Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). With an exposure of 5.6 ton-years, the sensitivity of Panda X-4 T could reach a minimum spin-independent dark matter-nucleon cross section of 6 × 10^(-48) cm^2 at a dark matter mass of 40 Ge V/c^2.HongGuang Zhang Abdusalam Abdukerim Wei Chen Xun Chen YunHua Chen XiangYi Cui BinBin Dong DeQing Fang ChangBo Fu Karl Giboni Franco Giuliani LinHui Gu XuYuan Guo ZhiFan Guo Ke Han ChangDa He ShengMing He Di Huang XingTao Huang Zhou Huang Peng Ji XiangDong Ji YongLin Ju ShaoLi Li Yao Li Heng Lin HuaXuan Liu JiangLai Liu YuGang Ma YaJun Mao KaiXiang Ni JinHua Ning XiangXiang Ren Fang Shi AnDi Tan AnQing Wang Cheng Wang HongWei Wang Meng Wang QiuHong Wang SiGuang Wang XiuLi Wang XuMing Wang Zhou Wang MengMeng Wu ShiYong Wu JingKai Xia MengJiao Xiao PengWei Xie BinBin Yan JiJun Yang Yong Yang ChunXu Yu JuMin Yuan JianFeng Yue Dan Zhang Tao Zhang Li Zhao QiBin Zheng JiFang Zhou Ning Zhou XiaoPeng Zhou 2019Science China(Physics,Mechanics & Astronomy)2019,62,3:6
Alignment of social and ecological structures increased the ability of river management显示文摘Large hydrologic basins involve multiple stakeholders, and coupled dynamic social and ecological processes. Managing such basins has long been a challenge. Balancing the demand for water from nature against that from humans is always difficult, particularly in arid watersheds. Here, we analyze potential institutional causes of ecological degradation and how it can be reversed by introducing new forms of governance. The framework and assumptions are illustrated using China’s second-largest endorheic basin, where empirical evidence shows that the introduction of a new governing authority connecting midstream and downstream actors facilitated the establishment of a new governance regime that is better aligned with the biophysical scales of the watershed. A trans-regional water allocation project initiated by the new higher-level authority successfully rescued downstream oases and restored a dried terminal lake. These outcomes suggest that when social and ecological structures are better aligned our ability to manage the interplay between social and ecological processes increases. However, the lack of direct connection between the actors of the middle and lower reaches resulted in the paradox of an increase in water demand. We therefore suggest that measures to stimulate the emergence of horizontal social ties linking different critical groups of actors across the watershed could further the alignment of the institutional and biophysical structures-without these changes sustainable management of river basins and other common pool resources will remain problematic.Shuai Wang Bojie Fu Orjan Bodin Jianguo Liu Mengmeng Zhang Xiaoyan Li 2019Science Bulletin2019,64,18:6
Catalysis under shell: Improved CO oxidation reaction confined in Pt@h-BN core-shell nanoreactors显示文摘Core-shell nanostructures consisting of active metal cores and protective shells often exhibit enhanced catalytic performance, in which reactants can access a small part of the core surfaces through the pores in the shells. In this study, we show that Pt nanoparticles (NPs) can be embedded into few-layer hexagonal boron nitride (h-BN) overlayers, forming Pt@h-BN core-shell nanocatalysts. The h-BN shells not only protect the Pt NPs under harsh conditions but also allow gaseous molecules such as CO and O2 to access a large part of the Pt surfaces through a facile intercalation process. As a result, the Pt@h-BN nanostructures act as nanoreactors, and CO oxidation reactions with improved activity, selectivity, and stability occur at the core-shell interfaces. The confinement effect exerted by the h-BN shells promotes the Pt-catalyzed reactions. Our work suggests that two-dimensional shells can function as robust but flexible covers on nanocatalyst surfaces and tune the surface reactivity.Mengmeng Sun Qiang Fu Lijun Gao Yanping Zheng Yangyang Li Mingshu Chen Xinhe Bao 2017Nano Research2017,10,4:5
