1Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. 2Antioxidant properties of magnesium lithospermate B contribute to the cardioprotection against myocardial ischemia/reperfusion injury in vivo and in vitro显示文摘OBJECTIVE: To determine the cardioprotective effect of magnesium lithospermate B (MLB) on myocardial ischemia/reperfusion (MI/R) injury and to investigate the antioxidant potential in vivo and in vitro. METHODS: MI/R injury was induced by the occlusion of left anterior descending coronary artery for 30 min followed by reperfusion for 3 h in rats. After reperfusion, hearts were harvested to assess infarct size, histopathological damages, the levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH) and malondialdehyde (MDA). Blood samples were col- lected to determine serum levels of creatine kinase-MB (CK-MB), cardiac troponin (cTnI) and lactate dehydrogenase (LDH). Furthermore, simulatedischemia/reperfusion (SI/R) injury in vitro was established by oxygen and glucose deprivation (OGD) for 2 h followed by 24-hour recovery period in cardiomyocytes. The activity of LDH in the cultured supernatant and the levels of intracellular reactive oxygen species (ROS), SOD and MDA in cardiomyocytes were also measured. Finally, cardiomyocytes apoptosis was determined with flow cytometry. RESULTS: MLB significantly limited infarct size, ameliorated histopathological damages and prevented leakage of CK-MB, cTnI and LDH. Additionally, SOD, CAT, GPx and GSH activities were notably increased by MLB, along with the MDA content decreased as compared with the model group in rats. In vitro study, MLB also decreased LDH activity in the cultured supernatant, increased SOD activity in cardiomyocytes, reduced intracellular ROS and MDA levels, and significantly suppressed cardiomyocytes apoptosis. CONCLUSION: MLB possessed remarkably cardioprotective effects on MI/R injury in vivo and in vitro. 3Medical expenditures for colorectal cancer diagnosis and treatment: A 10-year high-level-hospital-based multicenter retrospective survey in China, 2002-2011显示文摘Objective: Colorectal cancer(CRC) causes a substantial burden of disease in China and the evidence of economic burden triggered is fundamental for priority setting. The aim of this survey was to quantify medical expenditures and the time trends for CRC diagnosis and treatment in China.Methods: From 2012 to 2014, a hospital-based multicenter retrospective survey was conducted in 13 provinces across China. For each eligible CRC patient diagnosed from 2002 to 2011, clinical information and expenditure data were extracted using a uniform questionnaire. All expenditure data were reported in Chinese Yuan(CNY)using 2011 values.Results: Of the 14,536 CRC patients included, the average age at diagnosis was 58.2 years and 15.8% were stageI cases. The average medical expenditure per patient was estimated at 37,902 CNY [95 % confidence interval(95%CI): 37,282-38,522], and the annual average increase rate was 9.2% from 2002 to 2011(P for trend <0.001), with a cumulative increase of 2.4 times(from 23,275 CNY to 56,010 CNY). The expenditure per patient in stages Ⅰ, Ⅱ, Ⅲ and Ⅳ were 31,698 CNY, 37,067 CNY, 38,918 CNY and 42,614 CNY, respectively(P<0.001). Expenditure significantly differed within various subgroups. Expenses for drugs contributed the largest proportion(52.6%).Conclusions: These conservative estimates illustrated that medical expenditures for CRC diagnosis and treatment in tertiary hospitals in China were substantial and increased rapidly over the 10 years, with drugs continually being the main expense by 2011. Relatively, medical expenditures are lower for CRC in the earlier stages. 