| 1 | Key residues of the receptor binding motif in the spike protein of SARS-CoV-2 that interact with ACE2 and neutralizing antibodies显示文摘Coronavirus disease 2019(COVID-19),caused by the novel human coronavirus SARS-CoV-2,is currently a major threat to public health worldwide.The viral spike protein binds the host receptor angiotensin-converting enzyme 2(ACE2)via the receptor-binding domain(RBD),and thus is believed to be a major target to block viral entry.Both SARS-CoV-2 and SARS-CoV share this mechanism.Here we functionally analyzed the key amino acid residues located within receptor binding motif of RBD that may interact with human ACE2 and available neutralizing antibodies.The in vivo experiments showed that immunization with either the SARS-CoV RBD or SARS-CoV-2 RBD was able to induce strong clade-specific neutralizing antibodies in mice;however,the cross-neutralizing activity was much weaker,indicating that there are distinct antigenic features in the RBDs of the two viruses.This finding was confirmed with the available neutralizing monoclonal antibodies against SARS-CoV or SARS-CoV-2.It is worth noting that a newly developed SARS-CoV-2 human antibody,HA001,was able to neutralize SARS-CoV-2,but failed to recognize SARS-CoV.Moreover,the potential epitope residues of HA001 were identified as A475 and F486 in the SARS-CoV-2 RBD,representing new binding sites for neutralizing antibodies.Overall,our study has revealed the presence of different key epitopes between SARS-CoV and SARSCoV-2,which indicates the necessity to develop new prophylactic vaccine and antibody drugs for specific control of the COVID-19 pandemic although the available agents obtained from the SARS-CoV study are unneglectable. | Chunyan Yi Xiaoyu Sun Jing Ye Longfei Ding Meiqin Liu Zhuo Yang Xiao Lu Yaguang Zhang Liyang Ma Wangpeng Gu Aidong Qu Jianqing Xu Zhengli Shi Zhiyang Ling Bing Sun | 2020 | Cellular & Molecular Immunology2020,17,6: | 13 |

| 4 | Secalonic acid D induces cell apoptosis in both sensitive and ABCG2-overexpressing multidrug resistant cancer cells through upregulating c-Jun expression显示文摘Secalonic acid D(SAD) could inhibit cell growth in not only sensitive cells but also multidrug resistant(MDR) cells. However, the molecular mechanisms need to be elucidated. Here, we identified that SAD possessed potent cytotoxicity in 3 pairs of MDR and their parental sensitive cells including S1-MI-80 and S1,H460/MX20 and H460, MCF-7/ADR and MCF-7 cells. Furthermore, SAD induced cell G2/M phase arrest via the downregulation of cyclin B1 and the increase of CDC2 phosphorylation. Importantly, JNK pathway upregulated the expression of c-Jun in protein level and increased c-Jun phosphorylation induced by SAD, which was linked to cell apoptosis via c-Jun/Src/STAT3 pathway. To investigate the mechanisms of upregulation of c-Jun protein by SAD, the mR NA expression level and degradation of c-Jun were examined. We found that SAD did not alter the mR NA level of c-Jun but inhibited its proteasome-dependent degradation. | 11 | Multiple processes affecting surface seawater N_2O satura-tion anomalies in tropical oceans and Prydz Bay, Antarc-tica显示文摘We analyzed the N2O content of surface seawater sampled from Prydz Bay, Antarctica, on a cruise track between 30°S and 30°N during the twenty-second Chinese National Antarctic Research Expedition during austral summer, 2006. The surface water showed an average pN 2 O value of 311.9±7.6 nL·L -1 (14.1±0.4 nmol·L -1 ), which was slightly undersaturated. The air-sea N 2 O flux in the region was -0.3±0.8 μmol·m -2 ·d -1 ; however, N 2 O in the surface water was oversaturated in most stations along the cruise track. Saturation anomalies were greater than 10%, with a maximum of 54.7% being observed at the Equator, followed by 31% at 10°N in the Sulu Sea. The air-sea fluxes at these locations were 12.4 and 4 μmol·m -2 ·d -1 , respectively. Overall, the results indicated that surface water in Prydz Bay was near equilibrium with atmospheric N 2 O, and that ocean waters in lower latitudes acted as a N 2 O source. | 16 | Responses of potential ammonia oxidation and ammonia oxidizers community to arsenic stress in seven types of soil显示文摘Soil arsenic contamination is of great concern because of its toxicity to human,crops,and soil microorganisms.However,the impacts of arsenic on soil ammonia oxidizers communities remain unclear.Seven types of soil spiked with 0 or 100 mg arsenic per kg soil were incubated for 180 days and sampled at days 1,15,30,90 and 180.The changes in the community composition and abundance of ammonia oxidizing bacteria(AOB)and ammonia oxidizing archaea(AOA)were analyzed by terminal restriction fragment length polymorphism(TRFLP)analysis,clone library sequencing,and quantitative PCR(qPCR)targeting amoA gene.Results revealed considerable variations in the potential ammonia oxidation(PAO)rates in different soils,but soil PAO was not consistently significantly inhibited by arsenic,probably due to the low bioavailable arsenic contents or the existence of functional redundancy between AOB and AOA.The variations in AOB and AOA communities were closely associated with the changes in arsenic fractionations.The amoA gene abundances of AOA increased after arsenic addition,whereas AOB decreased,which corroborated the notion that AOA and AOB might occupy different niches in arsenic-contaminated soils.Phylogenetic analysis of amoA gene-encoded proteins revealed that all AOB clone sequences belonged to the genus Nitrosospira,among which those belonging to Nitrosospira cluster 3a were dominant.The main AOA sequence detected belonged to Thaumarchaeal Group 1.1b,which was considered to have a high ability to adapt to environmental changes.Our results provide new insights into the impacts of arsenic on the soil nitrogen cycling. | Yanan Wang Xibai Zeng Yang Zhang Nan Zhang Liyang Xu Cuixia Wu | 2023 | Journal of Environmental Sciences2023,,5: | 0 |

| 18 | Engineered human spinal cord-like tissues with dorsal and ventral neuronal progenitors for spinal cord injury repair in rats and monkeys显示文摘Transplanting human neural progenitor cells is a promising method of replenishing the lost neurons after spinal cord injury (SCI), but differentiating neural progenitor cells into the diverse types of mature functional spinal cord neurons in vivo is challenging. In this study, engineered human embryonic spinal cord-like tissues with dorsal and ventral neuronal characters (DV-SC) were generated by inducing human neural progenitor cells (hscNPCs) to differentiate into various types of dorsal and ventral neuronal cells on collagen scaffold in vitro. Transplantation of DV-SC into complete SCI models in rats and monkeys showed better therapeutic effects than undifferentiated hscNPCs, including pronounced cell survival and maturation. DV-SC formed a targeted connection with the host’s ascending and descending axons, partially restored interrupted neural circuits, and improved motor evoked potentials and the hindlimb function of animals with SCI.
