3Humoral immune response to circulating SARS-CoV-2 variants elicited by inactivated and RBD-subunit vaccines显示文摘SARS-CoV-2 variants could induce immune escape by mutations on the receptor-binding domain(RBD)and N-terminal domain(NTD).Here we report the humoral immune response to circulating SARS-CoV-2 variants,such as 501Y.V2(B.1.351),of the plasma and neutralizing antibodies(NAbs)elicited by CoronaVac(inactivated vaccine),ZF2001(RBD-subunit vaccine)and natural infection.Among 86 potent NAbs identified by high-throughput single-cell VDJ sequencing of peripheral blood mononuclear cells from vaccinees and convalescents,near half anti-RBD NAbs showed major neutralization reductions against the K417N/E484K/N501Y mutation combination,with E484K being the dominant cause.VH3-53/VH3-66 recurrent antibodies respond differently to RBD variants,and K417N compromises the majority of neutralizing activity through reduced polar contacts with complementarity determining regions.In contrast,the 242–244 deletion(242–244Δ)would abolish most neutralization activity of anti-NTD NAbs by interrupting the conformation of NTD antigenic supersite,indicating a much less diversity of anti-NTD NAbs than anti-RBD NAbs.Plasma of convalescents and CoronaVac vaccinees displayed comparable neutralization reductions against pseudo-and authentic 501Y.V2 variants,mainly caused by E484K/N501Y and 242–244Δ,with the effects being additive.Importantly,RBD-subunit vaccinees exhibit markedly higher tolerance to 501Y.V2 than convalescents,since the elicited anti-RBD NAbs display a high diversity and are unaffected by NTD mutations.Moreover,an extended gap between the third and second doses of ZF2001 leads to better neutralizing activity and tolerance to 501Y.V2 than the standard three-dose administration.Together,these results suggest that the deployment of RBD-vaccines,through a third-dose boost,may be ideal for combating SARS-CoV-2 variants when necessary,especially for those carrying mutations that disrupt the NTD supersite.Yunlong Cao Ayijiang Yisimayi Yali Bai Weijin Huang Xiaofeng Li Zhiying Zhang Tianjiao Yuan Ran An Jing Wang Tianhe Xiao Shuo Du Wenping Ma Liyang Song Yongzheng Li Xiang Li Weiliang Song Jiajing Wu Shuo Liu Xuemei Li Yonghong Zhang Bin Su Xianghua Guo Yangyang Wei Chuanping Gao Nana Zhang Yifei Zhang Yang Dou Xiaoyu Xu Rui Shi Bai Lu Ronghua Jin Yingmin Ma Chengfeng Qin Youchun Wang Yingmei Feng Junyu Xiao Xiaoliang Sunney Xie 2021Cell Research2021,31,7:12

5Presenilin 2 deficiency facilitates Aβ-induced neuroinflammation and injury by upregulating P2X7 expression显示文摘Accumulating evidence suggests that β-amyloid (Aβ)-induced neuroinflammation plays a prominent and early role in Alzheimer’s disease (AD). In this study, we demonstrated that Presenilin 2 (PS2) deficiency facilitates Aβ-induced neuroinflammation and injury by upregulating P2X7 expression both in vitro and in vivo. PS2 knockout mice demonstrated increased cognitive impairments and cerebral injury. PS2 deficiency increased the expression of P2X7 both in neurons and microglial cells.Furthermore, extracellular ATP also increased in both Aβ-treated and untreated PS2 knockout microglial cells. Notably, Aβ-induced classical proinflammatory cytokines such as IL-1β, IL-1α and TNF-α were increased in PS2 knockout microglial cells, suggesting a potential role for PS2 in the regulation of neuroinflammation. The expression of P2X7 clearly increased in PS2 knockdown BV2 cells. Consistent with in vivo data, Aβ-induced IL-1β production was also clearly enhanced in PS2 knockdown BV2 cells. Additionally, expression of the transcription factor Sp1 was increased in PS2 knockdown cells. When we treated PS2 knockdown cells with the specific Sp1 inhibitor MIT, we observed that enhanced P2X7 expression was significantly rescued. 