3Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 4Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. 6Flexible high energy density zinc-ion batteries enabled by binder-free MnO_(2)/reduced graphene oxide electrode显示文摘We demonstrate a rechargeable zinc-ion battery with high energy density and cyclability using MnO_(2)and reduced graphene oxide(MnO_(2)/rGO)electrode.The flexible and binder free electrode,with high MnO_(2)mass ratio(80 wt%of MnO_(2)),is fabricated using vacuum filtration without any additional additives other than rGO.Compared to batteries with conventional MnO_(2)electrodes,the Zn-MnO_(2)/rGO battery shows a significant enhanced capacity(332.2 mAh g^(-1)at 0.3 A g^(-1)),improved rate capability(172.3 mAh g^(-1)at 6 A g^(-1))and cyclability.The capacity retention remains 96%after 500 charge/discharge cycles at 6 A g^(-1).The high MnO_(2)mass ratio makes MnO_(2)/rGO electrode advantageous when the capacity is normalized to the whole electrode,particularly at high rates.The calculated gravimetric energy density of Zn-MnO_(2)/rGO battery is 33.17Wh kg^(-1),which is comparable to the existing commercial lead-acid batteries(30-40Wh kg^(-1)).Furthermore,the discharge profile and capacity of our Zn-MnO_(2)/rGO battery shows no deterioration during bending test,indicating good flexibility.As a result,zinc-ion battery is believed to be a promising technology for powering next generation flexible electronics.Yuan Huang Jiuwei Liu Qiyao Huang Zijian Zheng Pritesh Hiralal Fulin Zheng Dilek Ozgit Sikai Su Shuming Chen Ping-Heng Tan Shengdong Zhang Hang Zhou 2018npj Flexible Electronics2018,2,1:4
7Distribution and origin of biologically available phosphorus in the water of the Meiliang Bay in summer显示文摘The investigation and continuous monitoring with an innovative iron oxide embedded cellulose acetate membrane (FeO/CAM) on the concentrations of biologically available phosphorus (BAP) were conducted in the Meiliang Bay of the Taihu Lake during summer in 2004. The results showed that the concentrations of dissolved (FeO-DP), particulate (FeO-PP) and total bioavailable phosphorus (FeO-P) had similar horizontal distribution. The BAP concentrations were the highest in those estuaries in the northern bay. With the decrease of the distance to the estuary or long shore,there was little difference between BAP concentrations in an open lake area. During the observation period, algal blooms occurred in most waters of the northern bay, which was reflected from the high concentrations of chlorophyll a (Chl-a). While they were not highest in the estuarine waters of those major rivers, this is the case for the BAP concentrations. The concentrations of Chl-a had a significantly positive correlation with those of bioavailable phosphorus in the open area of the Meiliang Bay.With the sediment resuspension induced by wind and wave, BAP concentrations increased in a short-term, indicating that the riverine P inputs mainly contribute to the concentrations of BAP in the estuarine water while internal P release was the major source of BAP in the open lake area. In the eutrophic shallow lake, the blooms of alga may cause pH increase and further result in internal P release. 9Site-Specifc Gene Targeting Using Transcription Activator-Like Effector(TALE)-Based Nuclease in Brassica oleracea显示文摘Site-specifc recognition modules with DNA nuclease have tremendous potential as molecular tools for genome targeting.The type III transcription activator-like effectors(TALEs)contain a DNA binding domain consisting of tandem repeats that can be engineered to bind user-defned specifc DNA sequences.We demonstrated that customized TALE-based nucleases(TALENs),constructed using a method called'unit assembly',specifcally target the endogenous FRIGIDA gene in Brassica oleracea L.var.capitata L.The results indicate that the TALENs bound to the target site and cleaved double-strand DNA in vitro and in vivo,whereas the effector binding elements have a 23 bp spacer.The T7 endonuclease I assay and sequencing data show that TALENs made double-strand breaks,which were repaired by a non-homologous end-joining pathway within the target sequence.These data show the feasibility of applying customized TALENs to target and modify the genome with deletions in those organisms that are still in lacking gene target methods to provide germplasms in breeding improvement.Zijian Sun Nianzu Li Guodong Huang Junqiang Xu Yu Pan Zhimin Wang Qinglin Tang Ming Song Xiaojia Wang 2013Journal of Integrative Plant Biology2013,55,11:3
10Impact of dissolved humic acid on the bioavailability of acenaphthene and chrysene assessed by membrane-based passive samplers显示文摘Dissolved organic carbon (DOC) is known to reduce the bioavailability of hydrophobic organic compounds (HOCs) in aqueous environments. This reduction occurs as a result of adsorption to DOC, apparently reducing the freely dissolved concentration of HOCs. In the present study, triolein-embedded cellulose acetate membrane (TECAM) and Japanese medaka (Oryzias latipes) were used to measure the uptake of acenaphthene and chrysene in the presence of commercial humic acid (HA) at different concentrations (0―15 mg C·L-1) under controlled laboratory conditions. Apparent uptake rate constants for PAHs in TECAM and medaka were compared and DOC-water partition coefficients (KDOCs) of two PAHs were calculated with different sampling methods by model fit. Results showed that HA present in water significantly reduced the uptake of PAHs in TECAM and medaka. The obtained values of log KDOC of acenaphthene and chrysene measured by TECAM were 4.63 and 5.83, respectively, whereas biologically determined values were 4.52 and 5.76, respectively. These log KDOC values were comparable to earlier published KDOCs toward commercial HA, thereby indicating that TECAM accumulated only the freely dissolved fraction of chemicals and uptake PAHs in a manner similar to that of fish. 11Overview to the Hard X-ray Modulation Telescope (Insight-HXMT) Satellite显示文摘As China’s first X-ray astronomical satellite, the Hard X-ray Modulation Telescope (HXMT), which was dubbed as Insight-HXMT after the launch on June 15, 2017, is a wide-band(1-250 ke V) slat-collimator-based X-ray astronomy satellite with the capability of all-sky monitoring in 0.2-3 Me V. 13Design of Pd{111}-TiO_2 interface for enhanced catalytic efficiency towards formic acid decomposition显示文摘Supports are commonly implemented in the industrial application of heterogeneous catalysts to improve the stability and recyclability of catalysts.The supported catalysts often show the enhanced activity and selectivity in various catalytic reactions.However,the specific contributions of electronic and steric effects to a catalytic system often remain elusive due to the lack of well-defined model systems.In this work,two types of uniform Pd nanocrystals covered by{111}facets in tetrahedral and octahedral shapes,respectively,are synthesized with identical chemical environment and loaded on Ti O_2supports to form hybrid structures(Pd{111}-Ti O_2)towards the application of formic acid decomposition.Our observation suggests that the polarization effect at the interface of Pd-Ti O_2enhances its activity in formic acid decomposition.Moreover,the Pd tetrahedrons-Ti O_2hybrid structure whose Pd{111}-Ti O_2interface possesses a larger angle shows higher catalytic activity,owing to the reduced steric effect as compared to Pd octahedrons-Ti O_2.This study reveals the nature of interface effects in formic acid decomposition,and provides a guidance for the related catalyst design.Yang You Hao Huang Song Xia Zijian Cai Panyiming Liu Chengming Wang Ran Long Li Song Yujie Xiong 2018Science China Chemistry2018,61,9:2