Effects of Technological Parameters on Carbothermal Reduction and Nitridation of Zircon显示文摘Carbothermal reduction and nitridation(CRN)of zircon(ZrSiO4)permits obtaining different composites of oxides and nitrides such as ZrO2-Si2N2O and ZrN-Si3N4.The effects of technological parameters(carbon source, reaction temperature, and carbon content)on the reaction rate and product phase composition of CRN of zircon were investigated by TGA and XRD.The results show that:(1)carbon source is an important factor for a rapid reaction, and activated carbon is chosen as the carbon source considering the expect products and reaction rate;(2)reaction temperature has vital effect on reaction rate and product. In case of carbon content above 20%,the zircon phase and m-ZrO2 phase decrease with increasing temperature, while the Zr7N8O4 phase increases firstly and then decreases, and the ZrN phase increases continually;(3)different carbon contents result in different reaction products. The higher the carbon content, the lower the starting temperature for the CRN of zircon.YU Renhong FU Yadong ZHANG Mengmeng WANG Lizhi HONG Quanquan ZHANG Ruohua 2011China's Refractories2011,20,1:3
Structural design for electrocatalytic water splitting to realize industrial-scale deployment: Strategies, advances, and perspectives显示文摘The green hydrogen generation powered by renewable electricity promises the potential decarbonization of the hard-to-abate sector and is essential for the fulfillment of the Paris Agreement that attempts to limit the global average temperature rise in the range of 1.5–2.0 ℃ above the pre-industrial level by the end of this century. Tremendous efforts have been devoted to the optimization of the electrocatalytic performance of the catalysts under industrial-relevant current densities via rational structure design, which induces a preferential electron distribution that favors the adsorption/desorption behavior of the key intermediates, thus accelerating the reaction kinetics. In this review, a brief introduction of the current energy status will be first presented to necessitate the importance of green hydrogen.Followed by the basic concepts and fundamental understanding of the reaction mechanisms, we present efficient strategies for the enhancement of the electrocatalytic performance of the catalysts to meet the rigorous requirement under industrial conditions and the in-depth understanding behind the reinforcement will be briefly discussed next. Then the recent advances regarding the rational design of electrocatalysts operating at an industrial scale will be summarized. Finally, the challenges and perspectives in this thriving field will be proposed from our point of view.Xianwei Fu Ruijuan Shi Shilong Jiao Mengmeng Li Qiuye Li 2022Journal of Energy Chemistry2022,31,7:2

Propagation properties of foreshock cavitons: Cluster observations显示文摘Foreshock cavitons are transient phenomena observed in the terrestrial foreshock region. They are characterized by a simultaneous depression of magnetic field magnitude and plasma density, which are bounded with enhancements of these two parameters and surrounded by ultralow frequency(ULF)waves.Previous studies focused on the interplanetary magnetic field(IMF)conditions, solar wind(SW)conditions, and the growth of the foreshock waves related to the generation of foreshock cavitons.Previously,a multipoint spacecraft analysis method using Cluster data was applied to analyze only two foreshock cavitons, and this method did not consider uncertainties. In this study, multipoint spacecraft analysis methods, including the timing method, the minimum directional derivative(MDD)method, and the spatiotemporal difference(STD)method are applied to determine the velocity in both spacecraft and solar wind frames. The propagation properties show good agreement with previous results from simulations and observations that most