8Co-Co304@carbon core-shells derived from metal-organic framework nanocrystals as efficient hydrogenevolution catalysts显示文摘Controllable pyrolysis of metal-organic frameworks (MOFs) in confined spacesis a promising strategy for the design and development of advanced functionalmaterials. In this study, Co-C0304@carbon composites were synthesized viapyrolysis of a Co-MOFs@glucose polymer (Co-MOFs@GP) followed by partialoxidation of Co nanoparticles (NPs). The pyrolysis of Co-MOFs@GP generated acore-shell structure composed of carbon shells and Co NPs. The controlled partialoxidation of Co NPs formed Co-Co304 heterojunctions confined in carbon shells.Compared with Co-MOFs@GP and Co@carbon-n (Co@C-n), Co-C0304@carbon-n(Co-Co304@C-n) exhibited higher catalytic activity during NaBH4 hydrolysis.Co-C0304@C-II provided a maximum specific H2 generation rate of5,360 mL.min-l.gco-1 at room temperature due to synergistic interactionsbetween Co and C0304 NPs. 11Pseudocapacitance boosted N-doped carbon coated Fe7S8 nanoaggregates as promising anode materials for lithium and sodium storage显示文摘Herein,the core-shell structured N-doped carbon coated Fe7S8 nano-aggregates(Fe7S8@NC)were controllably prepared via a simple three-step synthesis strategy.The appropriate thickness of N-doped carbon layer outside Fe7S8 nano-aggregates can not only inhibit the particle pulverization induced by the big volume changes of Fe7S8,but can increase the electron transfer efficiency.The hierarchical Fe7S8 nano-aggregates composed of some primary nanoparticles can accelerate the lithium or sodium diffusion kinetics.As anode materials for Li-ion batteries(LIBs),the well-designed Fe7S8@NC nanocomposites exhibit outstanding lithium storage performance,which is better than that of pure Fe7S8,Fe3O4@NC and Fe7S8@C.Among these nanocomposites,the N-doped carbon coated Fe7S8 with carbon content of 26.87 wt.%shows a high reversible specific capacity of 833 mAh·g^−1 after 1,000 cycles at a high current density of 2 A·g^−1.The above electrode also shows excellent high rate sodium storage performance.The experimental and theoretical analyses indicate that the outstanding electrochemical performance could be attributed to the synergistic effect of hierarchical Fe7S8 nanostructure and conductive N-doped carbon layer.The quantitative kinetic analysis indicates that the charge storage of Fe7S8@NC electrode is a combination of diffusion-controlled battery behavior and surface-induced capacitance behavior.Yanli Zhou Ming Zhang Qi Wang Jian Yang Xingyun Luo Yanlu Li Rong Du Xinsheng Yan Xueqin Sun Caifu Dong Xiaoyu Zhang Fuyi Jiang 2020Nano Research2020,13,3:4
12Performances of conventional fusion methods evaluated for inland water body observation using GF-1 image显示文摘Satellite remote sensing of inland water body requires a high spatial resolution and a multiband narrow spectral resolution, which makes the fusion between panchromatic(PAN) and multi-spectral(MS) images particularly important. Taking the Daquekou section of the Qiantang River as an observation target, four conventional fusion methods widely accepted in satellite image processing, including pan sharpening(PS), principal component analysis(PCA), Gram-Schmidt(GS), and wavelet fusion(WF), are utilized to fuse MS and PAN images of GF-1.The results of subjective and objective evaluation methods application indicate that GS performs the best,followed by the PCA, the WF and the PS in the order of descending. The existence of a large area of the water body is a dominant factor impacting the fusion performance. Meanwhile, the ability of retaining spatial and spectral informations is an important factor affecting the fusion performance of different fusion methods. The fundamental difference of reflectivity information acquisition between water and land is the reason for the failure of conventional fusion methods for land observation such as the PS to be used in the presence of the large water body. It is suggested that the adoption of the conventional fusion methods in the observing water body as the main target should be taken with caution. 