| 2 | Hyperglycemia induces protein nonenzymeatic glycosylation in brain neurons of diabetic rats at early stage显示文摘BACKGROUND: Protein nonenzymatic glycosylation is supposed to be one of mechanisms for chronic complications development in diabetes mellitus, and therefore, might play an important role in the neuronal degeneration. OBJECTIVE: To study the protein nonenzymatic glycosylation in brain neurons of diabetic rats, and to analyze the pathway of neuronal degeneration at the early stage of hyperglymecia. DESIGN: Randomized controlled animal experiment. SETTING: Department of Endocrinology, First hospital Affiliated to General Hospital of Chinese PLA and Beijing Laboratory for Brain Aging, Xuanwu Hospital Affiliated to Capital Medical University. MATERIALS: Thirty-five male Wistar rats (grade Ⅱ), aged 3 months old, and 11 male purebred Kunming mice (grade Ⅲ) without special pathogen, aged 3 months old, were provided by the Animal Room of Capital Medical University. METHODS: This experiment was carried out in the Beijing Laboratory for Brain Aging, Xuanwu Hospital Affiliated to Capital Medical University in 1998. The rats in the diabetic model group were intraperitoneally injected into 10 g/L STZ according to 60 mg/kg to establish rat models of diabetes mellitus. The blood glucose and body mass of rats in each group were determined respectively at 1, 2 and 3 months after modeling. The antibodies of advanced glycosylation end products (AGEs) of bovine serum albumin (anti-BSA) were self-prepared: ①The antigen of AGEs-BSA was prepared.②Eleven male Kuming mice (grade Ⅱ) of 3 months old without special pathogen were selected to inoculate AGEs-BSA. ③ The animals were immunized. ④Primary purification and detection of poly-antibodies of AGEs: the AGEs were performed immunohistochemical examination at 1 month after diabetic modeling by ELISA method. MAIN OUTCOME MEASURES: ① Detection results of blood glucose and body mass of rats in two groups at different time points. ② Determination of polyclonal antibody titer of AGEs-BSA. ③ Changes in immunohistochemical image of AGEs in brain tissue of rats in two groups. RESULTS: Thirteen rats in the diabetic model group and fifteen rats in the normal model group entered the stage of final analysis. ①Changes of blood glucose and body mass: At 1, 2 and 3 months after modeling, the blood glucose of rats in the diabetic model group were respectively(28.8±2.8),(23.1±5.5),(25.4± 5.1) mmol/L, which were significantly higher than those in the normal control group [(6.2±0.9),(6.1±0.8), (6.1±0.7) mmol/L,P < 0.01]; At 1, 2 and 3 months after modeling, the body mass of rats in the diabetic model group were respectively (250.1±52.2),(263.8±50.0),(261.5±42.6) g, which were significantly lower than those in the normal control group [(422.6±36.2),(462.6±39.0),(485.0±28.8) g,P < 0.01]. ②Determination of antibody titer of immune serum: The mice were treated by AGEs-BSA of different concentrations twice. After that, the titer of AGEs -BSA was determined, and the results of which indicated that a higher absorbance existed at 1∶1 000. ③Determination of antigen concentration: The final titer of antibody in the abdominal dropsy was determined, and the results of which suggested that there was a much higher absorbance in the AGEs-BSA at the concentration of 5–50 mg/L. ④Determination of antibody titer in abdominal dropsy: The antibody titer in abdominal dropsy was detected by ELISA method with antigen at 20 mg/L, which indicated that the maximum absorbance (1.265±0.039) existed at 1∶4 000, and very larger absorbance (0.982±0.067) at 1∶20 000. The polyclonal antibody of AGEs-BSA was successfully prepared. ⑤Immunohistochemical detection results: The immunohistochemical staining of AGEs showed there were positive neurons in the first month in the diabetic model group, whereas it was not significant in the normalcontrol group. The positive substances were found mainly in the cytoplasm. CONCLUSION: Hyperglycemia at the early stage of diabetes mellitus (1 month after modeling) can lead to protein nonenzymeatic glycosylation in brain neurons, and no obvious reactions mentioned above are found in the normal control group. | 4 | Fault diagnosis of bearings based on deep separable convolutional neural network and spatial dropout显示文摘Bearing pitting,one of the common faults in mechanical systems,is a research hotspot in both academia and industry.Traditional fault diagnosis methods for bearings are based on manual experience with low diagnostic efficiency.This study proposes a novel bearing fault diagnosis method based on deep separable convolution and spatial dropout regularization.Deep separable convolution extracts features from the raw bearing vibration signals,during which a 3×1 convolutional kernel with a one-step size selects effective features by adjusting its weights.The similarity pruning process of the channel convolution and point convolution can reduce the number of parameters and calculation quantities by evaluating the size of the weights and removing the feature maps of smaller