cavitons move sunward in the solar wind frame, with the velocities larger than the Alfvén speed. The propagation properties of foreshock cavitons support the formation mechanism of cavitons in previous simulations, which suggested that cavitons are formed due to the nonlinear evolution of compressive ULF waves. We find that there is clear decreasing trend between the size of cavitons and their velocity in the solar wind frame. In addition, the timing method considering errors has been applied to study the evolution properties by comparing the velocities with errors of the leading and trailing edges, and we identify three stable cavitons and one contracting caviton, which has not been studied before. Most cavitons should remain stable when they travel toward the Earth’s bow shock. The relationship between the size of foreshock cavitons and their distance from the bow shock is also discussed.WANG MengMeng YAO ShuTao SHI QuanQi ZHANG Hui TIAN AnMin DEGELING Alexander William ZHANG Shuai GUO RuiLong SUN WeiJie LIU Ji BAI ShiChen SHEN XiaoChen ZHU XiaoQiong FU SuiYan PU ZuYin 2020Science China(Technological Sciences)2020,63,1:1

The genomic and bulked segregant analysis of Curcuma alismatifolia revealed its diverse bract pigmentation显示文摘Compared with most flowers where the showy part comprises specialized leaves(petals)directly subtending the reproductive structures, most Zingiberaceae species produce showy“flowers”through modifications of leaves(bracts)subtending the true flowers throughout an inflorescence.Curcuma alismatifolia, belonging to the Zingiberaceae family,a plant species originating from Southeast Asia, has become increasingly popular in the flower market worldwide because of its varied and esthetically pleasing bracts produced in different cultivars. Here,we present the chromosome-scale genome assembly of C.alismatifolia“Chiang Mai Pink”and explore the underlying mechanisms of bract pigmentation.Comparative genomic analysis revealed C.alismatifolia contains a residual signal of whole-genome duplication.Duplicated genes, including pigment-related genes, exhibit functional and structural differentiation resulting in diverse bract colors among C.alismatifolia cultivars. In addition,we identified the key genes that produce different colored bracts in C.alismatifolia, such as F3′5'H,DFR,ANS and several transcription factors for anthocyanin synthesis,as well as chlH and CAO in the chlorophyll synthesis pathway by conducting transcriptomic analysis, bulked segregant analysis using both DNA and RNA data, and population genomic analysis. This work provides data for understanding the mechanism of bract pigmentation and will accelerate breeding in developing novel cultivars with richly colored bracts in C.alismatifolia and related species. It is also important to understand the variation in the evolution of the Zingiberaceae family.Xuezhu Liao Yuanjun Ye Xiaoni Zhang Dan Peng Mengmeng Hou Gaofei Fu Jianjun Tan Jianli Zhao Rihong Jiang Yechun Xu Jinmei Liu Jinliang Yang Wusheng Liu Luke RTembrock Genfa Zhu Zhiqiang Wu 2022aBIOTECH2022,3,3:1
Mediating role of resilience in the relationship between COVID-19 related stigma and mental health among COVID-19 survivors:a cross-sectional study显示文摘Background The global coronavirus disease 2019(COVID-19)has caused many negative effects on physical and mental health of patients who have survived COVID-19.Apart from some long-lasting physical sequelae, those COVID-19 survivors are also suffering stigma and discrimination at different levels around the world. The current study aims to assess the role resilience played in stigma and mental disorders among COVID-19 survivors. Methods The cross-sectional study was carried out among former COVID-19 patients in Jianghan District(Wuhan,China)from June 10 to July 25,2021.The demographic questions, the Impact of Events Scale-Revised,the Generalized Anxiety Disorder Questionnaire, the Patient Health Questionnaire, the