| 1 | Nanoparticle–Cartilage Interaction: Pathology-Based Intra-articular Drug Delivery for Osteoarthritis Therapy显示文摘Osteoarthritis is the most prevalent chronic and debilitating joint disease,resulting in huge medical and socioeconomic burdens.Intra-articular administration of agents is clinically used for pain management.However,the effectiveness is inapparent caused by the rapid clearance of agents.To overcome this issue,nanoparticles as delivery systems hold considerable promise for local control of the pharmacokinetics of therapeutic agents.Given the therapeutic programs are inseparable from pathological progress of osteoarthritis,an ideal delivery system should allow the release of therapeutic agents upon specific features of disorders.In this review,we firstly introduce the pathological features of osteoarthritis and the design concept for accurate localization within cartilage for sustained drug release.Then,we review the interactions of nanoparticles with cartilage microenvironment and the rational design.Furthermore,we highlight advances in the therapeutic schemes according to the pathology signals.Finally,armed with an updated understanding of the pathological mechanisms,we place an emphasis on the development of“smart”bioresponsive and multiple modality nanoparticles on the near horizon to interact with the pathological signals.We anticipate that the exploration of nanoparticles by balancing the efficacy,safety,and complexity will lay down a solid foundation tangible for clinical translation. | Xu Li Bingyang Dai Jiaxin Guo Lizhen Zheng Quanyi Guo Jiang Peng Jiankun Xu Ling Qin | 2021 | Nano-Micro Letters2021,13,10: | 5 |

| 3 | Tumor-conditional IL-15 pro-cytokine reactivates anti-tumor immunity with limited toxicity显示文摘IL-15 is a promising cytokine to expand NK and CD8'T cells for cancer immunotherapy,but its application is limited by dose-limiting,on-target off-tumor toxicity.Here,we have developed a next-generation IL 15 that is activated inside the tumor microenvironment(TME).This pro-IL-15 has the extracellular domain of IL-15RP fused to the N-terminus of slL-15-Fc through a tumor-enriched Matrix Metalloproteinase(MMP)cleavable peptide linker to block its activity.Unlike slL-15-Fc,pro-IL-15 does not activate the peripheral expansion of NK cells and T cells,thus reducing systemic toxicity,but it still preserves efficient anti-tumor abilities.In various mouse tumors,the anti-tumor effect of pro-IL-15 depends on intratumoral CD8'T cells and IFN-y-Pro-IL-15 increases the stem-like TCF1^(+)Tim-3 CD8^(+)T cells within tumor tissue and helps overcome immune checkpoint blockade(ICB)resistance.Moreover,pro-IL-15 synergizes with current tyrosine kinase inhibitor(TKI)targeted-therapy in a poorly inflamed TUBO tumor model,suggesting that pro-IL-15 helps overcome targeted-therapy resistance.Our results demonstrate a next-generation IL-15 cytokine that can stimulate potent anti-tumor activity without severe toxicity. | Jingya Guo Yong Liang Diyuan Xue Jiao Shen Yueqi Cai Jiankun Zhu Yang-Xin Fu Hua Peng | 2021 | Cell Research2021,31,11: | 2 |

| 9 | Study on temperature field and settlement of thawing soil under static and dynamic loading显示文摘A series of tests were conducted to analyze temperature field distribution and thawing settlement of a thawing soil under static and dynamic loading at various cooling and thawing temperatures. | 10 | A Deep Reinforcement Learning Based Car Following Model for Electric Vehicle显示文摘Car following (CF) models are an appealing research area because they fundamentally describe longitudinal interactions of vehicles on the road, and contribute significantly to an understanding of traffic flow. There is an emerging trend to use data-driven method to build CF models. One challenge to the data-driven CF models is their capability to achieve optimal longitudinal driven behavior because a lot of bad driving behaviors will be learnt from human drivers by the supervised learning manner. In this study, by utilizing the deep reinforcement learning (DRL) techniques trust region policy optimization (TRPO), a DRL based CF model for electric vehicle (EV) is built. The proposed CF model can learn optimal driving behavior by itself in simulation. | 11 | Recognition for avian influenza virus proteins based on support vector machine and linear discriminant analysis显示文摘Total 200 properties related to structural characteristics were employed to represent structures of 400 HA coded proteins of influenza virus as training samples. Some recognition models for HA proteins of avian influenza virus (AIV) were developed using support vector machine (SVM) and linear discriminant analysis (LDA). The results obtained from LDA are as follows: the identification accuracy (Ria) for training samples is 99.8% and Ria by leave one out cross validation is 99.5%. Both Ria of 99.8% for training samples and Ria of 99.3% by leave one out cross validation are obtained using SVM model, respectively. External 200 HA proteins of influenza virus were used to validate the external predictive power of the resulting model. | 13 | FUS-mediated HypEVs: Neuroprotective effects against ischemic stroke显示文摘Few studies have investigated the properties and protein composition of small extracellular vesicles (sEVs) derived from neurons under hypoxic conditions. Presently, the extent of the involvement of these plentiful sEVs in the onset and progression of ischemic stroke remains an unresolved question. Our study systematically identified the characteristics of sEVs derived from neurons under hypoxic conditions (HypEVs) by physical characterization, sEV absorption, proteomics and transcriptomics analysis. The effects of HypEVs on neurites, cell survival, and neuron structure were assessed in vitro and in vivo by neural complexity tests, magnetic resonance imaging (MRI), Golgi staining, and Western blotting of synaptic plasticity-related proteins and apoptotic proteins. Knockdown of Fused in Sarcoma (FUS) small interfering RNA (siRNA) was used to validate FUS-mediated HypEV neuroprotection and mitochondrial mRNA release. Hypoxia promoted the secretion of sEVs, and HypEVs were more easily taken up and utilized by recipient cells. The MRI results illustrated that the cerebral infarction volume was reduced by 45% with the application of HypEVs, in comparison to the non- HypEV treatment group. Mechanistically, the FUS protein is necessary for the uptake and neuroprotection of HypEVs against ischemic stroke as well as carrying a large amount of mitochondrial mRNA in HypEVs. However, FUS knockdown attenuated the neuroprotective rescue capabilities of HypEVs.