weights.The spatial dropout regularization method focuses on bearing signal fault features,improving the independence between the bearing signal features and enhancing the robustness of the model.A batch normalization algorithm is added to the convolutional layer for gradient explosion control and network stability improvement.To validate the effectiveness of the proposed method,we collect raw vibration signals from bearings in eight different health states.The experimental results show that the proposed method can effectively distinguish different pitting faults in the bearings with a better accuracy than that of other typical deep learning methods. | Jiqiang ZHANG Xiangwei KONG Xueyi LI Zhiyong HU Liu CHENG Mingzhu YU | 2022 | Chinese Journal of Aeronautics2022,35,10: | 1 |

| 8 | Vercytochalasins A and B: Two unprecedented biosynthetically related cytochalasins from the deep-sea-sourced endozoic fungus Curvularia verruculosa显示文摘Vercytochalasins A(1) and B(2), two biosynthetically related cytochalasins featuring novel structure and substituents, were isolated from the endozoic fungus Curvularia verruculosa which was associated with the deep-sea squat lobster Shinkaia crosnieri collected from the cold seep environment in South China sea. Their structures were elucidated by detailed interpretation of NMR spectroscopic and mass spectrometric data. The absolute configurations were confirmed by NOESY experiments as well as by DP4+ and ECD calculations. Differed from common cytochalasins, compound 1 is an uncommon secocytochalasin featuring the ester group cleaved between C-9 and C-23, and incorporating an additional oxygenated C4 unit which coupled with C-20 and C-22 to form a new substituted cyclohexenone moiety, while compound 2 contains an unusual 2–hydroxy-3-oxobutan-2-yl unit at C-22. Both compounds are distinctive from the commonly described cytochalasins. Compound 1 exhibited potent activity against angiotensinI-converting enzyme(ACE) whereas compound 2 showed antibacterial activity. | 9 | Study on potential evapotranspiration and wet-dry condition in the seasonal frozen soil region of northern Tibetan Plateau显示文摘This study was based on the CEOP/CAMP-Tibet observed data at AWS (Automatic Weather Station) of MS3478 in the seasonal frozen soil region of northern Tibetan Plateau from March 2007 to February 2008.The variation characteristics of PE (potential evapotranspiration) were analyzed based on the Penman-Monteith method recommended by FAO (the Food and Agriculture Organization of the United Nations).The contributions of dynamic,thermal and water factors to PE were discussed,and the wet-dry condition of the plateau region was further studied.The results indicated that daily PE was between 0.52 mm and 6.46 mm for the whole year.Monthly PE was over 107 mm from May to September,but decreased to less than 41 mm from November to February.Annual PE was 1,037.8 mm.In the summer,thermal PE was significantly more than dynamic PE,but conversely in the winter.Annual variation of thermal PE was of sine wave pattern.In addition,drought and semi-drought climate lasted for a long time while semi-humid climate was short.The effect of water and dynamic factors on PE varied considerably with the seasons.Annual variation of thermal PE was of sine wave pattern. | HuiGen He ZeYong Hu XueYi Xun Jun Sun Li Hao LiJiao Xu Wen Peng | 2011 | Research in Cold and Arid Regions2011,3,2: | 1 |

| 13 | Mucus-penetrating nonviral gene vaccine processed in the epithelium for inducing advanced vaginal mucosal immune responses显示文摘Establishment of vaginal immune defenses at the mucosal interface layer through gene vaccines promise to prevent infectious diseases among females.Mucosal barriers composed of a flowing mucus hydrogel and tightly conjugated epithelial cells(ECs),which represent the main technical difficulties for vaccine development,reside in the harsh,acidic human vaginal environment.Different from frequently employed viral vectors,two types of nonviral nanocarriers were designed to concurrently overcome the barriers and induce immune responses.Differing design concepts include the charge-reversal property(DRLS)to mimic a virus that uses any cells as factories,as well as the addition of a hyaluronic acid coating(HA/RLS)to directly target dendritic cells(DCs).With a suitable size and electrostatic neutrality,these two nanoparticles penetrate a mucus hydrogel with similar diffusivity.The DRLS system expressed a higher level of the carried human papillomavirus type 16 L1 gene compared to HA/RLS in vivo.Therefore it induced more robust mucosal,cellular,and humoral immune responses.Moreover,the DLRS applied to intravaginal immunization induced high IgA levels compared with intramuscularly injected DNA(naked),indicating timely protection against pathogens at the mucus layer.These findings also offer important approaches for the design and fabrication of nonviral gene vaccines in other mucosal systems. | Qunjie Bi Xu Song Yangyang Zhao Xueyi Hu Huan Yang Rongrong Jin Yu Nie | 2023 | Acta Pharmaceutica Sinica B2023,13,3: | 0 |