Resilience Style Questionnaire and the Short Version of COVID-19 Stigma Scale of 12 items were used to collect relevant information of the participants.Descriptive analyses,Pearson correlation analysis and Structural Equation Modeling were used to make data description and analysis. Results A total of 1541 out of 1601 COVID-19 survivors(887 females and 654 males)were included in the analysis.Perceived stigma of those COVID-19 survivors correlates significantly with anxiety(r=0.335,P<0.001),depression(r=0.325,P<0.001)and post-traumatic stress disorder(PTSD)(r=0.384,P<0.001).It has a direct effect on COVID-19 survivors’anxiety(β=0.326,P<0.001),depression(β=0.314,P<0.001),PTSD(β=0.385,P<0.001)and their resilience(β=−0.114,P<0.01).Resilience partially mediated the association between perceived stigma and anxiety(β=0.020,P<0.01),depression(β=0.020,P<0.01),and PTSD(β=0.014,P<0.01)among COVID-19 survivors. Conclusion Stigma has a significant negative impact on mental health, while resilience plays a mediator role in the relationship between stigma and mental health among COVID-19 survivors.Based on our study,we suggested that when designing psychological interventions for COVID-19 survivors, consideration should be taken into account to reduce stigma and improve resilience.Weijun Xiao Xiaoyang Liu Hao Wang Yiman Huang Zhenwei Dai Mingyu Si Jiaqi Fu Xu Chen Mengmeng Jia Zhiwei Leng Dan Cui Winnie W.S.Mak Liming Dong Xiaoyou Su 2023Infectious Diseases of Poverty2023,12,2:0
An improved evaluation of the neutron background in the PandaX-Ⅱ experiment显示文摘In dark matter direct detection experiments, neutron is a serious source of background, which can mimic the dark matter-nucleus scattering signals. In this paper,we present an improved evaluation of the neutron background in the PandaX-II dark matter experiment by a novel approach.Instead of fully relying on the Monte Carlo simulation, the overall neutron background is determined from the neutron-induced high energy signals in the data.In addition, the probability of producing a dark-matter-like background per neutron is evaluated with a complete Monte Carlo generator, where the correlated emission of neutron(s)andγ(s)in the(α,n)reactions and spontaneous fissions is taken into consideration. With this method, the neutron backgrounds in the Run 9(26-ton-day)and Run 10(28-ton-day)data sets of PandaX-II are estimated to be(0.66±0.24)and(0.47±0.25)events,respectively.QiuHong Wang Abdusalam Abdukerim Wei Chen Xun Chen YunHua Chen XiangYi Cui YingJie Fan DeQing Fang ChangBo Fu LiSheng Geng Karl Giboni Franco Giuliani LinHui Gu XuYuan Guo Ke Han ChangDa He Di Huang Yan Huang YanLin Huang Zhou Huang Peng Ji XiangDong Ji YongLin Ju YiHui Lai Kun Liang HuaXuan Liu JiangLai Liu WenBo Ma YuGang Ma YaJun Mao Yue Meng Parinya Namwongsa KaiXiang Ni JinHua Ning XuYang Ning XiangXiang Ren ChangSong Shang Lin Si AnDi Tan AnQing Wang HongWei Wang Meng Wang SiGuang Wang XiuLi Wang Zhou Wang MengMeng Wu ShiYong Wu JingKai Xia MengJiao Xiao PengWei Xie BinBin Yan JiJun Yang Yong Yang ChunXu Yu Jumin Yuan Dan Zhang HongGuang Zhang Tao Zhang Li Zhao QiBin Zheng JiFang Zhou Ning Zhou XiaoPeng Zhou 2020Science China(Physics,Mechanics & Astronomy)2020,63,3:0
Constraining self-interacting dark matter with the full dataset of PandaX-II显示文摘Self-interacting dark matter(SIDM)is a leading candidate proposed to solve discrepancies between predictions of the prevailing cold dark matter theory and observations of galaxies. Many SIDM models predict the existence of a light force carrier that mediates strong dark matter self-interactions.If the mediator couples to the standard model particles,it could produce characteristic signals in dark matter direct detection experiments. We report searches for signals of SIDM models with a light mediator using the full dataset of the PandaX-II experiment, basing on a total exposure of 132 tonne-days.No significant excess over background is