4Effects of chemical modification on physicochemical properties and adsorption behavior of sludge-based activated carbon显示文摘This study aimed to explore the adsorption performance of sludge-based activated carbon(SBC)towards dissolved organic matters(DOMs)removal from sewage,and investigated the modification effect of different types of chemicals on the structure of synthesized SBC.Waste activated sludge(WAS)was used as a carbon source,and HCl,HNO 3,and Na OH were used as different types of chemicals to modify the SBC.With the aid of chemical activation,the modified SBC showed higher adsorption performances on DOMs removal with maximum adsorption of 29.05 mg/g and second-order constant(k)of 0.1367(L/mol/sec)due to the surface elution of ash and minerals by chemicals.The surface elemental composition of MSBC suggested that the content of C-C and C-O functional groups on the surface of modified sludge-based activated carbon(MSBC)played an important role on the adsorption capacities of MSBC towards DOMs removal in sewage.Additionally,the residual molecular weight of DOMs in sewage was investigated using a 3-dimension fluorescence excitationemission matrix(3 D-EEM)and high-performance size exclusion chromatography(HP-SEC).Results showed that the chemical modification significantly improved the adsorption capacity of MSBC on humic acids(HA)and aromatic proteins(APN),and both of Na OH-MSBC and HCl-MSBC were effective for a wide range of different AMW DOMs removal from sewage,while the HNO 3-MSBC exhibited poorly on AMW organics of 2,617 Da and 409 Da due to the reducing content of macropore.In brief,this study provides reference values for the impact of the chemicals of the activation stage before the SBCs application.Chunxu Wu Lanfeng Li Hao Zhou Jing Ai Hongtao Zhang Jialin Tao Dongsheng Wang Weijun Zhang 2021Journal of Environmental Sciences2021,33,2:7
5Effects of Shuanghuanglian oral liquids on patients with COVID-19:a randomized,open-label,parallel-controlled,multicenter clinical trial显示文摘We conducted a randomized,open-label,parallel-controlled,multicenter trial on the use of Shuanghuanglian(SHL),a traditional Chinese patent medicine,in treating cases of COVID-19.A total of 176 patients received SHL by three doses(56 in low dose,61 in middle dose,and 59 in high dose)in addition to standard care.The control group was composed of 59 patients who received standard therapy alone.Treatment with SHL was not associated with a difference from standard care in the time to disease recovery.Patients with 14-day SHL treatment had significantly higher rate in negative conversion of SARS-CoV-2 in nucleic acid swab tests than the patients from the control group(93.4%vs.73.9%,P=0.006).Analysis of chest computed tomography images showed that treatment with high-dose SHL significantly promoted absorption of inflammatory focus of pneumonia,which was evaluated by density reduction of inflammatory focus from baseline,at day 7(mean difference(95%CI),−46.39(−86.83 to−5.94)HU;P=0.025)and day 14(mean difference(95%CI),−74.21(−133.35 to−15.08)HU;P=0.014).No serious adverse events occurred in the SHL groups.This study illustrated that SHL in combination with standard care was safe and partially effective for the treatment of COVID-19.Li Ni Zheng Wen Xiaowen Hu Wei Tang Haisheng Wang Ling Zhou Lujin Wu Hong Wang Chang Xu Xizhen Xu Zhichao Xiao Zongzhe Li Chene Li Yujian Liu Jialin Duan Chen Chen Dan Li Runhua Zhang Jinliang Li Yongxiang Yi Wei Huang Yanyan Chen Jianping Zhao Jianping Zuo Jianping Weng Hualiang Jiang Dao Wen Wang 2021Frontiers of Medicine2021,15,5:7

