| 3 | Isolation and characterization of H7N9 viruses from live poultry markets--Implication of the source of current H7N9 infection in humans显示文摘On March 31, 2013, the National Health and Family Planning Commission announced that human infections with a previously undescribed influenza A (H7N9) virus had occurred in Shanghai and Anhui Province, China. To investigate the possible origins of the H7N9 viruses causing these human infections, we collected 970 samples, including drinking water, soil, and cloacal and tracheal swabs of poultry from live poultry markets and poultry farms in Shanghai and Anhui Province. Twenty samples were positive for the H7N9 influenza virus. Notably, all 20 viruses were isolated from samples collected from live poultry markets in Shanghai. Phylogenetic analyses showed that the six internal genes of these novel human H7N9 viruses were derived from avian H9N2 viruses, but the ancestor of their HA and NA genes is uncertain. When we examined the phylogenetic relationship between the H7N9 isolates from live poultry markets and the viruses that caused the human infections, we found that they shared high homology across all eight gene segments. We thus identified the direct avian origin of the H7N9 influenza viruses that caused the human infections. Importantly, we observed that the H7N9 viruses isolated from humans had acquired critical mutations that made them more 'human-like'. | 4 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 7 | High expression of ACE2 receptor of 2019-nCoV on the epithelial cells of oral mucosa显示文摘It has been reported that ACE2 is the main host cell receptor of 2019-nCoV and plays a crucial role in the entry of virus into the cell to cause the final infection. To investigate the potential route of 2019-n Cov infection on the mucosa of oral cavity, bulk RNA-seq profiles from two public databases including The Cancer Genome Atlas(TCGA) and Functional Annotation of The Mammalian Genome Cap Analysis of Gene Expression(FANTOM5 CAGE) dataset were collected. RNA-seq profiling data of 13 organ types with para-carcinoma normal tissues from TCGA and 14 organ types with normal tissues from FANTOM5 CAGE were analyzed in order to explore and validate the expression of ACE2 on the mucosa of oral cavity. Further, single-cell transcriptomes from an independent data generated in-house were used to identify and confirm the ACE2-expressing cell composition and proportion in oral cavity. The results demonstrated that the ACE2 expressed on the mucosa of oral cavity. Interestingly, this receptor was highly enriched in epithelial cells of tongue. | 8 | Black carbon aerosols and their radiative properties in the Pearl River Delta region显示文摘The climatic and environmental effects of atmospheric aerosols are a hot topic in global science community, and radiative properties of the aerosols are one of the important parameters in assessing climatic change. Here we studied the black carbon concentration and absorption coefficient measured with aethalometers, scattering coefficient measured with nephelometers, and single scattering albedo derived at an atmospheric composition watch station in Guangzhou from 2004 to 2007. Our main results are as follows. The data of black carbon concentration and absorption coefficients measured with instruments cannot be directly used until they are measured in parallel with internationally accepted instruments for comparison, calibration, and reduction. After evaluation of the data, the result shows that the monthly mean of BC concentration varies 3.1―14.8 μg·m-3 and the concentration decreases by about 1 μg·m-3 in average over the four years; It is higher in the dry season with a multi-year mean of 8.9 μg/m3 and lower in the rainy season with a multi-year mean of 8.0 μg·m-3; The extreme maximum of monthly mean concentration occurred in December 2004 and extreme minimum in July 2007, and a 4-year mean is 8.4 μg·m-3. It is also shown that monthly mean scattering coefficient derived varies 129 -565 Mm-1, monthly mean absorption coefficient 32-139 Mm-1, and monthly mean single scattering albedo 0.71-0.91, with annual mean values of 0.80, 0.82, 0.79 and 0.84 for 2004, 2005, 2006 and 2007, respectively. Three instruments were used to take simultaneous measurements of BC in PM10, PM2.5, and PM1 and the results showed that PM2.5 took up about 90% of PM10 and PM1 accounted for about 68% of PM2.5, and BC aerosols are mainly present in fine particulates. The variability of BC concentrations is quite consistent between the Nancun station (141 m above sea level) and the Panyu station (13 m above sea level), which are 8 km apart from each other. | 9 | Potential toxic risk of heavy metals from sediment of the Pearl River in South China显示文摘Based on the monitoring of five heavy metal elements in the surface sediments of the Pearl River in South China, potential toxicity of the heavy metals was assessed using consensus-based sediment quality guidelines (SQGs) method and geo-accumulation (Igeo) index method. The monitoring results showed the heavy metal concentrations were significantly and positively correlated with each other, demonstrating a common trend in variation of concentration in the surface sediments. The assessment using the consensus-based SQGs method showed the potential toxicity of Cu was the highest, and Cd was the lowest. The evaluation based on mean probable effect concentration (PEC) quotient showed the region was seriously polluted with high toxicity heavy metals. Correlation analysis revealed a significant and positive correlation between the mean PEC quotient and the average of Igeo with a correlation coefficient of 0.926 (n = 23, P < 0.01). | 16 | Combination of Total Astragalus Extract and Total Panax Notoginseng Saponins Strengthened the Protective Effects on Brain Damage through Improving Energy Metabolism and Inhibiting Apoptosis after Cerebral Ischemia-Reperfusion in Mice显示文摘Objective:To explore the effects and molecular mechanisms of the combination between total Astragalus extract(TAE) and total Panax notoginseng saponins(TPNS) against cerebral ischemiareperfusion injury.Methods:C57BL/6 mice were randomly divided into sham-operated group,model group,TAE(110 mg/kg) group,TPNS(115 mg/kg) group,TAE-TPNS combination group and Edaravone(4 mg/kg) group,treated for 4 days,then,cerebral ischemia-reperfusion injury was established by bilateral common carotid artery(CCA) ligation for 20 min followed by reperfusion for 1 and 24 h.Results:TPNS could increase adenosine triphosphate(ATP) level,TAE and TAE-TPNS combination increased ATP,adenosine diphosphate(ADP) contents and Na+-K+-ATPase activity,and the effects of TAE-TPNS combination were stronger than those of TAE or TPNS alone after reperfusion for 1 h.After reperfusion for 24 h,TAE,TPNS and TAE-TPNS combination significantly increased neurocyte survival rate and decreased the apoptosis rate as well as down-regulated the expression of phosphorylated c-June N-terminal kinase1/2(p-JNK1/2),cytochrome C(Cyt C),cysteine aspartic acid-specific protease(Caspase)-9 and Caspase-3.Furthermore,the effects in TAE-TPNS combination were better than those in TAE or TPNS alone.Conclusion:The combination of TAE 110 mg/kg and TPNS 115 mg/kg could strengthen protective effects on cerebral ischemia injury,the mechanism underlying might be related to improving jointly the early energy metabolism,and relieving the delayed apoptosis via inhibiting the mitochondrial apoptosis pathway of JNK signal transduction. | HUANG Xiao-ping TAN Hua CHEN Bei-yang DENG Chang-qing | 2017 | Chinese Journal of Integrative Medicine2017,23,6: | 46 |

| 20 | Tree ring based streamflow reconstruction for the Upper Yellow River over the past 1234 years显示文摘We established a Juniperus przewalski tree ring width chronology, based on tree ring cores collected from the A’nyêmaqên Mountains. Statistical analysis showed that the chronology was highly correlated with instrumental streamflow records from previous August to current July from the Tangnaihai Station in the upper reaches of the Yellow River, with a correlation coefficient of 0.656. Streamflow for the upper reaches of the Yellow River was reconstructed for the past 1234 years. Low flow periods for the 11-year averaged streamflow reconstruction were definite as lower than mean plus 1 standard deviation, and high flow periods were higher than mean minus 1 standard deviation. Over the past 1234 years, high flows occurred 18 times, and low flows occurred 12 times. The main low flow periods were identified as AD 1140–1156, AD 1295–1309, AD 1473–1500, and AD 1820–1847, and the main high flow periods were identified as AD 846–873, and AD 1375–1400. Extremely low streamflow over the reconstruction period was seen during the late 15th century, coinciding with a widespread drought phenomenon, which took place in the northeastern Tibetan Plateau over the same period. Reconstructed streamflow shows significant low-frequency variability, which is in line with drought variability of neighboring regions, as inferred from tree rings and other proxies.