found, and our likelihood analysis leads to a strong upper limit on the dark matter-nucleon coupling strength. We further combine the PandaX-II constraints and those from observations of the light element abundances in the early universe, and show that direct detection and cosmological probes can provide complementary constraints on dark matter models with a light mediator.Jijun Yang Abdusalam Abdukerim Wei Chen Xun Chen Yunhua Chen Chen Cheng Xiangyi Cui Yingjie Fan Deqing Fang Changbo Fu Mengting Fu Lisheng Geng Karl Giboni Linhui Gu Xuyuan Guo Ke Han Changda He Shengming He Di Huang Yan Huang Ran Huo Yanlin Huang Zhou Huang Xiangdong Ji Yonglin Ju Shuaijie Li Qing Lin Huaxuan Liu Jianglai Liu Xiaoying Lu Wenbo Ma Yugang Ma Yajun Mao Yue Meng Nasir Shaheed Kaixiang Ni Jinhua Ning Xuyang Ning Xiangxiang Ren Changsong Shang Guofang Shen Lin Si Andi Tan Anqing Wang Hongwei Wang Meng Wang QiuHong Wang Siguang Wang Wei Wang Xiuli Wang Zhou Wang Mengmeng Wu Shiyong Wu Weihao Wu Jingkai Xia Mengjiao Xiao Xiang Xiao Pengwei Xie Binbin Yan Yong Yang Chunxu Yu Hai-Bo Yu Jumin Yuan Ying Yuan Xinning Zeng Dan Zhang Tao Zhang Li Zhao Qibin Zheng Jifang Zhou Ning Zhou Xiaopeng Zhou 2021Science China(Physics,Mechanics & Astronomy)2021,64,11:0

The effects of plant resource inputs on the energy flux of soil nematodes are affected by climate and plant resource type显示文摘The relative abundance of different components of the soil food web can vary tremendously in response to plant resource inputs. However, little is known about the mechanisms that plant resource regulates the energy fluxes and soil community composition. Here,we experimentally reduced litter and root inputs for two years in China at low-,mid-,and high-latitude forests to explore the effects of plant-derived resource inputs on the nematode energy flux and community composition.Litter reduction at high and mid latitudes and root removal at low latitudes reduced nematode richness but did not alter nematode abundance.Besides, litter reduction reduced energy fluxes of bacterial-feeding nematodes at mid latitudes and energy fluxes of plant-feeding,bacterial-feeding and omnivorous-predatory nematodes at low latitudes, thus reducing the energy fluxes of total nematodes in mid-and low-latitude forests.By contrast, root removal reduced energy fluxes and relative energy flux of plant-feeding nematodes in high-and low-latitude forests. In most cases, nematode diversity in different trophic groups increased with increasing energy flux to nematodes.Taken together, our results suggest that the effects of plant resource inputs on nematode energy flux are affected by climate and plant resource type, which improves our understanding of plant-soil interactions.LINA ZHAO BINBIN YU MENGMENG WANG Jie Zhang Zhifeng Shen Yang Cui Junyong Li Ji Ye Weizhong Zu Xiaojing Liu Zongji Fan Shenglei Fu Yuanhu Shao 2021Soil Ecology Letters2021,3,2:0

Non-proteolytic ubiquitination of OTULIN regulates NF-κB signaling pathway显示文摘NF-κB signaling regulates diverse processes such as cell death, inflammation, immunity, and cancer. The activity of NF-κB is controlled by methionine 1-linked linear polyubiquitin, which is assembled by the linear ubiquitin chain assembly complex(LUBAC)and the ubiquitin-conjugating enzyme UBE2L3.Recent studies found that the deubiquitinase OTULIN breaks the linear ubiquitin chain, thus inhibiting NF-κB signaling.Despite the essential role of OTULIN in NF-κB signaling has been established, the regulatory mechanism for OTULIN is not well elucidated. To discover the potential regulators of OTULIN,we analyzed the OTULIN protein complex by proteomics and revealed several OTULIN-binding proteins, including LUBAC and tripartite motif-containing protein 32(TRIM32).TRIM32 is known to activate NF-κB signaling, but the mechanism is not dear.Genetic complement experiments found that TRIM32 is upstream of OTULIN and TRIM32-mediated NF-κB activation is dependent