7The pathogenicity on legumes of Cucumber mosaic virus was determined by 243 nucleotides on 2a polymerase gene of viral RNA2显示文摘We had isolated and identified two Cucumber mosaic virus (CMV) isolates, the CMV red bean (CMV-RB) isolate and the CMV pea (CMV-P1) isolate. CMV-RB induces necrotic local lesions on inoculated leaves of broad bean, pea, cowpea and bean, and could not infect these hosts systemically. However, CMV-P1 was able to infect these legumes systemically. To study the difference of pathogenic-ity on the legumes induced by these two CMV isolates, the full-length infectious cDNA clones of CMV-Fny, which induced similar symptoms as CMV-RB in the four legumes, were used. The 243 nucleotides fragment, which encodes highly conserved GDD amino acid motif on 2a replicase gene of CMV-Fny RNA2, was replaced with that of CMV-P1. The constructed chimeric virus FP could infect these legumes systemically. 8The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 11Identification and Characterization of Tonoplast Sugar Transporter (TST) Gene Family in Cucumber显示文摘Tonoplast sugar transporters(TSTs)play essential roles in regulating plant growth,development,and response to various biotic and abiotic stresses.In this study,a total of three TST genes were identified by a genome-wide analysis in cucumber.Phylogenetic analysis showed that TST proteins from cucumber and other plant species can be classified into five groups,and nearly all TST members in the same groups displayed similarmotif distributions,transmembrane(TM)domains,and gene structures.All of the three CsTST genes possess a number of development-,stress-,and hormone-related cis-elements in the promoter sequences.Meanwhile,qRT-PCR assays revealed that the CsTST1 was expressed in fruits,flowers,leaves,and other tissues,and its expression varied significantly under various abiotic stresses such as cold,salt,drought(PEG),and abscisic acid(ABA).Finally,functional analysis of CsTST1 in yeast revealed that it was able to complement the deficiency in galactose,mannose and sucrose transport.These results revealed that CsTST1 can act as a functional sugar transporter to play important roles in cucumber growth and response to abiotic stress probably through affecting carbohydrate distribution.Weifeng Huang Bin Hu Jialin Liu Yong Zhou Shiqiang Liu 2020Horticultural Plant Journal2020,6,3:4
12Isoflavones’effects on pharmacokinetic profiles of main iridoids from Gardeniae Fructus in rats显示文摘Gardeniae Fructus(GF)and Semen Sojae Praeparatum(SSP)are both medicine food homologies and widely used in Chinese clinical prescriptions together.The research investigated the pharmacokinetics of four iridoids in normal rats and isolfavones-fed rats,which were administered with isolfavones from SSP for 7,14,21 and 28 consecutive days.A validated LC-MS/MS method was developed for determining shanzhiside,genipin-1-gentiobioside,geniposide and their metabolite genipin in rat plasma.Plasma samples were pretreated by solid-phase extraction using paeoniflorin as the internal standard.The chromatographic separation was performed on a Waters Atlantis T3(4.6 mm×150 mm,3 mm)column using a gradient mobile phase consisting of acetonitril and water(containing 0.06%acetic acid).The mass detection was under the multiple reaction monitoring(MRM)mode via polarity switching between negative and positive ionization modes.The calibration curves exhibited good linearity(r>0.997)for all components.The lower limit of quantitation was in the range of 1 e10 ng/m L.The intra-day and inter-day precisions(RSD)at three different levels were both less than 12.2%and the accuracies(RE)ranged fromà10.1%to 16.4%.The extraction recovery of them ranged from 53.8%to 99.7%.Pharmacokinetic results indicated the bioavailability of three iridoid glycosides and the metabolite,genipin in normal rats was higher than that in rats exposed to isoflavones.With the longer time of administration of isoflavones,plasma concentrations of iridoids decreased,while genipin sulfate,the phase II metabolite of genposide and genipin-1-gentiobioside,appeared the rising exposure.The pharmacokinetic profiles of main iridoids from GF were altered by isoflavones.Ruirui Chang Jialin Liu Yusha Luo Taohong Huang Qiang Li Jun Wen Weidong Chen Tingting Zhou 2020Journal of Pharmaceutical Analysis2020,10,6:2
