| 1 | Expression of nuclear factor-KB in hepatocellular carcinoma and its relation with the X protein of hepatitis B virus显示文摘AIM In this study we investigated therelationship of the X protein of HBV and nuclearfactor-KB (NF-κB) and the expression of NF-KB inhuman hepatocellular carcinoma tissues.METHODS Immunohistochemistry SP methodwas used to detect the expression of NF-κB and the X protein of HBV in human hepatocellularcarcinoma tissues of 52 cases. Gene transfectionmediated by lipofectamine was used to transfectthe eukaryotic expression vector pCDNA3. 1-HBXof HBV x gene into human hepatocellularcarcinoma cell line HCC-9204 and NF-κB wasdetected.RESULTS NF-κB was widely expressed inhuman hepatocellular carcinoma tissues in atotal of 52 cases and its expression was relatedto the X protein of HBV. NF-KB was localizedboth in the cytoplasm and . The nuclei ofhepatocellular carcinoma cells in 11 cases whichwere positive for the X protein of HBV while in 41cases negative for the X protein of HBV, NF-Kbwas only localized in the cytoplasm ofhepatocellular carcinoma cells but translocatedto the nuclei of hepatocellular carcinoma cellsafter the eukaryotic expression vectorpCDNA3.1-HBX was transfected into HCC-9204cells.CONCLUSION This study strongly suggeststhat the nuclear factor NF-KB is widely expressedin hepatocellular carcinoma tissues in differentstyles according to the expression of the Xprotein of HBV. NF-κB is abnormally activated inhepatocellular carcinoma, which is probablyrelated to the X protein of HBV. | 11 | First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. | 12 | Downregulation of ceramide synthase 1 promotes oral cancer through endoplasmic reticulum stress显示文摘C18 ceramide plays an important role in the occurrence and development of oral squamous cell carcinoma.However,the function of ceramide synthase 1,a key enzyme in C18 ceramide synthesis,in oral squamous cell carcinoma is still unclear.The aim of our study was to investigate the relationship between ceramide synthase 1 and oral cancer.In this study,we found that the expression of ceramide synthase 1 was downregulated in oral cancer tissues and cell lines.In a mouse oral squamous cell carcinoma model induced by 4-nitroquinolin-1-oxide,ceramide synthase 1 knockout was associated with the severity of oral malignant transformation.Immunohistochemical studies showed significant upregulation of PCNA,MMP2,MMP9,and BCL2 expression and downregulation of BAX expression in the pathological hyperplastic area.In addition,ceramide synthase 1 knockdown promoted cell proliferation,migration,and invasion in vitro.Overexpression of CERS1 obtained the opposite effect.Ceramide synthase 1 knockdown caused endoplasmic reticulum stress and induced the VEGFA upregulation.Activating transcription factor 4 is responsible for ceramide synthase 1 knockdown caused VEGFA transcriptional upregulation.In addition,mild endoplasmic reticulum stress caused by ceramide synthase 1 knockdown could induce cisplatin resistance.Taken together,our study suggests that ceramide synthase 1 is downregulated in oral cancer and promotes the aggressiveness of oral squamous cell carcinoma and chemotherapeutic drug resistance. | Wen Chen Chenzhou Wu Yafei Chen Yuhao Guo Ling Qiu Zhe Liu Haibin Sun Siyu Chen Zijian An Zhuoyuan Zhang Yi Li Longjiang Li | 2021 | International Journal of Oral Science2021,13,1: | 3 |
| 13 | Chemical Fingerprint for Identification and Quality Control of Saccharides in Danhong Injection Based on HPLC-ELSD with Chemometrics显示文摘Chemical fingerprinting has been widely used for the quality control of complex natural products including traditional Chinese medicines and botanical drugs. However, there is still lack of appropriate method to monitor the batch-to-batch consistency of hydrophilic constituents, such as saccharides and oligosaccharides. In the present study, we developed a novel approach based on high performance liquid chromatography with evaporative light scattering detector(HPLC-ELSD) for establishing saccharide fingerprinting of Danhong Injection(DHI), which is a widely used botanical drug for treating cardiovascular diseases. Major saccharides in DHI were isolated and four compounds including fructose, glucose and two oligosaccharides were identified. The structures of two novel saccharides named glycerol-1-O-galactfpyranosyl-(1→4)-O-arabmofuranoside, and glycerol-1-O-galactpyranosyl-(1→4)[O-arabmfuranosyl-(1→3)]-O-arabinofuranoside were confirmed by NMR, HR-ESI-MS and GC-MS for the first time. The establishment approach was successfully applied to distinguishing 12 batches of DHI from other botanical drugs with the aid of principle component analysis as well as to evaluating batch-to-batch consistency with the help of calculating similarity of tmgerprints. | 14 | Measurement of the neutron total cross section of carbon at the Back-n white neutron beam of CSNS显示文摘To verify the performance of the neutron total cross-sectional spectrometer, the neutron total cross section of carbon is initially measured in the energy range of 1 eV to 20 MeV using the time-of-flight method. The measurement is performed at the Back-n white neutron source with a 76-m time-of-flight path using the China Spallation Neutron Source. A multilayer fast fission chamber with 235U and 238U is employed as the neutron detector. The diameter and thickness of the natural graphite sample are 70 mm and 40 mm, respectively. Signal waveforms are collected using a data acquisition system. Off-line data processing was used to obtain the neutron time-of-flight spectra and transmissions. The uncertainty of the counting statistics is generally approximately 3% for each bin in the energy range of 1–20 MeV. It is determined that the results for the neutron total cross section of carbon obtained using ^235U cells are in good agreement with the results obtained using 238U cells within limits of statistical uncertainty. | 20 | High energy and high brightness laser compton backscattering gamma-ray source at IHEP显示文摘Based on the LINAC of BEPCII, a high-polarized, high bightness, energy-tunable, monoenergetic laser compton backscattering (LCS)gamma-ray source is under construction at IHEP. The gamma-ray energy range is from 1 MeV to 111 MeV. It is a powerful and hopeful researchplatform to reveal the underlying physics of the nuclear, the basic particles and the vacuum or to check the exist basic physical models, quantumelectrodynamic (QED) theories. In the platform, a 1.064 mm Nd:YAG laser system and a 10.6 mm CO_(2) laser system are employed. All the triggersignals to the laser system and the electron control system are from the only reference clock at the very beginning of the LINAC to make sure thetemporal synchronization. Two optical transition radiation (OTR) targets and two charged-couple devices (CCD) are used to monitor and to alignthe electron beam and the laser beam.
