| 1 | Blockade of Tim-3 Pathway Ameliorates Interferon-γ Production from Hepatic CD8^+ T Cells in a Mouse Model of Hepatitis B Virus Infection显示文摘T cell immunoglobulin-and mucin-domain-containing molecule-3(Tim-3) has been reported to participate in the pathogenesis of inflammatory diseases. However,whether Tim-3 is involved in hepatitis B virus(HBV) infection remains unknown. Here,we studied the expression and function of Tim-3 in a hydrodynamics-based mouse model of HBV infection. A significant increase of Tim-3 expression on hepatic T lymphocytes,especially on CD8+ T cells,was demonstrated in HBV model mice from day 7 to day 18. After Tim-3 knockdown by specific shRNAs,significantly increased IFN-γ production from hepatic CD8+ T cells in HBV model mice was observed. Very interestingly,we found Tim-3 expression on CD8+ T cells was higher in HBV model mice with higher serum anti-HBs production. Moreover,Tim-3 knockdown influenced anti-HBs production in vivo. | 2 | Geochronology and Geochemistry of Metallogenetic Porphyry Bodies from the Nongping Au-Cu Deposit in the Eastern Yanbian Area, NE China: Implications for Metallogenic Environment显示文摘The metallogenetic porphyry bodies in the Nongping Au-Cu deposit, in the eastern Yanbian area, mainly include porphyritic granodiorite and biotite granodiorite porphyry. They are featured with high silicon and enrichment in sodium, and classified into sodic rocks of low-K tholeiitic basalt series. Except slightly low Sr content, the rock basically has the geochemical characteristics of the adakite: relatively high A12O3 content, relatively low MgO content, depletion in Y and Yb; relative enrichment in large ion lithophile elements (LILEs) and light rare-earth elements (LREEs), relatively low content of high field strength elements (HFSEs); positive Eu anomaly or weak negative Eu anomaly. In situ zircon dating technology LA-MC-ICP-MS was used to conduct single-grain zircon dating of biotite granodiorite porphyry, and the results show that the age of metallogenetic porphyry body is 100.04±0.88 Ma, indicating that the porphyry bodies were emplaced in the late Cretaceous period. According to the regional tectonic setting and the comparison with the same kind of deposits, we think that the metallogenetic porphyry bodies in the Nongping Au-Cu deposit have a close genetic connection with the subduction of the Pacific plate in the late Yanshanian period. | 3 | Bone Loss Induced by Simulated Microgravity,Ionizing Radiation and/or Ultradian Rhythms in the Hindlimbs of Rats显示文摘Objective To better understand the pathological causes of bone loss in a space environment, including microgravity, ionizing radiation, and ultradian rhythms. Methods Sprague Dawley(SD) rats were randomly divided into a baseline group, a control group, a hindlimb suspension group, a radiation group, a ultradian rhythms group and a combined-three-factor group. After four weeks of hindlimb suspension followed by X-ray exposure and/or ultradian rhythms, biomechanical properties, bone mineral density, histological analysis, microstructure parameters, and bone turnover markers were detected to evaluate bone loss in hindlimbs of rats. Results Simulated microgravity or combined-three factors treatment led to a significant decrease in the biomechanical properties of bones, reduction in bone mineral density, and deterioration of trabecular parameters. Ionizing radiation exposure also showed adverse impact while ultradian rhythms had no significant effect on these outcomes. Decrease in the concentration of the turnover markers bone alkaline phosphatase(b ALP), osteocalcin(OCN), and tartrate-resistant acid phosphatase-5 b(TRAP-5 b) in serum was in line with the changes in trabecular parameters. Conclusion Simulated microgravity is the main contributor of bone loss. Radiation also results in deleterious effects but ultradian rhythms has no significant effect. | 5 | Substantial transition to clean household energy mix in rural China显示文摘The household energy mix has significant impacts on human health and climate,as it contributes greatly to many health-and climate-relevant air pollutants.Compared to the well-established urban energy statistical system,the rural household energy statistical system is incomplete and is often associated with high biases.Via a nationwide investigation,this study revealed high contributions to energy supply from coal and biomass fuels in the rural household energy sector,while electricity comprised∼20%.Stacking(the