on OTULIN.Mutagenesis of the E3 ligase domain showed that the E3 ligase activity is essential for TRIM32-mediated NF-κB activation.Further experiments found that TRIM32 conjugates polyubiquitin onto OTULIN and the polyubiquitin blocks the interaction between HOIP and OTULIN,thereby activating NF-κB signaling.Taken together,we report a novel regulatory mechanism by which TRIM32-mediated non-proteolytic ubiquitination of OTULIN impedes the access of OTULIN to the LUBAC and promotes NF-κB activation.Mengmeng Zhao Kun Song Wenzhuo Hao Lingyan Wang Girish Patil Qingmei Li Lingling Xu Fang Hua Bishi Fu Jens C.Schwamborn Martin E.Dorf Shitao Li 2020Journal of Molecular Cell Biology2020,12,3:0
Arsenic removal from acidic industrial wastewater by ultrasonic activated phosphorus pentasulfide显示文摘Arsenic is one of the main harmful elements in industrial wastewater. How to remove arsenic has always been one of the research hotspots in academic circles. In the process of arsenic removal by traditional sulfuration, the use of traditional sulfurizing agent will introduce new metal cations, which will affect the recycling of acid.In this study, phosphorus pentasulfide (P_(2)S_(5)) was used as sulfurizing agent, which hydrolyzed to produce H_(3)PO_(4) and H_(2)S without introducing new metal cations. The effect of ultrasound on arsenic removal by P_(2)S_(5) was studied.Under the action of ultrasound, the utilization of P_(2)S_(5) was improved and the reaction time was shortened. The effects of S/As molar ratio and reaction time on arsenic removal rate were investigated under ultrasonic conditions.Ultrasonic enhanced heat and mass transfer so that the arsenic removal rate of 94.5%could be achieved under the conditions of S/As molar ratio of 2.1:1 and reaction time of 20 min.In the first 60 min, under the same S/As molar ratio and reaction time, the ultrasonic hydrolysis efficiency of P_(2)S_(5) was higher. This is because P_(2)S_(5) forms ([(P_(2)S_(4))])^(2+)under the ultrasonic action, and the structure is damaged, which is easier to be hydrolyzed. In addition, the precipitation after arsenic removal was characterized and analyzed by X-ray diffraction, scanning electron microscope-energy dispersive spectrometer,X-ray fluorescence spectrometer and X-ray photoelectron spectroscopy. Our research avoids the introduction of metal cations in the arsenic removal process, and shortens the reaction time.Bo Yu Guang Fu Xinpei Li Libo Zhang Jing Li Hongtao Qu Dongbin Wang Qingfeng Dong Mengmeng Zhang 2023Chinese Journal of Chemical Engineering2023,60,8:0
Clinical Epidemiology,Illness Profiles, and the Implication of COVID-19 Before and After the Nationwide Omicron Outbreak During the Winter of 2022显示文摘China.In this study,we aimed to investigate the clinical characteristics and outcomes of patients with coronavirus disease 2019(COVID-19)in the Beijing region. Methods:In this retrospective study,we enrolled inpatients admitted for COVID-19 in the Fifth Medical Center of Chinese PLA General Hospital in Beijing between November 10,2022,and January 30,2023.Demographic and clinical features and treatment outcomeswere comprehensively analyzed. We used logistic regression and linear regression analyses to explore the risk factors associated with disease severity and time of nucleic acid conversion, respectively. Results:A total of 1010 hospitalized patients with COVID-19 were enrolled. The median age was 43.0 years(interquartile range,28.0–63.0),and patients aged<60 years and≥60 years comprised 71.2%and 28.8%of total included patients, respectively. The clinical classification of mild(74.6%,753/1010),moderate(21.0%,212/1010),severe(2.7%,27/1010),and unidentified(1.8%,18/1010)was separately recorded;1005 patients were discharged, and 5 patients died in the hospital. The outbreak of the emerging epidemic witnessed an evident increase in the proportion of moderate(42.9%vs.16.4%)and severe(10.3%vs.1.1%)cases after December 