and the fire location on the maximum ceiling gas temperature.The research results can be utilized for providing an initial understanding of the smoke propagation in a subway train fire.Wei Cong Long Shi Zhicheng Shi Min Peng Hui Yang Xudong Cheng 2022Building Simulation2022,15,4:0