use of multiple sources of energy)is significant,and the average number of energy types was 2.8 per household.Compared to 2012,the consumption of biomass and coals in 2017 decreased by 45%and 12%,respectively,while the gas consumption amount increased by 204%.Increased gas and decreased coal consumptions were mainly in cooking,while decreased biomass was in both cooking(41%)and heating(59%).The time-sharing fraction of electricity and gases(E&G)for daily cooking grew,reaching 69%in 2017,but for space heating,traditional solid fuels were still dominant,with the national average shared fraction of E&G being only 20%.The non-uniform spatial distribution and the non-linear increase in the fraction of E&G indicated challenges to achieving universal access to modern cooking energy by 2030,particularly in less-developed rural and mountainous areas.In some non-typical heating zones,the increased share of E&G for heating was significant and largely driven by income growth,but in typical heating zones,the time-sharing fraction was<5%and was not significantly increased,except in areas with policy intervention.The intervention policy not only led to dramatic increases in the clean energy fraction for heating but also accelerated the clean cooking transition.Higher income,higher education,younger age,less energy/stove stacking and smaller family size positively impacted the clean energy transition. | Guofeng Shen Rui Xiong Yanlin Tian Zhihan Luo Bahabaike Jiangtulu Wenjun Meng Wei Du Jing Meng Yuanchen Chen Bing Xue Bin Wang Yonghong Duan Jia Duo Fenggui Fan Lei Huang Tianzhen Ju Fenggui Liu Shunxin Li Xianli Liu Yungui Li Mu Wang Ying Nan Bo Pan Yanfang Pan Lizhi Wang Eddy Zeng Chao Zhan Yilin Chen Huizhong Shen Hefa Cheng Shu Tao | 2022 | National Science Review2022,9,7: | 4 |

Till date, a total of 84 unique genotypes of AdVs have been identified and classified as human Mastadenovirus species A to G, and specific types are often associated with certain clinical symptoms, epidemiological settings, and demographic risk groups[2]. Among these species, members of the species HAdV-B (HAdV-3, HAdV-7, HAdV-11, HAdV-16, HAdV-21, HAdV-34, HAdV-35, HAdV-50, etc), HAdV-C (HAdV-1, HAdV-2, HAdV-5, and HAdV-6), and HAdV-E (HAdV-4) have been generally associated with respiratory infections[3]. | 9 | Ginsenoside Rb1 Reduces D-GalN/LPS-induced Acute Liver Injury by Regulating TLR4/NF-κB Signaling and NLRP3 Inflammasome显示文摘Background and Aims:The effect of ginsenoside Rb1 on D-galactosamine(D-GalN)/lipopolysaccharide(LPS)-induced acute liver injury(ALI)is unknown.The aim of this study was to evaluate the effect of ginsenoside Rb1 on ALI and its underlying mechanisms.Methods:Mice were pretreated with ginsenoside Rb1 by intraperitoneal injection for 3 days before D-GalN/LPS treatment,to induce ALI.The survival rate was monitored every hour for 24 h,and serum biochemical parameters,hepatic index and histopathological analysis were evaluated to measure the degree of liver injury.ELISA was used to detect oxidative stress and inflammatory cytokines in hepatic tissue and serum.Immunohistochemistry staining,RT-PCR and western blotting were performed to evaluate the expression of toll-like receptor 4(TLR4),nuclear factorkappa B(NF-κB),and NLR family,pyrin domain-containing 3 protein(NLRP3)in liver tissue and Kupffer cells(KCs).Results:Ginsenoside Rb1 improved survival with D-GalN/LPS-induced ALI by up to 80%,significantly ameliorated the increased alanine and aspartate transaminase,restored the hepatic pathological changes and reduced the levels of oxidative stress and inflammatory cytokines altered by D-GalN/LPS.Compared to the control group,the KCs were increased in the D-GalN/LPS groups but did not increase significantly with Rb1 pretreatment.D-GalN/LPS could upregulate while Rb1 pretreatment could downregulate the expression of interleukin(IL)-1β,IL-18,NLRP3,apoptosis associated specklike protein containing CARD(ASC)and caspase-1 in isolated KCs.Furthermore,ginsenoside Rb1 inhibited activation of the TLR4/NF-κB signaling pathway and NLRP3 inflammasome induced by D-GalN/LPS administration.Conclusions:Ginsenoside Rb1 protects mice against D-GalN/LPS-induced ALI by attenuating oxidative stress and the inflammatory response through the TLR4/NF-κB signaling pathway and NLRP3 inflammasome activation. | Yimei Liu Ninghua Liu Yujing Liu Hongyu He Zhe Luo Wenjun Liu Nan Song Minjie Ju | 2022 | Journal of Clinical and Translational Hepatology2022,10,3: | 2 |