7,2022.Patients with a moderate/severe classification had higher levels of procalcitonin,IL-6,serum ferritin,C-reactive protein, lactic dehydrogenase, serum urea nitrogen, and D-dimer and lower counts of CD4+T,CD8+T,and B cells(all P<0.001).Multivariable regression analysis revealed that increased odds of disease severity were associated with the following factors:age≥60 years,IL-6>7 pg/mL,lactic dehydrogenase level>245 U/L,cough,and fever at admission.Age≥80 years and chronic lung diseasewere independent risk factors in the nonmild group in elderly patients. In addition, the duration for nucleic acid to turn negativewas approximately 5.0 d(interquartile range,3.0–7.0).Prolonged time of nucleic acid conversion was associated with age≥60 years, serum urea nitrogen level>8.2 mmol/L,neutrophil count>7×10^(9)/L,and the presence of a chronic lung disease or carcinoma. Finally, unvaccinated patients accounted for 37.3%of enrolled patients;children and the elder people accounted for approximately half of that. The univariable analysis found that booster doses reduced disease severity and shortened the time of nucleic acid conversion in elderly patients. Conclusions:The outbreak ofOmicron rapidly increased the number of patientswith COVID-19 in Beijing. In elderly patients, booster doses may reduce disease severity and shorten the time of nucleic acid conversion.Healthcare systems should be optimized before an emerging epidemic outbreak.Yuming Guo Zhe Xu Wen-Xin Wang Cheng Zhen Jinhua Hu Jinsong Mu Chengcheng Ji Xin Yuan Ruonan Xu Lei Huang Lei Shi Fanping Meng Junliang Fu Shuangnan Zhou Siyu Wang Fengyi Li Bo Tu Dawei Zhang Huihuang Huang Yufeng Mao Wen Xu Chao Zhang Xiuying Mu Jun Zhao Bo Jin Haibin Su Yinying Lu Yongqian Cheng Dong Ji Shaoli You Jinghui Dong Changchun Liu Mengmeng Zhang Yuanyuan Li Tianjun Jiang Yonggang Li Fu-Sheng Wang 2023Infectious Diseases & Immunity2023,3,4:0
Gut microbiota-based pharmacokinetic-pharmacodynamic study and molecular mechanism of specnuezhenide in the treatment of colorectal cancer targeting carboxylesterase显示文摘Specnuezhenide(SNZ)is among the main components of Fructus Ligustri Lucidi, which has antiinflammation,anti-oxidation,and anti-tumor effect. The low bioavailability makes it difficult to explain the mechanism of pharmacological effect of SNZ.In this study, the role of the gut microbiota in the metabolism and pharmacokinetics characteristics of SNZ as well as the pharmacological meaning were explored.SNZ can be rapidly metabolized by the gut microbiome, and two intestinal bacterial metabolites of SNZ,salidroside and tyrosol, were discovered. In addition, carboxylesterase may be the main intestinal bacterial enzyme that mediates its metabolism. At the same time,no metabolism was found in the incubation system of SNZ with liver microsomes or liver homogenate, indicating that the gut microbiota is the main part involved in the metabolism of SNZ.In addition, pharmacokinetic studies showed that salidroside and tyrosol can be detected in plasma in the presence of gut microbiota.Interestingly, tumor development was inhibited in a colorectal tumor mice model administered orally with SNZ,which indicated that SNZ exhibited potential to inhibit tumor growth, and tissue distribution studies showed that salidroside and tyrosol could be distributed in tumor tissues. At the same time,SNZ modulated the structure of gut microbiota and fungal group, which may be the mechanism governing the antitumoral activity of SNZ.Furthermore,SNZ stimulates the secretion of short-chain fatty acids by intestinal flora in vitro and in vivo.In the future, targeting gut microbes and the interaction between natural products and gut microbes could lead to the discovery and development of new drugs.Hang Yu Hui Xu Xinyu Yang Zhengwei Zhang Jiachun Hu Jinyue Lu Jie Fu Mengmeng Bu Haojian Zhang Zhao Zhai Jingyue Wang Jiandong Jiang Yan Wang 2023Journal of Pharmaceutical Analysis2023,13,9:0