13Idiopathic Orbital Inflammation Associated with Panuveitis in an Adult and a Review of the Literature显示文摘Purpose:To report an unusual case of idiopathic orbital inflammation associated with panuveitis. Case report:We report a case of a 47-year-old female with a history of uveitis who presented with abrupt and painful prop tosis with complete vision loss unresponsive to antibiotic therapy.B-scan ultrasonography and magnetic resonance imaging both showed retinal detachment and an orbital space-occupying mass. The patient underwent orbitotomy and the diagnosis of idiopathic orbital inflammation was confirmed by histopathology. Anterior uveitis is rarely seen in idiopathic orbital inflammation in adults. We report for the first time an unusual case of idiopathic orbital inflammation associated with panuveitis. Conclusion:Inflammation involving both the eye and the orbit is rarely seen in adults. 15Genotypic variation in spatiotemporal distribution of canopy light interception in relation to yield formation in cotton显示文摘Background: Within-canopy interception of photosynthetically active radiation(PAR) impacts yield and other agronomic traits in cotton(Gossypium hirsutum L.). Field experiments were conducted to investigate the influence of 6 cotton varieties(they belong to 3 different plant types) on yield, yield distribution, light interception(LI), LI distribution and the relationship between yield formation and LI in Anyang, Henan, in 2014 and 2015.Result: The results showed that cotton cultivars with long branches(loose-type) intercepted more LI than did cultivars with short branches(compact-type), due to increased LI in the middle and upper canopy. Although loose-type varieties had greater LI, they did not yield significantly higher than compact-type varieties, due to decreased harvest index. Therefore, improving the harvest index by adjusting the source-to-sink relationship may further increase cotton yield for loose-type cotton. In addition, there was a positive relationship between reproductive organ biomass accumulation and canopy-accumulated LI, indicating that enhancing LI is important for yield improvement for each cultivar. 18Preliminary investigation on cytotoxicity of fluorinated polymer nanoparticles显示文摘As well-known persistent organic pollutants(POPs), organofluorine pollutants such as perfluorooctane sulfonate(PFOS) have been proven to be bioaccumulated and harmful to health. However, toxicological assessment of organofluorinated nanoparticles, which have emerged as a novel tool for biomedical and industrial applications, is lacking, to the best of our knowledge. To assess the biological effects and health risk of fluorinated nanoparticles,trifluoroethyl aryl ether-based fluorinated poly(methyl methacrylate) nanoparticles(PTFEPMMA NPs) were synthesized with various fluorine contents(PTFE-PMMA-1 NPs 12.0 wt.%,PTFE-PMMA-2 NPs 6.1 wt.% and PTFE-PMMA-3 NPs 5.0 wt.%), and their cytotoxicity was investigated in this study. The in vitro experimental results indicated that the cytotoxicity of PTFE-PMMA NPs was mild, and was closely related to their fluorine(F) contents and Fcontaining side chains. Specifically, the cytotoxicity of PTFE-PMMA NPs decreased with increasing F content and F-containing side chains. After exposure to PTFE-PMMA NPs at a sublethal dose(50 μg/m L) for 24 hr, the phospholipid bilayer was damaged, accompanied by increasing permeability of the cell membrane. Meanwhile, the intracellular accumulation of reactive oxygen species(ROS) occurred, resulting in the increase of DNA damage, cell cycle arrest and cell death. 20Generation of transgenic cucumbers with expression of a ten-tandem repeat long-acting GLP-1 analogue and their biological function on diabetic rats显示文摘Glucagon-like peptide-1 (GLP-1) is a 30-amino acid peptide hormone, which has the regulatory function of stimulating the secretion of insulin to balance blood glucose levels. In this study, marker-free expression vector pX6 with a fusion gene of ten tandem repeated GLP-1 analog ([Ser8, Gln26 and Asp34]-GLP-1) genes was constructed and transformed into cucumbers by Agrobacterium tumefaciens-mediated transformation method. Four transgenic lines were obtained by PCR and Southern blotting analysis and two transgenic lines successfully expressed the fusion protein (named GLP-T), which was revealed by Western blotting analysis. The biological activity test results showed that the serum glucose level of diabetic rats was significantly decreased after oral administration of the fusion protein GLP-T extracted from the transgenic cucumber fruitage.