13Directed migration of human neural progenitor cells to interleukin-1βis promoted by chemokines stromal cell-derived factor-1 and monocyte chemotactic factor-1 in mouse brains显示文摘Background:Neurogenesis,including the proliferation,migration and differentiation of neural progenitor cells(NPCs),is impaired in HIV-1 associated dementia(HAD).We previously demonstrated HIV-1-infected macrophages(HIV-MDM)regulate stromal cell-derived factor 1(SDF-1)production in astrocytes through Interleukin-1β(IL-1β).Chemokines are known to induce NPC migration;however,it remains unclear how chemokines produced in inflammation regulate NPC migration.Methods:The secretion of SDF-1 and Monocyte chemotactic preotein-1(MCP-1)in astrocytes upon IL-1βstimulation was measured by ELISA assay.Human NPCs were injected parallel along with IL-1β,SDF-1 or MCP-1 intracranially into basal ganglion 1 mm apart in SCID mice,and immunofluorescent staining was used to study the survival and migration of injected human NPCs.Results:SDF-1 and MCP-1 are secreted by astrocytes upon IL-1βstimulation in a time-dependent manner.Injected human NPCs survived in SCID mice and migrated towards sites of IL-1β,SDF-1 and MCP-1 injection.Conclusions:In conclusion,chemokines SDF-1 or MCP-1 secreted by astrocytes in the presence of IL-1βinjection are attractive to NPCs injected into SCID mouse brains,suggesting that SDF-1 and MCP-1 play important roles in NPC migration during neuroinflammation.Yumei Wu Qiang Chen Hui Peng Huanyu Dou You Zhou Yunlong Huang Jialin C Zheng 2012Translational Neurodegeneration2012,1,1:1

19Magnetocaloric effect of (Gd_(1-x)Nd_x)Co_2 alloys in low magnetic field显示文摘The phases and magnetocaloric effect in the alloys (Gd1-xNdx)Co2 with x = 0, 0.1, 0.2, 0.3, and 0.4 were investigated by X-ray diffraction analysis and magnetization measurement. The samples are single phase with a cubic MgCu2-type structure. The Tc decreases obviously with increasing Nd content from 404 K of the alloy with x = 0 to 272 K of the alloy with x = 0.4; for x = 0.3, the Tc is 296 K, which is near room temperature. In the samples (Gd1-xNdx)Co2 with x = 0.0, 0.1, 0.2, 0.3, and 0.4, the maximum magnetic entropy change is 1.471, 1.228, 1.280, 1.381 and 1.610 J·kg-1·K-1, respectively, in the applied field range of 0-2.0 T. 20Polydopamine/polyethyleneimine co-crosslinked graphene oxide for the enhanced tribological performance of epoxy resin coatings显示文摘To enhance the wear resistance of graphene oxide(GO)nano-filled epoxy resin(EP)coatings with ex-cellent dispersion,GO were modified using a co-crosslinking strategy between polydopamine(PDA)and polyethyleneimine(PEI)through an environmentally friendly,simple and low-cost process.High-density amino-branched PEI and highly adhesive PDA formed covalent bond cross-links with GO(PDA/PEI-GO)through the Michael addition and Schiffbase reactions.With the introduction of amino active sites,the PDA self-polymerization reaction time was significantly shorted from 24 to 6 h.The covalently cross-linked network increased the lamellar spacing of GO and enhanced the interfacial bonds between the PDA/PEI-GO lamellae and EP matrix.Due to the improved dispersion and surface activity of GO,the PDA/PEI-GO nano-filled EP coating exhibited excellent wear resistance.Reciprocating friction with GCr15 stainless steel ball,the average friction coefficient and wear rate of the coating were reduced by 31.9%and 62.7%,respectively,compared to the pure EP coating.In addition,the wear mechanism of the PDA/PEI-GO/EP coating changed from adhesive and fatigue wear to abrasive wear,and it maintained good wear resistance by forming a GO lubrication transfer film on the counterpart surface.Therefore,this study may provide a new strategy for improving the dispersion of graphene in wear-resistant polymer materials for practical applications.Shaofeng Zhou Jin Yan Jialin Chen Huimin Yan Yin Zhang Jin Huang Guizhe Zhao Qiaoxin Zhang Yaqing Liu 2023Journal of Materials Science & Technology2023,,5:1
